  • Liposoluble Constituents from the Flowers of Calotropis Gigantea and Their Antimicrobial Activity

    [Objectives] To study the liposoluble constituents and their antimicrobial activity from the flowers of Calotropis gigantea.[Methods]The liposoluble constituents from the flowers of Calotropis gigantea were analyzed by gas chromatography-mass spectrometry(GCMS) method and their relative content was determined by area normalization method; and their antimicrobial activity against 2 pathogens was tested by agar disc diffusion method. [Results]31 compounds were separated and identified,amounting to 85. 14% of the total constituents,27 compounds were reported for the first time in the study of the liposoluble constituents from Calotropis gigantea. The results indicated that the liposoluble constituents from the flowers of Calotropis gigantea exhibited weak antimicrobial activity against Staphylococcus aureus and Canidia albicans. [Conclusions] The research results provided scientific foundation for exploitation and utilization of Calotropis gigantea.

  • Analysis of Low Melting Point Fat Constituents of Black Soybeans by HS-SPME-GC-MS

    [Objectives] To analyze the low melting point fat constituents in the black soybeans with green and yellow heart and their relative content,and compare the differences in the low melting point fat constituents between different kinds of black soybeans. [Methods] Using HS-SPME-GC-MS,the qualitative analysis was performed on the low melting point fat constituents of black soybeans; using peak area normalization method,the relative content of constituents was calculated. [Results]A total of 42 peaks were identified from the low melting point fat constituents of the black soybeans with yellow heart,and 18 kinds of chemical constituents were identified,accounting for 81.39% of total relative content of low melting point fat constituents; a total of 37 peaks were identified from the low melting point fat constituents of the black soybeans with green heart,and 15 kinds of chemical constituents were identified,accounting for 83.24% of total relative content of low melting point fat constituents. There were 9 kinds of common chemical constituents for the two kinds of black soybeans,and 5-allylguaiacol had the highest relative content,followed by hexanol. [Conclusions] There was no significant difference in the low melting point fat constituents between two kinds of black soybeans.

  • Study of Cuscuta chinensis Lam. in Nuhuang Fuzheng Oral Liquid by Thin Layer Chromatography

    [Objectives] To better control the quality of Nuhuang Fuzheng Oral Liquid and study the main component Cuscuta chinensis Lam.by Thin Layer Chromatography. [Methods] Through changing the treatment methods of the test sample solution,proportion of the developing solvent and sample application volume,taking the spot resolution,definition,and Rf value,optimal Thin Layer Chromatography conditions were screened for Cuscuta chinensis Lam. [Results] After the test sample solution passing the neutral alumina column,it was extracted two times using the ethyl acetate. Methanol was added to dissolve. Benzene-ethyl acetate-formic acid( 5∶5∶2.5) was used as developing solvent.And ammonia fumigation was carried out to develop color. In the thin layer chromatograph obtained through these conditions,Nuhuang Fuzheng Oral Liquid test sample solution showed the same stripe in the same position of the control drug chromatogram and there was no obvious tailing phenomenon and the spot was clear. [Conclusions] The thin layer chromatography identification conditions can be used as the method for quality control of Cuscuta chinensis Lam. in Nuhuang Fuzheng Oral Liquid.

  • Content Determination of Total Flavonoids in Tetracera asiatica from 10 Different Production Areas in Guangxi

    [Objectives] To determine the content of total flavonoids in Tetracera asiatica from different production areas in Guangxi and provide a basis for improving the quality standards of crude drug. [Methods] Using 50% ethanol as extractor,NaNO_2-Al(NO_3)_3-NaOH as chromogenic system and rutin as the reference substance,the content of total flavonoids from the crude drug was determined. [Results] A method for determining the content of total flavonoids was established,the content of total flavonoids was 11.35-21.35 mg·g~(-1),and the average content was 17.64 mg·g~(-1). [Conclusions] The method had high repeatability and stability,and the method was simple,rapid and sensitive and suitable for quantitative analysis.

