  • A Study on the Chemical Constituents of Pilea sinofasciata C. J.Chen

    [Objectives] To study the chemical constituents of Pilea sinofasciata C. J. Chen. [Methods] Column chromatography,such as silica gel,Sephadex LH-20 and preparative HPLC were used to isolate and purify the compounds. Spectroscopic methods like MS,1H NMR and~(13)C NMR and physical constants were used to elucidate their structure. [Results]Ninteen compounds were isolated from 95% ethanol extracts of Pilea sinofasciata C. J. Chen,including α-tocopherol(1),stigmasterol(2),epihernandulcin(3),hernandulcin(4),benzoic acid(5),ethyl linolenate(6),ethyl hexadecanoate(7),α-amyrin(8),palmitic acid(9),behenic acid(10),adenosine(11),indole-3-carboxylic acid(12),protocatechuate(13),gallic acid(14),betulinic acid(15),oleanolic acid(16),potassium nitrate(17),diosmetin7-O-β-D-glucopyranoside( 18),3-O-β-D-xylopyranosyl(1→2)-β-D-glucopyranosyl-28-O-β-D-glucopyranosyl oleanolic acid(19). [Conclusions] All of the compounds were isolated from the plant for the first time.

  • Main Components of Polyphenols of Trillium tschonoskii Maxim. and Changes under Different Se Concentrations

    [Objectives] To analyze main components of polyphenols of Trillium tschonoskii Maxim. and study changes under different Se concentrations. [Methods] In this experiment,the Trillium tschonoskii Maxim. was used and spectrophotometry was applied to measure the total phenols in stems,leaves and rhizomes of Trillium tschonoskii Maxim. [Results]Total phenols in stems and leaves and rhizomes of Trillium tschonoskii Maxim. were 3. 026% and 0. 513% respectively,the content of total phenols in stems and leaves was 5. 9 times of that in rhizomes; total phenol extracting solution was analyzed through UV-Vis scanning,HPLC detection,and LC-MS; 3 main polyphenol compounds were obtained: chlorogenic acid,caffeic acid and rutin,in which chlorogenic acid and caffeic acid were firstly discovered in Trillium tschonoskii Maxim. and rutin was firstly discovered in stems and leaves of Trillium tschonoskii Maxim. HPLC method was used to measure the content of chlorogenic acid,caffeic acid,and rutin in stems and leaves of Trillium tschonoskii Maxim. under different Se concentrations. [Conclusions]The content of chlorogenic acid in stems and leaves of Trillium tschonoskii Maxim. increases with the increase of Se concentration,showing Se promotes Se accumulation and the maximum content of chlorogenic acid is 2. 58%; the content of caffeic acid in stems and leaves of Trillium tschonoskii Maxim. increases slightly with the Se concentration,showing the promotion of Se to accumulation of caffeic acid is not significant;the content of rutin in stems and leaves of Trillium tschonoskii Maxim. firstly increases then declines with the increase in Se concentration,at25 mg/kg Se concentration,rutin reached the maximum value,showing the promotion of Se to rutin accumulation is not significant.

  • GC-MS Analysis of Volatile Components Extracted from Fruit of Artemisia argyi

    [Objectives] To analyze the volatile components extracted from fruit of Artemisia argyi.[Methods] The volatile components from fruit of Artemisia argyi were extracted by steam distillation( SD) and headspaces solid-phase microextraction( HS-SPME). The components were separated and analyzed by gas chromatography-mass spectrometry( GC-MS) with the area normalization method to determine the relative percentage content of each component.[Results]33 compounds were isolated and 29 compounds were identified by SD accounting for about93. 02% of the total volatile components; and 35 compounds were isolated and 30 compounds were identified by HS-SPME accounting for about99. 01% of the total volatile components. 11 kinds of the volatile components were identified by the two methods together and the content of β-thujone was highest.[Conclusions] There were some differences between SD and HS-SPME,and the two methods were applied to the extraction of different types of volatile components,and combination of the two methods to analyze the volatile components in fruit of Artemisia argyi could help us to get more comprehensive information.

