Chemical Composition of Natural Medicine

  • Determinations of Five Kinds of Secondary Metabolites from Different Parts of Piper sarmentosum Roxb.

    [Objectives] To establish the determination methods for the contents of total flavonoids,total alkaloids,total saponins,total phenolics and total tannins from different parts of P.sarmentosum.[Methods] The content of total flavonoids was measured by the NaNO_2-Al(NO_3)_3-NaOH colorimetry; the content of total alkaloids was measured by the bromocresol green acid dyes colorimetric assay; the content of total spaonins was measured by the vanillin- acetic acid acid dyes colorimetric assay; the content of total phenolics was measured by the FolinCiocalteu assay; the content of total tannins was measured by the phospho molybdenum tungstic acid- casein colorimetry.[Results]The average recovery rate of rutin was 98.11% and RSD was 0. 59%,the contents of total flavonoids from roots,stems and leaves of P.sarmentosum were 0. 25%,0.33% and 1.18% respectively; the average recovery rate of berberine hydrochloride was 99.62% and RSD was 1.52%,the contents of total alkaloids from roots,stems and leaves of P.sarmentosum were 0. 01%,0.03% and 0.19% respectively; the average recovery rate of ginsenoside Re was 99.21% and RSD was 1.06 %,the contents of total saponins in roots,stems and leaves of P.sarmentosum were 4.13%,1.71% and 4.33% respectively; the average recovery of gallic acid was 97.95% and RSD was 0.88 %,the contents of total phenol in roots,stems and leaves of P.sarmentosum were 0.68%,1.08% and 2.42% respectively; the average recovery of gallic acid was 97.92% and RSD was 3.14%,the contents of total tannins in roots,stems and leaves of P.sarmentosum were 0.72%,1.21% and 1.69% respectively.[Conclusions] These five proposed methods are precise,accurate and reproducible,and they are suitable for the determinations of total flavonoids,total alkaloids,total saponins,total phenolics and total tannins in P.sarmentosum.

  • A Study on the Ultrasonic Extraction and Physicochemical Properties of Selfheal Pigment

    [Objectives] To explore a new method of ultrasound-assisted extraction of selfheal pigment as well as pigment stability.[Methods] Taking ultrasound-assisted extraction of selfheal pigment by single-factor and orthogonal experiment,the optimum process conditions were determined,and the stability of pigment was studied.[Results] The optimal extraction conditions were as follows: ultrasound-assisted power of 140W; extraction time of 40 min; extraction temperature of 70℃; distilled water as extraction reagent; liquid-solid ratio of 1: 20(g:mL). Selfheal pigment stability under light was good;thermal stability is poor; food additives had small effect on the stability; it was stable at room temperature,and the degradation was accelerated with rising temperature; the resistance to reduction is good while the resistance to oxidation is poor; K~+,Na~+,Mg~(2+) and Ga~(2+) almost had no effect on the pigment,while Zn~(2+),Ba~(2+),Fe~(3+) and Cu~(2+) affected the stability to some extent.[Conclusions]The method shortened the extraction time of pigment,and it was more effective and economical.

  • Comparison of Contents of Alkaloids and Flavonoids in Sophora flavescens from Different Habitats of Guizhou Province

    [Objectives] To provide theoretical foundation for protection and utilization of Sophora flavescens in Guizhou. [Methods] Contents of alkaloids and flavonoids in S.flavescens from different habitats of Guizhou Province were studied by ultraviolet spectrophotometry.[Results]The content of alkaloids in S.flavescens was 15.60% in Dafang County,Bijie City and 15.20% in Sinan County,Tongren City,obviously higher than that of other habitats. The content of flavonoids in S.flavescens from Laoyingshan Town,Liupanshui City was the highest,reaching 15.70%. [Conclusions] There were big differences between various habitats in the contents of alkaloids and flavonoids in S.flavescens.

  • Optimization of Ultrasonic Extraction Process of Glycyrrhizic Acid and Liquiritin from Extractum Glycyrrhizae by Response Surface Method

    [Objectives] To optimize the extraction technique for glycyrrhizic acid and liquiritin from Xinjiang extractum glycyrrhizae by response surface method. [Methods] Based on the single-factor tests on ethanol concentration,ultrasonic wave time and solid-liquid ratio,response surface method of three factors and three levels was adopted according to the Box-Behnken central composite design principle. The technological condition of glycyrrhizic acid and liquiritin was optimized by ultrasonic wave. [Results]The optimal extraction process for glycyrrhizic acid and liquiritin was as follows: 60 % ethanol concentration,30 min ultrasonic time,2. 6 g/L solid-liquid ratio. Under these conditions,the one-time extraction rate of glycyrrhizic acid and liquiritin was 8. 12 % and 0. 736 %,respectively. [Conclusions]The optimal extraction technology for glycyrrhizic acid and liquiritin from extractum glycyrrhizae was established,which could provide theoretical foundation for the intensive processing of extract and the application of glycyrrhizic acid and liquiritin in food and medicine.

