  • Systematic Review and Meta-analysis of Tanshinone Capsule in the Treatment of Polycystic Ovary Syndrome

    [Objectives] To study the efficacy and safety of tanshinone capsule in the treatment of polycystic ovary syndrome(PCOS). [Methods] A randomized controlled trial or semi-randomized controlled trial of tanshinone capsule in the treatment of polycystic ovary syndrome was included. Two researchers screened the retrieved literature, extracted the data, assessed the risk of bias of the included literature, and then carried out meta-analysis. [Results] A total of 5 articles were included, involving 350 patients. The results of meta-analysis showed that compared with the control group, tanshinone capsule could reduce total cholesterol[MD=-

In this article, the effective chemical constituents and pharmacological activity of Pholidota spp. are reviewed comprehensively in order to provide reference for the further rational development of Pholidota spp. and development of new drugs.

Chemical Composition of Natural Medicine

  • Study on Chemical Constituents of Ethanol Extract from Yao Medicine Cissampelopsis spelaeicola

    [Objectives] The research aimed to study the chemical constituents of ethanol extract from Yao medicine Cissampelopsis spelaeicola. [Methods] The petroleum ether, ethyl acetate and n-butanol fractions from 75% ethanol extract of C. spelaeicola were isolated and purified by silica gel, SephadexLH-20 gel column and AB-8 macroporous resin column, etc. The structures of the compounds were analyzed and identified by physicochemical properties and spectral data(mass spectrometry, hydrogen spectrum and carbon spectrum). [Results] Twelve compounds were isolated and identified from 75% ethanol extract of C. spelaeicola. β-sitostero(I) and stigmasterol(II) were isolated from petroleum ether fraction; p-hydroxybenzoic acid(III), β-daucossterol(IV), protocatechuic acid(V), 6β-hydroxyeremophi-7(11)-en-12, 8β-olide(VI), 10β-hydroxyeremophil-7(11)-en-8, 12-olide(VII), 10β-hydroxyeremophi-7(11), 8(9)-dien-8, 12-olide(VIII), quercetin(IX), hyperin(X) and 4α-hydroxy-eudesman-11-ene(XI) were isolated from ethyl acetate fraction; quercetin-3-O-robinobioside(XII) was isolated from n-butanol fraction. Compounds I-XII were isolated from C. spelaeicola for the first time. [Conclusions] The study can lay material foundation for activity study of C. spelaeicola.

  • Comparison of Polysaccharides Content among Common Fruits

    [Objectives] Polysaccharides in common fruits were determined and compared. [Methods] Polysaccharides in fresh bananas, apples, grapes and strawberries purchased from local markets were extracted using water extraction-alcohol precipitation method and determined using 3,5-dinitrosalicylic acid(DNS) method. [Results] The content of polysaccharides in banana was relatively high. The optimal conditions for DNS colorimetry were as follows: measurement wavelength of 510 nm, color developer of 1.0 mL, color development temperature of 90 ℃, color development time of 5 min and stabilization time within 30 min. [Conclusions] The reproducibility and recovery rate of this method are ideal. It shows that the method is highly precise and is helpful for the research of fruit polysaccharides. This study provides a certain reference value for the development and application research of fruit polysaccharide resources in China.

  • Determination of Bisoprolol Fumarate by Fluorescence Quenching Method

    [Objectives] To establish a new method for indirect determination of bisoprolol fumarate based on fluorescence quenching technology. [Methods] In ammonia water and ammonium chloride buffer solution at pH=9.2, when λ_(excitation)=277 nm and λ_(emission)=596 nm, with the increase of C_(Cu~(2+)), the fluorescence signal intensity of bisoprolol fumarate weakened, and the difference between the fluorescence intensity of bisoprolol fumarate itself and the fluorescence intensity of the test solution after the quencher Cu~(2+) was added(ΔF) and C_(bisoprolol fumarate) showed a good linear relationship. [Results] In the range of 15.39-76.93 μg/mL, ΔF=146.7 C_(bisoprolol fumarate)+482.1, r=0.998 8, and the detection limit is 0.139 1 μg/mL. [Conclusions] The fluorescence quenching method has been applied to the determination of actual samples with a recovery rate of 99.9% and an RSD of 2.7%. The results are satisfactory.

