  • Research Progress on Biological Activity of Cytisine

    Cytisine(Cyt) is a kind of quinolizidine alkaloids mainly existing in seeds of plants from Laburnum of Leguminosae sp. In recent years, various biological activities of Cyt have been explored gradually, and it has the effects of quitting smoking, promoting breathing, analgesia, and anti-cancer. At anti-cancer aspect, Cyt could induce apoptosis of cancer cells, and has been widely applied to inhibit proliferation of multiple tumor cells. At quitting smoking aspect, Cyt could be taken as selective local agonist to decline smokers' dependence on nicotine, and it is expected to be widely used in quitting smoking. At analgesia aspect, Cyt could regulate reflective exciting breathing and has been widely used in anesthesia of patients in clinical operation. In this paper, through consulting and verifying a large number of documents, biological activities of Cyt and its related action mechanisms are summarized, to provide theoretic basis for the development and utilization of Cyt.

  • Determination and Multidimensional Analysis of 22 Kinds of Inorganic Elements in Leontopodium franchetii Beauv. from Different Production Areas

    [Objectives]To determine the content of inorganic elements in Leontopodium franchetii Beauv. from different production areas, and provide a reference for quality control, safety assessment, and clinical application of L. franchetii Beauv.. [Methods] The contents of Al, B, Ba, Ca, Cd, Cu, Fe, K, Mn, Mo, Na, Ni, P, Pb, Si, Sn, Sr, Ti, V, Zn, Hg, Cr in the sample of L. franchetii Beauv. were determined by the wet digestion and technique of inductively coupled plasma optical emission spectrometry(ICP-OES), and the principal component analysis was carried out for the determination results. [Results] A total of 20 inorganic elements were detected from the samples of L. franchetii Beauv. from different production areas, among which the contents of Ca, Al, P and Fe were higher. There were significant differences among different elements, and samples of different production areas show similar distribution patterns of main elements. Correlation analysis shows that 17 elements had an extremely significant positive correlation and 19 elements had a significant positive correlation. According to the principal component analysis, B, Ca, K, Mn, Mo, Pb, Sn, Sr, Fe, Ni, Ti, V, Zn, Al, and Cd were characteristic inorganic elements of L. franchetii Beauv. [Conclusions] This method is simple, rapid and accurate, and can be used for the analysis and determination of inorganic elements in L. franchetii Beauv., and provides a theoretical basis for better development and utilization of L. franchetii Beauv.

  • Density Functional Theoretical Study on Antioxidant Activity of Four Anthocyanins from Purple Sweet Potato

    This study aimed to analyze the antioxidant mechanism of four different anthocyanins in purple sweet potato. The DPPH test and the lipid peroxidation inhibition ability test were used to analyze the in vitro antioxidant activity of four anthocyanins, and then the optimal structure, bond dissociation energy(BDE) and ionization potential(IP) of the anthocyanins and their simplified molecular models were analyzed by density functional theoretical study, and the antioxidant mechanisms were explored. The results showed that OH-4′ phenolic hydroxyl group of the four anthocyanins has the highest activity, and its bond dissociation energy is less than that of resveratrol. The theoretical calculation results of antioxidant activity are consistent with the results of in vitro antioxidant tests. The results showed that anthocyanins of purple sweet potato have good antioxidant activity, and the DFT method provides a powerful theoretical basis for the development of anthocyanin antioxidant activity.

  • Extraction Process of Basil Essential Oil by Steam Distillation

    [Objectives] To optimize volatile oil extraction from Ocimum basilicum with the water vapor distillation. [Methods] An orthogonal design was carried out to determine the volume of volatile oil by 3 factors: the immersion time, distillation time, amount of water. The ratio of the oil in inclusion complex was used to evaluate the technology based on the orthogonal design. [Results] The best volatile oil extraction condition was to add 400 mL of water into the mixture of crude drugs, and to extract the herbal medicine for 2 h with advanced soaking for 6 h with water. [Conclusions] The process is stable, reasonable, and feasible.

