  • Effects of Different Inter-cropping Density on Growth, Development, Yield and Income of Glechoma longituba and Zea mays L.

    [Objective] The research aimed to study the effects of different inter-cropping density on growth, development, yield and income of Glechoma longituba and Zea mays L. [Method] 5 treatments on Zea mays L. were designed with 3 times repeated by the randomized block design and the planting density were 1.20 × 104, 1.95 × 104, 2.70 × 104, 3.45 × 104, 4.20 × 104 plants per hectare respectively. The planting density of Glechoma longituba was 15.95 × 104 plants per hectare. [Result] When the planting density of Zea mays L. was 1.95 × 104 -2.70 × 104 plants per hectare, the growth, development of Glechoma longituba was good, its quality, yield and income were high. [Conclusion] The study can provide scientific basis for solving the problems on meeting the creeping growth and shade-loving and light-loving simultaneously, and can provide guidance for changing the traditional planting way of herb.

  • Screening of the Optimum Medium Formula for Radix gentianae

    [Objective] The research aimed to screen the optimum medium formula for Radix gentianae. [Method] The medium formulae for Radix gentianae composed of grass peat, vermiculite and soil according to a volume ratio were selected to study the water-absorption capacity of medium and its effects on seedling characteristics of Radix gentianae. [Result] The different medium formulae added with grass peat and vermiculite could improve the water-absorption capacity and was helpful to the emergence of seedling and growth of Radix gentianae. The water-absorption capacity of the media of grass peat∶ soil (3∶2) and grass peat: vermiculite: soil (3∶2∶5) were higher than that of other media, and the rates of seedling emergence of Radix gentianae on the two media were more than 80.0%. The seedling qualities on the two medium formulae were better than that on the medium formula composed of dryland soil. [Conclusion] The study can provide the theoretic guidance for solving the problems on the seedling culture of Radix gentianae and its industrial production.

  • Microwave-assisted Extraction of Flavonoids from the Fruits of Clausena dentata (Willd.) Roem.

    [Objective] This study was to optimize the parameters for the extraction of flavonoids from the fruits of Clausena dentata (Willd.) Roem. [Method] By employing both single factor and orthogonal designs, technological conditions for microwave-assisted extraction of flavonoids including solvent concentration, solid-to-liquid ratio, microwave power and microwave treatment time, were optimized. [Results] The optimum conditions of microwave-assisted extraction of flavonoids from the fruits of C. dentata (Willd.) Roem. were as follows: 60% of alcohol concentration, 1∶50 of solid-to-liquid ratio, 240 W of microwave power and 60 s of microwave extraction time. Under these conditions, flavonoids content extracted from the fruit of C. dentata (Willd.) Roem. reached 3.629%, highly better than that extracted by conventional method (3.216%). [Conclusion] The microwave-assisted extraction was superior to the conventional method in the extraction of flavonoids from the fruit of C. dentata (Willd.) Roem.

  • Ecological Suitability Evaluation and Regionalization of Swertiam mussotii Franch.

    [Objective] Suitable ecological factors of Swertiam mussotii Franch. were discussed, suitable growth regions of S. mussotii in China were analyzed, which provided a reference for standardizing planting site, as well as science and rational planning of production lay out of S. mussotii. [Method] Based on the field survey, specimens inspection and literature review, ranges of ecological factors of main producing areas of S. mussotii nationwide were determined by using Geographic Information System for Traditional Chinese Medicine (TCMGIS), and according to this, the ecological suitability of S. mussotii in China were analyzed. [Result] The comprehensive ecological factors of main producing areas for S. mussotii were determined. Analysis results showed that the suitable areas with high similarities of main producing areas of S. mussotii were mainly located in Tibet, Sichuan, Gansu and Qinghai Provinces. Ecological suitability area of S. mussotii Franch can be divided into three sub-regions, including western Sichuan plateau, mountains in eastern Tibet-Sanjiang middle, highland and valley in southern Tibet . [Conclusion] The study demonstrates that TCMGIS is a powerful tool for mapping suitable growth regions for S. mussotii Franch. and can be applied to assess bioclimatic suitability for other medicinal plants. In addition, it is a demonstration study to introduce the numerical quantitative evaluation to the regionalization study for the traditional medicinal plants.

