  • Exploration of the Source of Heavy Metal Elements Accumulated in a Famous-region Drug RHIZOMA CHUANXIONG in Sichuan Province

    [Objective] This study was to detect and analyze the contents of heavy metals accumulated in RHIZOMA CHUANXIONG materials sampled from Xudu Town of Dujiangyan City and in the soils from the sampling plots,so as to find out the source of heavy metal elements accumulated in RHIZOMA CHUANXIONG by comparing the content differences between the drug materials and soils tested.[Methods] Atomic absorption spectroscopy(AAS) was employed to measure the contents of Cu,Pb and Cd in RHIZOMA CHUANXIONG samples and soil.[Results] In the soil samples,the contents of Cu and Pb both accorded with China's National Second Class Standard for Soil Environmental Quality,while the content of Cd exceeded the average soil background value of China.The content of Cu in RHIZOMA CHUANXIONG samples was up to the National GAP Medicinal Plants Standard while the Pb content was slightly over the standard.The contents of Cu and Pb were both lower than that in soil;the content of Cd in RHIZOMA CHUANXIONG was higher than that in soil and seriously far beyond the GAP Medicinal Plants Standard.[Conclusion] The high content of Cd in plant samples is correlated with the higher Cd content in soil,suggesting that RHIZOMA CHUANXIONG has an enrichment effect to Cd in soil.

  • Analysis of Galium verum L. SPE-CO_2 Extract by GC-MS

    [Object] To analyze the compositions of SPE-CO2 extract from Galium verum L.by GC-MS and provide scientific references for further development of Galium verum L.. [Method] The whole plant of Galium verum L.was extracted by SPE-CO2 and the chemical compositions of the extract were analyzed by GC-MS. [Result] 45 components were identified. [Conclusion] The identified components were obtained from plant for the first time.

  • Effect of Chitooligosaccharides Plant Growth Regulators on the Growth and Secondary Metabolites of Salvia miltiorrhiza Bge.

    [Objective] To study the effect of chitooligosaccharides plant growth regulators on the growth and secondary metabolites of Salvia mitiorrhiza Bge.. [Method] Seed-dressed and foliar-sprayed with different concentrations of oligochitosan plant growth regulator, the growth of Salvia mitiorrhiza Bge. and the accumulation of its secondary metabolites tanshinone IIA were observed. [Result] The content of tanshinone IIA and the growth of Salvia mitiorrhiza Bge.were significantly improved by chitooligosaccharides plant growth regulator with optimum concentration. [Conclusion] The study was of great value and significance to expose the formation and accumulation mechanism of oligochitosan plant growth regulators on active ingredient,improve the theoretical and technological system for the quality regulation of Chinese herbal medicines and the cultivation of medicinal plants, establish standardized cultivation techniques of high-quality Salvia mitiorrhiza Bge., and promote the standardized and qualified cultivation of Chinese medicinal herbs.

  • Determination of Ursolic Acid and Oleanolic Acid in the Root of Agastache rugosa by RP-HPLC

    [Objective] To establish a quantitative method for simultaneous determination of ursolic acid and oleanolic acid in the roots of Agastache rugosa by RP-HPLC, so as to provide basis for the development of Agastache rugosa. [Method] C18 chromatographic column (4.6 mm×250 mm, 5 μm), mobile phase of acetonitrile-methanol-0.3% H3PO4 (V ∶V∶V=60∶30∶10), flow rate of 0.8 ml/min, detective wavelength of 210 nm, room temperature. [Result]The good linear relationship was obtained under the above conditions. The recovery rates of the ursolic acid and oleanolic acid were 99.6% and 98.9% with RSD of 3.12% and 1.99% respectively. The average contents of ursolic acid and oleanolic acid in the roots of Agastache rugosa were 0.138 mg/g and 0.147 mg/g respectively. [Conclusion]The method is rapid, precise and reliable, which can be used to determine the contents of ursolic acid and oleanolic acid in the roots of Agastache rugosa. This case would offer theoretical evidence for developing medicinal value of Agastache rugosa.