  • Experimental Study on Analgesia and Hemostasis of Paris polyphylla,Scutellaria baicalensis and Their Compatibility

    [Objectives] The analgesic and hemostatic effects of Paris polyphylla, Scutellaria baicalensis, and their compatibility were studied. [Methods](i) In the experiment on the writhing of mice induced by acetic acid, 60 Kunming SPF mice were randomly selected, and were randomly divided into 6 groups according to the male-to-female ratio of 1∶1, including blank control group, P. polyphylla group, S. baicalensis group, and three P. polyphylla and S. baicalensis compatibility groups(with the ratio of 1∶2, 1∶1 and 1∶2). There were 10 mice in each group. In the experimental groups, the mice were given 1 g/mL Chinese medicine extract according to a dose of 20 mL/kg. In the control group, the mice were given 0.9% normal saline in equal volumes. Gavage was performed one time every 24 h and lasted for 14 d. 1 h after the gavage on the 14 th day, they were given 0.5% glacial acetic acid solution via intraperitoneal injection.(ii) In the hot plate experiment, 60 Kunming SPF mice were selected, and the ratio of male to female, grouping, administration and gavage were the same as those of the glacial acetic acid-induced writhing experiment. Gavage was lasted for 14 d. 1 h after the gavage on the 14 th day, the mice were placed on a hot plate apparatus at(55±0.5) ℃, and the time of licking hindfoot was measured.(iii) The hemostatic effect was explored through three experimental methods of tail hemostasis, femoral artery hemostasis and liver hemostasis. 60 Kunming SPF mice were taken in each of the three experiments, and the male-female ratio and grouping were the same as above. The powder was covered on the surface of the wound sites, and the bleeding was observed. The hemostatic time was recorded, and the hemostatic time was recorded as 3 min if it exceeded 3 min. [Results](i) In the acetic acid-induced writhing experiment, compared with the blank control group, the pain of mice in the experimental groups was inhibited. Among them, P. polyphylla group, S. baicalensis group, compatibility 1∶2 group, compatibility 1∶1 group, and compatibility 2∶1 group had significant effects(P<0.05), and the inhibition rate of writhing was 20.43%, 28.32%, 52.30%, 32.79 %, and 39.02%, respectively. The analgesic effect of the compatibility 1∶2 group was the most obvious.(ii) In the hot plate experiment, compared with the blank control group, the experimental groups had analgesic effect. Among them, P. polyphylla group, S. baicalensis group, compatibility 1∶2 group, compatibility 1∶1 group, and compatibility 2∶1 group had significant effects(P<0.05), and the increase rate of pain threshold was 29.17%, 47.83%, 61.54%, and 50.61%, and 53.83%, respectively. The analgesic effect of the compatibility 2∶1 group was the most significant.(iii) In the hemostatic experiment, there was a significant difference in hemostatic time between the compatibility 1∶2 group and the blank control group, P. polyphylla group, and S. baicalensis group(P<0.05). The hemostatic effect of the compatibility 1∶2 group was the best. [Conclusions] P. polyphylla, S. baicalensis, and its compatibility had good analgesic and hemostatic effects, and the best compatibility ratio was 1∶2.

