  • PCA-HCA Chemical Pattern Recognition of CORTEX MOUTAN

    [Objective] To establish a chemical pattern recognition for CORTEX MOUTAN. [Methods] CORTEX MOUTAN in 20 different production areas were detected by RP-HPLC to obtain the fingerprint. Chemical pattern recognition was carried out by Principal Component Analysis( PCA) and Hierachical Cluster Analysis( HCA). [Results] The 20 batches of CORTEX MOUTAN were divided into two categories: one was Anhui production area,and the other was Shandong and Henan production areas. According to the chemical component contents,md-019,md-020,md-017,md-018 and md-004 had relatively high quality. [Conclusions] Pattern recognition technique on chemical level was established for the quality evaluation of CORTEX MOUTAN,which provided references for the classification and quality evaluation of CORTEX MOUTAN.

  • Content Detection of Total Flavonoids in Different Parts of Rumex acetosa L.

    [Objective] To establish the detection method of total flavonoids contents in Rumex acetosa L. by UV spectrophotometry,and to detect the total flavonoids contents in different parts of R. acetosa. [Methods] Effects of different factors on the extraction rate of total flavonoids from R. acetosa were researched by single factor test,which were extraction solvent,extraction method,extraction time,solid-liquid ratio and volume fraction of methanol. The optimal extraction technology of total flavonoids from R. acetosa was screened. Contents of total flavonoids were detected in different parts of R. acetosa.[Results] The optimal extraction technology of total flavonoids from R. acetosa was as follows: 70% methanol,1∶ 40 solid-liquid ratio,and hot reflux extraction for 60 min. Contents of total flavonoids in R. acetosa were the maximum in flowers and fruits.[Conclusions]The detection method for total flavonoids in R. acetosa was simple and accurate. Except the roots of R. acetosa,flowers and fruits also had high contents of total flavonoids,which could be used as the medicinal parts for the extraction of total flavonoids.

  • Detection of 21 Elements in CORDYCEPS Grown in Xinghai County by ICP-OES / ICP-MS

    A total of 487 CORDYCEPS were collected from Xinghai County,Hainan Tibetan Autonomous Prefecture,Qinghai Province. After washing and freeze-drying of CORDYCEPS,the coated soil was removed from CORDYCEPS. Freeze-dried powders of CORDYCEPS was digested by HNO3 and H2O2. Contents of 21 elements in CORDYCEPS were detected by ICP-MS( Inductively coupled plasma mass spectrometry)and ICP-OES( Inductively Coupled Plasma Optical Emission).

  • Volatile Components Analysis of FOLIUM ARTEMISIA ARGYI Grown in Different Areas Based on HS-SPME / GC-MS

    [Objective] To establish rapid analysis method for the volatile components in FOLIUM ARTEMISIA ARGYI,and to research the contents and distribution characteristics of volatile components in FOLIUM ARTEMISIA ARGYI grown in different areas. [Methods]Volatile components in FOLIUM ARTEMISIA ARGYI were analyzed by HS-SPME / GC-MS. Relative content of each component was calculated by peak area normalization method. Data were analyzed by principal component analysis and cluster analysis. [Results]A total of 84 chemical components were preliminarily identified,which were mainly the ketones,alkenes and alcohols. There were certain differences in the volatile components of FOLIUM ARTEMISIA ARGYI. Principal component analysis and cluster analysis could effectively isolate the FOLIUM ARTEMISIA ARGYI gown in different areas. [Conclusions]This method was stable and reliable,which could be used for the rapid analysis of the volatile components in FOLIUM ARTEMISIA ARGYI,and provided certain scientific references for the quality evaluation of the volatile components in FOLIUM ARTEMISIA ARGYI.

