  • GC-MS Fingerprint of Volatile Oil from Artemisia argyi

    [Objective] To establish the GC-MS fingerprints for Artemisia argyi volatile oil as a basis for the identification and quality of Artemisia argyi volatile oil evaluation. [Methods]15 batches of Artemisia argyi samples were collected,and different batches of Artemisia argyi volatile oil was extracted by steam distillation. And the temperature at feed inlet was 230 ℃,the carrier gas was nitrogen,and the carrier gas flow rate was 0. 8ml / min. GC-MS of volatile oil in 15 batches of Artemisia argyi was analyzed by temperature programmer. The GC-MS fingerprint was established and fingerprint similarity was analyzed. [Results] P-cymene,eucalyptol,terpene alkene,thujone,camphor,borneol,1-caryophyllene,caryophyllene oxide and other 23 characteristic components were calibrated as common characteristic peaks to establish Artemisia argyi GC-MS fingerprint. The relative retention time and RSD of relative peak of total peak had smaller differences,and 15 batches of Artemisia argyi volatile oil similarity was greater than 0. 90. [Conclusions]The study established the GC-MS fingerprint of Artemisia argyi volatile oil and determined 23 characteristic peaks of Artemisia argyi volatile components,which improved Artemisia argyi essential oil quality evaluation and control system,and provided overall objective experimental evaluation for Artemisia argyi quality.

  • Microwave-assisted Hydrodistillation of Essential Oil from Thyme (Thymus Vulgaris)

    [Objective]To extract essential oil from the leaves of thyme( Thymus Vulgaris). [Methods]Microwave-assisted hydrodistillation was used as an effective method in extraction of thyme essential oil. [Results]Extraction conditions were optimized to increase the yield of essential oil. The essential oil composition of thyme leaves were investigated by GC-MS and 52 volatile compounds were identified,representing96. 13% of as-obtained essential oil. Carvacrol,thymol,borneol,2-methoxy-3-( 2-propenyl)-phenol and spathulenol were found to be the major components,respectively. [Conclusions]The essential oil obtained was mainly composed of active monoterpene phenols,monoterpene hydrocarbon and sesquiterpenes. MAHD is a simple,rapid,energy-saving and inexpensive extraction method that deserves to be considered as an alternative technique in extraction of volatile compounds from other plants.

  • HS-SPME-GC-MS Analysis of Volatile Components in Semen Cassiae

    [Objective] To analyze the composition and content of volatile components in Semen Cassiae. [Methods]The volatile components were extracted with headspace solid phase micro extraction( HS-SPME),and their content and name were confirmed by GC-MS. [Results]54chromatographic peaks were isolated from volatile components of Semen Cassiae,and 31 kinds of compounds were identified,accounting for 51.96% of total volatile substances. [Conclusions] HS-SPME-GC-MS technology was used to analyze volatile components of Semen Cassiae,which can provide a reference for further development and use of Semen Cassiae.

  • Comparative Study on Volatile Components of Flos Caryophmlli from Different Habitats by HS-SPME-GC-MS

    [Objective] To analyze the volatile chemical components of Flos Caryophmlli from different Habitats. [Methods] The volatile components were analyzed by HS-SPME-GC / MS and quantified by peak area normalization method. Besides,the data was analyzed by principal component analysis( PCA) and hierarchical cluster analysis( HCA). A total of 72 kinds of volatile components were identified from the 13 batch of Flos Caryophmlli.[Results]The volatile components of the Flos Caryophmlli from different habitats are different. And Flos Caryophmlli from Madagascar,Guangdong,Guangxi and Indonesia can be identified effectively by PCA and HCA.[Conclusion]The HS-SPME-GC-MS technique combined with PCA and HCA can be used for identifying volatile components of Flos Caryophmlli from four habitats. This study is expected to provide a new approach for the comparison and quality assessment of Flos Caryophmlli from different habitats.