  • Comparative Study on the Effect of Mongolian Medicine Euphorbia pekinensis and Pharbitidis Semen (Black)

    [Objectives] To explore the difference of purgative effect of Mongolian medicine Euphorbia pekinensis and Pharbitidis Semen(black) by animal experiment, taking Euphorbia pekinensis and Pharbitidis Semen(black) as the research object. [Methods] Sixty Kunming mice were randomly divided into blank group(n=20), Euphorbia pekinensis group(n=20) and Pharbitidis Semen(black) group(n=20). After fasted but provided with free acess to drinking water for 3 d, the mice were intragastrically administered with drinking water, Euphorbia pekinensis test solution and Pharbitidis Semen(black) test solution, respectively. The mice were then placed separately in a mouse cage covered with filter paper for purgation test and intestinal propulsion test. [Results] The time of the first purgation in the Euphorbia pekinensis group and the Pharbitidis Semen(black) group was longer than that in the blank group(P<0.05). The total times of purgation and defecation within 5 h in the Euphorbia pekinensis group and the Pharbitidis Semen(black) group were higher than those in the blank group(P<0.05); the purgation index and intestinal propulsion rate in the Euphorbia pekinensis group and the Pharbitidis Semen(black) group were higher than those in the blank group(P<0.05). [Conclusions] The Mongolian medicine Euphorbia pekinensis was similar to Pharbitidis Semen(black) in purgation test and intestinal propulsion test, and could play a substitute role.

  • Effects of Different siRNA Transfection Methods on Expression of ARID4B in HepG2 Cells

    [Objectives] This study aimed to optimize the siRNA transfection method for ARID4B gene and lay a foundation for further study on the effect of ARID4B low expression on the biological function of liver cancer cells HepG2. [Methods] HepG2 cells were cultured for 0, 4 and 24 h, respectively and then transfected with ARID4B targeting siRNA. The mRNA expression of ARID4B gene was detected by RT-qPCR, and the protein expression was detected by Western blot. [Results] When HepG2 cells were transfected at 0 h after seeding, the expression of ARID4B was the lowest in both mRNA and protein levels, that is, the inhibition effect was the best. [Conclusions] Transfection at different time points after seeding of HepG2 cells had different inhibition effect on the expression of ARID4B. Transfection at 0 h after seeding showed the best inhibition effect.

  • Expression and Significance of MicroRNA-21 and P-gp in Colon Cancer Cells

    [Objectives] To investigate the expression and significance of miR-21 and P-gp in colon cancer cell line HCT-15. [Methods] RT-qPCR was used to detect the miR-21 and ABCB1 mRNA expression level of P-gp coding genes in normal colon mucosal cell NCM460 and colon cancer cell HCT-15, and Western Blot was used to detect the expression of P-gp in these two cell lines. [Results] The results of RT-qPCR showed that the expression level of miR-21 and ABCB1 mRNA in colon cancer cell HCT-15 was significantly higher than that in normal colon cell NCM460, and the difference was statistically significant(P<0.05). Western Blot results showed that the expression level of P-gp in HCT-15 was significantly higher than that in NCM460, and the difference was statistically significant(P<0.05). [Conclusions] The expression level of miR-21 and ABCB1 mRNA/P-gp in colon cancer cell HCT-15 was higher than that in normal colonic mucosal cell NCM460, and the relationship between miR-21 and multidrug resistance in colon cancer might be related to P-gp.

  • Effects of Ethanol Extract of Laggera alata (D. Don) Sch. Bip. ex Oliv on Cirrhotic Rats with Ascites