  • Simultaneous Determination of Scopoletin and Scoparone in HERBA ARTEMISIAE ANNUAE by Three-dimensional Fluorescence Coupled with Alternating Trilinear Decomposition Method

    [Objective] To simultaneously detect scopoletin and scoparone contents in HERBA ARTEMISIAE ANNUAE by three-dimensional fluorescence coupled with alternating trilinear decomposition method. [Methods] The contents and recovery rates of scopoletin and scoparone were detected in HERBA ARTEMISIAE ANNUAE under unknown disturbances by using three-dimensional fluorescence coupled with alternating trilinear decomposition method. Component number was estimated by core consistency diagnostic( CORCONDIA) test. [Results] As for standard curve method,scopoletin and scoparone contents were 0. 080 4% and 0. 024 8% in HERBA ARTEMISIAE ANNUAE,respectively.As for second-order standard addition method,scopoletin and scoparone contents were 0. 082 7% and 0. 023 4%,respectively; and their recovery rates were 97. 9% and 102. 9%,respectively. The resolved spectra and real spectra of scopoletin and scoparone matched quite well with each other. [Conclusions] This method was simple and effective,and solved the problem of analyze interested components under complex background.

  • Optimization of Extraction Technology of the Active Components from PORIA

    [Objective] To optimize the extraction technology of the active components from PORIA. [Methods] With pachymaran and pachymic acid contents as the investigation indexes,factors and levels were screened based on single factor test. Extraction technology of pachymaran and pachymic acid was investigated based on orthogonal test. [Results] The optimal extracting technology for pachymic acid was as follows: 85% ethanol with six times of the water volume was used for the extraction of pachymic acid; the extraction was carried out under55 ℃ for 1 h. At the same time,Ten times of water volume was used for the extraction of pachymaran for three times,with 2 h for each extraction. [Conclusions]This method was rational,stable and feasible,and provided theoretical foundation for the further industrialization of PORIA.

  • Optimization of the Extraction Technology of Lentinan from Lentinus edodes( berk. ) sing by Response Surface Method

    [Objective] To optimize the extraction technology of lentinan from Lentinus edodes( berk.) sing by response surface method.[Methods]With the fruiting body of L. edodes as the research materials,the extraction time,extraction temperature and solid-liquid ratio were optimized during the extraction of lentinan based on single factor test. Regression equation with the extraction rate of lentinan as the target function was obtained by Design-expert software. [Results]Based regression analysis,the optimal technology was 3. 8 h extraction time,92 ℃extraction temperature and 1∶ 43 solid-liquid ratio. Under this optimal condition,the mean value of the extraction rate of lentinan was 7. 865%in verification test. [Conclusions] The research result was close to the predicted value( 7. 893%),showing that this regression model had good fitting degree.

  • Pharmacodynamics of Paclitaxel Long-circulating Thermo-sensitive Liposome in Mice Bearing Lewis Lung Carcinoma Cells

    [Objective] To investigate the tumor inhibition effects of paclitaxel long-circulating thermo-sensitive liposome( PLTL) on mice bearing Lewis lung carcinoma cells. [Methods] A tumor-bearing mice model was established and divided into five groups randomly,which were control group,heating group,paclitaxel( PTX) group,paclitaxel thermo-sensitive liposome( PTL) group and PLTL( long-circulating thermo-sensitive liposome) group. Local hyperthermia was adopted; the survival status of mice was observed; volume and weight of tumor were detected; HE staining of tumor section was carried out; morphological changes of tumor cells were observed; and cell apoptosis was detected by flow cytometry. [Results]Tumor inhibition rates of PTX,PTL and PLTL groups were 48. 87%,57. 22% and 78. 87%,respectively. And the cell apoptosis rates were( 42. 7 ± 3. 8) %,( 54. 6 ± 2. 9) % and( 69. 7 ± 5. 0) %,respectively. [Conclusions] Compared with PTX and PTL,PLTL had evident thermo-sensitive characteristics,and increased the anticancer effect of paclitaxel remarkably when combined with local hyperthermia.