  • Identification and Determination of Total Flavonoids from Prinsepia utilis Royle

    [Objective] To establish the methods for the identification and determination of flavonoids components in Prinsepia utilis Royle.[Methods] Ethanol reflux extraction method was adopted to extract total flavonoids from Prinsepia utilis Royle,the physiochemical properties of which were identified by color reaction and thin-layer chromatography(TCL),the content of total flavonoids was determined by Al(NO3)-NaNO2 complexometry.[Results] The maximum absorption wavelength of sample solution was 510 nm and the solution was stable within 15 min.There was a good linear relationship between the concentration of rutin control solution(x) and absorbance value(y) with the linear regression equation of y=-2.571×10-3+13.163x,r=0.999 8;the average recovery rate was in the range of 98.00%-104.00%,101.16%was taken as average with RSD=2.15.It is detected that the flavonoids content in Prinsepia utilis Royle was 7.20%-7.30% with an average of 7.25%±0.05%,RSD=0.69%;the flavonoids content in oil meal of Prinsepia utilis Royle was 0.37%-0.38% with an average of 0.38%±0.01%,RSD=1.54%;The yield rate of flavonoids was 5.17%-5.25% with an average of 5.20%±0.04%,RSD=0.80%.[Conclusion] The method was simple,rapid,accurate and reliable,which could be applied to the determination of flavonoids in Prinsepia utilis Royle.

  • Components Analysis of HHT Produced by Endophytic Fungi Strain gyzy-20 and Optimization of Fermentation Conditions

    [Objective] To explore the application value of endophytic fungi isolated from Cephalotaxus oliveri Mast comprehensively.[Method] The fermentation products of strain gyzy-20 were extracted and analyzed by HPLC,and the producing process was optimized by orthogonal test.[Result] The metabolites of strain gyzy-20 contained homoharringtonine(HHT) whose content was affected by extraction duration largely.The optimized producing processes were as follows:carbon source of glucose,ultrasonic extraction duration of 5 min after fermenting for 10 d and extraction duration of 3 h after dichloromethane of twice the volumes was added in.The content of HHT reached as high as 0.26 μg/ml in fermentation broth under the optimum fermentation and extraction methods,which increased by more than 1.5 times.[Conclusion] The study can provide probability for producing homoharringtonine through microbial fermentation.

  • Study on the Antioxidant Activity of Mongolian Medicine FRUCTUS CHOEROSPONDIATIS in Vitro

    [Objective] To study the antioxidant activity of extracts from FRUCTUS CHOEROSPONDIATIS and provide theoretical basis for acknowledging the mechanism on strengthening the cardiovascular system. [Method] The antioxidant activities of extracts from FRUCTUS CHOEROSPONDIATIS were tested in vitro by pyrogallol autoxidation method, potassium ferricyanide reduction method and Fenton method. [Result] Both the crude extracts and ion exchange chromatographic extracts exhibited certain antioxidant capacity, among which the crude extracts had better capacity on scavenging ·OH and O2-·, the ion exchange chromatographic extracts had strong scavenging ability on ·OH while with low scavenging ability on O2-·. [Conclusion] Extracts from Monglian Medicine FRUCTUS CHOEROSPONDIATIS by different methods had different antioxidant activities,and the antioxidant activity mechanisms of different extracts varied as well.

  • Study on the Extraction Process of Total RNA from Dioscorea opposita Leaves

    [Object] To extract high quality total RNA from Dioscorea opposita leaves. [Methods] Leaves of Dioscorea opposite as tested materials, the guanidinium isothiocyanate method and modified CTAB method were introduced for extracting total RNA from Dioscorea opposita leaves, the integrity and purity of total RNA were analyzed by agarose gel electrophoresis and UV-spectrophotometer respectively. [Result] The 28S and 18 SrRNA bands of total RNA extracted by modified CTAB method were clear and visible, A260/A280 was in the range of 1.7-2.0. [Conclusion] Modified CTAB method was suitable for extracting total RNA from Dioscorea opposita Leaves.

