  • Analysis on Nutritional and Active Components of Rhizoma Dioscoreae

    [Objective] The nutritional components and active components of Rhizoma Dioscoreae were analyzed,to provide basis for further development and use of Rhizoma Dioscoreae. [Methods]44 Rhizoma Dioscoreae samples were collected and taken as research objects,conventional detection method was used to analyze main nutritional and active components. [Results] There are differences in the content of main components between 44 Rhizoma Dioscoreae samples,and the difference is up to extremely significant level. Henan Dioscorea opposita Thunb contains 12. 65% adhesive protein; Shandong Short Rhizoma Dioscoreae contains 27. 19% crude polysaccharides and 42. 89% dry matters;Guangdong Shenzhen Purple Rhizoma Dioscoreae contains 40. 68% polyphenol; Hubei Shiyan Maojian District Dujiawan No. 1 Rhizoma Dioscoreae contains 23. 72% amylose and 60. 48% starch; Zhejiang Taizhou Huangyan District purple Rhizoma Dioscoreae contains 39. 94% amylopectin; Henan Dioscorea opposita Thunb( planted in Enshi Prefecture of Hubei Province) contains 0. 88% allantoin; Shiyan Maojian District Dujiawan No. 6 contains 0. 0509% saponins. [Conclusion]Components of Rhizoma Dioscoreae are influenced by environment of planting area and field management factors including gene control,soil,climatic conditions,and cultivation and management methods,as well as harvesting time. This study is expected to provide theoretical reference for developing health products using Rhizoma Dioscoreae.

  • Extraction and Determination of Total Saponins in Tender Leaves of Acanthopanax giraldii Harms

    [Objective] To establish a method of extracting total saponins in tender leaves of Acanthopanax giraldii Harms. [Methods] Orthogonal experiment was carried out to optimize the extraction process of total saponins in tender leaves of A. giraldii Harms. Besides,single factor experiment was carried out to study the effects of extraction solvent,extraction methods,solid to liquid ratio,and reflux time on content of total saponins in tender leaves of A. giraldii Harms,so as to screen out optimal extraction process. [Results]The optimal extraction process conditions obtained from single factor experiment for total saponins in tender leaves of A. giraldii Harms: the content of total saponins was the highest at 80% methanol volume fraction,1: 35 solid to liquid ratio,and 150 min reflux time. [Conclusion]The method was simple and accurate in determining total saponins in tender leaves of A. giraldii Harms,and the content of total saponins in tender leaves of A. giraldii Harms was higher.

  • Chemical Components and Their Antioxidant Activities in vitro of Volatile Oils from Leaves of Artemisia argyi: A Case Study of Qiai

    [Objective] To determine chemical components of volatile oils from leaves of Artemisia argyi and study their antioxidant activities in vitro. [Methods]Volatile oils were extracted from leaves of Artemisia argyi by the steam distillation( SD) method and chemical components were analyzed and identified by gas chromatography-mass spectrometry( GC-MS) method. Through the method of scavenging DPPH( 1,1-diphenyl-2-picrylhydrazyl) organic free radicals,functions of volatile oils scavenging free radicals were studied. Ultraviolet-Visible Spectrophotometry was applied to determine and compare the clearance rate of volatile oils,antioxidant BHT( 2,6-Di-tert-butyl-4-methylphenol) and Ve( Vitamin E). [Results]40 chemical compounds were detected in volatile oils of leaves of Artemisia argyi. 27 of them were preliminarily identified,accounting for 89. 12% of total volatile compounds. Major compounds were thujone( 25. 17%),eucalyptol( 23. 42%) and alcanfor( 8. 58%),α-Thujone( 7. 76%),terpinen-4-ol( 4. 7%),and borneol( 3. 34%). Antioxidant activity in vitro experiment indicated that the ability of scavenging DPPH free radicals is BHT > volatile oils of Artemisia argyi > Ve. [Conclusion]Volatile oils of Artemisia argyi mainly consist of ketones and terpenes,which have excellent in vitro antioxidant activity.

