  • Mechanism of Pinelliae Rhizoma in Treatment of Insomnia Based on Network Pharmacology

    [Objectives] To explore the mechanism of Pinelliae Rhizoma in the treatment of insomnia. [Methods] Using the Traditional Chinese Medicine Systems Pharmacology(TCMSP) database, the effective components of Pinelliae Rhizoma were explored, to predict its targets, and screen out the targets of insomnia through GeneCards, OMIM and other databases. With the aid of Cytoscape software, a "component-target-pathway-disease" network for the treatment of insomnia by Pinelliae Rhizoma was built. Then, a target protein-related interaction network was created through the String database, and biological function annotation and pathway analysis of key targets were performed. [Results] The 13 active components of Pinelliae Rhizoma take action in the treatment of insomnia by intervening 33 targets, 59 GO biological processes, and 10 main biological pathways. [Conclusions] The active components of Pinelliae Rhizoma may treat insomnia by such pharmacological effect as regulating G protein-coupled amine receptor activity, adrenergic receptor activity, catecholamine binding, and neurotransmitter receptor activity.

Based on the theory of intermingled phlegm, blood stasis and toxin of heart disease put forward by Master Lei Zhongyi, the dialectical treatment thinking of COVID-19 patients from the concept of damage of phlegm, blood stasis and toxin to the heart were discussed. During the diagnosis, critical stage and recovery period of COVID-19, expectorant and blood-activating agents, heat and detoxification agents can be added to promote lung and asthma, free Bizheng and remove blood stasis, calm the heart and calm the mind, and promote the recovery of cardiopulmonary functions.

Chemical Composition of Natural Medicine

  • Determination and Multidimensional Analysis of 29 Inorganic Elements in Cardamine tangutorum O. E. Schulz of Different Origins

    [Objectives] This study aimed to determine the contents of inorganic elements in Cardamine tangutorum O. E. Schulz samples from different origins to provide a scientific reference for the quality control, safety evaluation and clinical medicinal use of C. tangutorum O. E. Schulz. [Methods] The contents of Al, As, B, Ba, Ca, Cd, Co, Cr, Cu, Fe, Hg, K, Mg, Mn, Mo, Na, Ni, P, Pb, Pd, Rb, Se, Si, Sn, Sr, Ti, Tl, V and Zn in the C. tangutorum O. E. Schulz samples were determined simultaneously by wet digestion and inductively coupled plasma optical emission spectroscopy(ICP-OES), and the determination results were analyzed by principal component analysis. [Results] A total of 28 inorganic elements were detected in the samples of C. tangutorum O. E. Schulz from different origins. The contents of heavy metals did not exceed the limits. Among the elements, the determined values of Hg in all samples were below the detection limit, so it was not detected. The contents of Ca, Al, P and Fe were relatively high, and they were essential nutrients in the human body. There were significant differences among different elements. Samples from different origins show similar patterns in the distribution of main elements. Correlation analysis shows that 15 pairs of inorganic elements out of 24 elements had extremely significant positive correlations, and 26 pairs of elements had significant positive correlations. The principal component analysis determined Al, As, Ba, Ca, Cd, Fe, Mn, Ni, P, Pb, Pd, Se, V and Zn were the characteristic inorganic elements in the samples of C. tangutorum O. E. Schulz. [Conclusions] The contents of toxic heavy metals in C. tangutorum O. E. Schulz samples from different origins met the edible requirements. The method was simple, rapid and accurate, and could be used to analyze and determine the inorganic elements in the samples of C. tangutorum O. E. Schulz, providing a theoretical basis for the better development and utilization of C. tangutorum O. E. Schulz.