  • Study on Extraction Technology and Antioxidant Activity of Total Flavonoids from FOLIUM MENTHAE HAPLOCALYCIS

    [Objective] The research aimed to optimize the extraction technology of total flavonoids from Folium Menthae haplocalycis and study its antioxidant activity, and to provide a basis for extraction and utilization of total flavonoids from FOLIUM MENTHAE HAPLOCALYCIS. [Method] Taking the extraction rate of total flavonoids as index, L9(34) orthogonal test with four factors and three levels was used to optimize the extraction process of total flavonoids from FOLIUM MENTHAE HAPLOCALYCIS by ultrasound-assisted extraction method. The ·OH scavenging activities of water and ethanol extracts from FOLIUM MENTHAE HAPLOCALYCIS were further studied. [Result] The results showed that the optimal extraction technology of total flavonoids from Folium Menthae haplocalycis was as follows: volume fraction of ethanol 40%, solid-liquid ratio 1∶30, ultrasonic power 400 W, extraction temperature 50 ℃, and extraction time 40 min. Under this condition, the extraction rate of total flavonoids was 6.58%. The water extracts from FOLIUM MENTHAE HAPLOCALYCIS had a good scavenging effect on ·OH, and its ·OH scavenging rate was gradually increased with the raising total flavonoids content. The maximum scavenging rate of water extracts was 44.23%. The scavenging effect of ethanol extracts on ·OH was increased slowly first, then decreased quickly and continuously with the increasing of total flavonoids content, but less than the effect of its water extracts in whole. [Conclusion] The method of extracting total flavonoids by using ethanol as the solvent assisted with ultrasonic wave was simple, and had strong maneuverability. The total flavonoids from FOLIUM MENTHAE HAPLOCALYCIS had strong scavenging effect on ·OH, and can be developed and utilized.

  • Effect of SPICA PRUNELLAE Decoction on the Contraction of Frog Heart in Vitro

    [Objective] To study the effects of SPICA PRUNELLAE water extract on the systolic function of frog heart in vitro.[Method] The effects of different concentrations of SPICA PRUNELLAE extract on the isolated frog heart were observed by perfusion method through BL-420F biological experimental system.[Result] The myocardial tension and the heart rate of the isolated frog heart can be enhanced by lower concentration of SPICA PRUNELLAE decoction(0.000 01-0.000 08 g/ml)(P<0.01),while negatively affected by high concentration of SPICA PRUNELLAE decoction(0.01-0.08 g/ml);cardiac arrest even took place when the concentration of SPICA PRUNELLAE decoction up to 0.1 g/ml(P<0.01).[Conclusion] It was concluded that SPICA PRUNELLAE has bidirectional adjust effect on the heart.

  • Determination of Linarin in RADIX et RHIZOMA JATAMANSI VALERIANAE by RP-HPLC

    [Objective] To determine the content of linarin in RADIX et RHIZOMA JATAMANSI VALERIANAE by RP-HPLC method.[Method] The HPLC conditions were as follows:Zorbax SB C18 column as separation column(150 mm×4.6 mm,5 μm),mobile phase of methanol-0.1% acetic acid(V:V=70:30),flow rate of 1.0 ml/min,column temperature of 30 ℃ and detective wavelength of 326 nm.[Result] The linear relationship between linarin concentration and peak area was good in the range of 0.010-0.080 μg,(r=0.999 7).The average recovery rate of linarin was 98.2% with RSD value of 1.78%(n=5).[Conclusion] The RP-HPLC method established in this study is favorable for the determination of linarin in Valeriana wallichii DC.,which would provide reference for the quality control of Valeriana wallichii DC..