    [Objectives] This study aimed to investigate the intervention effects of ethanol extract of Laggera alata(D. Don) Sch. Bip. ex Oliv on cirrhotic rats with ascites. [Methods] A total of 100 male SD rats of SPF grade were used. Rat models of cirrhosis and ascites were established. After the last administration, the amount of peritoneal fluid was measured, and the blood of each rat tested was collected for determination of serum AST, ALT and ALB levels, serum K~+ concentration and plasma Ald level. [Results] In ascitic cirrhotic rats, the livers showed obvious enlargement, the surface of the livers was uneven, and nodules were visible. Compared with the blank group, the amount of peritoneal fluid in the model group increased significantly(P<0.01); and compared with the model group, the amounts of peritoneal fluid in the high and medium-dose ethanol extract groups reduced significantly(P<0.01). Compared with the blank group, the serum ALT and AST levels in the model group increased significantly(P<0.01); and compared with the model group, the serum ALT and AST levels in the high-dose ethanol extract group decreased significantly(P<0.05). Compared with the blank group, the serum K~+ concentration in the positive group reduced(P<0.05); no significant difference was found in serum K~+ concentration between the ethanol extract administration groups and the blank group(P>0.05). Compared with the blank group, the plasma Ald levels in the high-dose ethanol extract group and the positive group reduced(P<0.05); and there was no significant difference in serum K~+ concentration between the rest groups and the blank group(P>0.05). [Conclusions] In the ethanol extract administration groups, the amounts of peritoneal fluid significantly reduced, the serum ALT and AST reduced, and the liver functions improved to a certain extent, indicating that the ethanol extract of L. alata(D. Don) Sch. Bip. ex Oliv has a better correction effect on electrolyte disturbances in cirrhotic rats with ascites.

  • Lignans from Seeds of Herpetospermum caudigerum Wall. Protect Against Alcoholic Liver Injury via TGF-β/Smads Pathway in Rats

    [Objectives] This study aimed to investigate the effects of lignans from seeds of Herpetospermum caudigerum Wall.(SHC) on expression of TGF-β/Smads in liver tissue of hepatic alcohol-injuried rats, and to explore its protective mechanism. [Methods] A total of 60 SD rats were randomly divided into six groups. The rats in all the groups except those in the normal group were given with white spirit by gavage for 8 weeks to establish alcoholic liver injury models. After rat models were established successfully, they were administered intragastrically with 400, 200 and 100 mg/kg of lignans, respectively. The rats in the normal group were administered intragastrically with 50 mg/kg of silymarin. The administration lasted for 8 weeks, once a day. The changes in the general state, liver tissue pathology and collagen deposition of the rats were observed. The expression of TGF-β, TGF-β1 receptor 1(TβR-I), TGF-β1 receptor 2(TβR-II), Smad2/p-Smad2 and Smad3/p-Smad3 in the hepatic tissue was detected. [Results] The expression levels of TGF-β1, Smad2, p-Smad2, and p-Smad3 significantly declined, and the expression levels of TβR-I, TβR-II and Smad3 did not change significantly in the liver tissue of rats in the lignans groups and the expression levels of TGF-β, Smad2, and p-Smad3 significantly declined. Meanwhile, the expression levels of TβR-I, TβR-II, p-Smad2 and Smad3 did not change significantly in the silymarin group. [Conclusions] The lignans from SHC have significant intervention effects on alcoholic livery injury. The mechanism may be related to the inhibition of hepatic stellate cell(HSC) activation, TGF-β secretion and p-Smad2, p-Smad3 expression in the signaling pathway.

  • Effects of Zingber mioga Aqueous Extract on Hepatic Anti-alcoholism in Mice

    [Objectives] To study the anti-alcoholism effects of Zingber mioga(Thunb) Rosc aqueous extract(ZME) in drunken mice. [Methods] After all animals were administered with red star Erguotou by gavage to drunk, orally given ZME [0.4 mL/(10 g·BW)] or isometric saline. Neptunus Drinking [0.5 mL/(10 g·BW)] was used as positive control to compare the anti-alcoholism function. Then the time of righting reflex and climbing test, the activity of ADH and ALDH were measured. [Results] ZME(0.4 mL/10 g) can accelerate ethanol metabolism, also reduce the sober time and extend the time of climbing significantly. Meanwhile, the activity of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) were obviously increased in mouse liver. The lower ethanol and acetaldehyde concentrations in the assay systems after enzyme reactions mean higher ADH and ALDH activities. [Conclusions]ZME can prevent drunkenness by enhancing the activities of ADH and ALDH in liver.