  • Analysis of the Volatile Components in Ardisia chinensis Benth. by GC-MS

    [Objective] The research aimed to analyze the volatile components of Ardisia chinensis Benth. by GC-MS. [Method] The volatile components of essential oil in Ardisia chinensis Benth. were extracted by steam distillation and identified by GC-MS. [Result] 56 compounds have been isolated from Ardisia chinensis Benth. which representing 84.1% of the total oil. The major components were found to be alkenes, ketones, alcohols, esters and acids and so on. [Conclusion] The study can provide scientific basis for acknowledging the relationship between the chemical components of Ardisia chinensis Benth. and pharmacologic efficiency as well as reasonable exploring and utilizing in future.

  • Study on Antibacterial Effects of Elaeagnus macrophylla Thunb. Stems

    [Objective] The research aimed to study the antibacterial effects of Elaeagnus macrophylla Thunb. stems. [Method] The antimicrobial effects of ethanol extract from Elaeagnus macrophylla Thunb. stems were studied. [Result] The antibacterial experiments showed that Elaeagnus macrophylla Thunb. stems had strong antimicrobial effects on Pseudomonas aeruginosa, Staphyloccocus aureus Rosenbach, Klebsiella pneumoniae, Escherichia coli and Bacillus subtilis in vitro and the minimal inhibitory concentration of which was 3.332-6.667 mg/ml crude drugs. With the changes of pH value, the antibacterial effects of Elaeagnus macrophylla Thunb. stems were different. [Conclusion] The study can provide the scientific basis for better applying Elaeagnus macrphylla Thunb. in clinical research.

  • Effects of Barax, Ammonium molybdate and Lanthanum nitrate Trace Fertilizers on Blossom and Yields of Schisandra chinensis (Turcz.) Baill.

    [Objective] The research aimed to use barax, ammonium molybdate and lanthanum nitrate trace fertilizes to regulate the blossom of Schisandra chinensis (Turcz.) Baill. and analyze their influences on yield. [Method] The completely randomized block design of 9 treatments was introduced in this study that including low, middle and high concentration treatments of barax, ammonium molybdate, and lanthanum nitrate trace fertilizers, concentration of which were 2 000, 4 000 and 6 000 mg/L, 1 000, 3 000 and 5 000 mg/L, 100, 300 and 600 mg/L respectively, distilled water as control. [Result] The high concentration treatment of lanthanum nitrate trace fertilizer was the best in improving the total number of flowers of Schisandra chinensis (Turcz.) Baill., increasing by 53.02% compared with CK, yet it could not effectively regulate the proportion of male and female flower. The middle concentration treatment of barax could raise the number of female flowers and the yield of Schisandra chinensis (Turcz.) Baill. most effectively, increasing by 151.76% and 65.43% respectively than CK. Ammonium could increase the total number of flowers, female flower and the yield compare with CK. But the increasing quantity was not obvious as borax and lanthanum nitrate. [Conclusion] The study can provide effective technological measures for improving the female flower rate and yield of Schisandra chinensis (Turcz.) Baill.

  • Study on Antibacterial Activity of Arnebia euchroma (Royle) Johnst and Isatis indigotica Fort Extracts in Vitro

    [Objective] The research aimed to study the effects of Arnebia euchroma (Royle) Johnst and Isatis indigotica Fort extracts with different concentration on the bacteriostasis and minimal inhibitory concentrations (MIC) of 6 strains food-borne microorganism and pathogenic bacteria (Escherichia coli, Salmonella typhimurium, Staphyloccocus aureus, Listeria monocytogenes, Streptococcus faecalis, Bacillus anthra). [Method] The filter paper dispersion method was used. [Result] The results showed that Arnebia euchroma (Royle) Johnst extracts had a strong inhibition on S. faecalis, L. monocytogenes, B. anthracis and S. aureus, and their minimal inhibitory concentrations(MIC) were 0.03, 0.03, 0.08 and 0.11 mg/ml, respectively. While Arnebia euchroma (Royle) Johnst extracts had a weak inhibition on S. typhimurium and E. coli. Isatis indigotica Fort extracts had a strong inhibition on S. aureus, L. monocytogenes, S. typhimurium and B. anthracis, and their MIC were 0.08, 0.20, 0.19 and 0.56 mg/ml respectively. While Isatis indigotica Fort extracts had a weak inhibition on the S. faecalis and E. coli with MIC of 1.08 and 1.16 mg/ml respectively. [Conclusion] The study can provide reference for developing and applying Arnebia euchroma (Royle) Johnst and Isatis indigotica Fort extracts as functional natural preservative.