  • Study on Anti-fatigue Effects of Opuntia Milpa Alta Extracts in Rats

    [Objective] To discuss the anti-fatigue effects of Opuntia Milpa alta(OMA) extracts in rats.[Method] The rats were subdivided into two groups randomly,and all the rats in each group were given Opuntia Milpa alta extracts via gastrogavage as the ratio of 400 milligrams per kilogram of the mouse weight everyday,using the rats infused normal saline as control.Parameters of anti-fatigue were determined 21 d later.[Result] The Opuntia Milpa alta extracts could delay rats' swimming duration(P<0.01)as the mice were exhausted.It could also decrease the depletion of rats' liver and muscle glycogen(P<0.05),and descend the content of blood lactic acid and serum urea nitrogen(P<0.05 or P<0.01)in rats after exercise.[Conclusion] Opuntia Milpa alta extracts are endowed with distinct efficacies on anti-fatigue of rats.

  • Comparative Analysis of Nutritional Compositions of Eight Kinds of Wild Vegetables from Guizhou

    [Objective] To comparatively analyze the major nutritional components of 8 wild vegetables from Guizhou Province like Cryptotaenia japonica Hassk., Geum japonicum Thunb., Mentha spicata Linn., Chenopodium album L., Aster sublatus Michx., Nasturtium officinale R.Br., Gnaphalium affine D. Don and Oenanthe javanica (Bl.) DC., so as to provide reference for the development and utilization of them. [Method] By employing conventional methods, the contents of basic nutrient components and mineral elements in 8 wild vegetables were determined. [Result] Contents of nutritional components in 8 kinds of wild vegetables varied with moisture content of 78.10%-93.40%, ash content of 8.74%-19.70%, fat content of 1.90%-0.90%, protein content of 0.31%-1.15%, crude fiber content of 10.20%-32.20%, polysaccharide content of 0.007%-0.054% and Vc content of 5.2-35.1 mg/kg. The content of K, Mg and Ca in Chenopodium album L. were the highest, being 8.6%, 2.20% and 3.23% respectively. The content of Na in Nasturtium officinale R.Br. was 6.15%, and that of Mn in Gnaphalium affine D. Don reached a peak (583.3 mg/kg). In addition, the content of Fe and Zn in Oenanthe javanica (Bl.) DC. were 0.24% and 131.8 mg/kg respectively, being the highest. [Conclusion] These 8 wild vegetables from Guizhou Province are rich in protein, fat, fiber, polysaccharide, vitamins and mineral elements, which are endowed with high exploring value.

  • Separation and Preparation of Gentiopicroside and Sweroside from the Extracts of Swertia franchetiana H.Smith by HSCCC

    [Objective] To separate and prepare gentiopicroside and sweroside from the extracts of Swertia franchetiana H. Smith by high speed counter-current chromatography (HSCCC). [Method] Chloroform∶ methanol∶ water=4∶4∶2 (V/V/V) as solvent system, the upper one was stationary phase while the lower one was mobile phase; ultrasonic degassed for 20 min; the rotating velocity of 850 r/min; mobile phase flow rate of 2 ml/min; the detective wavelength of 254 nm; the purity of product was identified by polarimeter. [Result] 14 mg gentiopricroside and 27 mg swersoside were obtained from 100 mg crude extracts of Swertia franchetiana H. Smith, the specific rotation [a]tD of which were -196.3° and -126.6°respectively, which suggested the products were of high purity. [Conclusion] The study would lay a feasible way for the large scale preparation of gentiopicroside and sweroside by HSCCC.

  • Identification of Guizhou Fructus schizandrae by UV Spectrophotometry

    [Objective] The research aimed to establish the ultraviolet spectrum group draft of Guizhou Fructus schizandrea, and confirm its belonging to which genus. [Method] Guizhou Fructus schizandrea was extracted by the ultrasonic extraction method with petroleum ether, chloroform, methanol and water as solvent, respectively. The extracts were determined by ultraviolet spectroscopy. It was the same with control medicinal materials of Fructus schisandrea Sphenantherae and Fructus schisandrea Chinensis. [Result] The results showed that the 3 kinds of medicinal herbs had good spectral characteristics so that it was easily to distinguish Fructus schisandrea Chinensis from Fructus schisandrea Sphenantherae according to the comparison of shape and position of absorption peaks. In addition, ultraviolet spectrum group draft of the extracts from Guizhou Fructus schisandrea was more similar to Fructus schisandrea Sphenantherae than to Fructus schisandrea Chinensis. [Conclusion] It could be inferred that the herb Guizhou Fructus schisandrea might belong to the genus Kadsura.