  • Optimization of Extraction Process and Content Determination of Total Caffeoyl Quinic Acid Esters in Erigeron multiradiatus

    [Objective] To determine the content of total caffeoyl quinic acid esters( TCaf) in Erigeron multiradiatus and optimize the extraction process of total caffeoyl quinic acid esters in Erigeron multiradiatus. [Methods]Ultraviolet-Visible Spectrophotometry was used. 1,3-Dicaffeoylquinic acid was used as reference substance to determine the content of total caffeoyl quinic acid esters in Erigeron multiradiatus. Single factor experiment was carried out to study influences of ethanol mass fraction,extraction time,extraction temperature,and solid-to-liquid ratio on extraction rate of total caffeoyl quinic acid esters. And L9( 34) orthogonal experiment was carried out to optimize extraction process of total caffeoyl quinic acid esters in Erigeron multiradiatus,and determine content of total caffeoyl quinic acid esters in Erigeron multiradiatus samples. [Results] The optimal extraction process is ethanol volume fraction 55%,extraction temperature 60℃,extraction time 100 min,solidto-liquid ratio 1∶ 14. Under this condition,the extraction rate of total caffeoyl quinic acid esters in Erigeron multiradiatus is 73. 52 mg / g; there is significant difference in the content of total caffeoyl quinic acid esters between different plant positions,and leaves contain the highest total caffeoyl quinic acid esters. [Conclusion]The optimized extraction process is reasonable and feasible and can be used in extraction of total caffeoyl quinic acid esters in Erigeron multiradiatus.

  • Effect of Shade on Chemical Constituents of Turmeric Volatile Oil

    [Objective]To study the effect of shade on chemical constituents of the volatile oil in turmeric( Curcuma longa L.).[Methods]The volatile oil of turmeric under different degrees of the shade was extracted by steam distillation. The chemical constituents were analyzed by gas chromatography-mass spectrometry( GC-MS).[Results]34 constituents from turmeric under different degrees of shades were identified,accounting for 64. 10%,64. 33% and 70. 05% of total constituents,respectively,and the content of them decreased or increased with shading. Ar-turmerone is the main chemical constituent of the volatile oil,accounting for 44. 83%,47. 95% and 51. 90% of the total volatile oil,respectively,and the content was gradually increased with the increase in degree of shade.[Conclusions]There was a difference in the content of components of turmeric volatile oil under different degrees of shade,and increasing the degree of shade is conducive to the synthesis of arturmerone and other major active components in volatile oil.

  • Analysis on Chemical Components of Volatile Oils from Alpiniae Oxyphyllae Fructus in Guangdong

    [Objective] To analyze chemical components of volatile oil from Alpiniae Oxyphyllae Fructus in Guangdong by the gas chromatography mass spectrometry( GC-MS). [Methods] Volatile oils were extracted from Alpiniae Oxyphyllae Fructus by steam distillation method;chemical components of volatile oils from Alpiniae Oxyphyllae Fructus were determined by GC-MS; relative percentage content of chemical components was determined by normalization method. [Results] 45 kinds of chemical components were identified. Major components included Nootkatone( 20. 500%),Alloaromadendrene( 6. 133%),7-Isopropenyl-1,4a-dimethyl-4,4a,5,6,7,8-hexahydro-3H-naphthalen-2-one( 3. 044%),Naphthalene,1,2,3,5,6,7,8,8a-octahydro-1,8a-dimethyl-7-( 1-methylethenyl)-,[1S-( 1. alpha.,7. alpha.,8a. alpha.) ]-( 2. 816%),2( 1H) Naphthalenone,3,5,6,7,8,8a-hexahydro-4,8a-( 2. 309%),and Caryophyllene oxide( 1. 372%),etc. [Conclusion] This method is high efficient and rapid in separation of chemical components,and thus can be applied in quality control of Alpiniae Oxyphyllae Fructus.

  • Determination of Cadmium Content in CHUANXIONG RHIZOMA by Microwave Digestion—Graphite Furnace Atomic Absorption Method

    [Objective] To establish an analysis method for accurately determining the cadmium content in CHUANXIONG RHIZOMA.[Methods]Tuberous roots of CHUANXIONG RHIZOMA were pulverized and 0. 1g of the powder was digested by microwave digestion. After25 min of digestion,the acid was driven away by heating and diluted to 100 m L. The cadmium content of the sample solution was detected by graphite furnace atomic absorption method. 1. 0 g / L phosphoric acid was used as the matrix modifier and the ashing temperature was increased to 550 ℃. [Results]The detection limit was 0. 08 mg / kg,and the intraday precision( % RSD,n = 6) was 0. 2%- 6. 5% in the linear range of 0. 08- 2. 0 μg / L. 48 samples of CHUANXIONG RHIZOMA in 5 planting regions were measured with the recovery rate of 96. 0%-109. 6%. [Conclusion] The method established in this study is sensitive,accurate,high efficient and easy to be popularized. It will provide a reliable quantitative method for determining the distribution of cadmium content in CHUANXIONG RHIZOMA.

