  • Research Progress of Chemical Composition and Biological Activity of Genus Dendropanax

    Most Dendropanax(Araliaceae) plants are used as traditional folk herbal medicines and have functions of dispelling wind and eliminating dampness,invigorating blood circulation and relieving swelling. They have been collected in Chinese Materia Medica,The Dictionary of Medicinal Plant,The National Assembly of Chinese Herbal Medicine,She-national Medicine,and etc. Through comprehensively consulting CNKI,Wanfang,CqVip,Web of science,Sci Finder,and Pubmed databases,it made literatures review,analysis,and summary of chemical composition and biological activity of Dendropanax plants. Chemical composition of Dendropanax plants mainly includes polyacetylenes,phenylpropanoids,flavonoids,and triterpenes. They have biological activity of anti-inflammation,anti-complement,anti-tumor,and anti-oxidation.Besides,Dendropanax has close relation with Panax,so it belongs to medicinal plant with high potential development value. However,at present,chemical composition and biological activity were studied on D. dentiger,D. proteus,D. arboreus,D. morbifera,D. trifidus,and one uncertain species D. cf querceti,and there are few literatures about clinical efficacy. Therefore,it is urgent to study chemical composition and biological activity of Dendropanax plants,and elaborate material foundation of medical efficacy and acting mechanism in combination with clinical efficacy.

Chemical Composition of Natural Medicine

  • Analysis of Volatile Components from ACANTHOPANACIS CORTEX,PERIPLOCAE CORTEX and LYCII CORTEX by HS-SPME-GC-MS

    [Objectives] To analyze volatile components in ACANTHOPANACIS CORTEX,PERIPLOCAE CORTEX and LYCII CORTEX.[Methods]Headspace solid phase micro-extraction( HS-SPME) combined with gas chromatography and mass spectrometry technique( GCMS) were used to analyze volatile components in ACANTHOPANACIS CORTEX,PERIPLOCAE CORTEX and LYCII CORTEX. [Results]The results indicated that a total of 63 different volatile components were identified in the extracts,among which 34 compounds were detected,accounting for 89. 65% of the total volatile components; 50 kinds of volatile components were detected in PERIPLOCAE CORTEX,among which 19 kinds were identified,accounting for 93. 93% of the total volatile components; 37 kinds of volatile components were detected in LYCII CORTEX,among which 22 kinds were identified,accounting for 87. 37% of the total volatile components. [Conclusions]There are great differences between species and content of volatile constituents in ACANTHOPANACIS CORTEX,PERIPLOCAE CORTEX and LYCII CORTEX. Three kinds of herbs contain unique volatile components. It is expected to provide certain reference for rapid identification.

  • Determination of Chemical Composition of Volatile Oils from Cnidium monnieri(L.) Cuss. in Hebei by GC-MS

    [Objective] To determine chemical composition of volatile oils from Cnidium monnieri(L.) Cuss. in Hebei Province by GC-MS.[Methods]Volatile oils were extracted from Cnidium monnieri(L.) Cuss. by steam distillation method; chemical composition of volatile oils from Cnidium monnieri(L.) Cuss. was determined by GC-MS; relative percentage content of chemical composition was determined by normalization method. [Results]42 major components were identified,including alpha-pinene(18. 19%),limonene(17. 88%),borneyl acetate(11.05%),and camphene(7.81%),etc. [Conclusion]This method is simple and fast and highly efficient. This study can be applied for determining chemical composition of volatile oils of traditional Chinese medicines,and is expected to provide basis for scientific quality control of Cnidium monnieri(L.) Cuss.

  • Determination of Hyperoside in Acanthopanax setchuenensis and A. leucorrhigus var. fulvescens by RP-HPLC

    [Objective] Hyperoside in Acanthopanax setchuenensis Harms ex Diels and A. leucorrhigus var. fulvescens Harms & Rehd was identified qualitatively by LC-MS,and a RP-HPLC method was established to determine the content of hyperoside in their different medicinal parts. [Methods] The analysis was carried out at 30 ℃ on a Kromasil C18 column eluted with a mobile phase consisted of methanol(A) and 0.5% phosphate in water(B). The detection wavelength was 358 nm,and the flow rate was 1.0 mL/min. [Results] The calibration curve was linear in the concentration range of 2.50-25.00 μg / mL,the regression equation was Y = 4 × 10~6 X + 1. 327 × 10~4( r = 0. 999 9). The average recovery was 100. 25%,with RSD of 0. 96%( n = 9). [Conclusions] This method was simple,accurate and replicable,and it was useful for the study of the quality control of A. setchuenensis Harms ex Diels and A. leucorrhigus var. fulvescens Harms & Rehd.