  • Chemical Constituents and Antitumor Activities of Walnut Green Husk

    [Objectives] To identify chemical constituents and antitumor activities of walnut green husk. [Methods] The column chromatography and recrystallization were used to isolate 13 compounds from walnut green husk, and their structures were identified by modern spectroscopy. [Results] The 13 kinds of compounds isolated from walnut green husk were identified as 3, 5-dimethoxy-4-hydroxy benzene carbonic-7-O-β-D-pyranglucose(1), myricananin F(2), β-stitosterol(3), oleanolic acid(4), regiolone(5), asiatic acid(6), hederagenin(7), β-daucosterin(8),(4 R)-4,8-dihydroxy-α-tetralone-4-O-β-D-glucopyranoside(9), 3β, 23-dihydroxy-urs-12-en-28-oic acid(10), ursolic acid(11), syringc acid(12), and Juglanoside B(13). Among these, compounds 1, 2, 6, 7, 9, and 10 were all isolated from this genus of plant for the first time. [Conclusions] Through screening for antitumor activity in vitro, it was found that compound 4 showed strong growth inhibitory activity against the proliferation of human hepatoma cells(HepG-2), and compound 9 had certain growth inhibitory activity against the proliferation of human breast cancer cells(MCF-7).

  • Cytological Observation of Pollen Abortion of Lycium barbarum Haploids from Anther Culture

    To reveal the sterility characteristics of Lycium barbarum haploids, cytological observations were made on the anthers of Ningqi No.1 and its haploids obtained from anther culture. The results showed that there were no significant differences in anther development between Ningqi No.1 and its haploids at the stage of pollen mother cell, and tetrads were formed successfully after the meiosis stage. The tetrads of Ningqi No.1 could release microspores. At the same time, the tapetal cells can provide nutrition for the development of the microspores, which eventually developed into mature pollen grains. Although the haploids could also release microspores at the tetrad stage, the tapetal cells degraded in advance, which made the released microspores unable to develop into mature pollen grains, resulting in pollen abortion of haploids.

  • Lipase-catalyzed Synthesis of 6-O-acyl-D-galactose Ester for Surface Modification of Cantharidin Liposomal Nanoparticles

    [Objectives] To prepare and characterize a novel liver targeted drug carrier. [Methods] 6-O-acyl-D-galactose esters were enzymatically synthesized as a targeting molecule and incorporated into liposomes to prepare liposomal nanoparticles surface-modified with galactose. [Results] Liposomes were characterized by morphology, particle distribution size, zeta potential and encapsulating efficiency of drugs. [Conclusions] This novel approach for the liposomes surface-modifed with galactose by enzymatic synthesis is expected to provide potential application as drug carrier for active targeted delivery to hepatocytes.

  • Analysis of Amino Acids in Houttuynia Cordata Thunb. by High Performance Liquid Chromatography-Diode Array Detector (HPLC-DAD)

    [Objectives] The biologically active compounds of Houttuynia cordata Thunb. as a medicinal and edible plant are well documented. However, amino acid as an important indicator of the value of H. cordata is little reported. [Methods] Amino acids of the H. cordata were analysed by pre-column derivatization with 2,4-dinitrochlorobenzne(CDNB) coupled to HPLC/DAD. The optimized conditions of the amino acids derivatized by CDNB were followed: 110 ℃ of hydrolysis temperature, 1∶12 of the equivalent proportion between amino acid and CDNB, 80 min of derivatization reaction time and 80 ℃ of derivatization reaction temperature. The method was used to determine the amino acids of H. cordata samples from 11 different geographical sites in Guizhou. [Results] The results suggested that the contents of 17 amino acids were quite variable at different H. cordata samples. [Conclusions] This study will provide a basis for the quality evaluation of H. cordata nutrition.

