  • Extraction of Polysaccharides from Black Soybean by Enzyme Hydrolysis Method

    [Objective]To select the optimal extraction technology for the extraction of polysaccharides from black soybean by enzyme hydrolysis method. [Methods] The optimal enzyme was selected among cellulase,pectinase and papain. Effects of extraction temperature,extraction time,pH value and enzyme concentration were investigated. With the extraction rate of polysaccharides in black soybean as the evaluation index,the extraction parameters were optimized by asterion optimization design. [Results]Among the papain,cellulase and pectinase,cellulase showed the maximum extraction rate. The optimal extraction condition was as follows: pH 6. 0,0. 03 mg / mL enzyme concentration,50 ℃ extraction tempeature and 120 min extraction time. [Conclusions]Under the optimal condition,the extraction rate of polysaccharides from black soybean was 0. 321 4%,which had 99. 41% degree of closeness to the theoretical value. Factors affecting the extraction rate of polysaccharides from black soybean were in the order of pH value > extraction time > extraction tempeature.

  • Extraction Technology of Total Flavonoids from the Aerial Roots of Ficus microcarpa by Orthogonal Test

    [Objective]To optimize the extraction technology of total flavonoids from the aerial roots of Ficus microcarpa L. f. [Methods] With the content of total flavonoids as the detection index,the solid-liquid ratio,extraction time and extraction times were taken as the factors. The extraction technology of total flavonoids from the aerial roots of F. microcarpa was optimized based on single factor test and orthogonal test. [Results]The optimal extraction technology of total flavonoids from the aerial roots of F. microcarpa was as follows: 1∶12 solid-liquid ratio, 2 h extraction time and 3 extraction times. Under this condition,the average extraction content of total flavonoids from the aerial roots of F. microcarpa was 13. 78 mg / g. [Conclusions] This extraction technology was stable and was suitable for the extraction of total flavonoids from the aerial roots of F. microcarpa.

  • Optimization the Method of Microwave Assisted Extraction Technology of Acid in Star Anise by Orthogonal Test

    [Objective]To explore the method of microwave assisted extraction technology of shikimic acid in star anise. [Methods] Using single factor experiment with orthogonal experiment method,to optimize the extraction conditions for studying the effect of different factors on the shikimic acid extraction rate. [Results] The best technology of microwave extract anise shikimic acid is material liquid ratio of 1 ∶ 20( g/mL),under the condition of temperature of 65 ℃ for 10 min,power 400 W,the anise shikimic acid content is 37. 45 g/mg. [Conclusions]the method of microwave assisted extraction technology is provided with advantages as simple operation,high extraction efficiency,repeatability is good and is worthy application.

  • Study on the Content of Indirubin in the Leaves of Baphicacanthus cusia (Nees) Bremek. in Different Illumination Intensities

    [Objective]This research aims to study the influence of different illumination intensities on the content of indirubin in the leaves of Baphicacanthus cusia( Nees) Bremek. and to provide a scientific evidence for carrying out standardized planting of Baphicacanthus cusia( Nees) Bremek.[Methods]Three groups of plants grown in different illumination intensities were set up,which were plants grown in shade, plants grown in half shade,and plants grown in sunshine. The growth status of plants was observed after plants were fixed in March. Leaves of Baphicacanthus cusia were picked up from May to next March. The content of indirubin in leaves of Baphicacanthus cusia was detected through high pressure liquid chromatograms( HPLC). [Results]the content of indirubin in leaves of Baphicacanthus cusia from three groups of plants grown in different illumination intensities had basically similar variation tendency during their growth cycles except comparatively lower content of indirubin in Baphicacanthus cusia leaves picked up in several specific months.[Conclusions]Although three illumination intensities are all suitable for the plantation of Baphicacanthus cusia,full shade and half shade were far more appropriate after comprehensively considering the influence of illumination on the content of indirubin in Baphicacanthus cusia leaves,growth of plants and output.