  • Study on Anti-tumor Activity of Ginseng Fiber Particles in Vitro and Vivo

    [Objective] The aim was to observe the particles of ginseng fiber in vitro and in vivo anti-tumor activity and its impact on immune organs. [Method] MTT was adopted to estimate the anti-tumor effects of particles of ginseng fiber on tumor strain CNE-2Z in vitro; by using tumor weight, inhibition rate to detect the anti-tumor of particles of ginseng fiber to mice transplanted S180 tumor; through observing the thymus weight, liver weight, spleen weight, white blood cell count the other indicators to estimate the effect on the immune organs. [Result] Particles of ginseng fiber have significant anti-tumor effects on tumor strain CNE-2Z in vitro and S180 tumor-bearing mice in vivo. [Conclusion] The particles of ginseng fiber have a certain anti-tumor activity in vitro and in vivo, and the inhibitory action on immune function in vivo has not been found.

  • Optimization and Purification of Expression Condition of Recombinant Tyrosol Glycosyltransferase

    [Objective]The aim was to study the preparation of salidroside basing on modern biotechnology. [Method] Basing on SDS-PAGE method, the expression condition of the recombinant gene was optimized by investigating the effects of induction temperature, concentration of IPTG and induction time on the gene expression. [Result]The optimum induction conditions were as follows: induction time of 5 h, IPTG concentration of 0.6 mmol/L and induction temperature of 37 ℃. The target protein with purity of 99% was obtained by purifying the fusion protein with Ni-chromatography column. [Conclusion]The study will provide successful experience for the purification of tyrosol glycosyltransferase.

  • Identification and LuxAB Biochemical Mark of A Certain Bacterium Contributing to Settle Continuous Cropping Obstacles of Rehmannia glutinosa

    [Objective]The research aimed at studying the characteristics and its effects of a certain bacterium which was benefit to settle continuous cropping obstacles of Rehmannia glutinosa. [Method]Combining with morphological observation, gram stain and comparison of physiological and biochemical characteristics, conservative sequence alignment of bacterial 16S rDNA was used to identify the No. 43 strain. The recombinant strain was obtained by marking with LuxAB, antibiotic selection and caprimadehyde detection in the dark room. Finally, the effects of No. 43 and No. C-43 strain on cultured Rehmannia glutinosa seedlings in the medium including dried rhizome of Rehmannia glutinosa extracts were studied. [Result]No. 43 strain was Pseudomonas sp., and the characteistics of No. 43 strain and the recombinant No. C-43 strain were similar, which revealed that No. C-43 strain was still benefit to settle continuous cropping obstacles of Rehmannia glutinosa. [Conclusion]The recombinant No. C-43 strain can be used for the following study of the field experiments.

  • Study on the Anti-oxidation of Polyphenol from the Seeds of Toona sinensis (A. Juss) Roem in Vitro

    [Objective] The research aimed to study the anti-oxidation of polyphenol from the seeds of Toona sinensis in vitro. [Method] The reducing power of total polyphenol extracts from the seeds of T. sinensis was measured by using reducing K3Fe(CN)6 method. The antioxidant activities of polyphenols from seeds of Toona sinensis (A. Juss) Roem were evaluated by ferric reducing antioxidant power assay, hydroxyl ( ·OH) free radical scavenging capacity assay, superoxide anion (O2·) free radical scavenging capacity assay,LO· and LOO·peroxidation reaction of PUFA of yolk LDL induced by Fe2+ scavenging capacity assay. [Result] The polyphenol from the seeds of T. sinensis had good reducing power and had obvious inhibition effects on · OH, O2-·, LO · and LOO ·, with certain dosage-effect correlation. The scavenging capacity of polyphenol from the seeds of T. sinensis on free radicals was weaker than ascohic acid, but its total reducing power was stronger ascorbic acid. [Conclusion] The polyphenol from seeds of T. sinensis in vitro had obvious anti-oxidation.