  • Study on Enrichment and Purification Procedures of Total Glycosides from Acanthopanax giraldii Harms with Macroporous Resin

    [Objective] To establish the technical parameters,enrichment and purification procedures of total glycosides from Acanthopanax giraldii Harms with macroporous resin. [Methods]A RP-HPLC method was adopted to determine the content of total glycosides in A. giraldii Harms,and with the elution efficiency of syringin and oleanolic acid as the index,the adsorption characteristics and elution parameters of the enrichment and purification procedures were tested from 10 kinds of macroporous resin. [Results] The method was good for determining the content of total glycosides. X-5 was the best of macroporous resin; the optimal concentration of total glycosides solution extracted from A. giraldii Harms was 0. 30 g crude powder / m L; the best p H of the powder solution was 10; static adsorption lasted for 8 h; the best dosage of 80%ethanol was 4 BV. [Conclusions]These results were accurate and repeatable; X-5 macroporous resin was the best one for enrichment and purification procedures of total glycosides from A. giraldii Harms.

  • GC Determination of Limonene in Bergamot Volatile Oil

    [Objective] To establish a method to control the quality of bergamot volatile oil. [Methods] GC method was used to determine the limonene content of bergamot volatile oil. [Results]The separation was carried out on a 5% Phenyl Methyl Siloxan column( 30 m × 0. 32 mm × 0. 25 μ m); the detector was FID,and the temperatures of the injection port and detector were 250 ℃ and 280 ℃,respectively; temperature programming started at 65 ℃,holding for 1 min,then increased to 100 ℃ at a rate of 2 ℃·min- 1,kept for 1 min; finally,the column temperature increased to 250 ℃ at a rate of 50 ℃·min- 1,kept for 5 min; the carrier gas was nitrogen with flow rate of 0. 8 m L·min- 1,split radio was 40∶ 1,and injection volume was 1 μL. [Conclusions] The calibration curve had good linearity over the range of 0. 613 1 ~ 30. 654μg( r = 0. 999 9). The average recovery of limonene was 98. 96%. The method is simple,sensitive and accurate,with good repeatability and recovery,and it can be used for the quality control of bergamot volatile oil.

  • Determination of Mogroside V,Liquiritin and Glycyrrhizic Acid in Luomai Granules by HPLC

    [Objective] To establish the methods for determining the contents of Mogroside V,Liquiritin and Glycyrrhizic acid in Luomai granule by HPLC.[Methods] The chromatographic conditions were as follows: the chromatographic column was ZORBAX SB-C18( 4. 6 mm× 250 mm,5 μm); the mobile phases were acetonitrile-water and 0. 1% phosphoric acid-acetonitrile respectively,and the flow rate was 0. 80 ml / min; the column temperature was 30 ℃,and the detection wavelength were 203 nm and 237 nm separately. [Results] The experiment proved that there was good linear relation when injection amount were in a range from 0. 95 μg to 4. 75 μg for Mogroside V( r = 0. 999 6),from0. 152 μg to 1. 064 μg for liquiritin( r = 0. 999 8) and from 0. 074 μg to 0. 518 μg from glycyrrhizic acid( r = 0. 999 8). The average recovery rate was 100. 06%,99. 96%,100. 08% respectively and the RSD were 1. 01%,1. 03%,1. 02%( n = 6) respectively.[Conclusions] This method is simple,accurate and highly reproducible,so it can be used to determine the contents of Mogroside V,Liquiritin and glycyrrhizic acid in Luomai granules,and can be applied to control the quality of Luomai granules.

