[Objective] To identify germplasm quality of cultivated FRUCTUS CANNABIS from different producing areas,and to explore germplasm evaluation method of traditional Chinese medicine.[Method] 21 copies of FRUCTUS CANNABIS collected from different cultivar of Cannabis sativa L.in China were planted in field by randomized block design.After harvesting,quality evaluation of FRUCTUS CANNABIS was carried out.According to its unique characteristics,some key indicators were adopted to comprehensively evaluate the quality of FRUCTUS CANNABIS by weight grade method.[Result] Among all collected FRUCTUS CANNABIS,there were 3 copies of cultivars with comprehensive evaluation index of more than 80 points performing excellent germplasm quality.[Conclusion] FRUCTUS CANNABIS germplasm with comprehensive evaluation index of more than 80 points was excellent,which could be used as materials for breeding fine varieties.
2011年04期 v.2 1-3+6页 [查看摘要][在线阅读][下载 239K] [Objective] To determine the content of ferulic acid in FRUCTUS TOOSENDAN and investigate the variation of ferulic acid contents in different seasons.[Method] Contents of ferulic acid in FRUCTUS TOOSENDAN were determined by HPLC,and the chromatographic conditions were as follows:ultrasphere TM-ODS(4.6 mm×150 mm,5 μm)column,acetonitrile-0.1% phosphoric acid(15∶85,V/V)as mobile phase with flow rate of 1.0 ml/min,the detection wavelength was 322 nm and column temperature was 25 ℃.[Result]Content of ferulic acid of FRUCTUS TOOSENDAN didn't change significantly in different seasons,and which was the highest in January.[Conclusion] The method is simple and fast,which could provide scientific evidence for determining the optimal collecting period for FRUCTUS TOOSENDAN.

[Objective] To explore effect of different sowing depth on seedling emergence of Scutellaria baicalensis Georgi,so as to provide a basis for ensuring a good cultivation in large scale.[Method] 7 treatment groups were employed in this study,sowing depths of which were 0,0.5,1,1.5,2.0,2.5 and 3.0 cm,respectively.Randomized block design with three replications was adopted in this study.[Results] Emergence rate of Scutellaria baicalensis Georgi in groups with sowing depth at 0(0 cm soil coverage)and 0.5 cm(0.5 cm soil coverage)was higher than that in other groups,and they were up to 69% and 60% respectively.Followed by groups with sowing depth at 1 and 1.5 cm,their emergence rates were 46% and 16%.The others were less than 5%.[Conclusion] Along with the increase in sowing depth,the emergence rate of Scutellaria baicalensis Georgi decreased,in other words,the shallower the sowing depth was,the higher emergency rate would be and its speed would be faster.Therefore,the most appropriate sowing depth for Scutellaria baicalensis Georgi in large field was 0 or 0.5 cm.
2011年04期 v.2 7-8+11页 [查看摘要][在线阅读][下载 316K] [Objective] To analyze volatile constituents in fresh leaves of Pelargonium odoratissimum(L.)L 'Her.ex Ait..[Method] Volatile constituents were isolated from fresh leaves of Pelargonium odoratissimum(L.)L 'Her.ex Ait.and identified by headspace solid-phase microextraction(HS-SPME)coupled with gas chromatography-mass spectrometry(GC-MS),contents of which were determined by area normalization method.[Result] 68 chromatographic peaks were isolated from fresh leaves of Pelargonium odoratissimum(L.)L 'Her.ex Ait.,among which 61 were identified,including D-limonene(61.18%),linalool(4.09%),germacrene(3.38%),4-carene(2.92%),eucalyptol(1.31%),α-pinene(2.19%),sabinene(2.15%),terpinolene(2.86%)and 1,7,7-trimethylbicyclo [2.2.1] hept-2-yl acetate(1.98%).[Conclusion] The study on the volatile constituents could provide a test basis for further development and utilization of Pelargonium odoratissimum(L.)L 'Her.ex Ait.resources.