  • Study on HPLC Fingerprint and Simultaneous Determination of Three Active Components of Dipsaci Radix Collected in Different Time

    [Objectives] To establish methods of HPLC fingerprint and simultaneous determination of the three bioactive components(namely, asperosaponin VI, loganin and sweroside) of Dipsaci Radix, and explore the quality situation of this crude medicine collected in various months in autumn. [Methods] The analysis was performed on Thermo BDS Hypersil C_(18)(4.6 mm×250 mm, 5 μm) column with a mixture of acetonitrile and 0.1% phosphoric acid as mobile phase in gradient mood at a flow rate of 1.0 mL/min, the column temperature was set at 30 ℃, and the detection wavelength was 220 nm. [Results] The analysis methods for HPLC fingerprint and determination of the three components in Dipsaci Radix had been established. The results showed that 12 batches of samples, which were collected in four different places from September to November, possessed the similarities of greater than 0.976. Through the quantitative analysis of asperosaponin VI, loganin and sweroside in Dipsaci Radix, it was found that the quality variation of this herbal medicine and the different collected months of autumn showed a low correlation. [Conclusions] The established methods of HPLC characteristic fingerprint and simultaneous determination of three glycosides provided a new way for quality analysis of Dipsaci Radix. It was preliminarily indicated that collecting this plant medicine in different months of autumn would not affect its quality.

Pharmacological and Toxicological Analyses

  • Protective Effect of Paeonol on Diabetic Cardiomyopathy in Rats

    [Objectives] The protective effect of paeonol on DCM was further confirmed by observing the changes of myocardial structure and function in rats. [Methods] Rats were fed high-sugar and high-fat diet and injected intraperitoneally with streptozotocin(STZ) for two consecutive days to prepare animal model of type II diabetes. The model rats were randomly divided into model group, low-dose paeonol group(30 mg/kg), middle-dose paeonol group(60 mg/kg), high-dose paeonol group(120 mg/kg) and metformin group(157 mg/kg). The detected indicators included heart weight index(HWI), blood glucose(FBG), body mass, triglyceride(TG), and total cholesterol(TC). Myocardial pathological changes were observed by HE staining. The extent of myocardial fibrosis was observed by Masson staining. [Results] In the model group, the body weight of the rats declined significantly, and the phenomena of overdrinking, overeating and polyuria occurred. Compared with the model group, the body weight of rats in the paeonol treatment groups increased to different extents. Compared with the normal group, the HWI, FBG, LDH, AST, and TG increased in the model group, and decreased in different degrees in the paeonol treatment groups(P<0.05), suggesting that paeonol has certain protective effect on diabetic cardiomyopathy in rats. After HE staining and fixation, the myocardial tissue of rats was observed by optical microscope. The morphology of myocardial cells and myocardial fibers in the normal group were normal;Compared with the normal group, the model group has hypertrophy of myocardial cells, disordered arrangement of myocardial fibers, fiber breakage and dissolution, uneven staining between myocardial cells, and nucleus breakage or even disappearance;Compared with the model group, the pathological changes of myocardium in the paeonol treatment groups and DMBG group have been improved to different degrees. The arrangement of myocardial fibers is regular and the coloration is uniform. The results of Masson staining showed that in the normal group, and there was no significant increase of myocardial collagen fibers in the myocardial interstitium. Compared with the normal group, myocardial cells and interstitial collagen fibers(blue) in the model group were significantly increased. Compared with model group, myocardial collagen fiber hyperplasia in the paeonol treatment groups and DMBG group showed different degrees of improvement, among which the Pae-H group had the least blue bands and the most obvious improvement of collagen fiber. [Conclusions] Paeonol has different degrees of improvement effects on myocardial fibrosis in rats with diabetic cardiomyopathy, suggesting that paeonol has a protective effect on diabetic cardiomyopathy in rats.