  • Study on Pharmacological Effects of Peikun Pills

    [Objectives] To explore the effects of Peikun Pill on blood pressure of healthy adult male rats. [Methods] The duodenum of anesthetized rats was given different doses of Peikun Pills, normal saline was used as a blank control group, and captopril was used as a positive western medicine control group. The systolic blood pressure, diastolic blood pressure, mean blood pressure and pulse pressure difference(systolic blood pressure-diastolic blood pressure), and heart rate were measured, myocardial tension time index was calculated, and statistical processing was performed. [Results] Peikun Pills have a significant pressure boosting effect, but have no effect on the myocardial tension time index. Captopril has the effects of lowering blood pressure, slowing down the heart rate and reducing the myocardial tension time index, and significantly increasing the pulse pressure difference. [Conclusions] Peikun Pills can increase blood pressure of rats. Peikun Pills are suitable for women with hypotension accompanied by anemia. However, patients with hypertension are prohibited to use.

  • Safety Evaluation of Tongchui Baihua Capsule

    [Objectives] The study aimed to evaluate the safety of Tongchui Baihua capsule. [Methods] Long-term toxicology experiment was adopted, the rats were randomly divided into control group and administration group according to the gender of male and female; the drug administration group was given different doses of Tongchui Baihua capsules, and the drug was administered by continuous gavage for 13 weeks; the general conditions, haematological indexes, blood biochemical indexes and visceral coefficients of the rats were detected at the 13 th week and 4 weeks after the drug withdrawal respectively, and the toxic symptoms were observed. [Results] The rats were given Tongchui Baihua capsule in different doses by gavage for 13 weeks, and no obvious toxicity was observed in weight, haematological indexes, blood biochemical indexes and histopathology. [Conclusions] Under the condition of long-term toxicity experiment, Tongchui Baihua capsule was safe for rats.

  • Investigation on the Resources of Paris Polyphylla var. yunnanensis

    In this study, the resource distribution, morphological characteristics, artificial cultivation and market circulation of Paris Polyphylla var. yunnanensis and its polygerm varieties in Yunnan were investigated by referring to the research literature, field investigation and our planting experience. It is found that P. polyphylla var. yunnanensis is widely distributed in Yunnan Province, realizing large-scale artificial cultivation, while the wild resources of polygerm varieties are almost on the verge of extinction and are in urgent need of protection.

Processing of Traditional Chinese Medicine and Quality Control

  • Effect of Different Drying Processes on Active Ingredients of Rheum officinale Baill.

    [Objectives] To determine the drying process of different functional rhubarbs. [Methods] The contents of tannins, bianthrone, free anthraquinones and combined anthraquinones in medicinal rhubarb were determined by HPLC. The results were analyzed by two-way analysis of variance. [Results] Under different drying conditions, the contents of tannins, bianthrone, free anthraquinones and combined anthraquinones in medicinal rhubarb were significantly different. Taking the tannins content as the index, the optimum drying condition of the rhizome is 1 cm thick, dried at 60 ℃, the root is cut 1 cm thick, and dried at 30 ℃; the bianthrone content is used as an indicator, the rhizome is optimally dried. The condition is to cut 1 cm thick, dry at 60 ℃, the root is cut to 5 cm thick, and dried at 40 ℃; the free anthraquinones content is used as an index, and the optimum drying condition of the rhizome is 3 cm thick and dried at 50 ℃. The root is cut to a thickness of 3 cm and dried at 30 ℃. The combine anthraquinones content is used as an indicator. The optimum drying conditions for the rhizome are 5 cm thick, dried at 40 ℃, and the root is cut to 5 cm thick and dried at 70 ℃. [Conclusions] Different functional components of rhubarb have different directional processing methods. The drying process can reduce the drying temperature or increase the thickness of the slice, and the directional processing of the diarrhea-type rhubarb can be processed. The drying process can be carried out by increasing the drying temperature or reducing the thickness of the slice directed processing of heat-clearing and purging-fire rhubarb.