[Objective] To study the effects of ethanol extract of POLLEN TYPHAE(EEPT)on immune function in C57BL/6 tumor bearing mice.[Method] Taking tumor inhibition rate as index,antitumor activity of EEPT in vivo was investigated by establishing transplanted Lewis tumor mice.The tumor inhibition rate,spleen index and thymus index were respectively determined.The proliferation of spleen lymphocyte and contents of IL-2 and TNF-α in tumor bearing mice were respectively determined by MTT assay and ELISA method.[Result] 100 mg/kg·d EEPT had a significant inhibitory action on the growth of tumor,whose tumor inhibition rate was 48.3%.The three dose groups of EEPT could raise spleen index and thymus index.Meanwhile,100 and 200 mg/kg·d EEPT could significantly increase not only proliferation of spleen lymphocyte,but also the content of IL-2 and TNF-α.[Conclusion] EEPT could promote the function of humoral immunity and cellular immunity in C57BL/6 tumor bearing mice.
2011年04期 v.2 12-14页 [查看摘要][在线阅读][下载 232K] [Objective] To study the anti-gout effects of CORTEX PHELLODENDRI and its different processed products.[Method] Effects of CORTEX PHELLODENDRI and its processed products on concentration of uric acid and creatinine in the serum and joint swelling index were investigated in rats by injection under gavage with corresponding extracts.[Result] Compared with positive control group,CORTEX PHELLODENDRI and its processed products could significantly reduce contents of uric acid and creatinine in model rats of hyperuricemia,and resist joint swelling in model rats of acute gouty arthritis.[Conclusion] CORTEX PHELLODENDRI and its processed products showed varying degrees of anti-gout effects which were little influenced by different processing methods.

[Objective] To analyze the in vitro antioxidant activities of fruit pedicels extracts from Lycium barbarum L.by different solvents,and to determine the contents of total flavonoids.[Method] By employing colorimetric method,contents of total flavonoids in different extracts were determined.The free radical scavenging activity of each extract was measured by DPPH method.[Result] Different extract fractions of fruit pedicels from Lycium barbarum L.were provided with DPPH free radical scavenging activities,of which the n-butanol extracts were highest in scavenging activities and showed no significant differences to that of Vc.In addition,total flavonoid content in n-butanol extracts was significantly higher than that inothers,which reached as high as 61.26 mg/g.[Conclusion] The antioxidant activity of fruit pedicels extracts from Lycium barbarum L.was related to the content of total flavonoids in it.
2011年04期 v.2 18-19+22页 [查看摘要][在线阅读][下载 294K] [Objective] To determine the in vitro anti-oxidation and antitumor activities of the fermentations from Lepista sordida.[Method] The secondary metabolites of L.sordida were extracted with different solvents,and the antioxidant and antitumor activities were determined by DPPH method and MTT assay,respectively.[Result] The ethyl acetone(EA)extracts of the solid fermentations exhibited the highest activity in DPPH radical inhibition assays,followed by the ethyl acetone(EA)extracts of the liquid fermentations.The ethyl acetone(EA)extracts of the liquid fermentations showed the highest antitumor activity and the IC50 value was about 50 μg/ml on the human cervix epitheloid carcinoma cell lines(Hela).The extracts of the mycelia also showed stronger antioxidative and antitumor activities.[Conclusion]The above-mentioned results provided new scientific basis for further study and exploitation of L.sordida.