  • Effects of Different Processed Products of Ilex hainanensis Merr on Rats with Hyperlipidemia

    [Objectives] This study aimed to investigate the hypolipidemic effects of different processed products of Ilex hainanensis Merr to provide experimental basis for the research on the hypolipidemic mechanism of I. hainanensis Merr. [Methods] SD rats were fed high-fat diet to establish hyperlipidemia rat models. The hyperlipidemia rats were administered with different processed products of I. hainanensis Merr by gavage to investigate the preventive and therapeutic effects of the medicinal material. [Results] The levels of total cholesterol(TC), triglyceride(TG), low density lipoprotein cholesterol(LDL-C) in the rats fed high-fat diet were significantly higher than those in of the control group(P<0.01), indicating successful modeling. Among different processing methods, the hypolipidemic effects were in the order as baking=traditional processing=frying=microwaving>drying in shade. Among different baking times, the hypolipidemic effects ranked as 180 ℃>200 ℃=220 ℃. Among different baking times, the hypolipidemic effects ranked as 20 min=40 min>30 min. The hypolipidemic effects of processed product of I. hainanensis Merr with different amounts of active ingredients were different. [Conclusions] The processed products of I. hainanensis Merr all had hypolipidemic effects, but the effects were different. This study will provide a scientific basis for expanding the resources of I. hainanensis Merr.

  • In vitro Antioxidant Activity of Leontopodium franchetii Beauv.

    [Objectives] To study the in vitro antioxidant activity of L. franchetii Beauv. [Methods] The DPPH and ABTS~+ scavenging rate and reducing power of L. franchetii Beauv. were detected by ultraviolet spectrophotometry. [Results] The 70% ethanol extract of L. franchetii Beauv. had strong antioxidant capacity, and its antioxidant capacity increased with the increase of mass concentration. [Conclusions] The 70% ethanol extract of L. franchetii Beauv. has higher DPPH and ABTS~+ scavenging rate and reducing power.

Processing of Traditional Chinese Medicineand Quality Control

  • Application of Fingerprint Combined with QAMS in Quality Evaluation of Zhenrongdan Mixture

    [Objectives] The research aimed to evaluate the quality of Zhenrongdan mixture by fingerprint combining QAMS. [Methods] The quality evaluation method was established and validated with echinacoside as internal reference to determine the contents of other components(ferulic acid, salvianolic acid B, and icariin) according to the relative correction factor. The accuracy and feasibility of QAMS were evaluated by comparison on the results between the measured value and calculation value by external standard method and QAMS. [Results] A common pattern of characteristic fingerprint of Zhenrongdan mixture by HPLC was established. Thirteen common peaks were identified, and they account for 91% of the total peak area, and four components were verified in five batches of Zhenrongdan mixture. Good similarities with correlation coefficients higher than 0.99 were found in the fingerprints. There was no significant difference between the quantitative results of the four ingredients in the five batches by external standard method and QAMS. [Conclusions] The method of fingerprint combined with QAMS could be used for the quality control of multiple components determination and fingerprint chromatography for Zhenrongdan mixture.

    [Objectives] Via systematic comparative study on Paris daliensis and Paris dulongensis, the research aimed to provide test data for their character and microscopic identification. [Methods] Original plants, characters, microscopic identification of cross section and powder of rhizome were used. [Results] Microscopic characteristics of cross section and powder of rhizome were basically consistent with identification results of literature, and parenchyma cell of P. dulongensis had brown-red mass. [Conclusions] The research could provide reference basis for identification of authenticity and establishment of quality standards for original plants and medicinal materials of P. daliensis and P. dulongensis, and their further research and development.