  • Optimization of Ultrasonic-enzymatic Extraction of Total Flavonoids from Cardamine tangutorum O. E. Schulz by Response Surface Methodology and Content Determination

    [Objectives] This study aimed to optimize the extraction process of total flavonoids in Cardamine tangutorum O. E. Schulz. [Methods] On the basis of single-factor experiments, the ultrasonic-enzymatic extraction process of total flavonoids in C. tangutorum O. E. Schulz was optimized by Box-Behnken design from aspects of ethanol volume fraction, extraction time, extraction temperature and enzyme addition, with yield of total flavonoids as the evaluation index. [Results] The optimal extraction process of total flavonoids in C. tangutorum O. E. Schulz was as follows: ethanol volume fraction of 70%, ultrasonic extraction time of 43 min, ultrasonic extraction temperature of 37 ℃ and cellulose addition of 0.07 mg. Under the optimal conditions, the yield of total flavonoids from C. tangutorum O. E. Schulz reached 75.90 mg/g. [Conclusions] The optimized extraction process was reasonable and feasible, and it could provide a reference for the extraction of total flavonoids in C. tangutorum O. E. Schulz. This study provided a foundation for the quality control of food/medicinal materials and the development and research of big health products of C. tangutorum O. E. Schulz.

  • Analysis on Quality Control of Argyreia acuta Lour

    [Objectives] To establish a method for caffeic acid content determination and thin-layer chromatography identification of the Zhuang medicine Argyreia acuta Lour. [Methods] Silica gel GF254(thin-layer plate), toluene-formic acid-ethyl acetate(V∶V∶V=5∶1.2∶3)(developing agent) and 365 nm ultraviolet light were used for thin-layer chromatography identification. Under the chromatographic conditions of column of Thermo SCIENTIFIC Hypersil C_(18)(5 μm, 4.60 mm × 250 mm), mobile phase of methanol-0.1% phosphoric acid(23∶77), detection wavelength of 320 nm, column temperature of 30 ℃, sample size of 10 μL and flow rate of 1.0 mL/min, the content of caffeic acid in A. acuta Lour was determined. [Results] Caffeic acid can be detected in thin layer chromatography, with strong specificity and clear spots. When the sample size of caffeic acid is within the range of 0.015 1-0.453 0 μg(R~2=0.999 9), the content of caffeic acid showed a relatively good linear relationship with the peak area. The average recovery rate of A. acuta Lour was 99.94%(RSD=2.68%), 97.56%(RSD=1.57%) and 99.79%(RSD=2.05%), respectively. [Conclusions] This method can effectively identify A. acuta Lour, and can accurately determine the content of caffeic acid in A. acuta Lour. It has characteristics of high accuracy, high precision, good color rendering stability, good reproducibility and fast analysis speed.

  • Evaluation on Quality and Utilization Value of Bupleurum falcatum Linne

    [Objectives] This study aimed to evaluate the quality of Bupleurum falcatum Linne interplanted with walnut in Ganluo and study the development and utilization value of other parts. [Methods] The contents of saikosaponins a and d were determined with RP-HPLC, and the contents of 7 kinds of harmful elements were determined by ICP-MS. The quality of B. falcatum Linne was compared with that of other Bupleurum species. [Results]The total saikosaponin content in Radix Bupleurum Falcatum(15.894 mg/g) was higher than that in Radix Bupleuri of other origins(8.748 mg/g). The total saikosaponin content in leaves of B. falcatum Linne(7.518 mg/g) is more than twice the limit promulgated by Chinese Pharmacopoeia. The contents of Al, Cr, Cu, As, Cd, Pb and Hg in B. falcatum Linne were all lower than the limits promulgated by the pharmacopoeia. In short, the quality of Radix Bupleurum Falcatum was better than that of Radix Bupleurum Marginatum(S3, S4) and Radix Bupleurum Sachalinense(S5). The leaves of B. falcatum Linne contained more saikosaponins and less harmful elements. [Conclusions] The method and technology of interplanting B. falcatum Linne with walnut in Ganluo are mature. The quality of the medicinal materials produced is superior, and the leaves are also rich in saikosaponins a and d, and can be used as the raw material for extracting saikosaponins a and d. This study provides a basis for further in-depth research on the cultivation of B. falcatum Linne in the domestic market.