[Objective] To determine the contents of trace elements in FLOS SOPHORAE of Kaili.[Method] Contents of trace elements including K,Ca,Zn,Fe,Pb and Cd in medical and edible FLOS SOPHORAE of Kaili were determined by combining microwave-assisted sample digestion with flame atomic absorption spectrometry.[Results] FLOS SOPHORAE was rich in K,Ca,Zn and Fe that were essential to human body;4 trace elements decreased according to content in the following order,Fe,Ca,K,Zn.Harmful heavy metal elements Cd and Pb were not detected in tested samples.The recovery and relative standard deviation(RSD)of these trace elements were 94.1%-110.0% and 0.54%-1.73%,respectively.[Conclusion] This research can provide scientific basis to assess medical value of local FLOS SOPHORAE.
2011年04期 v.2 23-24+27页 [查看摘要][在线阅读][下载 177K] [Objective] To determine the contents of amino acids in flower of Lilium brownii var.viridulum.[Method] Contents of 17 amino acids in flower of Lilium brownii var.viridulum were determined using OPA-HPLC method,in which o-phthalaldehyde(OPA)and 9-fluore-nylmetloy chorofomnate(FMOC)were selected as pre-column derivative reagent and VWD detector was used.[Result] The Lilium brownii var.viridulum from Shaoyang was rich in Asp,Glu,Ser,His,Gly,Thr,Arg,Ala,Tyr,Cys,Val,Met,Phe,Iso,Leu,Lys and Pro.The contents of essential amino acids for adult and for children were 49.7 and 30.6 mg/g,respectively,which were 31.20% and 19.20% of total contents of amino acids individually.Content of Glu and Arg in samples ranked the first and second,respectively,which were 28.7 and 27.5 mg/g respectively,taking up 18.03% and 17.30% of total amino acids.[Conclusion] The method does not require proprietary analyzers and is simple in operation,high in accuracy and accurate in determining result.Flower of Lilium brownii var.viridulum is rich in amino acids,which is of high developing value.

[Objective] To determine the content of total flavonoids in Dendrobium officinale Kimura et Migo from Yueqing.[Method] Using rutin as reference substance,contents of total flavonoids in Dendrobium officinale Kimura et Migo from Yueqing were measured at 510 nm by UV spectrophotometry colored with NaNO2-Al(NO3)3-NaOH.[Result] Rutin concentration was linear with absorbance in the range of 0.011 1 to 0.066 7 mg/ml(R2=0.999 2);sample solution was stable within 12 h with RSD value of 0.15%;the established method was good in repeatability with RSD value of 3.12%.[Conclusion] The method was simple,accurate and stable,which could be used to determine the content of total flavonoids in Dendrobium officinale Kimura et Migo.
2011年04期 v.2 28-29页 [查看摘要][在线阅读][下载 154K] [Objective] To compare effects of different polar parts of Galium aparine L.extracts on anti-leukemia activity.[Method] Galium aparine L.was refluxed with 95% ethanol and the obtained extracts were separated into different polar parts by Soxhlet extractor.The relationship between polar parts and biological activity was investigated through leukemia cells-K562 in vitro antitumor activity test.[Result] The content of extractum was the highest in normal butanol part,followed by dichloromethane part.The extractum and polar parts had a certain inhibitory activity on the growth of K562 cells,in which the latter was better than the former.Ether,dichloromethane and ethyl acetate polarity components had the stronger inhibitory activity,while normal butanol phase's inhibition was weak.[Conclusion] Extracts of Galium aparine L.contained some anti-cancer components that could be used in developing new anticancer drugs.

