  • Advance in Pathogenesis of Gout and Related Gene Polymorphism

    Lamei ZHOU;Wen XU;Chunqing LI;

    Gout is an independent risk factor for hypertension, diabetes mellitus, hyperlipidemia, coronary heart disease and cerebral infarction. At present, the studies on the mechanism of gout at home and abroad have mainly focused on immune inflammation, gene polymorphism and related studies. Uric acid deposition or crystal precipitation activates phagocytes, fibroblasts and mast cells in synovium, produces IL-1β, TNF and chemokine IL-8(CXCL8), thereby activating neutrophils, urate crystal polyanion surface can be coated with immunoglobulins and other serum proteins as substrates for complement activation, complement substitution pathways and classical complement pathways to activate complements. Uric acid crystals activate inflammation-related signal transduction pathways including the activation of inflammation-related signal transduction pathway by uric acid crystals and TLRS/MyD88 signal transduction pathways. Gene polymorphism is related to inflammation and signaling pathway, EGF gene is closely related to gout inflammation, which may be involved in the regulation of gout inflammation, among which NLRP3 inflammatory signaling pathway and gene polymorphism have been deeply studied in the pathogenesis of gout, which is the main therapeutic target of anti-inflammatory and uric acid lowering.

    [Objectives] This study aimed to investigate the inhibiting effect of Obazema on proliferation of gastric cancer BGC-823 cells and its mechanism. [Methods] BGC-823 cells were treated with high, medium and low concentrations of drug-containing serum(0.316%, 0.158% and 0.079%) for 0, 48, 72 and 96 h, respectively. Then, the proliferation of the cells was detected with CCK-8 method, and the expression of related proteins, B lymphocytoma-2(Bcl-2), phosphorylated protein kinase B(p-Akt), protein kinase B(Akt) and glyceraldehyde-3-phosphate dehydrogenase(GAPDH), was detected using Western blotting. [Results] The proliferation of the BGC-823 cells was significantly inhibited with different doses of Boenninghausenia albiflora(Hook.) Reichb. ex Meisn. var. albiflora(CH) and B. sessilicarpa Lévl.(S)(P<0.01), in dose-dependent and time-dependent manners. The inhibition of high-dose S on cell proliferation was similar to that of CTX 48 h after administration; the inhibition of high-dose CH on cell proliferation was significantly stronger than that of CTX(P<0.01); different doses of drug administration groups significantly inhibited the expression of p-Akt and Bcl-2 in the BGC-823 cells; the inhibition of high-dose CH on the expression of P-Akt and Bcl-2 and the inhibition of medium-dose CH on the expression of Bcl-2 were significantly stronger than that of CTX(P<0.05, P<0.01), in a certain dose-dependent manner; at the same dose, the inhibition of CH on the expression of the proteins was stronger than that of S(P<0.05, P<0.01); administration of S and CH significantly inhibited the expression of GAPDH compared with CTX(P<0.05, P<0.01). [Conclusions] Obazema has the capacity to inhibit the proliferation of BGC-823 cells. The mechanism may be achieved by inhibiting the expression of p-Akt and Bcl-2, and GAPDH may be the target gene of its anti-tumor mechanism. The inhibiting effect of CH on BGC-823 cells was more significantly than that of S.

    [Objectives] This study aimed to explore the protective effect of Oxalis coriniculata L. on rats with acute liver injury induced by carbon tetrachloride(CCl_4) and related mechanism by regulating oxidative stress and the TLR-2 TLR-2/NF-κB signaling pathway. [Methods] A total of 48 female rats were randomly and evenly divided into normal group, model group, silymarin group(0.12 g/kg), and high(16 g/kg), middle(8 g/kg) and low-dose(4 g/kg) O. coriniculata L. groups. The rats in the groups were intragastrically administered with 5 mL/kg of corresponding drugs(equal-volume distilled water for normal group and control group), respectively. The administration was conducted twice a day, for 10 consecutive days. After 2 h of the last administration, the rats in all the groups except the normal group were intraperitoneally injected with 12% carbon tetrachloride(CCl_4) olive oil solution(5 mL/kg), respectively to establish liver injury rat models. After 16 h, the eyeball blood of the rats was collected, and their liver tissues were collected for preparation of HE sections. The biochemical indicators detected included aspartate aminotransferase(AST), alanine aminotransferase(ALT), total superoxide dismutase(T-SOD) and glutathione peroxidase(GSH-Px) activity and malondialdehyde(MDA) content in the serum. The contents of tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β) and interleukin-6(IL-6) in the serum were detected by ELISA. The expression of Toll-like receptor-2(TLR-2) and nuclear factor-κB(NF-κB) in liver tissue was detected using Western blotting. The pathological changes of liver were observed under light microscope. [Results] Compared with the normal group, the ALT, AST activity and MDA, IL-1β, IL-6, TNF-α levels in rat serum significantly increased(P<0.01), the GSH-Px, T-SOD activity in rat serum significantly decreased(P<0.01), and the expression of TLR-2 and NF-κB in liver tissue was up-regulated(P<0.01) in the model group. Compared with the model group, the ALT, AST activity and MDA, IL-1β, IL-6 and TNF-α levels in rat serum reduced(P<0.05, P<0.01), the GSH-Px and T-SOD activity in rat serum increased(P<0.05, P<0.01), and the expression of TLR-2 and NF-κB in liver tissue was down-regulated(P<0.05, P<0.01) in the O. coriniculata L. administration groups. Pathological sections show that O. coriniculata L. had an improving effect on rats with acute liver injury induced by CCl_4. [Conclusions] O. coriniculata L. has a good protective effect on rats with acute liver injury induced by CCl_4. Its mechanism may be related to inhibition of oxidative stress, inhibition of inflammatory response and regulation of the TLR-2/NF-κB signaling pathway.

[Methods] The in vitro cytology studies were carried out to confirm the expression of the AURKB gene in HCT 116 cells and make clear its role in cell activity, cell cycle control and apoptosis, and investigate the effect of AURKB gene in colorectal cancer(CRC). Quantitative reverse transcription/polymerase chain reaction(PCR) analysis and immunofluorescence(IF) staining for markers of AURKB gene were used to examine the effect of AURKB on HCT 116. The AURKB gene target was examined using western blot analysis. In addition, inhibition of AURKB expression was examined using RNA interference(RNAi) on HCT 116 cells in vitro. [Results] HCT 116 cells infected with AURKB shRNA virus suppressed expression of AURKB in vitro. AURKB gene knockdown HCT 116 cells showed reducing cell apoptosis in vitro. Finally, it demonstrated that AURKB function can induce apoptosis of HCT cells. [Conclusions] AURKB is a key regulator of colorectal cancer. AURKs are potential novel molecular targets for the prevention of cancer cell proliferation.