  • Determination of Oleanolic Acid in Clematis hexapetala by HPLC

    [Objective] To determine the content of oleanolic acid in the roots and stem of Clematis hexapetala, so as to provide theoretical evidence for the further development of Clematis hexapetala resources. [Method] HPLC conditions were as follows: YW-C18 column (4.6 mm×150 mm, 5 μm), mobile phase of methanol-water (V ∶ V=90∶ 10), detection wavelength of 220 nm, sampling amount of 10 μl, flow rate of 1.0 ml/min and column temperature of 25 ℃. [Result] The content of oleanolic acid in roots and stem of Clematis hexapetala were 0.175 2% and 0.096 3% respectively. [Conclusion] Simple and rapid in operations and accurate in determination, the method established in this study is favorable for the determination of oleanolic acid in Clematis hexapetala and provides reference for the assessment of the medicinal value of Clematis hexapetala stem, as well as offers scientific basis for the further development and quality control of Clematis hexapetala resources.

  • Optimization of the Extraction Technology of Total Flavonoids from Pteridum aquilinum var. latiusculum by Orthogonal Test

    [Objective] The research aimed to optimize extraction process of total flavonoids from Pteridum aquilinum var. latiusculum and provide a theoretical basis for development and utilization of total flavonoids from Pteridum aquilinum var. latiusculum. [Method] Taking the yield of total flavonoids as index, the orthogonal test L16 (44) with four factors and four levels was used to optimize the technological condition of total flavonoids including volume fraction of ethanol, boiling time, leaching time and solid-liquid ratio on the basis of the single-factor experiments. [Result] The results showed that the optimal extraction process of total flavonoids from Pteridum aquilinum var. latiusculum was as follows:volume fraction of ethanol 60%, boiling time 2.0 h, leaching time 1.0 h and solid-liquid ratio 1∶30. The yield of total flavonoids was 8.80% under the optimized extraction process. The total flavonoids contents in different organs of Pteridum aquilinum var. latiusculum were in order: fat-free leaves>leaves>fat-free petioles>petioles. [Conclusion] The extracting total flavonoids method established in this study could obtain higher yield comparing with traditional methods, which was simple, easy and to save time in practice.

  • The Effect of Grifola frondosa Polysaccharide on Immune Function in Mice

    [Objective] The research aimed to discuss the effects of Grifola frondosa polysaccharide on immune function in mice. [Method] The 30 healthy Kunming mice were randomly divided into three groups of 10 each: control group, low-dose group and high-dose group. Low-dose group and high-dose group mice were respectively fed with low or high doses of Grifola frondosa polysaccharides (300 or 400 mg/kg body weight). Control group mice were fed with distilled water. Mice of each group were allowed to feed on their dose once a day for 15 successive days to study the effect of Grifola frondosa polysaccharide on immune organ weight and blood biochemical indexes.[Result] The results showed that the thymus index in low-dose group was significantly lower than that in control group (P<0.05), and spleen index in low-dose group and high-dose group was very significantly and significantly higher than that in control group (P<0.01, P<0.05), respectively. The serum total protein and the serum globulin levels in low-dose group were significantly or very significantly higher than that in control group (P<0.05, P<0.01). The serum total cholesterols level in low-dose group was significantly lower than that in control group (P<0.05). The serum IgG in low-dose group was significantly higher than that in control group (P<0.05). [Conclusion] The results indicated that the Grifola frondosa polysaccharides in low-dose group could increase the spleen weight and serum antibody content and reduce total cholesterol level, which had a significant effect on enhancing immune function and reducing blood lipids in mice.