[Objective] To study the extraction method of polysaccharides from Pleurotus citrinopileatus Sing and its effects on immune function in animals.[Methods] Through the liquid culture,the polysaccharides were extracted from fermentation broth of Pleurotus citrinopileatus Sing by ethanol precipitation method,whose extraction rate was determined by combining anthrone colorimetry technology with 3,5-dinitrosalicylic acid method.Effects of different doses of polysaccharides from Pleurotus citrinopileatus Sing on immune function in mice were explored by weighing thymus and spleen and calculating thymus index and spleen index.[Results]The extraction rate of polysaccharides was 14.35%.With the dose of polysaccharides increasing,spleen weight gradually increased.The immunity in mice that were injected with polysaccharides was significantly higher than that injected with cyclophosphamide(CY),and lower than that were injected with normal saline(NS).[Conclusion] The polysaccharides from Pleurotus citrinopileatus Sing could play a role in CY attenuation and enhancing immune function in mice.
2011年04期 v.2 32-34页 [查看摘要][在线阅读][下载 197K] [Objective] To simultaneously determine contents of chlorogenic acid,rutin,quercetin and kaempferol in leaves of Eucommia ulmoides Oliver at different tree age by high performance liquid chromatography method(HPLC).[Method] Mixed solution of methanol and 0.4% phosphoric acid solution in different volume ratio were used as mobile phase in the gradient elution,whose flow rate was 1.0 ml/min.Using Shim-pack VP-ODS(4.6 mm×150 mm,5 μm)as chromatographic column,the bioactive constituents were analyzed qualitatively base on their retention times combination with ultraviolet absorption spectra.The quantization analysis was carried out by external standard method with detection wavelength at 360 nm and column temperature at 40 ℃.[Result] The linear ranges of chlorogenic acid,rutin,quercetin and kaempferol were 4.63-92.65(r=0.999 8),0.714-14.285(r=0.999 9),0.101-2.066(r=0.997 0)and 0.043-0.886 μg/ml(r=0.998 0),respectively.The recoveries of bioactive constituents were 98%-101.5%,96%-98.4%,94%-101% and 92%-96% respectively.[Conclusion] The contents of four kind's bioactive constituents in leaves of Eucommia ulmoides Oliver at different tree age were analyzed by way of variance analysis and multiple comparisons.Considering the content of chlorogenic acid,leaves of Eucommia ulmoides Oliver about three-year-old tree age was the best.

[Objective] To study the processing technology of instant powder from fresh fruits of Momordica grosvenori Swingle by enzymatic hydrolysis.[Method] Using seedless fruits of Momordica grosvenori Swingle as raw material,wet ultrafine grinding,enzymolysis,clarifying and spray drying were adopted to study the processing technology of instant powder through orthogonal test.[Result] Instant powders from fresh fruits of Momordica grosvenori Swingle whose moisture content was less than 3%,total mogroside and mogroside V contents were respectively 9.96% and 5.42% were prepared through the following processing:fresh fruit was crushed and ground by colloid mill,and hydrolyzed at 50 ℃ for 60 min with addition of 0.10 ml/L plant proteinase and 0.25 ml/L pectinase,and manufactured by spray drying under the optimum parameters of air-in temperature of 160 ℃,material-in flow of 15 ml/min and nozzle speed of 20 000 r/min.[Conclusion] The developed product which was sweet and mellow was used not only as instant drinks directly,but also as substitutions of sweetener in foods and drinks.

[Objective] To optimize the extraction technology of oil from Xinjiang Desi chickpea and study its chemical component,so as to provide scientific evidences for exploitation and development of Xinjiang Desi chickpea.[Method] Taking the extraction rate of oil as index,L9(34)orthogonal test was used to optimize extraction parameters of oil from Desi chickpea including extraction pressure,extraction temperature and extraction time by supercritical CO2 fluid extraction(SFE-CO2).Furthermore,chemical components of oil from Desi chickpea were analyzed by chemical and physical test and GC-MS method.[Result] The optimum extraction conditions of oil were as follows:extraction pressure of 30 MPa,extraction temperature of 40 ℃ and extraction time of 3 h.Under this condition,the extraction rate of oil was up to 18.19%.Through GC-MS analysis,the main chemical component of oil from Desi chickpea was linoleic acid.[Conclusion] SFE-CO2 method for extracting oil from Desi chickpea was proved to be stable and reliable with an extreme high application value.