  • Analysis and Identification on a Strain of Entomogenous Fungus in Tibet's Nyingchi

    [Objective] To conduct morphological and molecular identification on a strain of entomogenous fungus collected from Baji Village,Bayi Town,Nyingchi County,Tibet in July 2015. [Methods]The fungal ITS sequence identification and analysis were conducted. [Results]The degree of similarity in ITS sequence between the entomogenous fungus and Ophiocordyceps sinensis was 83%. The host insect was Thitarodes pui,with COI gene similarity of 99%. [Conclusions]Based on morphological analysis,it was found that this entomogenous fungus might fall within the group of Ophiocordyceps sinensis.

  • Thin Layer Chromatography for Specnuezhenide in LIGUSTRI LUCIDI FRUCTUS- ASTRAGALI RADIX Extraction Solution

    [Objective] To explore thin layer chromatography for specnuezhenide in LIGUSTRI LUCIDI FRUCTUS-ASTRAGALI RADIX extraction solution. [Methods] Ultrasonic was used to extract the active component specnuezhenide. Different extraction reagents were designed to obtain test sample solution. Specnuezhenide was taken as the reference substance. Ethyl acetate- acetone- formic acid- water were used to prepare different developing agents. 1% vanillin H2SO4 ethanol solution and 10% H2SO4 ethanol solution were used as color developing agent to conduct thin layer chromatography. [Results] The fourth extraction method indicated the best effect for the test sample solution of LIGUSTRI LUCIDI FRUCTUS- ASTRAGALI RADIX extraction solution. The developing agent was ethyl acetate- acetone- formic acid- water( 6∶ 3∶ 0. 5∶ 1). Color developing agent was 1% vanillin H2SO4 ethanol solution. In visible light,the test sample solution showed purple spot with the same color in corresponding position of the reference solution,and the developed color was clear. [Conclusion]The fourth method of thin layer chromatography for specnuezhenide in LIGUSTRI LUCIDI FRUCTUS- ASTRAGALI RADIX is used to prepare the test sample solution. The ethyl acetate-acetone-formic acid-water( 6∶ 3∶ 0. 5∶ 1) was used as the developing agent. After color developing using 1% vanillin H2SO4 ethanol solution,the same purple spot appears in the visible light and the corresponding position of the reference solution.

  • Microscopic and TLC Identification on Herbal Slice of Chinese Honeylocust Abnormal Fruit

    [Objective] To establish the microscopic and TLC method for the herbal slice of Chinese Honeylocust Abnormal Fruit in Standard on the Chinese Herbal Slice Preparation in Sichuan Province( 2015). [Methods] Conventional powder microscopic identification and TLC.[Results]The microscopic characteristics could be seen about exocarp cells,fibers,clusters of calcium oxalate,lignified parenchyma cells,polycrystalline fibers,calcium oxalate crystals and pitted vessels; the TLC conditions took chloroform-methanol-2 water-glacial acetic acid( 18∶ 1∶ 0. 6∶ 0. 2),toluene-ethyl acetate-formic acid( 14∶ 4∶ 0. 5),and chloroform-ethyl acetate-methanol-water( 15∶ 40∶ 22∶ 10) as developing agents,respectively,and 10% sulfuric acid and ethanol solution as chromogenic agent; the Chinese Honeylocust Abnormal Fruit sample and the control herbs shared the consistent corresponding spots,the color was clear,and the separation effect was good. [Conclusions]The microscopic characteristics of Chinese Honeylocust Abnormal Fruit powder showed strong specificity; the three kinds of TLC conditions had strong specificity controllability,easy operation and high accuracy,which could quickly identify Chinese Honeylocust Abnormal Fruit herbs.