  • Study on Medicinal Plant Resources and Their Sustainable Utilization In Yuechengling Area of Guangxi Province

    [Objective] To investigate the medicinal plant resources in Yuechengling area of Guangxi Province and study their sustainable utilization status. [Method] Through field survey, consulting references, market research and specimens identifying, the medicinal plants resources of Yuechenling area in Guangxi Province was preliminarily studied. [Result] Yuechengling area is rich in medicinal plants resources, including 201 families, 752 genus and 1424 species, among which there are 36 families, 61 genus and 106 species of pteridophyte, 7 families, 10 genus and 15 species of gymnosperm, 153 families, 673 genus and 1 285 species of angiosperm as well as 20 rare and endangered species. Based on the analysis of investigation, the medicinal plants were classified into 8 categories according to their medicinal parts and 17 categories according to their purpose; among which 10 species were characterized by large reserves and high utilization value. In terms of the current status of medicinal plant resources, some protection and development suggestions were put forward. [Conclusion] The study provided references for reasonable developing and protecting medicinal plant resources in Yuechengling area of Guangxi Province.

  • Optimization of Supercritical CO_2 Extraction Process of Anthraquinones from RADIX et RHIZOMA RHEI

    [Objective] To optimize the supercritical CO2 extraction process of anthraquinones from Rheum palmatum L. and provide references for exploring new extraction technologies of traditional Chinese medicine. [Method] On the basis of single factor test, the extraction conditions were optimized through the orthogonal test. [Result]The optimum extraction conditions were as follows: static extraction duration of 30 min, dynamic extraction duration of 15 min, absolute ethanol dosage of 2.0 ml, extraction temperature of 50 ℃, extraction pressure of 30 Mpa and CO2 flow rate of 6 ml/min. The yield of anthraquinones from Rheum palmatum L. reached as high as 1.12% under the optimal extraction conditions. [Conclusion] The optimum extraction conditions of anthraquinones from RADIX et RHIZOMA RHEI were obtained in this study, which would provide references for exploring new extraction process of traditional Chinese medicine.

  • Study on Repeated Selection of Rubus chingii Hu Superior Individuals and Their Demonstration and Extension

    [Objective] To provide scientific evidence for repeated selection of Rubus chingii Hu superior individuals and their extensive plantation. [Method] The plants characteristics, growth habit, flowering habit and fruiting habit of primary selected ten Rubus chingii Hu superior individuals were investigated, and the individuals that could not meet the standard of superior plants were eliminated. Then the superior plants at different maturation period were screened out by comparing different individuals. [Result] The selected superior individuals of Rubus chingii Hu were determined to be No.7 (late-maturation, long fruit), No.6 (early-maturation, round fruit) and No.4 (mid-maturation, round fruit). [Conclusion] The popularization and application of three superior Rubus chingii Hu individuals screened out in this study will be promising.

  • Preparation of High-Purity Flavonoids from Ginkgo biloba L.

    [Objective] To prepare highly purified flavonoids of Ginkgo biloba L., so as to explore a large-scale preparation process route for high purity flavonoids from Ginkgo biloba L.. [Method] Purified with chromatographic column with packing of ODS, effects of mobile phase ratio, concentration temperature and drying temperature on yield of flavonoids from Ginkgo biloba L. were investigated. [Result] The optimum preparation conditions were as follows: target fraction of methanol to 0.5% phosphoric acid ratio of 5∶5 and 7∶ 3, concentration temperature and drying temperature of 45 ℃. The content of flavonoids from Ginkgo biloba L. was more than 90%, and the yield was up to 78.3% under the optimal preparation process. [Conclusion] High-purity flavonoids from Ginkgo biloba L. are obtained, which provides an effective means for the preparation of highly purified Ginkgo biloba L. flavonoids.

  • Study on Solvothermal Extraction Process of Flavonoids from Zingiber officinale Rosc.