  • Effects of Volatile Components of Modified Wendan Decoction on Improving the Behavior of Autistic Rats

    [Objective] To research the effects of volatile components of Modified Wendan Decoction on improving the behavior of autistic rats. [Methods]Pregnant rats were randomly divided into two groups. On 12. 5 d after pregnancy,intraperitoneal injection of 600 mg / kg VPA was carried out so as to establish autistic rats model. In control group,the same volume of 0. 85% normal saline( NS) was intraperitonealy injected. The auxanology,behavior and morphology identification of rats were carried out. A total of 60 autistic rats and 20 rats in NS group were randomly collected on the 35 thday after pregnancy. And autistic rats were randomly divided into distilled water group( DW),Modified Wendan Decoction group( VPA-1) and Modified Wendan Decoction and volatile oil group( VPA-2). There were 20 rats in each group. The volatile components of Modified Wendan Decoction were analyzed by steam distillation method( SD) and GC-MS. [Results] VPA-1 group improved the behavior of autistic rats in certain degrees,but the effects were poorer than VPA-2 group. A total of 71 volatile components were detected.Among them,α-asarone,D-limonene and eugenol had relatively high content,accounting for 99. 89% of the total volatile components. [Results] The volatile oils in Modified Wendan Decoction enhanced the therapeutic effects of autism. Literatures showed that α-asarone,D-limonene,eugenol and other volatile components adjusted the central nervous system,improved cerebral function,had the functions of anticonvulsion,anti-epileptic and diazepam,and improved the symptoms of autism. This research provided methodological basis for the analysis of volatile components in Modified Wendan Decoction,and provided references for the medicinal efficacy research on curing autism.

  • The Anti-inflammatory Effects and Mechanism of the Stems and Leaves of Platycodon grandiflorum

    [Objective] To observe the anti-inflammatory effects of the stems and leaves of Platycodon grandiflorum( Jacq.) A. DC.,and to discuss its anti-inflammatory mechanism. [Methods] ICR mice were randomly divided into model control group,dexamethasone group( 0. 01 g / kg),ethanol extracts groups of P. grandiflorum stems and leaves( 9. 0,4. 5,3. 0 g / kg). Intragastrical administration was carried out once every day with continuous 15 days. On 30 min after final administration,anti-inflammatory effects were researched by observing the increase of blood capillary permeability in mice abdominal cavity induced by acetic acid. SD rats were randomly divided into model control group,dexamethasone group( 0. 01 g / kg),ethanol extracts groups of P. grandiflorum stems and leaves( 9. 0,4. 5,3. 0 g / kg). Intragastrical administration was carried out once every day with continuous 15 days. On 30 min after final administration,anti-inflammatory effects were researched by observing the pleurisy of rats induced by carrageenan. prostaglandins( PGE2),tumor necrosis factor-alpha( TNF-α) and interleukin-1β( IL-1β) contents were detected in exudates,as well as the TNF-α,IL-1β and MDA contents in lung tissues. [Results]Compared with model control group,ethanol extracts groups of P. grandiflorum stems and leaves( 9. 0,4. 5 g / kg) could significantly restrict the increase of blood capillary permeability in mice abdominal cavity induced by acetic acid. Ethanol extracts groups of P. grandiflorum stems and leaves( 9. 0,4. 5g / kg) restricted the increase of PGE2,TNF-α and IL-1β contents in exudates( p <0. 05). 9. 0 g/kg ethanol extracts group of P. grandiflorum stems and leaves restricted the increase of TNF-α,IL-1β and MDA contents in lung tissues( p < 0. 01); and 4. 5 g / kg ethanol extracts group restricted the increase of IL-1β( p < 0. 01). [Conclusions] Ethanol extracts from P. grandiflorum stems and leaves had anti-inflammatory effects; and the anti-inflammatory mechanism might be related to the antioxidation and the production interference of inflammatory mediator.

  • Protective Effects of Lycium ruthenicum Murr on Irradiated Mice

    [Objective] To research the protective effects of Lycium ruthenicum Murr on irradiated mice. [Methods] Except normal control group,mice were radiated by X-Rays with the radiation dosage being 5 Gy. The RBC,WBC,PLT and HGB of mice were detected. The thymus and spleen indices were calculated. The antioxidase activity,DNA,caspase-3 and caspase-6 contents were detected. [Results] L. ruthenicum water extracts significantly enhanced the hemogram of irradiated mice,increased the SOD activity,improved DNA content,reduced the CAT,GSH-PX and T-AOC activities,and lowered the caspase-3 and caspase-6 contents. Viscera index showed no statistical significance.[Conclusions]L. ruthenicum water extracts had certain protective effects on irradiated mice.