  • Analysis on Volatile oils from Curcuma longa Linn in Hainan Province in Different Harvest Period by GC-MS

    [Objective] To analyze volatile oils of Curcuma longa Linn in Hainan Province in different harvest period. [Methods] Taking Curcuma longa Linn in Hainan Province as experimental materials,this paper studied changes in volatile oils of Curcuma longa Linn in different harvest period by gas chromatography / mass spectrometry( GC-MS). [Results] At rhizome expanding stage( October 17),the content of volatile oils of Curcuma longa Linn is the highest,up to 80 m L / kg. Later,the content slightly declines,till the withering period( March 4),the content of volatile oils is 67 m L / kg. In different harvest period,chemical composition of volatile oils of Curcuma longa Linn is basically the same,but the relative content is different. In four harvest times,36,36,34,and 34 kinds of chemical components were identified separately.Main components were Ar-Turmerone,α-Tumerone,Zingiberenol,β-Bisabolene,2-epi-α-Cedrene,Ar-Curcumene,and other sesquiterpene compounds. Ar-Turmerone was the highest in different harvest times,accounting for 49. 259%,33. 794%,33. 610% and 44. 829%,respectively. [Conclusion] In different harvest period,chemical composition of volatile oils of Curcuma longa Linn is basically the same,but the relative content is different. Ar-Turmerone was the highest in different harvest times.

  • Regulation Effects of Coenzyme Q10 on the ox-LDL-induced Proliferation of Rat Vascular Smooth Muscle Cells

    [Objective] To study regulation effects of Coenzyme Q10( Co Q 10) on the oxidized low-density lipoprotein( ox-LDL)-induced proliferation of rat vascular smooth muscle cells( VSMCs). [Methods]After incubation of ox-LDL( 0,10,20,30,40,50,60,70,80 mg /L) and Co Q 10( 0,10,20,30,40,50,60,70,80 μmol / L) on logarithmic growth phase vascular smooth muscle cells for 24 hours,respectively,CCK-8 method was used to detect the proliferation index of VSMCs,and screen ox-LDL model concentration and cytotoxicity of Co Q 10.Cell were divided into the control group,model group,Co Q10 low concentration,medium concentration,and high concentration. After drug action for 24 hours,except the normal group,a1 l groups were given culture solution containing screened concentration of ox-LDL to reproduce proliferation model of VSMCs. Mitochondrial protein was extracted,and protein levels of Cytochrome C( Cyto-C) and apoptosis inducing factor( AIF) were detected by Western blotting technique. [Results]Compared with the control group,the model group had higher cell survival rate and protein levels of Cyto-C and AIF,and the difference showed statistical significance( p < 0. 05); compared with the model group,Co Q 10 low,medium,and high concentration groups had low cell survival rate,higher protein levels of Cyto-C and AIF,and the difference showed statistical significance( p < 0. 05). [Conclusion]Co Q 10 can inhibit abnormal proliferation VSMCs through promoting protein expression of Cyto-C and AIF.

  • Optimization of Extraction Process of Tannins in SANGUISORBAE RADIX

    [Objective] To find out the optimal extraction process for tannins in SANGUISORBAE RADIX. [Methods] Content of tannins and extract yielding rate of SANGUISORBAE RADIX were taken as evaluation indicators,Gallic acid was taken as reference solution. Medicinal material grain size,solvent consumption,permeability speed,seepage factor,and diameter-to-height ratio were optimized,to obtain optimal experimental conditions for extraction of tannins in SANGUISORBAE RADIX. [Results] After screening with no. 1 sieve,powder of SANGUISORBAE RADIX was put into percolation vat( the diameter-to-height ratio was 1∶ 1),6 times of 70% ethanol was added,finally obtained the optimal extraction process: 5 m L / min / kg,the average paste forming rate was 30. 5%,and extraction rate of tannins was 15. 5%.[Conclusions]This process is reasonable and feasible,highly reproducible. Using this process,it is able to obtain high extraction rate. Thus,it is expected to provide theoretical basis for preparing semi-finished products of tannins of SANGUISORBAE RADIX.