    [Objective] To establish a new extraction method of flavonoids from Zingiber officinale Rosc., so as to provide basis for defending the oxidation of flavonoids from Zingiber officinale Rosc. and new means for its industrial application. [Method] By employing the single factor test and L9 (33) orthogonal experiment, the optimum extraction process of flavonoids from Zingiber officinale Rosc. was confirmed. [Result] The optimal solvothermal extraction conditions were determined to be as follows: ethanol concentration of 70%, 0.5 mm of particle diameter, extraction duration of 2.0 h. Under the optimum extraction process, the yield of flavonoids from Zingiber officinale Rosc. was up to 92%. [Conclusion] Simple in test device, economical in solvent dosage and low in energy consumption, the solvothermal method for the extraction of flavonoids from Zingiber officinale Rosc. established in this study is of strong maneuverability.

  • Study on the Antioxidant Effect of Rutin and Its Esterification Products

    [Objective] To study the scavenging effects of Rutin and lipase Novozym 435 synthesized rutin fatty acid ester on ·OH and O2-·. [Method]The UV spectrophotometer was adopted to determine the scavenging effects on ·OH and O2-·. [Result] Rutin and its fatty acid ester had certain scavenging effects on ·OH and O2-·, the scavenging effect of rutin on ·OH and O2-·could achieve 50% with the concentrations of 1.711 mg/ml and 1.423 mg/ml respectively(EC50), While the concentrations of rutin fatty acid ester called for 0.893 3 mg/ml and 1.358 mg/ml to achieve the same effects. [Conclusion]Rutin fatty acid ester could scavenge ·OH and O2-· effectively, which was superior to mannitol under the same conditions.

  • Optimization of Cutting Propagation Conditions of Sapindus mukorossi Gaertn by Orthogonal Test

    [Objective] The study was performed to explore more optimal breeding technology by the cutting propagation experiments of Sapindus mukorossi Gaertn. [Method] Cutting propagation experiments of Sapindus mukorossi Gaertn were carried out to investigate the rooting rate, root number and root length, and calculate and analyze the root index and root effective index by using orthogonal design method with the cuttings different in source, length and treatment. [Result] It was the best rooting performance by using about 15 cm long tip of branch as cuttings, and direct cuttage with the treatment of 200 mg/L ABT (1) rooting powder solution. Under these conditions, the rooting rate, root number and average root length were the best with the values of 92.3%, 18 and 17.3 cm in this experiment, respectively. The root effective index could reach 16.72 under the above conditions. [Conclusion] There was a better survival rate and rooting rate by using the tip of branches with keeping sharp and 15 cm long as cuttings.

  • Study on the Extraction Technology of Alkali-soluble Polysaccharides from Atractylodis macrocephalae Koidz

    [Objective] This research aimed to optimize the extraction technology of alkali-soluble polysaccharides from Atractylodis macrocephalae Koidz.[Method] Using aqueous residue extracted from Atractylodis macrocephalae Koidz as raw material,the orthogonal test was used to optimize the parameters of alkali-soluble polysaccharides assisted alkali extraction including alkali concentration and extraction time and solid-liquid ratio based on the single-factor experiments.[Result] The optimal extracting conditions of alkali-soluble polysaccharides were as follows:the concentration of NaOH 0.9 mol/L,extraction time 5 h,solid-liquid ratio 1:30.Under this condition,the extracting rate of alkali-soluble polysaccharides was 8.38%.[Conclusion] The study can provide a scientific basis for development and utilization of alkali-soluble polysaccharides from Atractylodis macrocephalae Koidz.

  • Study on Kinetics and Thermodynamics of Hypericin Adsorbed by Macroporous Resin

    [Objective] To study the kinetics and thermodynamics characteristics of hypericin adsorbed by D101 macroporous resin.[Method] The pseudo-first-order kinetics model,pseudo-second-order kinetics model and Kannan equation of intra-particle diffusion were used to fit the kinetics process of hypericin adsorbed by D101 macroporous resin,and Langmuir model,Freundlich model,Temkin model and Origin soft were used to fit the thermodynamics process of hypericin.[Result] The adsorption of hypericin by D101 macroporous resin conformed to the pseudo-second-order kinetics equation,and it was demonstrated from the results of thermodynamics study that adsorption of hypericin by D101 macroporous resin conformed to Freundlich adsorption isotherm equation.The enthalpy changes △H<0 indicated that the absorption was an exothermic process,and |△H | < 20 kJ/mol revealed that the absorption process was physical adsorption.Meanwhile,the free energy changes △G<0 indicated that the absorption was a spontaneous process;the entropy changes △S>0 demonstrated that the absorption process was the process of entropy increase.[Conclusion] The study provides theoretical basis for industrial production of isolation and purification of hypericin through macroporous resin.