  • Content Determination of Kaempferol,Jaceosidin and Eupatilin in the Moxa Roll and Moxa Cone from Different Producing Areas by HPLC

    [Objectives] To determine the content of main active flavonoids in the moxa roll and moxa cone from different producing areas and provide a reference for the quality control of finished moxa roll and moxa cone. [Methods]Using HPLC,with kaempferol,jaceosidin and eupatilin as reference substance,there comparative analysis was done on the content of kaempferol,jaceosidin and eupatilin in the moxa roll and moxa cone from different producing areas. [Results]11 lots of samples contained kaempferol,jaceosidin and eupatilin,but the content differed significantly. [Conclusions] There was a large difference in the content of three kinds of flavonoids in the moxa roll and moxa cone from different producing areas,and the study provided a reference for the quality evaluation and quality control of moxa roll and moxa cone.

  • Simultaneous Determination of Six Active Components of Smilacis Glabrae Rhizome by UPLC-DAD

    [Objectives] To determine the content of neoastilbin,astilbin,neoisoastilbin,isoastilbin,engeletin,and resveratrol in Smilacis glabrae rhizome. [Methods] The ultra-high-performance liquid chromatography method with diode array detection( UPLC-DAD) was developed. Analysis conditions as follows: waters Acquity UHPLC HT3 C18 chromatographic column( 2.1 × 100 mm),column temperature 30 ℃,mobile phase acetonitrile( A)-0.1% phosphoric acid( B) gradient elution,flow rate 0.2 mL/min,and detection wavelength 327 nm. [Results]Results indicated that the content of astilbin,neoastilbin,isoastilbin,neoisoastilbin,engeletin,and resveratrol in 10 samples of Smilacis glabrae rhizome was 1.313-6.1755 mg,0.013-3.699 mg,0.146-0.975 mg,0.137-0.7 mg,0.147-0.797 mg,0.077-0.112 mg,respectively. [Conclusions]This method is high efficient,convenient and accurate,and can be used as the method for quality control of Smilacis glabrae rhizome.

  • Comparative Analysis of Antioxidants in Hippophae rhamnoides L. Leaves by UPLC

    [Objectives]The research aimed to compare antioxidation abilities of different extraction phases of Hippophae rhamnoides L. leaves extract and find its active marker. [Methods] H. rhamnoides L. leaves were extracted firstly by 75% of ethanol,and then were extracted by petroleum ether,chloroform,ethyl acetate and n-butanol in turn after decompression concentration. Via DPPH and ABTS~+ radical scavenging experiments and Fe~(3+) reduction experiment,antioxidation activity of each phase was evaluated. And chemical compositions related to its antioxidation activity were determined by using UPLC for qualitative identification. [Results] Antioxidation activity sequence of each phase was ethyl acetate phase≈n-butanol phase > alcohol extract of raw material > water phase > chloroform phase > petroleum ether phase. 33 kinds of chemical compositions were isolated from alcohol extract and extracts by using UPLC. [Conclusions] UPLC and antioxidation activity test were used simultaneously to analyze alcohol extract and extracts of H. rhamnoides L. leaves,which could fast and accurately determine extraction phase with the strongest antioxidation ability. The research is significant to study active ingredients and mechanism of antioxidation from H. rhamnoides L. leaves.

  • Content Determination about the Pharmacological Constituents of Salvia miltiorrhiza Cultivated in Jizhou District

    [Objectives] To evaluate the quality of Salvia miltiorrhiza cultivated in Jizhou District. [Methods]With Salvia miltiorrhiza cultivated in Jizhou District as test material,HPLC was used to study the content of its pharmacological constituents( tanshinone II A,tanshinone I and danshinolic acid B). [Results] For Salvia miltiorrhiza cultivated in Jizhou District,tanshinone II A content was 0. 014%,tanshinone I content was 0. 4212%,and danshinolic acid B content was 3. 018%,higher than the standard specified in Pharmacopoeia( 2015).[Conclusions]The quality of Salvia miltiorrhiza met the requirements of medication.