  • Study on the Protective Effect and Mechanism of Tibetan Medicine Ershiwuwei Songshi Pill on Acute Alcoholic Liver Injury in Rats

    [Objectives] To investigate the protective effect and mechanism of Ershiwuwei Songshi Pill on acute alcoholic liver injury in rats. [Methods] Sixty male SD rats were randomly divided into normal control group, model group, polyene phosphatidylcholine(82.08 mg/kg) positive control group and Ershiwuwei Songshi Pill high(180 mg/kg), medium(90 mg/kg) and low(45 mg/kg) dose groups. The rats in each group were given corresponding drugs by intragastric administration for 3 d and fed normally. From the 4 th d, all groups except the normal control group were fed with 15 mL/kg liquor(56% vol) for 7 d after 2 h of administration. After the last modeling, the levels of AST, ALT, TC, TG, SOD, GSH in serum and IL-1β, IL-6, TNF-α in liver tissue were measured. The protein expressions of Bax, Bcl-2 and Caspase3 were determined by Western blotting. HE staining was used to observe the pathological changes of liver tissue under light microscope. [Results] Compared with the model group, the body weight and the levels of SOD and GSH in the treatment group were significantly higher than those in the model group; liver index, serum ALT, AST, TC, TG, IL-1β, IL-6 and TNF-α levels decreased significantly(P<0.01 or P<0.05); HE staining showed that there was a small amount of collagen proliferation and lymphocyte infiltration in connective tissue around the hepatic portal area in the model group, multiple inflammatory foci could be seen in the lobules, and round fat vacuoles could be seen in the cytoplasm; in each administration group, the pathological condition of the liver was mild. [Conclusions] Ershiwuwei Songshi Pill had a certain protective effect on acute alcoholic liver injury in rats, and its mechanism might be related to reducing enzyme, promoting lipid metabolism and anti-inflammation.

  • Optimization of Extraction Process for Longan (Dimocarpus longan Lour.) Leaves Using Multi-indicator Comprehensive Evaluation Method

    [Objectives] This study aimed to optimize the extraction process of longan(Dimocarpus longan Lour.) leaves by multi-indicator comprehensive evaluation method. [Methods] The contents of gallic acid ethyl ester, astragalin, quercetin, luteolin and kaempferol in longan leaves were determined simultaneously by HPLC. Using the multi-indicator comprehensive evaluation method, the methanol concentration, solid to liquid ratio and extraction time for extraction of longan leaves were optimized by orthogonal test. [Results] The optimal extraction process of longan leaves are as follows: methanol concentration of 100%, solid to liquid ratio of 1∶5, and extraction time of 50 min. [Conclusions] The optimized process is simple and feasible, and it can be used to determine the contents of different ingredients in longan leaves.

[Results] K. coccinea was clearly distinguished from K. heteroclite in botanical characteristics, traits and microscopic morphology. [Conclusions] This study is of great significance to the identification and development of K. coccinea.

  • Optimization of Extraction of Total Flavonoids from HERBA BLUMEAE RIPARIAE Using Response Surface Methodology

    [Objectives] This study aimed to optimize the ultrasonic-assisted extraction process of total flavonoids from a Zhuang medicine HERBA BLUMEAE RIPARIAE using response surface methodology. [Methods] Using rutin as a reference substance, a standard curve was drawn at a wavelength of 292 nm. Content determination was performed through absorbance under different conditions. The effects of ultrasonic time, ethanol volume fraction and liquid-to-solid ratio on the yield of total flavonoids from HERBA BLUMEAE RIPARIAE were investigated by single-factor tests. By using Box-Behnken test, the ultrasonic-assisted extraction process of total flavonoids from HERBA BLUMEAE RIPARIAE was optimized. [Results] The optimal extraction process of total flavonoids from HERBA BLUMEAE RIPARIAE was as follows: ultrasonic time of 30 min, ethanol volume fraction of 50% and liquid-to-solid ratio of 40∶1(mL/g). Under the optimal extraction conditions, the average yield of total flavonoids from HERBA BLUMEAE RIPARIAE was 85.47 mg/g. [Conclusions] The optimized extraction process was convenient and feasible. This study will provide a reference for the extraction of total flavonoids from HERBA BLUMEAE RIPARIAE, and provide certain guidance and basis for further development and utilization of the plan resource of Blumea riparia(Bl.) DC.