  • Structural Characteristics of Lycium ruthenicum Community & Soil Properties on Different Types of Desert Rangeland in the Lower Reaches of Shiyang River

    [Objectives] To study structural characteristics of Lycium ruthenicum community and soil properties on different types of desert rangeland in the lower reaches of Shiyang River, and provide theoretical basis for ecological protection and management in the lower reaches of Shiyang River. [Methods] One-way ANOVA analysis and correlation analysis were carried out with the aid of SPSS19.0 software for different types of desert rangeland soils. [Results](i) Among the communities surveyed, there are 31 species belonging to 9 families and 28 genera, including 12 shrub plants, 8 perennial herbs and 11 annual herbs. There are more single genus, occupying 50% of total genus. L. ruthenicum occupies an important position in the community with relatively higher importance value. It plays a dominated role in the community.(ii) In the soils with depths of 0-10 cm, 10-20 cm, 20-40 cm and 40-60 cm in different types of desert rangeland, the organic carbon contents in the saline alkali soil were the highest, with the values of 1.08%, 0.98%, 0.89% and 0.972%, respectively. Compared with that in sandy land, soil organic carbon in saline alkali land(0-10 cm) had the significant difference(P<0.05), available soil potassium in saline alkali land(except the depth of 20-40 cm) and the total phosphorus content in saline alkali land(0-10 cm) had the significant difference(P<0.05), the total soil nitrogen, alkali-hydrolyzable nitrogen, available phosphorus and the total potassium content had no significant difference(P>0.05). Soil organic carbon, available soil phosphorus in saline alkali land, total soil potassium on sand dune and total soil phosphorus in saline alkali land and gravel in different soil layers of the same type of desert rangeland had distinct aggregation effect.(iii) The soil microbial biomass in different types of desert rangeland displayed from high to low as saline alkali land > sandy land > fixed and semi fixed sandy land > gravel. The discrepancy of soil microbial biomass varied in different types of desert rangeland. The four soil enzyme activities in salinization was the highest. Soil invertase activity in saline alkali land in different soil layers were the highest. For different soil layers in the same type of desert rangeland, soil microbial biomass and invertase decreased gradually with the deepening of soil layer.(iv) There was a significant positive correlation between soil organic matter and microbial biomass carbon, microbial biomass nitrogen, phosphatase and sucrase(P<0.01), and a significant positive correlation with urease and catalase(P<0.05). Phosphorus content was significant positively correlated with urease, phosphatase and invertase(P<0.01), and positively correlated with microbial biomass carbon(P<0.05). Soil available potassium content was significant positively correlated with microbial and phosphatase(P<0.01). [Conclusions] The species of L. ruthenicum community and soil properties on different types of desert rangeland in the lower reaches of Shiyang River clearly shows that most species belong to a few families, and a few species belong to most families, and many species are single genus or single family. Soil organic carbon, soil available phosphorus, total phosphorus, soil microbial biomass and enzyme activity in salinization land had distinct surface aggregation effects, so salinization land provides excellent growth environment for the growth of L. ruthenicum, which is of great significance for the protection of L. ruthenicum population.

  • Screening and Analysis of Germplasm Resources of Wild Coix lacryma-jobi L. with High Content of Coixin

    [Objectives] To screen and analyze germplasm resources of wild Coix lacryma-jobi L. with high content of coixin. [Methods] From the 150 kinds of collected wild C. lacryma-jobi L. germplasm resources, 24 kinds of excellent resources were screened and detected. Morphological statistics were carried out, and the content of coixin in the four organ of roots, stems, leaves and seeds were measured, and the approved cultivar white husk Liugu of C. lacryma-jobi L. was taking as the control. [Results] Among the organs of the plant of C. lacryma-jobi L., the roots had the highest content of coixin, so the roots of C. lacryma-jobi L. can be used as the medicinal parts of C. lacryma-jobi L. [Conclusions] The wild C. lacryma-jobi L.Y229, Y164, Y166, Y83, and Y84 stains had excellent yield, and their root coixin content was significantly higher than that of the control variety, so they can be used for cultivation as excellent germplasm, so as to obtain new variety(line) of C. lacryma-jobi L.with higher content of coixin.