  • Effects of Volatile Oils of RADIX CURCUMAE on the Activity and Secretion of Hepatic Stellate Cells

    [Objective] To observe the effects of volatile oils of RADIX CURCUMAE on the activity and secretion of hepatic stellate cells.[Methods]HSC-LX2 of hepatic fibrosis was incubated by the volatile oils of RADIX CURCUMAE for 48 h. The proliferation inhibition rate of cells was detected by Methylthiazol Tetraztlium( MTT) Colorimetry; DNA content and cell apoptosis at different cell cycles were detected by flow cytometry( PI staining). The impacts on secretion functions of collagens typeⅠand Ⅲ,MMP-1 and MMP-13 were observed by immunohistochemistry. [Results] Volatile oils of RADIX CURCUMAE at low,middle and high dosage could all restrict the proliferation of HSC-LX2 cells. The inhibitory rates on 48 h were 15. 4%,49. 1% and 58. 3%,respectively. The IC50 on 48 h was 45 μmol / L. HSC-LX2 cells were blocked to enter the DNA synthesis and proliferation cycle; and the early and late apoptosis of HSC-LX2 cells was induced. Percentages of total cell numbers at S stage + G2 / M stage were 70. 1%,63. 6% and 50. 2% on 48 h,respectively. The apoptosis rates were 13. 9%,18. 9% and23. 5%,respectively. The volatile oils at different concentrations could restrict the HSC-LX2 cells to secrete collagens typeⅠand Ⅲ,and enhance the secretion of MMP-1 and MMP-13. [Conclusions]Volatile oils of RADIX CURCUMAE restricted the proliferation of HSC-LX2 cells,blocked the cells to enter the DNA synthesis and proliferation cycle,induced the cell apoptosis,reduced the secretion of fibrosis protein in HSC-LX2 cells,enhanced the degradation of fibrosis protein,and interfered the hepatic fibrosis. All these showed a dose-dependant manner.And the abilities mentioned above in middle-and high-dosage groups were stronger than those in colchicine group and Compound Biejiaruangan Tablet group.

  • Effects of Lianmei Oral Liquid on Blood Index,Hormone Secretion and Antioxidase of Damp Heat and Diarrhea Piglet Model

    [Objective] To research the effects of Lianmei oral liquid on blood index,hormone secretion and antioxidase of damp heat and diarrhea piglet model. [Methods]A total of 60 Rongchang pigs were randomly divided into six groups. Under the simulated hot and humid environment,damp heat syndrome was established. Piglets were given Lianmei oral liquid by gavage,so as to detect the haematological index,biochemical index,hormone content and antioxidase activity of piglets. The effects on nutrition metabolism,hormone secretion and antioxidase were researched in damp heat and diarrhea piglet model. [Results] High- and middle-dosage Lianmei oral liquids could prevent damp heat and diarrhea of piglets with the healing rate being higher than 80%,enhanced the hemoglobin content,erythrocyte count,white blood cells count,lymphocytes count and monocytes count in damp heat and diarrhea piglet model,improved the contents of total protein,globulin,urea nitrogen,glucose,total cholesterol and triglyceride,increased the activities of glutamic-pyruvic transaminase,glutamic oxalacetic transaminase and alkaline phosphatase,and reduced the urea nitrogen and triglyceride contents,showing significant( P < 0. 05) differences or extremely significant( P < 0. 01) differences with those in model group. Lianmei oral liquid enhanced T3,T4 and GH contents,and reduced the GC secretion level,having significant differences with those in model group( P < 0. 05). Moreover,Lianmei oral liquid enhanced the T-SOD and GSHPx activities,and reduced the MDA content,CK and LDH activities,showing extremely significant differences( P < 0. 01) with those in model group. [Conclusions] Lianmei oral liquid adjusted the nutrient metabolism of damp heat and diarrhea piglet model,enhanced T3,T4 and GH contents,improved the endocrine hormone level of piglets,enhanced the T-SOD and GSH-Px activities,and strengthened the body's antioxidant function.