  • Extraction Process of Volatile Oil of Artemisiae Scopariae Herba

    [Objectives] To obtain the optimal extraction process parameters of volatile oil of Artemisiae Scopariae Herba. [Methods] The controlled variable method was used to take the extraction amount of volatile oil as the evaluation indicator, and the effects of single factor solid-to-liquid ratio, soaking time and extraction time on the yield rate of volatile oil were investigated. [Results] When the solid-to-liquid ratio reached 1∶12, the soaking time reached 3 h, and the extraction time reached 2 h, the yield of volatile oil was the highest. [Conclusions] The conditions of this process are stable and feasible, and it is expected to provide a certain reference for extracting volatile oil from Artemisiae Scopariae Herba.

Besides, it introduced the common changes in the learning and application of the decoction, to respond to various changes in the clinical diseases.

  • miR-23a Promoting Cell Proliferation of Human Tongue Squamous Cell Carcinoma Cell through Regulating PPP2R5E

    [Objectives] To investigate the mechanism of miR-23 a in the proliferation of human tongue squamous cell carcinoma cells. [Methods] Clinical tissue samples of tongue squamous cell carcinoma were collected and Tca8113 and CAL27 were cultured. Real-time quantitative PCR was performed to detect the expressions of miR-23 a and PPP2R5E in clinical tissue samples of tongue squamous cell carcinoma. MTT assay, colony formation assay and growth curve assay were used to detect the effect of miR-23 a and PPP2R5E on the proliferation of human tongue squamous carcinoma cell. Luciferase reporter assay verified the regulatory relationship between miR-23 a and PPP2R5E. [Results] The expression of miR-23 a in tongue squamous cell carcinoma was significantly up-regulated(P<0.01). miR-23 a promoted the proliferation of tongue squamous cell carcinoma cells. Bioinformatics prediction and luciferin reporting experiments showed that PPP2R5E was a direct target gene of miR-23 a. The expression of PPP2R5E was decreased in tongue squamous cell carcinoma. PPP2R5E inhibited the proliferation of tongue squamous cell carcinoma cells. Overexpression of PPP2R5E can reverse the proliferation promoting effect of miR-23 a on tongue squamous cell carcinoma cells. [Conclusions] miR-23 a can promote the proliferation of tongue squamous cell carcinoma cells through PPP2R5E and miR-23 a plays an oncogene role in the occurrence and development of tongue squamous cell carcinoma.

  • Comparative Study on the Effectiveness of Two Nickel-titanium Instruments in Removing Gutta-percha

    [Objectives] This study is to investigate the difference in the comprehensive effects of the two types of machine-used nickel-titanium(ni-ti) instruments in removing root canal filling materials to provide guidance for clinical applications. [Methods] We selected 60 single-tube premolars and randomly divided them into a P group(ProTaper ni-ti file group) and a K group(K3 ni-ti file group). The root canal was prepared according to the uniform standard crown cutting and manual k-file stepwise regression method. The root canal was fully filled and the X-ray film was taken. X-rays were taken again after re-treatment. The removal rate of the root filler material was obtained using Auto CAD 2000 image analysis software. [Results] We found no statistical difference in the clearance and total time of the two nickel-titanium devices in removing the gum filling material. Both the ProTaper ni-ti file and the K3 ni-ti file were able to remove most of the root canal filling during root canal retreatment. [Conclusions] There was no significant difference between the two devices in clinical applications.