  • Study on the Processing Technology of Gypsophila oldhamiana and Rubus idaeus Compound Beverage

    [Objective] Using the juice of Gypsophila oldhamiana and Rubus idaeus as the raw materials, the processing technology of Gypsophila oldhamiana and Rubus idaeus compound beverage was studied. [Method] The optimum extracting conditions, the optimum formula and the addition of composite stabilizer were obtained by orthogonal test. [Result] The results showed that the optimum extracting conditions were: solid-liquid ratio as 1∶ 1 (g/ml), pH value as 7, extracting time as 5 h, extracting temperature as 90 ℃. The optimum formula was: the addition amount of Gypsophila oldhamiana juice as 50% (V/V), the addition amount of Rubus idaeus juice as 30% (V/V), the addition amount of sugar as 45 g/L, the addition amount of citric acid as 0.5 g/L, the optimum proportion of composite stabilizer was: the xanthan gum as 10.04 mg/ml, the sodium citrate as 0.12 mg/ml by using this formula. [Conclusion] After the sensory evaluation, the physical, chemical indicators and microbiological indicators of the compound beverage produced in optimum conditions are qualified. Gypsophila oldhamiana and Rubus idaeus compound beverage has good promotional value because of its good taste, unique flavor, simple technology, medicinal value and health functions.

  • Comparative Study on Different Methods of Extracting Essential Oil from R. Cinnamomi

    [Objective] A new method for the industrial extraction of essential oil from Ramulus Cinnamomi was selected. [Method] With cinnamaldehyde as standard substance, the content analysis was conducted by TLC and HPLC. The yield and purity essential oil extracted from Ramulus Cinnamomi by supercritical-CO2 extraction (SFE-CO2) method were compared with that by steam distillation. [Result] In the concentration range of 0.524 9-2.099 4 μg/ml, cinnamaldehyde content showed a linear relationship with peak area. And their linear regression equation was Y=6.033 3×106x+0.457 3×106, r=0.999 8. The yield of essential oil from Ramulus Cinnamomi by supercritical-CO2 extraction (SFE-CO2) method increased 7.33 times than that by steam distillation. And the purity of essential oil from Ramulus Cinnamomi by supercritical-CO2 extraction (SFE-CO2) method was higher. [Conclusion] Extracting essential oil from Ramulus Cinnamomi by SFE-CO2 method could enhance the extracting yield.

  • Study on Ultrasonic-Microwave Synergistic Extraction of Total Flavonoids from Morus alba L. Leaves

    [Objective] The aim was to optimize the ultrasonic-microwave synergistic extraction conditions of total flavonoids from Morus alba L. leaves by exploring the extraction process. [Method] The single factor test was designed to confirm the main factors and the range of levels, then the orthogonal test was designed to confirm the optimized conditions. [Result] The results showed that the optimum conditions were as following: the extraction power of 400 W, material-liquid ratio of 1∶ 10 (W/V), extraction time of 12 min, and the ethanol concentration of 65% (V/V), under which the extraction efficiency of total flavonoids was 3.07%. [Conclusion] The optimum ultrasonic-microwave synergistic extraction conditions of total flavonoids from Morus alba L. was established in the experiment, which is simple, quick, efficient and free of pollution.

  • Study on Genomic DNA Extraction Method for Sophora Tonkinensis Gagnep. from Genuine Medicinal Material in Guangxi

    [Objective] The aim was to develop a simple and rapid method to extract the high-quality genomic DNA of Sophora Tonkinensis Gagnep. from genuine medicinal material in Guangxi. [Method] The modified Cetyltrimethyl Ammonium Bromide (CTAB) method was used. [Result] The high-quality and unbroken DNA was obtained using this method by which the interfering substances (i.e. polysaccharides) were eliminated. The obtained DNA can be digested completely by restriction enzymes of EcoRI and MseI, and the clear polymorphism band was gained by AFLP analysis. [Conclusion] This method is suitable for the extraction of genomic DNA in Sophora Tonkinensis Gagnep. and the DNA yielded can be further used for molecular biology experiments.