Pharmacological and Toxicological Analyses

  • Effects of Volatile Oils of Sichuan Citrisarcodactylis fructus on Proliferation and Apoptosis of MCF-7 Human Breast Cancer Cells

    [Objectives] To study the effects of volatile oils of Sichuan Citrisarcodactylis fructus on proliferation and apoptosis of MCF-7human breast cancer cells.[Methods]CCK-8 method was used to measure the cell proliferation,the flow cytometry was used to measure changes in cell cycle,Western blot was used to detect p53 and cyclin D1 activity changes,TUNEL method was used to measure percentage of apoptotic cells,inverted phase contrast microscope and transmission electron microscope were used to observe morphology of MCF-7 cells treated with different concentrations.[Results]When the concentration was 5-50 μmol/L,the cell proliferation inhibition rate increased significantly(P <0. 05),G2/M phase decreased significantly( P < 0. 05),eventually disappeared completely,G1 phase significantly increased with time and concentration( P < 0. 05),finally reached 90. 0%; the activity of cyclin D1 significantly declined( P < 0. 05),while the activity of p53 had no significant change( P > 0. 05). The apoptotic rate of MCF-7 cells significantly increased with the extension of time( P < 0. 05). At6 h,12 h and 24 h of action time,the apoptotic rate of MCF-7 cells significantly increased with the increase in volatile oil concentration(P <0. 05). Morphological observation showed that the apoptosis of MCF-7 cells was obvious.[Conclusions]The volatile oils of Citrisarcodactylis fructus have obvious inhibition of proliferation of MCF-7 cells and function of inducing apoptosis,and the effects took on the dose and time dependent. 5-50 μmol/L volatile oil of Sichuan Citrisarcodactylis fructus can inhibit the proliferation of MCF-7 cells and induce the apoptosis of MCF-7 cells through inhibiting the cyclin D1.

  • Regulation of Zishen Qinggan( Shuangyi) Prescription to Insulin Resistance of GK Rats

    [Objective] To study the effects of Zishen Qinggan( Shuangyi) Prescription on blood glucose,Hb A1c( glycosylated hemoglobin A1c),C-peptide( C-P),glucagon-like peptide( GLP-1),and α-glucosidase activity in GK( Goto-Kakizaki) rats. [Methods]Selected fasting blood glucose( FBG) > 11. 1mmol/L GK rats and randomly divided into groups: model group,Zishen Qinggan( Shuangyi) Prescription low dosage( ig,400 mg/kg/d),medium dosage( ig,800 mg/kg/d),and high dosage groups( ig,1600 mg/kg/d),and metformin group(ig,85 mg/kg/d). After continuous administration for 28 d,fasting blood glucose(FBG),oral glucose tolerance test(OGTT),Hb A1 c,C-P,GLP-1 and α-glucosidase activity were detected. [Results] Compared with the control group,FBG and Hb A1 c of the model group was significantly higher( P < 0. 01); compared with the model group,Zishen Qinggan( Shuangyi) Prescription could significantly reduce the levels of blood glucose,Hb A1 c,and the elevated blood glucose levels after 120 min of glucose loading in OGTT( P < 0. 05,P < 0. 01),and also can significantly improve the level of C-P,GLP-1( P < 0. 05,P < 0. 01),reduce Homa-IR,and inhibit α-glucosidase activity. [Conclusions]Zishen Qinggan( Shuangyi) Prescription can effectively reduce the blood glucose of GK rats,the action mechanism of which is possibly that it can improve insulin resistance,increase GLP-1 content and inhibit α-glucosidase activity.

  • Comparative Study on Tissue Distribution of Tibetan Medicines Renqing Mangjue Compatible with Zuota

    [Objectives]To explore the mechanism for the attenuate-synergistic effect of Zuota to Renqing Mangjue,a contrasted study on the tissue distribution of Tibetan medicines Renqing Mangjue compatible with Zuota was carried out. [Methods] The SD rats gastric ulcer model were made successfully,and then respectively were given Renqing Mangjue compatible with Zuota or not according to the dose of 0. 144 g/kg and normal saline by gavage administration,once a day. After 14 d of administration,various organs and tissue were isolated,including brain,heart,liver,spleen,lung,kidney,jejunum,skeletal muscle,fat,skin,submandibular gland,uterus and ovary( or testicle and epididymis).The LC-MS/MS methods were used for determining the contents of brucine and strychnine in various organs and tissue,and significant difference analysis,main target site analysis and clustering analysis were implemented. [Results]Between the Zuota group and non-Zuota group,there was extremely significant difference in the content of brucine in the liver,kidney,muscle,lung,ovary,and heart( P < 0. 01),significant difference in the jejunum( P < 0. 05),and no significant difference in the epididymis,testis,brain,skin and uterus( P > 0. 05); the content of brucine in the submandibular gland,spleen,liver and ovary increased significantly,but significantly reduced in the kidney,muscle,jejunum,lung and heart. And there was extremely significant difference in the content of strychnine in the liver,kidney,ovary,brain and heart( P < 0. 01),and no significant difference in the uterus( P > 0. 05); the strychnine content increased significantly in the liver,spleen and ovary,and significantly decreased in the kidney,muscle,testis,lung,brain and heart.[Conclusions] Compatibility of Zuota can affect the distribution of brucine and strychnine in some tissue and organs,so as to achieve attenuate-synergistic effect of Zuota to Renqing Mangjue.