  • Content Determination about Semi-finished Products of Astragalus Saponin

    [Objectives] For qualitative and quantitative investigation of Astragalus saponin semi-finished products,to establish the Astragalus content determination method,determine the content of Astragalus saponins in semi-finished products,and provide full guarantee for the followup preparation and product quality control.[Methods]With Astragaloside A as reference substance,TLC method was used for qualitative determination of the main components in semi-finished products of Astragalus saponin. Using colorimetry,the content was determined,and with sulfuric acid as chromogenic reagent,the wavelength was measured to be 320 nm.[Results]When the concentration of Astragaloside A was 1.04-8.32 μg·ml~(-1),there was a linear correlation between absorbance and concentration,and precision,stability and reproducibility were in line with standards. The content of Astragaloside A in semi-finished products of Astragalus saponin was measured to be 50- 60%. [Conclusions]The sulfuric acid chromogenic method could be used to determine the content of Astragaloside A in semi-finished products of Astragalus saponins,and it had good precision,stability and reproducibility.

  • Optimization of Extraction Process of Total Coumarins from Tibetan Herb Artemisia Subdigitata Mattf. by Response Surface Method

    [Objectives]To optimize the ultrasonic extraction conditions of total coumarins from Tibetan herb Artemisia subdigitata Mattf.[Methods]In single-factor tests,the effects of ethanol concentration,extraction time,extraction temperature,solid-liquid ratio and ultrasonic time in extraction procedure on the extraction rate of total coumarins in A.subdigitata Mattf. were researched,and the Box-Behnken experimental design was used to optimize the extraction of total coumarins in A.subdigitata Mattf. [Results]The optimum extraction conditions were as follows:53% ethanol,extraction time 30 min,extraction temperature 85 ℃,ratio of solid to liquid 1:52(g/mL) and ultrasonic time 20 min. Under the conditions,the average extraction yield of total coumarin reached 20.01 mg/g. [Conclusions]he optimized method was convenient and feasible,which could provide reference for the extraction of total coumarins from A.subdigitata Mattf.,and provide some guidance and basis for the further exploitation of A.subdigitata Mattf.

  • Effects of Brassinolide on Physiological Traits of Different Varieties of Salvia miltiorrhiza Bunge Seedlings under Drought Stress

    [Objectives] To study changes in SOD,POD,CAT,MAD and Pro content of S.miltiorrhiza Bunge seedlings and the effects of brassinolide( BR) on drought resistance of S.miltiorrhiza Bunge. [Methods] 6 varieties of S.miltiorrhiza Bunge produced in Zhongjiang County of Sichuan Province were used as research object,and the method of PEG-6000 simulated drought stress was applied. [Results]20% PEG stress 9 d was used as the optimal simulation condition of drought stress. Under the same degree of drought stress,tissue-cultured big leaf S.miltiorrhiza Bunge showed the highest SOD and POD activity,tissue-cultured small leaf S.miltiorrhiza Bunge showed highest CAT activity,long-stalked big leaf S.miltiorrhiza Bunge had lower MDA content than other varieties,and Zhongjiang wild S.miltiorrhiza Bunge had the highest Pro content. [Conclusions] BR can indirectly improve drought resistance ability of S.miltiorrhiza Bunge,and 0.1mg/L BR has the best action effect.

  • Effects of IBA on Hydroponics Rooting and Plant Height by Branch and Twig of Lysimachia christinae ‘Aurea’

    [Objectives] Effects of IBA( Indole Butyric Acid) on rooting and plant height by branch and twig of Lysimachia christinae‘Aurea'were studied in order to research its hydroponics propagation and provide theoretical and practical basis for its reproduction and culture.[Methods]The branch and twig of L. christinae‘Aurea'were treated with IBA in the concentrations of 0,10,50 and 100 mg/L and with 4 h of soaking time in this study.[Results] The rooting rate,rooting number,root developing index and the plant height of the twig and branch cuttings treated with three kinds of concentrations of IBA were higher than control. The rooting rate and number of branch treated with three kinds of concentrations of IBA were significantly higher than control,while the differences of number and length of adventitious root were not obvious.The rooting rate and number,the plant height of the branch cutting were larger than that of twig. The speed of root growing on the branch was faster than that of the twig among all the treatments. After the branch cuttings were treated with 50 mg/L of IBA,the rooting rate was 100% and the rooting developing index was 1.73,which were significantly higher than the other treatments.[Conclusions]The best treatment for the branch cutting rooting of L. christinae‘Aurea'was 50 mg/L of IBA solution with 4 h of soaking time.