  • Effects of Different Storage Conditions on Physiological and Biochemical Characteristics of Polygala tenuifolia Willd. Seeds

    [Objectives] Taking Polygala tenuifolia Willd. seeds as experimental material, the changes of physiological and biochemical characteristics of seeds under different storage temperatures and seed moisture contents were studied. [Methods] The seeds of P. tenuifolia Willd. with moisture contents of 3%, 5% and 15% were stored at-10 ℃, 0 ℃, 20 ℃ and 36 ℃, respectively, and the storage time was one year. The physiological and biochemical characteristics of the seeds, such as conductivity, SOD activity, POD activity, and soluble sugar content were studied.[Results] After one-year storage, as the storage temperature and seed moisture content increased, the relative conductivity, MDA content and soluble sugar content of P. tenuifolia Willd. seeds increased, while the SOD activity, POD activity, and protein content declined. This phenomenon was most obvious in the P. tenuifolia Willd. seeds with moisture content of 15%. [Conclusions] This study will provide reference for seed storage of medicinal plants.

    [Objectives] To analyze the similarities of Boenninghausenia sessilicarpa Levl. and Boenninghausenia albiflora(Hook.) Meiss by the common and variant peak ratio dual indicator sequence method. [Methods] Four different solvents(petroleum ether, chloroform, ethyl acetate and methanol) were used to extract the chemical components of different polar regions of B. sessilicarpa and B. albiflora. UV-visible spectrophotometry(second derivative method) was used to collect the fingerprints of different solvent extracts, and common and variant peak peak ratios were analyzed for the absorption peak data. [Results] The common peak ratio and variant peak ratio data of ground parts of B. albiflora and B. sessilicarpa was S2∶S5(46.2(54.2, 62.5)), compared with the data of other groups, the common peak ratio was the largest, thus the components of ground parts of B. albiflora and B. sessilicarpa were closest and had the largest similarities; the common peak ratio and variant peak ratio data of the components extracted by chloroform from B. albiflora and B. sessilicarpa was Y2∶Y6(54.2(38.5, 46.2)), compared with the data of other groups, the variant peak ratio was the smallest, thus, the chemical components near the chloroform polarity of two kinds of medicinal materials had the largest similarities and smallest differences. [Conclusions] This method is simple and easy to operate, and the ultraviolet fingerprint data of four different polar organic solvent extracts are used for comprehensive analysis, and the results have high specificity and high accuracy. Besides, there are certain similarities and also differences between the chemical components of B. sessilicarpa and B. albiflora. It is expected to provide a new evaluation method for the variety quality of B. sessilicarpa and B. albiflora.

  • Anti-inflammatory Activity of Phenolic Acids on HAECs Induced by TNF-α

    [Objectives] To study the anti-inflammatory activity and mechanism of salvianolic acid A(Sal A), salvianolic acid C(Sal C), and rosmarinic acid(RA) on HAECs induced by TNF-α. [Methods] VE was used as a positive control and TNF-α induced human aortic endothelial cells(HAECs) were selected as the inflammation model cells. The levels of IL-6, MCP-1 were determined using ELISA. The mRNA levels of IL-6, MCP-1, ICAM-1, P-selectin and NF-κB were analyzed by quantitative RT-PCR. The protein expression of NF-κB was determined by western blot. The adhesion of U937 to HAECs was assessed by BCECF/AM labeling assay. Immunohistochemistry detection was used for ICAM-1 and P-selectin expression levels in TNF-induced HAECs, and DHE detection for the ROS production in HAECs induced by TNF-α. [Results] TNF-α-induced over-expressions of IL-6, MCP-1, NF-κB, ICAM-1 and P-selectin in HAECs were down-regulated by Sal A, Sal C and RA both in mRNA or protein levels, respectively. Meanwhile Sal A, Sal C and RA could significantly inhibit the production of ROS and U937 cells adhesion to HAECs. [Conclusions] Phenolic acids could inhibit TNF-α-induced inflammatory responses on HAECs and the mechanism might be related to inhibition of the production of ROS and blocking NF-κB signaling pathway.