  • Extraction of Herba Lycopi Polysaccharide and Study of Its Antioxidant Activity by Cellulase Method

    [Objectives] To optimize the extraction process of Herba Lycopi polysaccharide by cellulase,and study its antioxidant activity.[Methods]The single factor test and orthogonal test were conducted to study the effect of p H,enzyme dosage,enzymolysis temperature and extraction time on the extraction of Herba Lycopi polysaccharide. And the test was done about its scavenging effect on DPPH radicals and superoxide anions,and the antioxidant activity was evaluated. [Results] The optimum extraction process by cellulase method was as follows: p H value of 7; enzyme dosage of 4 ml; enzymolysis temperature of 40 ℃; extraction time of 2. 5 h. The average extraction rate of Herba Lycopi polysaccharide was 10. 70%,and RSD was 0. 17%. The antioxidant activity test showed that Herba Lycopi polysaccharide had a significant scavenging effect on radicals. [Conclusions] Herba Lycopi polysaccharides had strong antioxidant activity,and could provide a scientific basis for the development and utilization of Herba Lycopi medicinal resources.

  • Pharmacognostic Identification of Yi Medicine Herba Boenninghauseniae Albiflorae

    [Objectives] To identify original plant and traits of Yi medicine Herba Boenninghauseniae Albiflorae,and provide experimental data for its identification and application. [Methods] The original plant,traits,and microscopic identification were used. [Results] Essential identification characteristics of Yi medicine Herba Boenninghauseniae Albiflorae: small leaf was 1-2. 5 cm long,while the gynophore was 4-8mm long when bearing fruit; traits mainly included characteristic odor,strong and bitter taste,and spicy flavor; microscopic exclusive identifying characteristics: rich aleurone grain in the root; leaf had lysigenous secretory cavity,and non-glandular hairs and oil droplets were common. [Conclusions] Yi medicine Herba Boenninghauseniae Albiflorae( Ebazema) has distinctive identifying characteristics in original plant,traits,and microscopic tissues. This study is expected to provide certain reference for identification of original plant and medicine and formulation of quality standard for Yi medicine Ebazema,and to provide basis for its further research and development.

  • Identification of Yi Medicine Mubaribo

    [Objectives] To identify original plant and traits of Yi medicine Mubaribo( Verbenae herba) and provide effective experimental data for its identification and application.[Methods]The original plant,traits,and microscopic identification were used.[Results]Stem was square,the edges of basal leaves usually had rough saw teeth and notches; most cauline leaves were three parted,edges of lobes have uneven saw teeth,both edges had hard hair; spikes were terminal and axillary,thin and delicate; corolla was light purple to blue. Epidermal cells of stem were long polygonal or quasi-rectangular,vertical wall was straight with pores; lower epidermal cells of leaves were vertical to wall and took on wavy bending; pores were infinitive or anisocytic; 3-5 subsidiary cells; 4 glandularscale head cells,stalk single cell,non-glandular single cell; pollen grains were quasi-circular or quasi-triangular,smooth surface,and 3 germination apertures. [Conclusions] Yi medicine Mubaribo has distinctive identifying characteristics in original plant,traits,and microscopic tissues. This study is expected to provide certain reference for identification of original plant and medicine and formulation of quality standard for Yi medicine Mubaribo,and to provide basis for its further research and development.

