  • Induction and Molecule Detection of Ginseng Hairy Roots

    [Objective]The research aimed to study the induction and molecule detection of Ginseng Hairy roots. [Method] The three-year-old Ginseng roots explants were infected by Agrobacterium rhizogenes A4. Ginseng hairy roots were induced in hormone free MS-culture medium, then detected in molecular level to prove that the rol B gene and rol C gene had been integrated into the Ginseng genome. [Result] The results showed that Ginseng hairy roots were selected, and the secondary culture of Ginseng hairy roots were obtained successfully in fluid medium and solid medium. The rol B gene and rol C gene had been integrated into the Ginseng genome. Comparing with the published genomic sequence of Agobacterium rhizgenses with Ri-plasmid, there was 99% amino acid identities between them. [Conclusion] The molecular mechanism of getting Ginseng hairy roots is that rol B gene and rol C gene have been integrated into the Ginseng genome.

  • Construction of Plant Expression Vectors of PMT, TR-I and H6H

    [Objective]To construct the plant expression vectors of putrescine N-methyltransferase (PMT), tropinone reductase-I(TR-I) and hyoscyamine 6-β-hydroxylase (H6H). [Method] The coding sequences of PMT, TR-I and H6H were obtained by the method of RT-PCR and ligated to the plant expression vector pCAMBIA1304+ (p1304+). [Result] The expression vectors p1304+-PMT, p1304+-TR-I, p1304+-H6H, p1304+-PMT-H6H, p1304+-TR-I -PMT and p1304+-TR-I-H6H were constructed and transferred to Agrobacterium successfully. Engineered bacteria were obtained that can meliorate Atropa belladonna L. [Conclusion] It establishes the base of improving production of scopolamine by genetic engineering technology.

  • Extraction of Essential Oil from Nepeta pratti Levl.Based on CO_2 Supercritical Extraction Technology

    [Objective]The research aimed to establish the best extraction technology of essential oil from Nepeta pratti Levl based on CO2 supercritical extract technology (CD2-SFE). [Method] The orthogonal experiment was used to optimize the extract technology. And the gas chromatography-mass spectrum(GC-MS) technology was used to analyze the main components and their contents of N. pratti Levl volatile oil, then compare them to that of volatile oils extracted by the steam distillation method and the solvent extraction method. [Result] The results showed that the main components of volatile oil extracted by CO2 supercritical extract technology were pulegone, menthone, caryophyllene, D-limonene, transcarveol, germacrene D, (2R-cis)-5-methyl-2-(1-methylethyl)-Cyclohexanone, isopulegone, limonene, 1-octen-3-ol, 2-cyclopentylcyclopen-tanone, caryophyllene oxide, dactylol, β-myrcene, and their content was respectively 45.21%, 19.90%, 1.89%, 1.80%, 1.49%, 1.07%,0 .81%, 0.78%, 0.65%, 0.51%, 0.51%, 0.50%, 0.47%, 0.40%, 0.21%. The terpenes content was the highest, up to 76.20%. Compared with the steam distillation method and the solvent extraction method, the main components from N. pratti Levl extracted by CO2 supercritical extract technology were the highest, and the kinds of extracted components were relatively more. [Conclusion] Supercritical extraction method is simple and feasible, with the characteristics of quick mass transfer speed, strong penetrability, high selectivity and extractive efficiency, low operating temperature and so on.

  • Purification of Total Flavones in Sarcandra glabra by Macroporous Adsorption Resin

    [Objective]To search the optimal macroporous adsorption resin for purifying total flavones in Sarcandra glabra.[Methods]The static absorption ratios and elution ratios of total flavones in Sarcandra glabra by 12 types of macroporous adsorption resin were compared,the HPD-600 resin was eluted with ethanol of different concentration the purity and yield of total flavones were calculated. [Results]The recovery rate and purity of flavones by HPD-600 resin were 87% and 68%. [Conclusion]The HPD-600 type macroporous adsorption resin shows better comprehensive adsorption property,which is available for the isolation and purification of total flavones in Sarcandra glabra.