In this paper,an overview was given on the development of surgical method of immediate dental implantation technology,application of bone augmentation technology,and soft tissue management,and a prediction was made about the development trend of immediate dental implantation technology in anterior teeth area.

Pharmacological and Toxicological Analyses

  • Rat Models of Fatty Liver due to Liver Depression and Spleen Deficiency

    [Objectives] To establish animal models of fatty liver due to liver depression and spleen deficiency that were suitable for activity discovery of traditional Chinese medicine( TCM) compounds,efficacy evaluation,new drug research and development and correspondence between prescription and syndrome of TCM. [Methods] A syndrome score scale that was suitable for evaluating the rat models was established according to the evolvement rules of etiology and pathogenesis of TCM and the modern clinical pathological mechanism. At the same time,bifendate pills and Sanqi Zhigan pills were selected as drug counterevidence for the models. Rats in model groups were given different proportions of high-fat and low-protein fodder and different concentrations of alcohol every day,and intraperitoneally injected with porcine serum twice a week. Drug groups were given with bifendate pills and Sanqi Zhigan pills( 8. 1 mg/kg,and 2. 7 g/kg) respectively for 14 consecutive days.During the experiment,general status,weight,daily fodder intake and daily water intake of the animals were observed,and TCM syndromes were scored. After the experiment,the levels of alanine aminotransferase( ALT),aspartate aminotransferase( AST),γ-glutamine transpeptidase( γ-GT),total cholesterol( TC),triglycerides( TG),low density lipoprotein-cholesterol( LDL-C),and high density lipoprotein-cholesterol( HDL-C) in serum were detected,and the pathological changes in liver tissue were observed. [Results] Compared with the control group,body weight,daily fodder and water intake of rats increased slowly in the model group,and the levels of ALT,AST,γ-GT,TC,TG and LDL-C increased significantly,while the level of HDL-C dropped. Pathological examination showed that steatosis and fat granule were observed in hepatocytes of rats in the model group. Behavioral observation found that main symptoms and minor symptoms of rats in the model group conformed to the syndrome manifestation of liver depression and spleen deficiency,which suggested that the three model groups were established successfully. Among them,three rats died in model group 1,and one rat died in model group 2. The manifestation of all the above lesions can be alleviated by drug counterevidence. [Conclusions]The animal model of fatty liver due to liver depression and spleen deficiency shows an obviously lower mortality and shorter duration and can be used for research of correlation between prescription and syndrome and its mechanism.

  • Neuroprotective Effect of Icaritin on Alzheimer's Disease of Fruit Fly Transforming Aβ42 Gene

    [Objectives] The research aimed to explore the prevention and control effects of icaritin( ICT) on fruit fly model of Alzheimer's disease( AD),and provide theoretical basis for the development and application of new clinical medicine. [Methods]AD fruit fly model was established,and ICT was used to respectively intervene normal fruit fly and the fly transforming Aβ42. The test was divided into four groups:normal control group( Norm. group,feeding normal food solvent control),ICT intervention group of normal fruit fly( Norm. + ICT group,0. 04 mg/m L of ICT intervention),model control group( Aβ42 group,feeding normal feed solvent control) and model feeding ICT group( Aβ42 + ICT group,0. 04 mg/m L of ICT intervention). HE dyeing of fruit fly brain tissue,climbing ability,lifetime were respectively detected at whole level,and ATP,SOD and MDA contents of brain tissue were measured. [Results]Compared with AD fruit fly,there was no obvious hollow in fruit fly brain tissue slice of normal control group with the same age. Climbing ability,average lifetime,ATP,SOD and MDA contents in AD fruit fly group feeding ICT in old age were significantly improved. Normal control group was compared with normal control feeding ICT group,climbing ability,ATP,SOD and MDA contents had no statistical difference,and average lifetime was improved in normal feeding ICT group. [Conclusions]ICT could improve the behavior and lifetime of AD fruit fly,increase ATP content of brain tissue,enhance the ability to scavenge free radicals and resist lipid peroxidation,and protect AD fruit fly.