Pharmacological and Toxicological Analyses

  • Studies on Anti-inflammatory Effects and Mechanism of Phellodendri Chinensis Cortex

    [Objective] To observe the anti-inflammatory effects of alcohol extract of Phellodendri Chinensis Corte and explore its mechanism.[Method]ICR mice were divided into 5 groups randomly,namely the model group,dexameathone group( 0. 01 g/kg),alcohol extraction of Phellodendri Chinensis Corte groups( 9. 0,4. 5 and 3. 0 g / kg). Each drug was administrated to the mice orally once a day for 15 days. After30 min of the last administration,its anti-inflammatory effect was observed by the method of acetic acid-induced permeability accentuation. SD rats were divided into 5 groups randomly,namely the model group,dexameathone group( 0. 01 g / kg),alcohol extraction of Phellodendri Chinensis Corte groups( 9. 0,4. 5 and 3. 0 g / kg). Each drug was administrated to the rats orally once a day for 15 days. After 30 min of the last administration,the rat model of carrageenan-induced acute pleurisy was also selected to observe the anti-inflamatory effect. The anti-inflammatory mechanism was investigated by detecting the contents of prostaglandins( PGE_2),tumor necrosis factor-alpha( TNF-α),and interleukin-1β( IL-1β) in the exudates,and the contents of TNF-α,IL-1β,and malondialdehyde( MDA) in the lung tissue were also detected.[Result]Compared with the model group,the alcohol extract of Phellodendri Chinensis Corte groups( 9. 0 and 4. 5 g/kg) had significant inhibitory effects on the acetic acid-induced permeability accentuation in the mice,while the alcohol extract of Phellodendri Chinensis Corte groups( 9. 0 and 4. 5g / kg) significantly reduced the levels of PGE_2,TNF-α and IL-1β in the exudates( P <0. 05),while the high-dose alcohol extract of Phellodendri Chinensis Corte group( 9. 0 g / kg) also reduced the contents of TNF-α,IL-1β and MDA in the lung tissue of the carrageenan treated rats( P < 0. 01),and the moderate-dose alcohol extract of Phellodendri Chinensis Corte group( 4. 5 g / kg) decreased the content of IL-1β in the lung tissue( P < 0. 01). [Conclusion]The alcohol extraction of Phellodendri Chinensis Corte possesses significant anti-inflammatory effects,which can be related to the inhibition of generation of the inflammatory mediators and its antioxidation.

  • Biological Activities of Several Kinds of Traditional Chinese Medicine Extracts in Chuxiong

    [Objective] To study antibacterial activity of several kinds of traditional Chinese medicinal plants common in Chuxiong,Yunnan Province. [Methods]4 common traditional medicinal plants in Chuxiong were selected. Different solvents were used for reflux extraction,and antibacterial activity was measured for extracts. [Results]Extracts of Teucrium viscidum Bl.,Impatiens noli-tangere L.,Leucas ciliata Benth,and Phryma leptostachya L. subsp. asiatica( Hara) Kitamura extracted by 50% alcohol heating reflux for three times have the best antibacterial effect. They have antibacterial activity in various degrees to 4 kinds of bacteria and 2 kinds of fungi. [Conclusions]These four kinds of medicinal plants have wide source,convenient extraction,and excellent antibacterial effect,so it is expected to provide theoretical basis for further development.