  • Experimental Study on the Resistance of Fructus cannabis Germplasm

    [Objective]The aim was to provide a scientific basis for the introduction and cultivation of Fructus cannabis by identifying the resistance of the germplasms from different habitats. [Method] The drought, cold and waterlogging tolerances of 21 representative Fructus cannabis germplasms resources from different parts of China were determined by applying pot culture to control the water and temperature. The performance of flea beetle resistance of Fructus cannabis germplasms was determined by surveying and observing the germplasms cultivated in field. The drought resistance, waterlogging tolerance, cold resistance and flea beetle resistance of 21 Fructus cannabis germplasms resources were comprehensively evaluated by calculating the resistance recovery index (RI) and population density of pests with the membership function, and the synthetical resistance was evaluated by weight grade method. [Result] The comprehensive resistance score of 21 Fructus cannabis was between 49.85 and 84.1. The comprehensive resistance of Fructus cannabis originated in Hohhot (Inner Mongolia), Jingbian (Shaanxi Province) and Lu feng (Yunnan Province) ranked the first, second and third respectively, and that originated in Xifeng, Guizhou Province ranked the last. [Conclusion] The germplasm resistance of Fructus cannabis from different habitats varies, for which the introduction and cultivation of Fructus cannabis should base on the local climatic conditions.

  • Mutagenicity Research on Ficus simplicissima Lour.

    [Objective]The research aimed to study the mutagenicity of Ficus simplicissima Lour. aqueous extracts. [Method] Using micronucleus test, the formation rates of micronucleus in liver cells,testicular germ cells,bone marrow polychromatic erythrocytes cells and periphral blood lymphocytes cells of mice caused by Ficus simplicissima Lour. aqueous extracts were measured. [Result] There was no difference in the formation rates of micronucleus liver cells,testicular germ cells,bone marrow polychromatic erythrocytes cells and periphral blood lymphocytes cells at 0.05 level; No significant difference between dose groups at 0.05 level; However, the dose groups and the negative control groups were different in the formation rates at 0.01 level [Conclusion]The aqueous extracts from Ficus simplicissima Lour.had no mutagenicity under the experimental conditions.

  • The Inhabiting Effect of Polygonum cuspidatum Sieb. et Zucc. on α-Glucosidase

    [Objective]The research aimed to screen α-glucosidase inhibitor from Polygonum cuspidatum Sieb. et Zucc. . [Method]The inhabiting effect of Polygonum cuspidatum Sieb. et Zucc. extract on α-glucosidase was studied and the active ingredients were isolated. [Result]The resultsshowed that the inhibition rate of 50 mg/ml extract to α-glucosidase was 68.5%,its IC50 was about 37 mg/ml. The Polygonum cuspidatum Sieb. et Zucc. extract was noncompetitive inhibitor of α-glucosidase,and the Ki was 33 mg/L. The crude inhibitor was purified through DEAE-cellulose DE-52 and identified as polysaccharides. [Conclusion]The study provides basis for developing botanical α-glucosidase inhibitor and has an importante ffect on finding new safety medicine with high activity that can reduce postprandial plasma glucose.

  • Cluster Analysis and FingerPrint Study of Inorganic Elements in Xinjiang Glycyrrhiza uralensis Fisch.

    [Objective]The aim is to investigate the regional differences and fingerprint characteristics, and provide the basis for the quality control and for the evaluation of Glycyrrhiza uralensis Fisch. in Xinjiang. [Method] The contents of thirteen inorganic elements in 15 Xinjiang Glycyrrhiza uralensis Fisch. samples were detected by FAAS, and the contents were studied by the cluster analysis.[Result] the Xinjiang Glycyrrhiza uralensis Fisch. samples are rich in inorganic elements. The results of cluster analysis indicate that there are significant regional differences in the Glycyrrhiza uralensis Fisch. samples from southern and northern Xinjiang if asked a high similarity, but no significant differences if asked a low similarity. From the distribution curves for the contents of 12 inorganic elements, we can conclude there are no significant regional differences in 15 Xinjiang Glycyrrhiza uralensis Fisch. samples, because they have the same peak shape. [Conclusion] Xinjiang Glycyrrhiza uralensis Fisch. is provided with obvious fingerprint characteristics from the inorganic elements.

  • Optimization on Extraction Process of Total Flavonoids in Poacynum hendersonii ( Hook. f. ) Woodson

    [Objective] The research aimed to optimize the extraction process of total flavonoids in the leaves of Poacynum hendersonii (Hook. f.) Woodson from Qinghai. [Method] The influence of different ethanol volume, extracting temperature, treatment time, solid-liquid ratio, extracting time, mesh, ultrasound power, pH on ultrasonic extraction of total flavonoids in the leaves of Poacynum hendersonii (Hook. f.) Woodson were studied. The extracting technology was optimized on the basis of single factor experiments through orthogonal design. [Result] The results showed the optimal conditions for the extraction of total flavonoids in Poacynum hendersonii (Hook. f.) Woodson. leaves were as follows: ethanol concentration 65%, extraction temperature 50 ℃, treatment time 2 h, ratio of material to solvent 1∶15, extraction times 3, mesh 20, ultrasonic power 100 W and pH value 9. Under the optimized conditions, the extraction rate of total flavonoids was the highest. [Conclusion] The study can provide the base data for deeper exploration and utilization of Poacynum hendersonii (Hook. f.) Woodson. resources.