  • GC-MS Analysis of Seeds from Different Pomegranate Varieties

    [Objectives] This study aimed to study and analyze the main components of the oil from seeds of 8 different pomegranate varieties. [Methods] The oil in sour and sweet pomegranate seeds was extracted by ultrasonic-assisted method, and the composition of the soil after methyl esterification was identified by GC-MS. [Results] The oil yield of the four sweet pomegranate varieties and the four sour pomegranate varieties ranged from 9.43%-16.97%. A total of 6 compounds were identified in the methyl esterified pomegranate seed oil by GC-MS. [Conclusions] There were no significant differences in oil yield between sweet and sour pomegranate varieties(P>0.05). The content of methyl punicate in the four sweet pomegranate varieties was 52.18%-78.06%, and that in the four sour pomegranate varieties was 78.25%-84.55%. The content of methyl punicate in sour pomegranate was slightly higher than that in sweet pomegranate.

  • Extraction Process and Content Determination of Total Flavonoids in Lonicera japonica

    [Objectives] To optimize the extraction process of total flavonoids of Lonicerae japonica and establish a method for its extraction and content determination. [Methods] The total flavonoids of L. japonica were extracted by reflux extraction method. Through a single factor experiment, the effects of extraction method, extraction solvent concentration, extraction volume and extraction time on the total flavonoids content of medicinal materials were investigated to select the optimal extraction process of the total flavonoids of L. japonica. [Results] The optimal extraction process of the total flavonoids of L. japonica was 70% ethanol, 1∶30 of solid-to-liquid ratio, and 1.0 h of reflux extraction time. [Conclusions] This method can effectively determine the content of total flavonoids in L. japonica and is expected to provide a certain scientific basis for the study of the quality standard of L. japonica. This method has high reproducibility. It is stable and feasible in extraction of total flavonoids from L. japonica.

  • Effects of Different Drying Methods on Content of Paederosidic Acid in Paederia scandens

    [Objectives] To establish a method to simultaneously determine the content of paederosidic acid in Paederia scandens dried by four different methods. [Methods] Reversed-phase high performance liquid chromatography(RP-HPLC) was adopted. The chromatographic column: Thermo SCIENTIFIC Hypersil GOLD Dim.(mm); mobile phase: acetonitrile-0.1% phosphoric acid aqueous solution; gradient elution; column temperature: 30 ℃; flow rate: 1 mL/min; detection wavelength: 236 nm; injection volume: 10 μL. [Results] The linear range of the detection injection volume of paederosidic acid was 0.64-9.60 μg(R=0.999 2); the limit of quantity(LOQ) was 5.10 ng, and the limit of detection(LOD) was 1.36 ng; the RSD of the precision, stability and reproducibility test was all less than 3%; the sample recovery rate was 98.87%, RSD<3.00, n=6. The results show that the content of paederosidic acid in the shade-dried P. scandens is the highest, and the order of the content is P. scandens(dry in shade)>P. scandens(dried at 50 ℃)>P. scandens(dried at 60 ℃)>P. scandens(dried at 70 ℃). [Conclusions] This method is sensitive, reliable and reproducible, and can be used to simultaneously determine the content of paederosidic acid in P. scandens dried by four different methods.

  • Extraction Optimization of Polysaccharides from Rhizoma Polygonati by Response Surface Methodology and Content Determination

    [Objectives] This study aimed to investigate the water extraction-alcohol precipitation technology and content determination method of polysaccharides in Rhizoma Polygonati. [Methods] Taking the content of polysaccharides in the extract as the indicator, the extraction process was optimized using single-factor tests and Box-Behnken Design. The content of polysaccharides extracted was determined by the anthrone-sulfuric acid method. Among the three factors in the test design, the solid/liquid ratio and extraction time had a significant effect(P<0.05, P<0.01), while the extraction temperature had no effect on the content of polysaccharides extracted(P>0.05). [Results] The optimal extraction process for polysaccharides in Rhizoma Polygonati was as follows: liquid/solid ratio 20∶1(mL/g), extraction time 2 h, and extraction temperature 85 ℃. Under the optimal extraction process, the content of polysaccharides in the extract reached 12.50%. [Conclusions] The extraction process optimized by response surface methodology for polysaccharides in Rhizoma Polygonati is reliable and has practical value, and can be used to guide production practices.