  • Isolation and Purification of Icraiin from Epimedium wushanense T. S. Ying by Polymide Column Chromatography

    [Objective] The aim was to investigate the technological parameters of the refining process of Icraiin from Epimedium wushanense T. S. Ying by polyamide. [Method] Polyamide chromatogram was adopted to separate and purify. Gradient elution used 10%-40% alcohol, and HPLC was adopted to undertake assaying of the Icraiin. [Result] The content of the Icraiin by adopting the gradient elution with 30% alcohol was 17.22%. Total yield reached 95.97%. [Conclution] The method is simple to operate and has good reproducibility. It can purify and enrich the Icraiin from E. wushanense effectively.

  • Discrimination of Padax Notoginseng (Sanqi) by Fourier Transform Infrared Spectroscopy

    [Objective] To seek a simple and rapid method of discriminating the raw materials of Chinese herb. [Method] The different grades dried flowers and dried roots of Padax Notoginseng were determined by fourier transform infrared spectroscopy. [Result]The results showed that the spectra of the flower and the root exhibited obvious differences in the range of 1 800-700 cm-1, and there were obvious differences of certain absorbance ratio between them too. The value of A1242/A2928, A1080/A2928 and A1020/A2928 of root were larger than that of the flower, while the A1643/A2928, A1416/A2928 and A1372/A2928 value of root were smaller than that of the flower. The spectral features and peaks of different grade Sanqi were very similar, some absorbance ratio differences were observed among characteristic absorption peaks. For the ratios of A1640/A2928, A1240/A2928, A1080/A2928, A1022/A2928 and A928/A2928, different grades of Sanqi presented the order as follows: 20 > 40 > 60 >80 >100 heads. [Conclusion]The containing of the flower and the root was almost the same, but the quality of which was different according to the different absorbance ratio. The FTIR is proved to be a rapid, simple and non-destructive method and it will be the standard method in identifying the grade and source of Chinese herb.

  • Study on Purification of Hydroxy Safflower Yellow A From Carthamus tinctorius L. by Macroporus Resins

    [Objective]The research aimed at obtaining the optimum conditions for the isolation and purification of hydroxy safflower yellow A (HSYA) from Carthamus tinctorius L. extracts using macroporous resins and providing basis for the effective industrial production of HSYA. [Method] Resins were screened through the determination of adsorption ratio and desorption ratio of HSYA. The yield and purity of HSYA were used as indices to investigate the purification conditions including the rate of adsorption, pH and concentration of test solution ass well as the flow rate and concentration of the eluent etc. [Result]With HPD-400 as the adsorbed resin, 50% ethanol as the eluent, adsorption rate of 6 BV/h, and the eluent flow rate of 8-10 BV/ h, the purification efficiency was the best, under which the yield and the purity of HSYA were 73.8% and 35.7% respectively. [Conclusion] The purification efficiency of HSYA is promoted greatly by purifying, which will provide the theoretical evidence for effective industrial production of Carthamus tinctorius L.

  • Protection Situation and Countermeasures of National Geographical Indication Products of Genuine Medicinal Materials in Anhui Province

    The modernization of Chinese medicine demands the intellectual property protection and standardized production of Chinese medicine materials resources. The protection of geographical indications is the important content of the intellectual property protection of agricultural products. There are 19 kinds of national geographical indications products in Anhui Province. Among which, there are 4 kinds of genuine materials, including Flos chrysanthemi, Huangshan Gongju, Paeonia ostii and D.huoshanense. The protection situation of the national products of geographical indications in Anhui Province was introduced. The present protection situation and existing problems of national geographical indications products in Anhui Province were analyzed from the kinds of products, protection scope of geographical indications, the special signs of geographical indications products, national standards of quality, trademarks of geographical indications, the certification of good agricultural practice for Chinese crude drugs and other aspects, in the meanwhile the development countermeasures were proposed accordingly.