  • Study on the Volatile Components in Different Parts of Michelia yunnanensis Flower

    [Objective] The research aimed to study the distribution of volatile components in different parts of Michelia yunnanensis flowers. [Method] The volatile components in different parts of M. yunnanensis flowers were extracted by headspace solid-phase micro-extraction(HS-SPME). And the composition was analyzed by gas chromatography-mass spectrometry(GC-MS). [Result] Twenty-eight compounds were identified from receptacle extracts. Among them, β-elemene(21.07%), caryophyllene(30.78%) and germacrene-D(15.41%) were main components. Twenty-seven compounds were identified from petal extracts. And the major components were butyl acetate(62.56%), ethyl acetate(6.36%), 2, 4-toluene diisocyanate(4.67%), 2, 4-dimethyl-5-formyl-3-nitrile pyrrole(4.04%) and 2-methyl-ethyl propionate(4.13%). Nineteen compounds were identified from filaments extracts. And the major components were 2-methyl-1-propanol(28.34%), butyl acetate(20.93%), 2, 4-toluene diisocyanate(18.58%) and 2, 4-dimethyl-5-formyl-3-nitrile pyrrole(16.29%). [Conclusion] The medicinal ingredients were mainly distributed in the receptacle of M. yunnanensis flowers and the aroma components were mainly distributed in the petals and filaments.

  • Effects of Different Extraction Methods on Extraction Rate of Polysaccharide from Sonchus oleraceus L.

    [Objective] The aim was to screen method for extracting polysaccharide from Sonchus oleraceus L. [Method] Ultrasonic wave, microwave irradiation and Ultrasonic-microwave synergistic extraction method were designed to compare with water bath method. With extraction rate of crude polysaccharide as an indicator, the methods were evaluated. [Result] Four extraction methods all have significant effects on extraction rate of S. oleraceus polysaccharide(P<0.05), the extraction rate of ultrasonic-microwave synergistic method was highest as 23.16 %,followed by microwave, water bath and ultrasonic wave, with extraction rate as 22.33 %, 20.98 % and 19.54 %. [Conclusion] Ultrasonic-microwave synergistic extraction method was best, accurately weigh 5.0 g S. oleraceus dry powder, add 100 ml distilled water, after mixing, put on ultrasound-microwave instrument to extract. Ultrasonic power was 50 w, frequency was 50 kHz; microwave frequency was 2 450 MHz. Under 80 ℃,extracting for 30 min, and through filtering, residue repeat extraction for 1 time, collecting filtrate, with 200 ml as constant volume, the polysaccharide content was determined. With glucose as standards, choosing anthrone-sulphuric acid colorimetry, the linear range of glucose concentration was 10-100 mg/ml, colorimetric wavelength was 620 nm, the average recovery rate was 100.29%, RSD value was 0.22% (n=5).

  • Determination of Eugenic Acid Content in Cornus officinalis Sieb. et Zucc. By HPCE

    [Objective] The research was to develop an HPCE method for the determination of Eugenic acid Content in Cornus officinalis Sieb. et Zucc. [Method] Capillary column, uncoated capillary(50 cm×50 μm, effective length 40 cm); 1×TBE buffer solution; separated voltage, 20 kV; detection wavelength, 280 nm; temperature, 15 ℃; sampling conditions, pressure differential injection, 10 s; height: 8 cm. Before analyzing, capillary column was rinsed with 5 min with 0.1 mol/L sodium hydroxide solution, re-distilled water and buffer solution. Before sampling, 0.1 mol/L sodium hydroxide, distilled water and running buffer rinsed with 3 min. [Result] Eugenol had a good linear relationship with the peak area of integral linearity at the range of 0.08-1.00 mg/ml, the regression equation Y=19 760.03X-19.02(R2=0.990 3), the average recovery was 97.5%, RSD was 1.46%(n=6). [Conclusion] The method has many advantages, such as simple operation, accuracy and good repeatability, which is contributed to quality control of C. officinalis.