  • Optimization of Extraction Process and Determination of Antioxidant Activity of Myrica ruba Leaf Pigment by Orthogonal Design

    [Objectives] To determine the optimum conditions and antioxidant activity of Myrica ruba leaf pigment,and provide reference for further development and use of Myrica ruba leaves. [Methods]Pigment of Myrica ruba leaves was extracted by ethanol extraction method. Absorbance was measured at wavelength of 300 nm. Effects of ethanol volume fraction,solid-to-liquid ratio,temperature,and extraction time were studied. L_9( 3~3) orthogonal experiment was carried out to optimize the extraction process,and DPPH scavenging ability was measured.[Results]The optimum extraction process of Myrica ruba leaf pigment was as follows: the ethanol volume fraction was 50%; the solid-to-liquid ratio was 1: 60( w/v),extraction temperature 70℃; extraction time was 60 min; after the concentration of purified Myrica ruba leaf pigment reached 0. 4455 mg/mL,the DPPH scavenging ability was basically the same as the same concentration Vc.[Conclusions] The experiment indicates that the optimized extraction process has a high yield rate,and Myrica ruba leaf pigment has high anti-oxidant activity.

[Conclusions] This study is expected to provide basis for pharmacognostical quality standard of stems and leaves of Campsis grandiflora( Thunb.) Schum.

  • Extraction Process Optimization of Total Flavonoids from Mallotus apelta Stems by Central Composite Design/Response Surface Method

    [Objectives] To optimize the extraction process of total flavonoids in stems of Mallotus apelta. [Methods]On the basis of singlefactor test,with volume fraction of ethanol,extraction time and ratio of solvent as independent variables,the content of total flavonoids as dependent variables,the completely secondary response surface regression fitting was conducted on the independent and dependent variables,and the Response Surface Method was used to optimize the optimum extraction process of total flavonoids in Mallotus apelta stems and predict the optimum process. [Results] The optimum extraction process of total flavonoids in Mallotus apelta was determined as follows: ethanol concentration of 71. 5%; extraction time of 154. 6 min; solid-liquid ratio of 1∶19. 2; total flavonoids content of 7. 060 mg/g; fitted binomial squared correlation coefficient R~2= 0. 8751.[Conclusions]Composite Design/Response Surface Method could be used in the extraction process optimization of total flavonoids in Mallotus apelta stems,the mathematical model established had high prediction accuracy,the method was simple and operability was good.

  • Improvement of TLC Identification of Matrine in Jin Weng Zhi Li Granule

    [Objectives] The aim of this study was to screen out the TLC conditions of matrine in Jin Weng Zhi Li Granule. [Methods]Referring to the method of Chinese Veterinary Pharmacopoeia,the composition,chromogenic agent and expansion conditions of the developer were optimized,and matrine was qualitatively identified by TLC. [Results] After optimizing TLC conditions of Jin Weng Zhi Li Granule,that is,the thin layer plate was pre-equilibrated in the expansion cylinder for 5 min after spotting,the separation effect of TLC of test samples was favorable. The spot color was clear,reproducibility was good,precision was high,specificity was better. [Conclusions] The method was of good feasibility,reproducibility and specificity,and could be used for qualitative detection of matrine in Jin Weng Zhi Li Granule dampnessheat and diarrhea disease.

  • Screening of TLC Identification of Forsythin in Jin Weng Zhi Li Granule

    [Objectives] To screen TLC conditions of forsythin contained in Jin Weng Zhi Li Granule. [Methods] According to the method described in Chinese Veterinary Pharmacopoeia,thin layer chromatography was used for qualitative identification of forsythin contained in Jin Weng Zhi Li Granule by improving the developing conditions. [Results]After optimizing TLC conditions of Jin Weng Zhi Li Granule,that is,the thin layer plate was pre-equilibrated in the expansion cylinder for 5 min after spotting,the separation effect of TLC of test samples was favorable. The spot color was clear,reproducibility was good,precision was high,specificity was better.