[Objective] To optimize the ultrasonic extraction process for extracting flavonoids from Bambusa Rigida leaves.[Method] Effect of solvent kind,alcohol concentration,extraction temperature,solid to liquid ratio and extraction time on extraction efficiency of flavonoids were investigated through single factor experiments and the extraction technology was optimized by orthogonal test.[Result] The factors influencing extraction of flavonoids with ultrasonic power at 600 W were in order as follows:alcohol concentration > extraction temperature > ratio of solid-to-liquid ratio > extraction time.The optimal extraction conditions were:alcohol concentration of 85%,extraction temperature of 75 ℃,extraction time of 90 min and solid to liquid ratio of 1∶30.Under this condition,the content of flavonoids from Bambusa Rigida leaves was 11.78 mg/g.[Conclusion] The optimal technology for extracting flavonoids from Bambusa Rigida leaves had obtained in this work,which provided references for industrial production of bamboo leaf flavonoids.

[Objective] The aim was to establish a method for the content determination of catechin in Embelia parviflora Wall..[Methods] UV Spectrophotometry was adopted to determine content of catechin at the detection wavelength of 280 nm.[Results] The absorbance and concentration of catechin showed a good linear relationship when the concentration ranged from 0.007 81 mg/ml to 0.046 86 mg/ml(r=0.999 2)with average recovery of 98.72% and RSD value of 0.09%(n=5).Mean content of catechin in Embelia parviflora Wall.was 2.88%.[Conclusion] This method was simple,rapid and accurate,and could be used for the quality control of catechin in Embelia parviflora Wall.

[Objective] To optimize the microwave-assisted extraction process for red pigment from Rosa rugosa Thunb.[Method] Using Yunnan edible rose as materials,the extraction conditions of red pigment were optimized by orthogonal test based on single factor test.[Result] The optimum extraction conditions were:microwave power of 560 W,extraction duration of 35 s,solid to liquid ratio of 1∶5 and extraction solvent of 40% ethanol.Under the optimal conditions,the extraction rate of red pigment reached as high as 81.6% and yield reached 3.6%.[Conclusion] Microwave-assisted extraction technology was favorable for the extracting of red pigment from Rosa rugosa Thunb.

[Objective] To optimize the ultrasonic extraction process of total flavonoids from litchi pericarp.[Method] The total flavnoids was extracted by ultrasonic method,and the optimum extraction process was optimized with orthogonal design based on single factor test.[Result]The optimum extraction conditions of flavonoids from litchi pericarp were as follows:alcohol concentration of 60%,solid-liquid ratio of 1∶50,extracting duration of 30 min,extraction for twice.Under these conditions,the extraction rate of flavonoids reached as high as 28.18 mg/g.[Conclusion] Ultrasonic technology is convenient and effective for the extraction of total flavonoids from litchi pericarp,which lays foundation for its further development.

[Objective] The aim was to obtain high-quality long silk gutta-percha and seed oil from key fruits of Eucommia ulmoides Oliv..[Method] Key fruits of Eucommia ulmoides Oliv.were taken as the raw materials.After treated by diluted alkaline and cellulase,the key fruits of Eucommia ulmoides Oliv.were extracted by petroleum ether,and petroleum ether combing with ultrasonic wave,respectively.And they were compared with the conventional method using petroleum ether directly after breaking up the key fruits.[Result] The optimal extraction condition for key fruits of Eucommia ulmoides Oliv.was:using 0.50% NaOH to remove stratum corneum under 70 ℃ and destroying the cell wall by cellulase with pH 4 under 40 ℃.In comparison with the conventional extraction process,the oil yield and gutta-percha yield were improved by 23.00% and 58.00%,respectively.After the treatment of alkali and enzymes and combining with the ultrasonic assisted extraction,the whole key fruits of Eucommia ulmoides Oliv.could produce more oils(42.00%)and gutta-percha(76.00%)than the process directly using petroleum ether to extract two materials.And the extraction time was reduced by 90.00%.[Conclusion] The suitable extracting process of seed oil and gutta-percha from key fruit of Eucommia ulmoides Oliv.could be selected according to the actual conditions.This research provided s