  • Inhibitory Effects of 30 Kinds of Chinese Herbal Medicine on Bacteria in Food

    [Objective] The research aimed to study the effects of 30 kinds of Chinese herbs on bacteria in food. [Method] 30 kinds of Chinese herbs and 3 common bacteria in foods were used as experiment material. The effects of water, ethanol absolute and petroleum ether extracts on bacteria were studied by filter paper method. [Result] The results showed that water extracts of Coptis chinensis Franch., Rheum officinale Baill., Magnolia officinalis Rehd.et Wils. and Syzygium aromaticum Thunb., the ethanol extracts of Coptis chinensis Franch., Magnolia officinalis Rehd.et Wils., Polygonum cuspidatum eb.et Zucc. and Syzygium aromaticum Thunb., the petroleum ether extracts of Allium sativum L., Magnolia officinalis Rehd.et Wils., Syzygium aromaticum Thunb. and Coptis chinensis Franch. had strong antimicrobial ability. [Conclusion] The study can provide the scientific basis for exploring natural food preservatives.

  • Content Measurement of Microelements in Cultivated Anoectochilus roxburghii in Wenshan of Yunnan Province

    [Objective] The research aimed to discuss the value of cultivated Anoectochilus roxburghii (Anoectochilus roxburghii (Wall) Lindl. and Anoectochilus chapaensis Gagnep)in Wenshan of Yunnan Province and provide the reference of the value of cultivated Anoectochilus roxburghii. [Method] The samples were treated by dry ash method to measure the contents of 9 microelements (Zn, Mn, Cu, Fe, Ni, Co, Cr, Cd, Pb) in cultivated Anoectochilus roxburghii by Flame Atomic Absorption Spectrometry. And the contents were compared with that in wild A. roxburghii and the contents in fresh samples was calculated. [Result] The contents of heavy metals in cultivated A. roxburghii were as follows:Fe>Mn>Zn>Cr>Cu>Co>Ni>Pb;The content of heavy metals in wild A. roxburghii was as follows:Fe>Mn>Zn>Co>Cu>Cr>Ni>Cd>Pb, and the recovery of the 9 microelements was 99.80%-103.70% with RSD of 0.66%-3.25%. Compared with that in cultivated A. roxburghii,the contents of Fe,Mn,Zn,Co and Cu in wild A. roxburghii were higher. But the content of Cr in cultivated A. roxburghii was higher than that in wild A. roxburghii. Both the cultivated and wild Anoectochilus roxburghii contained no or low contents of harmful elements. [Conclusion] Anoectochilus roxburghii is a rare wildlife resource, and artificial breeding has been successfully done. The contents of microelements in the cultivated Anoectochilus roxburghii were measured and compared with that in the wild A. roxburghii. It was thought that cultivated A. roxburghii had higher value and was worth popularizing.

  • Comprehensively Qualitative Investigation of Cultivated Dipsacus asperoides

    [Objective] This study was to reveal the effects of growth stage, age and process of refining on medicinal quality of Dipsacus asperoides. [Method] Employing high performance liquid chromatography (HPLC), the contents of saponin Ⅵ in cultivated Dipsacus asperoides of different harvest stages, different tissues and different processes of refining and the dynamic changes were determined. [Results] With saponin Ⅵ content as indicator, the optimized parameters for harvesting and processing Dipsacus asperoides materials were as follows: harvesting the three years old materials before flowering stage, drying at 105 ℃ for 30 min, then processing at 80 ℃. [Conclusion] The results provide experimental data for formulating the SOP for standardized cultivation of Dipsacus asperoides and production of medicinal industries related, and further facilitate the reasonable exploitation and sustainable use of Dipsacus asperoides resources.

  • HPLC Fingerprint of Isoflavonoid Content Distribution and Constituent in Heshui Pueraria thomsonii Root

    [Objective] The research aimed to study HPLC fingerprint of isoflavonoids content in Heshui Pueraria thomsonii Benth. root and provide theoretical basis for reasonable development and utilization of this resource. [Method] Puerarin was served as the standard substance,the contents of total isoflavonoids and puerarin,the constituent of the isoflavonoids in fibrous and in the head,middle,tail,epidermis,subcutaneous tissue of tuber root were studied by HPLC. [Result] The contents of total isoflavonoids and puerarin in epidermis of Heshui Pueraria thomsonii Benth. root were both greatly higher than that in subcutaneous tissue. The contents of total isoflavonoids and puerarin in tail tuber root were lower than that in head and middle parts. The content of total isoflavonoids in fibrous root was higher than that in the tuber root. But the content ratio of the puerarin in isoflavonoids was much lower than that in the tuber root. And there was an unknown isoflavonoid whose content was close to the puerarin. [Conclusion] The contents of total isoflavonoids and puerarin as well as the constituent of the isoflavonoids in different parts of Heshui Pueraria thomsonii Benth. root were different.