  • System Control Research of Multi-link Fructus Trichosanthis Quality

    It could not guarantee the quality of medicinal materials and decoction pieces to purely make quality standard of traditional Chinese medicine and carry out quality control. For the problems existed in each process and link of traditional Chinese medicine production,control techniques and measures of guaranteeing and improving quality should be proposed,thereby forming process control system for quality of each traditional Chinese medicine. Based on systemic research on Fructus Trichosanthis which is common traditional Chinese medicine more than30 years,the concept of establishing process control system of Fructus Trichosanthis quality is proposed firstly in this paper. The research aims to find quality problems from 10 processes and links( plant variety,cultivation and propagation,field pest control,fruiting and maturity degree of fruit,storage,regulator,chemical ingredient,quality standard,limited index and circulation),and form feasible techniques and measures solving problems in each process and link,thereby systemically guaranteeing the quality of Fructus Trichosanthis medicinal material and decoction pieces. It guarantees and improves clinical efficacy of Fructus Trichosanthis in Chinese medicine,and could provide reference for establishing process control system of quality of other traditional Chinese medicines.

  • Identification of Citric Acid in Wuhuang Oral Liquid by Thin Layer Chromatography

    [Objectives] To screen the identification conditions of citric acid in Wuhuang Oral Liquid by thin layer chromatography,and establish the quality control method for citric acid in Wuhuang Oral Liquid.[Methods] Different treatment methods were adopted for test sample,developing agent,and drying time,thin layer chromatography separation condition and spot definition were taken as indicators to conduct experiment,to select optimal thin layer identification method. [Results] Methanol was used as the extraction solvent,ultrasonic treatment,ether extraction,dissolution by anhydrous ethanol as treatment conditions of test sample; upper solution of butyl acetate-formic acid-water(4 ∶ 2 ∶ 2) after placing one hour was taken as developing agent; 0. 1% bromocresol green(BCG) as the developer; when developing the color in 3 hours after development,in thin layer chromatograph,there appeared the same strip in the same position of test sample of Wuhuang ORAL Liquid and control substance,no obvious trailing phenomenon,and the color was uniform and clear.[Conclusions]The thin layer chromatography identification conditions can be used as the method for quality control of Wuhuang Oral Liquid.

  • Production of Sagittaria sagittifolia Beverage and Technological Condition Optimization

    [Objectives] To develop a kind of antioxidant health drink which could also regulate blood sugar.[Methods]With Sagittaria sagittifolia,hawthorn and liquorice as raw materials,the orthogonal test was done to determine the optimum preparation ratio of each raw material,optimum flavor modulation ratio and optimum stabilizer type and amount.[Results]Under the conditions of 0. 4% crude polysaccharide powder amount of Sagittaria sagittifolia,10. 0 m L hawthorn extract,2. 5 m L liquorice extract,7. 0% white sugar amount,0. 5% citric acid amount,0. 2% cyclodextrine amount,0. 05 g/L sodium alginate amount,0. 5 g/L xanthan gum amount,2. 0 g/L agar amount and 20 MPa homogenization,the beverage had optimum flavor and stability.[Conclusions]The health drink obtained under these technological conditions had delicate sweet taste and rich nutrition.

  • Pharmacognosy Identification of Yi Medicinal Material Campanula colorata

    [Objectives] To study the original plant,traits and microscopic identification for Yi medicinal material Campanula colorata,and provide experimental data for its identification and medicinal use. [Methods] Plant identification,trait identification,powder identification and paraffin slice identification were adopted in the study. [Results]It was found that the original plant of Campanula colorata was characterized by stiff hairs,slightly thicker root than stem,serrated leaf margin,tubular bell-shaped corolla and blue-purple color; traits were featured by odorlessness,and bitter taste after slight sweetness; the microscopic identification characteristics were that the laticifer was sporadic,containing reddish brown or yellow brown materials,and the oil droplets and nonglandular hairs were common. [Conclusions] The original plant,trait and microscopic identification characteristics were obvious for Yi medicinal material Campanula colorata,which could provide a reference for developing quality standards and identifying the original plant of Campanula colorata,and provide the basis for further research and development.