  • Extraction Process and Content Determination of Total Flavonoids in Acanthopanax giraldii Harms

    [Objective] To establish Ultraviolet-Visible Spectrophotometry for determining total flavonoids in Acanthopanax girldii Harms.[Methods]Single factor experiment was carried out to find out the effects of extracted solvent,extraction method,and solid to liquid ratio on content of total flavonoids in A. girldii Harms,select the optimal condition,and determine total flavonoids in different parts of A. girldii Harms. [Results] The optimal condition of extraction process obtained from single factor experiment was reflux extracting for 2 hours at 1: 20 solid to liquid ratio using 60% methanol solution. [Conclusions] Ultraviolet-Visible Spectrophotometry was simple,accurate and mature for determining content of total flavonoids in A. girldii Harms. This method was high reproducibility and the experimental data were highly reliable. Thus,it could be provided data support for further development of different samples of A. girldii Harms.

  • Determination of Total Alkaloids in Solanum nigrum by Acid Dye Colorimetry

    [Objective] To study the determination method for total alkaloids in Solanum nigrum. [Methods]Acid dye colorimetry was used to determine total alkaloids in Solanum nigrum. [Results] Color conditions for acid dye colorimetry are 3. 5 m L p H 3. 6 buffer solution,2. 0m L bromocresol green solution,3 times of extraction using 10 m L chloroform,and settle the solution for 30 min. Total alkaloids in Solanum nigrum showed good linear relationship in the range of 5- 30 μg / m L,the average recovery rate was 99. 7%,and RSD = 1. 4%( n = 6).[Conclusions]Acid dye colorimetry is simple in operation,has high sensitivity,high accuracy and reproducibility,and can be used for determining total alkaloids in Solanum nigrum.

  • A Study on Cutting Propagation of Rabdosia ternifolia (D. Don) Hara

    [Objective] To examine the effect of different concentrations of IBA and NAA as well as different cutting time on cutting rooting of Rabdosia ternifolia( D. Don) Hara branch. [Methods] With the branches in different parts of Rabdosia ternifolia( D. Don) Hara as experimental material,different concentrations of growth hormones IBA and NAA were used for treatment. [Results]200 mg. L- 1IBA and NAA had better treatment effect than other concentrations of IBA and NAA,and the variance analysis results show that there was a significant difference in rooting rate between the treatments; IBA treatment had the greatest effect on rooting rate,the effect on root length and root number was not obvious,but it could significantly increase root length and root number. [Conclusions]During May cutting,the branches soaked with 200 mg·L- 1IBA and 200 mg·L- 1NAA for 12 h had the best rooting effect,and the rooting rate reached as high as 75. 8%.

  • Optimization of Extraction Technology for Insoluble Dietary Fiber from Seed-coat of Mung Bean by Response Surface Methodology

    [Objectives] To study influences of Na OH mass fraction,alkaline extraction time,alkaline extraction temperature,alkaline extraction liquid-to-material ratio,acid extraction temperature,acid extraction time,and acid extraction liquid-to-material ratio on extraction yielding rate and purity of insoluble dietary fiber( IDF),and to optimize the extraction process conditions. [Methods] Single factor experiment and response surface methodology( RSM) were applied. [Results]Results indicate that the optimal process conditions for acid and alkaline combined extraction of IDF from seed-coat of mung bean are Na OH mass fraction 1. 97%,alkaline extraction time 89 minutes,alkaline extraction temperature 61 ℃,alkaline extraction liquid-to-material ratio 15∶ 1( m L / g),acid extraction temperature 71 ℃,acid extraction time120 minutes,and acid extraction liquid-to-material ratio 15∶ 1( m L / g). [Conclusions] In this optimal process condition,the IDF yielding rate is up to 64. 23%,and IDF purity reaches 90. 12%,so this process is feasible. This study is expected to produce certain industrial value.Seed-coat of mung bean is main by-product of mung bean processing. Optimization of extraction technology for seed-coat of mung bean can increase added value of mung bean and accordingly raise economic benefits of mung bean processing industry.