  • Study on IR Fingerprint Spectra of Passiflora edulis Leaf

    [Objective]The research aimed to study the IR fingerprint spectra of Passiflora edulis. [Method] Two new indices including common peak ratio and variant peak ratio were applied and their values were calculated by means of the sequential analysis among different kinds of Passiflora edulis, in which each sample's IR fingerprint spectra was set up and the common peak ratio and variant peak ratio sequence were arranged in order of size comparing with the other samples. [Result] The IR fingerprint spectras of Passiflora edulis from Haikou, Danzhou, Wanning and Tunchang were similar, but stiu had some difference. [Conclusion] The study can provide the scientific basis for quickly classifying the similar Passiflora edulis samples.

  • Optimization of the Extraction Technology of Total Saponins in Lonicera fulvotomentosa Hsu. Et. S. C. Cheng by Orthogonal Design

    [Objective] The paper was to optimize the extraction technology for total saponins in Lonicera fulvotomentosa Hsu. Et. S.C. Cheng. [Method] Active ingredient content of α-hederasaponin content of total saponins was determined by HPLC, taking α-hederin content as the criterion. [Result] The order of effect factors was: decocting times > water-adding amount > soaking hours > decocting hours, that was, decocting times had the most important impact on the decocting conditions, followed by water-adding amount > soaking hours > decocting hours. The optimal solution was that L. fulvotomentosa did not need soaking, plus 10 times water and decocted three times, each time for 1.0 h. Analysis of variance results showed that water-adding amount and decocting times had important significance on content of α-hederasaponin (PA=0.034<0.05, PB =0.020<0.05), while soaking hours and decocting hours had no significance on the content of α-hederasaponin(PC=0.500>0.05, PD=0.264>0.05), which was consistent with the visual analysis. [Conclusion] This optimized process is simple, stable and efficient.

  • A Survey of Anti-tumor Plant Resources in Shilin County of Yunnan Province

    Based on the investigation of more than five years in Shilin Country, the identification of over 5000 samples collected there, and the reference to relevant files, the study presented, for the first time, that there are so many anti-tumor plants in the country, including 72 families, 123 genera and 132 species (and their varieties). These plants living environment, medicinal part, chemical components, sections for medicine, medical characters and functions, pharmaco dynamics and tumor-fighting power were further discussed. In addition, some suggestions for using and protecting these anti-tumor plant resources were put forward.

  • Study on Fatty Acid Components in Capparis spinosa L. Seed Oil by GC-MS

    [Objective] The research aimed to study the fatty acid components in Capparis spinosa L. seed oil. [Method] The fatty acid components were analyzed by GC-MS. [Result] The results showed that 6 kinds of fatty acids had been indentified, and the unsaturated fatty acid was 73.10% relatively, the essential fatty acid was 56.01% relatively. [Conclusion] The study can provide the scientific basis for deep development and utilization of Capparis spinosa L.

  • Effects of Elicitor Synergism on the Physiological and Biochemical Indices of Astragalus membranaceus ( Fisch) Bge. Calli

    [Objective] With the synergistic effects of salicylic acid and fungi elicitors, its effects on the physiological and biochemical indices of cell culture materials of Astragalus membranaceus (Fisch) Bge. were studied, so as to provide theoretical basis and technical system for producing useful secondary metabolites by using medicinal plants as bioreactors. [Method] Using tissue culture materials of Astragalus membranaceus (Fisch) Bge. as test materials, the bioactivities of POD, PAL and CAT in Astragalus membranaceus (Fisch) Bge. calli were determined. [Result] The results showed that the activities of peroxydase, polyphenol oxidase and catalase in Astragalus membranaceus (Fisch) Bge. calli were higher than those of control. The contents of hydrogen peroxide and nitric oxide were obviously higher than those of control. [Conclusion] The test results indicated that the synergistic effects of salicylic acid and fungi elicitors increased the resistance of Astragalus membranaceus (Fisch) Bge. calli. And it was helpful to the growth of Astragalus membranaceus (Fisch) Bge. callus. For example, greater economic values could be obtained in the production of adding elicitors.