  • Protective Effect and Acting Mechanism of Total Flavonoids of POLYGONI PERFOLIATI HERBA on Acute Alcohol-Induced Liver Injury in Mice

    [Objective]To study protective effect and acting mechanism of total flavonoids of POLYGONI PERFOLIATI HERBA on acute alcohol-induced liver injury in mice. [Methods]60 mice were randomly divided into normal group,model group,Bifendate positive group(150 mg / kg),and high( 600 mg / kg),medium( 400 mg / kg) and low( 200 mg / kg) dosage groups of PPTF,with 10 mice for each group. Except the normal group,fill the stomach with 12 mL/kg 56% liquor one time every morning for the rest groups and fill the corresponding dosage liquor in the afternoon,and fill the same volume of distilled water for normal group and model group. Continuously implement these operations for 14 days. After the end of the last time stomach filling,take fasting for 24 hours,anaesthetize them and take blood,and collect livers; measure the activity or content of alanine transaminase( ALT),aspartate transaminase( AST),malondialdehyde( MDA),and superoxide dismutase( SOD) in serum by biochemical method; measure the content of interleukin-4( IL-4),interleukin-6( IL-6),tumor necrosis factor-α( TNF-α) in liver tissue by enzyme-linked immunosorbent assay( ELISA) method; HE dyeing,and observe pathological changes of liver tissue by a microscope. [Results] Compared with the model group,all dosage groups of total flavonoids of POLYGONI PERFOLIATI HERBA can effectively reduce activity and content of ALT,AST and MDA in serum of mice,can improve SOD activity,and can reduce IL-6 and TNF-α in liver tissue,and increase IL-4 content. [Conclusions] Total flavonoids of POLYGONI PERFOLIATI HERBA have protective effect on acute alcohol-induced liver injury in mice and the mechanism is possibly related to anti-lipid peroxidation and inhibition of inflammation reaction in liver.

  • Quality Evaluation of Polygerm Paris polyphylla var. yunnanensis(Polygerm Varieties) in Yunnan Province Based on Moisture,Ashes and Extracts

    The moisture,total ashes,acid insoluble ashes and alcohol extracts of Paris polyphylla var. yunnanensis( polygerm varieties) and Paris polyphylla var. yunnanensis collected from different places in Yunnan Province were determined according to the procedures recorded in the General Rule 0832,2302 and 2201 of 2015 Chinese Pharmacopoeia respectively. The results showed that the moisture of the 22 samples was not higher than the limit of PARIDIS RHIZOMA of 12% specified in 2015 Chinese Pharmacopoeia; the total ashes of 21 samples were not higher than the limit of PARIDIS RHIZOMA of 6% specified in 2015 Chinese Pharmacopoeia,but that of the Haochang sample was 6. 85%; the acid insoluble ashes of 22 samples were not higher than the limit of PARIDIS RHIZOMA of 3% specified in 2015 Chinese Pharmacopoeia; the hot dip alcohol soluble extracts of 22 samples were not lower than 8%; The moisture,ashes and acid insoluble ashes were up to the limit of PARIDIS RHIZOMA specified in 2015 Chinese Pharmacopoeia in the 11 samples of Paris polyphylla var. yunnanensis( polygerm varieties) collected from Tengchong City area; the acid-insoluble ashes were lower than others,and the alcohol extracts were also higher and these four indicators was smaller changes compared with others. In conclusion,Paris polyphylla var. yunnanensis(polygerm varieties) of Tengchong City area was smaller changes in these four indicators and stable quality,so it was worthy of extension and in-depth researches.

  • Genetic Polymorphism and Puerarin Differences of Puoraria lobata(Willd) Ohwi in Three Gorges Reservoir Region

    [Objective]To conduct Inter- simple Sequence Repeat( ISSR) amplification and show the genetic polymorphism. [Methods]In natural growth conditions,Puoraria lobata( Willd) Ohwi was selected from 6 different growth places in Three Gorges Reservoir Region A comparative study was carried out on puerarin content,total flavonoids,Daidzin,and isoflavoues Aglycone. [Results] Results indicate that there are significant differences in puerarin content,total flavonoids,daidzin,and isoflavoues Aglycone among Puoraria lobata( Willd) Ohwi from 6 growth places. [Conclusion] There are significant differences in puerarin content,total flavonoids,daidzin,and isoflavoues Aglycone among Puoraria lobata( Willd) Ohwi from 6 growth places.

  • Construction of a Multi-functional T-DNA Insertion Vector

    [Objective] Plant mutants have been widely used in gene function analysis. T-DNA insertion is a promising approach for mutant library construction. This study aimed to construct a multifunctional T-DNA insertion vector for generating mutant libraries of Salvia miltiorrhiza Bunge.[Methods] By using pBI121 as a starting vector,an intron sequence from S. miltiorrhiza was introduced. Four enhancers were directionally connected by digestion with Ban I restriction endonuclease. GUS gene promoter was removed and GUS gene was moved forward to construct the recombinant vector pBI-INTRON-GUS-4E.[Results] The constructed recombinant vector pBI-INTRON-GUS-4E harbors an intron sequence from S.miltiorrhiza,four enhancers,GUS gene and NPTII gene,which exhibits multiple functions such as activation tagging,gene(promoter) trapping and insertional mutagenesis.[Conclusions]The successfully constructed multifunctional T-DNA insertion vector provided the basis for generating mutant libraries of S.miltiorrhiza and other plant species.