  • Efficacy of Temozolomide Combined with Whole Brain Radiotherapy in the Treatment of Cerebral Metastases from Lung Cancer

    [Objectives] To observe the clinical efficacy,adverse reaction and survival time of temozolomide combined with whole brain radiotherapy in the treatment of lung cancer. [Methods] A total of 43 patients with lung cancer and cerebral metastases were reviewed and analyzed. Three-dimensional conformal radiotherapy(3D-CRT) technique was used to perform whole brain radiotherapy,one time a day and5 times a week. At the same time of radiotherapy,temozolomide chemotherapy was performed,150 mg/( m2·d),continuous oral administration of 5 d,every 28 days were a cycle( those patients who continue receiving temozolomide chemotherapy did not receive other related antitumor therapy,such as systemic chemotherapy and molecular targeted therapy,etc.),and drugs were administered for 4-6 cycles according to tolerance of patients. Kaplan-Meier method was used to calculate the survival rate. [Results]The objective response rate of 43 patients was79. 0%(34/43),in which CR was 6/43,PR was 28/43,and 9 cases had PD. By December 31,2016,7 patients in 43 cases died,one patient died of cerebral hernia due to intracranial lesions and 6 patients died of failure of other important organs due to metastasis. The OS and PFS of the whole group of patients in one year were 49. 1% and 56. 9% respectively. The adverse reactions were mild and the patients could tolerate such treatment. [Conclusions] Temozolomide combined with whole brain radiotherapy in the treatment of lung cancer with cerebral metastases has excellent clinical efficacy,while patients can tolerate such treatment.

  • Expression of MMP-1 in the Scar Tissue of Skin and Oral Mucosa

    [Objectives] To investigate expression of MMP-1 in the scar tissue of skin and oral mucosa,and to explore if there is the significance of MMP-1 in the scar of skin and oral mucosa.[Methods]All samples were collected from surgical postoperative soft tissue from Department of Oral and Maxillofacial Surgery and Plastic Surgery in the Affiliated Qingdao Hiser Medical Center of Qingdao University Medical College,including 22 samples of dermal scar tissue,20 samples of normal skin tissue,and 13 samples of oral mucosal scar tissue,and 20 samples of normal oral mucosal tissue. HE staining was used to observe the morphology and collagen arrangement of fibroblasts in the scar tissue,the expression of MMP-1 in different tissue was observed by immunohistchemical staining. [Results] The positive expression rates of MMP-1 in skin scar tissue,oral mucousal scar tissue,normal skin tissue and normal mucosal tissue were 27. 27%,69. 23%,5. 00%,10. 00%. Comparison of positive expression of MMP-1 in skin scar tissue and oral mucosal scar tissue: the positive expression rate of MMP-1 in oral mucosal scar tissue was higher than that of MMP-1 in skin scar tissue,and the statistically significant difference was found between them( P < 0. 05).Comparison of positive expression of MMP-1 in skin scar tissue and normal skin tissue: the positive expression in skin scar tissue was higher than that in normal skin,and the statistically significant difference was found between them( P < 0. 05). Comparison of positive expression of MMP-1 in oral mucosal scar tissue and normal oral mucosal tissue: the positive expression rate of MMP-1 in oral mucosal scar tissue was higher than that in normal mucosal tissue,and the statistically significant difference was found between them( P < 0. 05). Comparison of positive expression of MMP-1 in normal oral mucosal tissue and normal skin tissue: the positive expression rate of MMP-1 in oral mucosal scar tissue was higher than that in normal mucosal tissue,and there was no statistically significant difference between them( P > 0. 05). [Conclusions] The positive expression of MMP-1 was different from that in the scar tissue of the oral mucosa,indicating that MMP-1 might be related to the severity of scar formation in different tissues.

  • Analysis of Clinical Treatment Effect on Oromaxillo-facial Multispace Infection

    [Objectives] To analyze the clinical treatment effect on oromaxillo-facial multi-space infection. [Methods] Based on the clinical data about 72 oromaxillo-facial multi-space infection cases from January 2016 to December 2016,the double-blind study was used to divide them into study group(36 cases) and control group(36 cases). As for the control group,36 patients with oromaxillo-facial multi-space infections were given the conventional treatment; as for the study group,36 patients were given the treatment of abscess incision and drainage combined with hyperbaric oxygen based on conventional treatment. The total effective rate and complication rate were compared between the two groups. [Results]The total effective rate and complication rate under the study group were higher than under the control group,and the difference was statistically significant( P < 0. 05). [Conclusions] The clinical treatment effect on oromaxillo-facial multi-space infection was very significant,and the treatment was worthy of promotion and application in clinical practice.