    [Objectives] To identify the species of purple flower of Allium in Jiucaiping,Hezhang County of Guizhou,and provide reference data for scientific and reasonable development of this plant resources. [Methods] Botanical origin was identified to the species of wild purple flower of Allium and morphological identification and microscopic identification were adopted to make pharmacognostic study. [Results] Wild purple flower of Allium was Allium wallichiii Kunth; in botanical origin,morphological and microscopic aspects,it had special and distinctive characteristics from Allium tuberosum Rottl. ex Spreng. [Conclusions]In botanical origin,morphological,and microscopic aspects,the identification characteristics were distinctive. This study was expected to provide reference for identification and formulation of quality standard for botanical origin and medicinal plant of Allium wallichii Kunth,and provide fundamental experimental data for scientific and reasonable use of this large area wild purple flower of Allium.

  • Protection of Dicliptera chinensis Polysaccharide Granule on Acute Liver Injury of Rat Caused by D-galactosamine

    [Objectives] By comparing D. chinensis polysaccharide granule with D. chinensis polysaccharide of direct use equivalent,it is determined if the dosage form could affect hepatoprotective effects and mechanisms of D. chinensis polysaccharide. [Methods] 50 SD rats are randomly divided into 5 groups: blank group,model group,biphenyl dimethylester positive group( 50 mg /( kg·d)),D. chinensis polysaccharide group( 200 mg /( kg·d)) and D. chinensis polysaccharide granule group( 700 mg /( kg·d),which is equivalent to 200 mg /( kg·d) of polysaccharide). Except blank group and model group,corresponding dosage of drug is irrigated to the stomach in other groups. After two weeks,except blank group,D-galactosamine( 400 mg / kg) is injected into abdominal cavity of each group to establish liver injury model of rat.After anaesthesia,blood is taken from abdominal aorta,and liver is collected and fixed by 4% paraformaldehyde. The levels of glutamic-pyruvic transaminase( ALT),AST,superoxide dismutase( SOD),glutathione peroxidase( GSH-PX),malonaldehyde( MDA),tumor necrosis factor-α( TNF-α),interleukin-1β( IL-1β) and interleukin-6( IL-6) in serum are detected,and microscopic observation of liver tissue pathological section is conducted. [Results]Compared with model group,the levels of ALT,AST,MDA,TNF-α,IL-1β and IL-6 in rat's serum from D.chinensis polysaccharide group and D. chinensis polysaccharide granule group significantly decline,while SOD activity significantly rises. Pathological changes of liver tissue are improved significantly,and their curative effects are equivalent. [Conclusions]D. chinensis polysaccharide granule has significant protection role on acute liver injury of rat caused by D-galactosamine; its utilization convenience is showed while not declining its hepatoprotective effect.

  • Molecular Cloning and Characteristics of Farnesyl Pyrophosphate Synthase in Fritillaria thunbergii

    [Objectives] To conduct molecular cloning and characteristic analysis of Farnesyl Pyrophosphate Synthase(FPS) so as to provide the foundation for molecular regulation of steroid alkaloid biosynthesis in Fritillaria thunbergii.[Methods]A pair of degenerate primers were designed based on the conservative sequences of the cloned FPS gene from other plant species,and this study used RT-PCR and Ta cloning technology as well as q PCR to analyze the bioinformation of FPS and the expression of FPS in different parts of Fritillaria thunbergii. [Results] A 414 bp gene was gained,and sequence analysis showed that the conserved sequence fragments of Fritillaria thunbergii FPS gene had high identity(93%- 76%) to other plants,and the deduced amino acid sequence was predicted to have high homology with other plants(96%-82%) and was closely related to Lilium longiflorum. Gene expression analysis revealed that the expression of FPS was highest in flower,and was followed by bulb and stem.[Conclusions]FPS gene of Fritillaria thunbergii was initially determined and named Ft FPS as the first reported FPS gene from Fritillaria thunbergii,laying a foundation for effective utilization of the gene.

  • Optimal Selection of Formation Process for Dicliptera chinensis Polysaccharide Granules

    [Objective] To select optimal formation process for Dicliptera chinensis polysaccharide granules. [Methods] Forming rate,flow,and solubility of granules were used as indicators. Single factor experiment and orthogonal experiment were carried out to optimize types and volume of auxiliary materials( filling agent and binding agent) used in granule prescription. Quality of formed granules was inspected.[Results]The optimal formation process was 1: 2. 5: 1% for Dicliptera chinensis polysaccharide: mannitol: 3% HPMC aqueous solution. The grain size,flow,and solubility of prepared granules met requirements of Chinese Pharmacopoeia. [Conclusion]The optimally selected formation process for Dicliptera chinensis polysaccharide granules is feasible and suitable for industrialized production.