  • Optimization of SFE-CO_2 Extraction Process of Total Vol atile Oil from Citrisarcodactylisfructus by Box-Behnken Design-Response Surface Methodology

    [Objective] To optimize the SFE-CO_2 extraction process of total volatile oil from Citrisarcodactylisfructus. [Methods] The influences of extraction temperature,extraction pressure,CO_2 flow,extraction time,acquisition pressure and acquisition temperature on the extraction rate of total volatile oil from Citrisarcodactylisfructus were investigated firstly. Moreover,four significant factors such as extraction temperature,extraction pressure,CO_2 flow and extraction time were selected to optimize the SFE-CO_2 extraction process by Box-Behnken Design-response surface methodology( BBD-RSM). [Results] The highest extraction rate of total volatile oil from Citrisarcodactylisfructus 1. 892% could be obtained when extraction pressure was 57 MPa; extraction temperature was 67 ℃; CO_2 flow was 22.0 L/h; extraction time was 4 h.It was verified by the further experiments and the relative standard deviation( RSD) between them was 2. 32%. [Conclusions] This experiment provides a reasonable method of optimizing the SFE-CO_2 extraction process of total volatile oil from Citrisarcodactylisfructus,which can promote the further development and utilization of volatile oil from Citrisarcodactylisfructus.

  • Optimization of Ultrasound-assisted Extraction of Polyphenols from Periploca forrestii Schltr and Its Anti-allergic Activities

    [Objective] To optimize the extraction of total polyphenols from Periploca forrestii Schltr. [Methods]Response surface methodology was applied. A three-level,three-variable Box-Behnken Design was employed to obtain the best possible combination of extraction solvent( ethanol concentration 40%-80%),extraction time( 10 min- 50 min) and ratio of solution to raw material(10∶ 1- 15∶ 1,v/w) for maximum extraction yield oftotal polyphenols. Besides,anti-allergic capacity of P. forrestii polyphenols was evaluated by initiation ratio of hyaluronidase decomposition of sodium hyaluronate. [Results]The results showed that optimized extraction conditions wereethanol concentration57.33%,extraction time 33. 33 min and ratio of solvent to raw material 16. 11∶ 1. Under these conditions,the experimental yield was 6. 03 ± 0.10%,which is well in close agreement with the value( 6. 1%) predicted by RSM model. [Conclusion]Pharmacological test showed that P.forrestii polyphenols have excellent anti-allergic activity.

  • Determination of Absorbed Component in Rat Plasma in Periploca forrestti Schltr. by High Performance Liquid Chromatography

    [Objective] To determine absorbed component in rat plasma in Periploca forrestti Schltr. [Methods] Ultra-performance liquid chromatography-mass spectrometry analysis technology was used. And L_9(3~4) orthogonal test and chlorogenic acid content determination method was applied. Range analysis and ANOVA analysis were carried out. [Results] Periploca forrestti Schltr. was an active component which could be absorbed into the rat blood. Through the range analysis and ANOVA analysis,the optimal extract conditions were obtained. Then,Periploca forrestti Schltr. was analyzed and chlorogenic acid was successfully separated by C18 reversed phase column( 4. 6 mm × 250 mm,5 μm) at wavelength of 327 nm and temperature of 30℃. With the mobile phase consisted of acetonitrile( A) and water( 0. 5% formic acid,B) in the volume ratio of 9∶ 91( v / v) and flow rate of 1. 0 m L / min,peaks showed good shape and separation. The linear equation of chlorogenic acid was y = 223. 8900 x- 0. 225 8,R~2= 0. 999 7 in the range of 8 μg / m L to 192 μg / m and the average recovery was 98. 569% which indicated the method was fine. [Conclusion]The discovery and the established analysis method of chlorogenic acid in Periploca forrestti Schltr. are expected to control the quality of this medicine better.