  • Optimization of Extraction Process of Quercetin from Flos Sophorae Immaturus Using Orthogonal Design

    [Objectives] This study aimed to optimize the ultrasonic-assisted extraction process of quercetin from Flos Sophorae Immaturus. [Methods] The natural product quercetin in Flos Sophorae Immaturus was extracted by ultrasonic-assisted method, and the content of quercetin in the extract was determined by ultraviolet spectrophotometry. The effect of ethanol concentration, solid/liquid ratio, temperature, time and ultrasonic power on the yield of quercetin in Flos Sophorae Immaturus was investigated by using single-factor experiments. Based on the results of single-factor experiments, an orthogonal experiment was designed to optimize the extraction process. The experimental data obtained were subjected to range analysis, analysis of variance and SSR test using SPSS 20.0 software to obtain the optimal extraction process. The results concluded were verified. [Results] The optimal ultrasonic-assisted extraction process for quercetin in Flos Sophorae Immaturus was as follows: temperature of 85 ℃, time of 30 min, solid/liquid ratio of 1∶20 g/mL, ethanol concentration of 50%. Under the optimal extraction conditions, the yield of quercetin in Flos Sophorae Immaturus was the highest. [Conclusions] This study provides a theoretical basis for the application of quercetin in Flos Sophorae Immaturus.

  • Thin-layer Identification, Extraction Process and Content Determination of Chlorogenic Acid in Laggera alata (D. Don) Sch. Bip. ex Oliv.

    [Objectives] This paper aims to establish thin-layer identification and content determination method for Laggera alata(D. Don) Sch. Bip. ex Oliv. with chlorogenic acid as the index component and compare the content of chlorogenic acid in L. alata from different places in Guangxi. [Methods] Silica gel GF254 thin-layer plate was used for identification under an ultraviolet lamp(365 nm), with butyl acetate-formic acid-water(V∶V∶V=7∶2.5∶2.5) as a developing agent. The content of chlorogenic acid was determined under the following chromatographic conditions: column, Inertsil ODS-3 C_(18) column(4.60 mm × 250 mm, 5 μm); mobile phase, methanol-0.1% phosphoric acid(28∶72); detection wavelength, 329 nm; flow rate, 1.0 mL/min; column temperature, 25 ℃; and injection volume, 10 μL. [Results] Chlorogenic acid can be detected by thin layer chromatography with clear spot and good specificity. Chlorogenic acid showed a good linear relationship in the injection amount range of 0.099-0.99 μg(R~2=0.999 9). The content of chlorogenic acid in L. alata varied greatly among the 10 different producing areas in Guangxi. L. alata produced in Dee Township, Longlin, Baise, Guangxi showed the highest chlorogenic acid content, and that produced in Shangsi County and Pingle County showed the lowest chlorogenic acid content. [Conclusions] This method can effectively identify L. alata and accurately determine the content of chlorogenic acid, thereby providing a scientific basis for the development and utilization of L. alata resources.

  • Prediction of Triptolide Targets in Colorectal Cancer Using Network Pharmacology and Molecular Docking

    [Objectives] To investigate the potential mechanisms of action of triptolide, an active component in the traditional Chinese medicine Tripterygium wilfordii Hook F, in colorectal cancer(CRC). [Methods] Public databases were first searched for genes and proteins known to be associated with CRC, as well as those predicted to be targets of triptolide, and then Ingenuity Pathway Analysis(IPA) was applied to identify enriched gene pathways and networks. Networks and pathways that overlapped between CRC-associated proteins and triptolide target proteins were then used to predict candidate protein targets of triptolide in CRC. [Results] The following proteins were found to be expressed in both CRC-associated networks and triptolide target networks: JUN, FOS, CASP3, BCL2, IFNG, and VEGFA. Docking studies suggested that triptolide can fit in the binding pocket of the four top candidate triptolide target proteins(CASP3, BCL2, VEGFA and IFNG). The overlapping pathways were activation of neuroinflammation signaling, glucocorticoid receptor signaling, T helper(Th) cell differentiation, Th1/Th2 activation, and colorectal cancer metastasis signaling. [Conclusions] These results show that network pharmacology can be used to generate hypotheses about how triptolide exerts therapeutic effects in CRC. Network pharmacology may be a useful method for characterizing multi-target drugs in complex diseases.