  • Optimization of Decoloration Process of PERICARPIUM CITRI RETICULATAE Polysaccharide

    [Objective] To optimize the decoloration conditions of PERICARPIUM CITRI RETICULATAE polysaccharide.[Method] The polysaccharide was extracted from PERICARPIUM CITRI RETICULATAE with hot water.The decoloration effects of ion exchange resins(D-201,DH-8) and macroporous resins(D-101,HPD-400,HPD-600) on tangerine peel polysaccharide were studied.And the optimum decoloration conditions of PERICARPIUM CITRI RETICULATAE polysaccharide were determined by orthogonal test.[Result] The decoloration effect of anion exchange resin was better than that of cation exchange resin and the macroporous resins.The influencing order on the decoloration rate of PERICARPIUM CITRI RETICULATAE polysaccharide was:resin dosage> rotation speed> temperature.The optimum conditions for decoloring tangerine peel polysaccharides were as follows:D-201 anion exchange resin(8:100 W/V),rotation speed of 75 r/min and 2 h treatment in 40 ℃ water bath.Under this condition,the decoloration rate and sugar retention rate were 62.3% and 37.8% respectively.[Conclusion] The study has provided scientific basis for the production of high-quality PERICARPIUM CITRI RETICULATAE polysaccharide.

  • Optimization of Extracting Pigments from FRUCTUS SCHISANDRAE CHINENSIS By Ultrasonic Assisted Method

    [Object] To optimize the extraction process of pigments from FRUCTUS SCHISANDRAE CHINENSIS by ultrasonic assisted method and provide basis for the industrialization of extracting pigments from FRUCTUS SCHISANDRAE CHINENSIS.[Method] Single factor test and orthogonal test were introduced for the optimization of extracting process.[Result] The optimum extracting process was as follows:ethanol concentration of 80%(V/V),extracting temperature of 40 ℃,extracting duration of 35 min,material-liquid ratio of 1:12(W/V).[Conclusion] The optimum extracting process of pigments from FRUCTUS SCHISANDRAE CHINENSIS has been obtained in this study,which would provide scientific basis for the industrialized extraction of it.

  • Study on Symbiotic Mechanism between Gastrodia elata Blume and Armillaria mellea in Tissue Culture System

    [Objective] To discuss the symbiotic mechanism between Gastrodia elata Blume and Armillaria mellea in tissue culture system.[Method] By culturing Gastrodia elata Blume in the medium with Armillaria mellea extracellular extracts,intracellular extracts and live Armillaria mellea in,respectively and analyzing the variation of the weight of Gastrodia elata Blume,the symbiotic mechanism between Gastrodia elata Blume and Armillaria mellea was investigated.[Result] Intracellular extracts of Armillaria mellea didn't promote the growth of Gastrodia elata Blume,and the extracellular extracts and live Armillaria mellea were beneficial to its growth,whereas the effect of live Armillaria mellea was stronger.[Conclusion]It is concluded that living Armillaria mellea could favor the growth of Gastrodia elata Blume more effectively.

    [Object] To optimize the extraction process of total flavonoids from Lonicera edulis Turcz..[Method] On the basis of single factor test,the effects of extraction duration,ethanol concentration,ethanol dosage,extraction times on the yield rate of total flavonoids from Lonicera edulis Turcz.have been investigated by orthogonal design.[Result] The optimum extraction process of total flavonoids from Lonicera edulis Turcz.was as follows:55% ethanol as extractant,extraction times of 2,1.5 h of each time,solid-liquid ratio of 1:2(W/V).[Conclusion] The method adopted in this study was of good reproducibility,which has preferable reference value for the further development of Lonicera edulis Turcz..