  • Effects of Quercetin Liposomes on Oxidative Stress and Expression of TGF- β1 in Kidney of Diabetic Rats

    [Objective] To observe the effects of different doses of quercetin liposomes( LQ) on oxidative stress and expression of TGF-β1 in kidney of diabetic rats. [Methods]The model of type 2 diabetic nephropathy rats was established by being fed on high-sugar and high-fat diet combined with intraperitoneal injection of streptozotocin( STZ). Then type 2 diabetic rats were randomly divided into six groups: diabetic model group( group DM),low level of LQ group( group LQ-L),medium level of LQ group( group LQ-M),high level of LQ group( group LQH),positive control group( group aminoguanidine,AG) and control group( group N). After 8 weeks,the serum levels of total cholesterol( TC),triglycerides( TG),superoxide dismutase( SOD),malondialdehyde( MDA),glutathione peroxidase( GSH-Px) were measured by biochemical methods. Immunohistochemistry and RT-PCR were used to detect expression of TGF-β1 gene and protein in kidney cortex of diabetic rats. [Results]Compared with the group N,the serum levels of SOD and GSH-Px decreased significantly in group DM,while the serum levels of TC,TG and MDA increased significantly and the expression levels of TGF-β1 gene and protein in kidney tissue increased significantly.Compared with the group DM,in each dose group of LQ have improved the above indicators to some extent and the group LQ-M was more obvious than others. [Conclusions] LQ reduces the blood lipid levels,protects antioxidant activity,inhibits lipid peroxidation of DM rats,and then reduces expression of TGF-β1 gene and protein in kidney cortex.

  • A Study on the in vitro Antioxidant Activity of Total Alkaloids in Abrus cantoniensis Hance Stems

    [Objective] To evaluate the antioxidant activity of total alkaloids in Abrus cantoniensis Hance stems. [Methods]The experiment of clearing super O_2~-·,clearing DPPH· and suppressing β-carotene-linoleic acid was conducted on Abrus cantoniensis Hance stems. [Results]The total alkaloids in Abrus cantoniensis Hance stems had strong antioxidant effect on β-carotene-linoleic acid oxidation system,and had the ability to scavenge super O_2~-· and DPPH· free radical. [Conclusions]Different in vitro antioxidant models showed that the total alkaloids in Abrus cantoniensis Hance stems had strong antioxidant activity.

Processing of Traditional Chinese Medicine and Quality Control

  • Orthogonal Design for Extraction Process and Anti- oxidant Activity Study of Polysaccharides in Abutilon indicum( Linn. ) Sweet

    [Objective] To extract polysaccharides in Abutilon indicum( Linn.) Sweet using cellulase,determine the optimal extraction process through orthogonal design,and treat the extracted polysaccharides and study the anti-oxidant activity. [Methods]Single factor experiment and orthogonal experiment were carried out to study effects of p H value of buffer solution,enzyme volume,enzymolysis temperature,and extraction time on extraction of polysaccharides in Abutilon indicum( Linn.) Sweet. Polysaccharides in Abutilon indicum( Linn.) Sweet was prepared and purified,DPPH and hydroxyl free radicals scavenging effect and Fe3 +reducing ability were measured,and anti-oxidant ability was evaluated according to experimental results. [Results]The conditions for optimal extraction process: p H 6. 5,enzyme volume 5 m L,enzymolysis temperature 50℃,extraction time 1. 5 h,average extraction rate 5. 223%,and RSD 1. 68%. Anti-oxidant experiment indicates that polysaccharides in Abutilon indicum( Linn.) Sweet has significant effect on scavenging of free radicals. [Conclusion]In certain range of concentration,there is dose-response relationship between polysaccharides in Abutilon indicum( Linn.) Sweet and scavenging rate of free radicals and the scavenging activity is significant. This study is expected to provide a certain scientific basis for comprehensive use and development of Abutilon indicum( Linn.) Sweet.

  • Effect of Different Concentrations of ABT-6 Rooting Agent on Hardwood Cutting of the New Species of Lycium barbarum var. implicatum

    [Objective] To develop the wild Chinese wolfberry resources and accelerate the propagation rate of hard branch cutting of Lycium barbarum var. implicatum.[Methods] The experiment about the effect of quick dipping of different concentrations of ABT-6 rooting agent on the hardwood cutting seedling formation of Lycium barbarum var. implicatum was carried out.[Results]The survival rate of Lycium. Barbarum var. implicatum was influenced mostly by 400 mg / L ABT-6 at 82. 33%; the number of shoots and number of new roots were the greatest,6. 6and 8. 9 per plant,respectively,significantly larger than under other treatments; the new shoots and new roots were also longest,with length of 39. 6 cm and 29. 8 cm,respectively,significantly greater than under other treatments.[Conclusions]The quick dipping of 400 mg / L ABT-6 rooting agent on the hardwood cutting of Lycium barbarum var. implicatum can significantly improve the survival rate,promote the growth of roots and shoots,and speed up the propagation.