    [Objectives] To establish HPLC fingerprints of wild and cultivated Millettia speciosa Champ., and identify medicinal materials combined with chemical pattern recognition methods, and provide a reference system for the identification and quality control of M. speciosa from different sources. [Methods] 20 batches of M. speciosa from different sources were determined by HPLC method, and the similarity analysis and evaluation were performed using the Similarity Evaluation System of Traditional Chinese Medicine Chromatographic Fingerprints(2012 edition). Principal component analysis(PCA) and least partial squares method-discrimination analysis(PLS-DA) were used to conduct chemical pattern recognition research on wild and cultivated M. speciosa. [Results] The HPLC fingerprints of wild and cultivated M. speciosa were established, 10 common peaks were calibrated, and the similarity of 20 batches of samples was greater than 0.9; PCA can better classify M. speciosa from different sources into 2 categories, and PLS-DA can completely distinguish between wild and cultivated M. speciosa. [Conclusions] The established M. speciosa fingerprint, combined with chemical pattern recognition methods, can effectively distinguish between wild and cultivated M. speciosa, so it can provide a reference for quality control and evaluation of M. speciosa.

  • Influence of Moxibustion of Complementary Acupoints on Analgesic Effect and Mast Cell Function in CIA Rats

    [Objectives] To explore the moxibustion effect and mechanism of "Complementary Acupoints" on rheumatoid arthritis(RA). [Methods] A total of 32 Wistar rats were randomly divided into 4 group: normal control(K), model group(M group), "the Complementary Acupoints" Yanglingquan and Yinlingquan group(AB group), Shenshu and Zusanli group(CD group), with 8 rats in each group. Moxibustion was performed using refined moxa floss(2 mg/pc) on acupoints in both sides every other day, 5 pieces for each acupoint, and implemented for 10 times. Before and after the modeling and after the intervention, the left foot sole thermal pain threshold of each group was measured. After the intervention, the rats' left paws were examined by X-ray to detect the number of mast cells(MCs) and degranulation in the affected area of the foot. [Results] The thermal pain threshold of the model group was lower than that of the K group(P<0.05). The number of MC in the affected area and the degranulation were increased. After the moxibustion intervention, the thermal pain threshold of the AB and CD groups was higher than that of the M group(P<0.05); the number of MCs and degranulation were decreased compared with M group, and the degree of reduction was better than that of CD group. The data of foot and claw showed that the foot and paw soft tissue swelling, bone and joint deformity and bone destruction were better in AB and CD groups were more relieved than that in M group. [Conclusions] The moxibustion of Complementary Acupoints can effectively intervene with RA of rats, and its mechanism may be closely related to the MC degranulation.

  • Anshen Babu Ointment Improves Sleep Function of Insomnia Mice and Its Mechanism

    [Objectives] This study aimed to study the sleep-improving effect of Anshen Babu ointment on insomnia mice and its mechanism of action. [Methods] Mouse models of insomnia were established by p-chlorophenylalanine(PCPA) injection. They were given high, middle and low doses of Anshen Babu ointment, for 7 consecutive days, and then, the spontaneous activity of insomnia mice and the effect on hypnotic time and subthreshold hypnotic dose of pentobarbital sodium were observed, and the levels of 5-HT, IL-6 and TNF-α in the hippocampus of mice were determined. [Results] High and middle-dose Anshen Babu ointment significantly reduced the spontaneous activity of insomnia mice and shorten the hypnotic time of pentobarbital sodium; high and middle-dose Anshen Babu ointment increased the 5-HT content and inhibited the IL-6 and TNF-α expression in the hippocampus of mice. [Conclusions] Anshen Babu ointment has hypnotic and sedative effect on insomnia mice. The mechanism may be achieved by increasing 5-HT and reducing IL-6 and TNF-α levels.

  • Acute Toxicity and Toxicokinetics of Tongguan Powder in Rats

    [Objectives] To evaluate the acute toxicity and toxicokinetics of Tongguan Powder in rats, and provide references for clinical safe medication. [Methods] The classical acute toxicity test method was used, rats were given different doses of Tongguan Powder through the mouth and nasal cavity to observe the symptoms of toxicity, and make a record of the food intake, weight changes, and death. After the medication, blood was taken from each group of rats at different time points, and the plasma levels of benzoyl aconitine(BA), benzoyl hypaconine(BH) and benzoyl mesaconine(BM) were determined by the liquid chromatography tandem-mass spectrometry(LC-MS/MS), and the toxicokinetic parameters were fitted with the aid of DAS software. [Results] Rats were given Tongguan Powder 3.75 g/kg(equivalent to 54 times the human daily dose) in the nasal cavity of rats. Rats were observed with reactions such as scratching and sneezing; rats were given Tongguan Powder LD_(50) and 95% confidence limit of 4.15(3.53-4.71) g/kg through the oral administration, which is equivalent to 60 times the human daily dose, rats showed slow weight gain, decreased food intake, decreased voluntary activities, prone, black stools, etc. One hour after nasal administration of Tongguan Powder, the plasma concentration of benzoyl aconitine and benzoyl hypaconine was below the lower limit of detection, and benzoyl mesaconine could not be detected at any time point; one hour after the oral administration of Tongguan Powder, the plasma concentration of the three components reached the maximum, the exposure level of benzoyl hypaconine was higher than that of benzoyl aconitine and benzoyl mesaconine; there was no gender difference in the kinetic parameters. [Conclusions] The toxicity of Tongguan Powder in nasal administration is much lower than that of intragastric administration. The target organs and mechanism of toxicity need to be further studied.

  • Expression of P-gp and miRNA-21 in Colorectal Cancer Tissues and the Clinical Significance

    [Objectives] To study the expression of P-gp and miRNA-21 in colorectal cancer tissues and the relationship between the expression of P-gp and clinicopathological features, and to analyze the correlation between the expression of miRNA-21 and P-gp. [Methods] Immunohistochemical SP method was used to detect the expression level of P-gp in the tissues of 40 cases of colorectal cancer and the normal mucosal tissues 5 cm away from the corresponding cancer tissues, and the expression level of miRNA-21 in the paraffin tissues of the 40 cases was detected by RT-qPCR. [Results] The positive expression of P-gp was localized in the cell membrane, and the expression in colorectal cancer tissues was significantly higher than that in the corresponding adjacent tissues(P<0.05); the positive expression of P-gp was closely related to the degree of tumor differentiation and the depth of invasion(P<0.05), and the comparison results of P-gp expression in the differentiation groups of different cancer tissues showed that the expression level of P-gp in the moderately and poorly differentiated groups was significantly higher than that in the highly differentiated group(P<0.05), and the expression level of the moderately and poorly differentiated groups was similar(P>0.05). The expression of miRNA-21 in all cancer tissues was higher than that in the corresponding adjacent tissues(P<0.05), and the expression levels of miRNA-21 in differentiation groups of different cancer tissues were 1.286, 1.445 and 1.992 times of those in the corresponding adjacent tissues, respectively; the comparison results of miRNA-21 between different differentiation groups showed that the expression level of miRNA-21 in the poorly differentiated group was significantly higher than that in the highly differentiated group and the moderately differentiated group(P<0.05), while the expression level of miRNA-21 in the highly differentiated group and the moderately differentiated group was similar(P>0.05). Spearman correlation analysis showed that there was a positive correlation on the expression between the two in colorectal cancer tissues(r=0.423, P<0.05). [Conclusions] The expression levels of miRNA-21 and P-gp are closely related to the differentiation degree of clinical colorectal cancer, the lower the differentiation degree, the higher the expression level of the two.