  • Research on the Sclerotium Yield of Poria cocos by Different Elicitation Region and Harvest Time

    [Objective]The influence of elicitation region and harvest time to sclerotium yield during the cultivation of Poria cocos is to be explored. [Methods] The research is to be carried out in Luotian- the major production area of Poria cocos in Hubei Province. All related factors is to be arranged. The production of sclerotia will be counted and the analysis of its variance is going to be conducted. [Results]The production of sclerotia in one part of the clog with elicited strain is 2. 5 times of that of sclerotia in the other part of the clog without elicited strain. In the end of October and November,these sclerotia are to be harvested. The average production of the first method is 0. 512 kg and that of the second one is 0. 568 kg. The analysis of variance is that F = 2. 337 6,less than F0. 05. This indicates that there are no obvious differences between the two types of treatment. [Conclusions] When transplanted into the part of the clog without elicited strain,the sclerotia are going to be affected by the climate factors more often than usual. As a result,the uncertainty of production will be improved. After becoming mature, the color of sclerotia is dark brown. The weight of sclerotia remains steady,so that harvest time can be arranged from the end of October.

  • Optimization of Submerged Fermentation Conditions for Intracellular Polysaccharide of Pleurotus tuber-regium

    [Objective]A submerged fermentation medium of Pleurotus tuber-regium was developed for efficient production of polysaccharide and biomass. [Methods] By employing single factor experiment and orthogonal design,the effects of carbohydrate and nitrogen sources and cations on the Pleurotus tuber-regium fermentation were investigated with the intracellular and extracellular polysaccharides as indices. [Results]Glucose was found to be the optimal carbohydrate source,favorable for cell growth,metabolite production of extracellular polysaccharide( EPS) and intracellular polysaccharide( IPS),the yield of polysaccharides per unit biomass. Yeast extract as nitrogen sources was suitable for the cell growth,mycelial biomass and the yield of extracellular polysaccharide. The type of cations source also affects polysaccharide production,and EDTA performed best in promoting cell growth and metabolites accumulation. Under the optimized conditions,the production of biomass,intracellular polysaccharide,extracellular polysaccharide,the yield of total polysaccharide and dried cell biomass was 3. 34 g / L,1. 92 g / L, 80. 34 mg / g,8. 13% and 23. 62 g / L,respectively. [Conclusions]The optimized conditions for submerged fermentation of Pleurotus tuber-regium provided scientific basis for the cultivation of Pleurotus tuber-regium.

  • Effect of MORINDAE OFFICINALIS Extract on the Behavior and Zymology of Senile Dementia Model Rats

    [Objective]To study the effect of MORINDAE OFFICINALIS extract on the behavior and zymology of senile dementia rat model. [Methods]Senile dementia rat model was prepared by intraperitoneal injection of D-galactose( 120 mg/kg) and sodium nitrite( 90 mg/kg) for 30 d. 60 rats were divided into 6 groups,normal control group,model group,nimodipine group,high-dosage group of aqueous extract of MORINDAE OFFICINALIS,medium-dosage group of aqueous extract of MORINDAE OFFICINALIS and low-dosage of aqueous extract of MORINDAE OFFICINALIS. The learning and memory ability was measured by rat step-down test. The activities of SOD,MDA and MAO-B were determined. The expression of MAO-B mRNA was determined by RT-PCR. [Results]After intraperitoneal injection of MORINDAE OFFICINALIS extract,the learning and memory abilities of model rats increased. The biochemical criterion of SOD,MDA and MAO-B was improved. The expression level of MAO-B mRNA decreased significantly. [Conclusions] Extract of MORINDAE OFFICINALIS could protect acute aging model rats at certain degree.

  • Effect of Extracting Solution of Euphorbia fischeriana Steud. on the Proliferation and Apoptosis of K562 Cell Lines

    [Objective]To observe the effect that extracting solution of radix euphorbiae lantu has on the proliferation and apoptosis of K562 cell lines. [Methods] The growth inhibition ratio of the extracting solution of Euphorbia fischeriana Steud. on K562 cell line was determined by MTT. Apoptosis and its morphology were observed by Hoechst fluorescence staining. Changes of cell membrane potential and cell cycle were detected by FCM after treated with extracting solution of Euphorbia fischeriana Steud. [Results]The extracting solution of Euphorbia fischeriana Steud. had significant inhibiting effect on the growth of K562 cell line. Observed by fluorescence microscope,we found that the extracting solution of Euphorbia fischeriana Steud. could change the morphology of K562 cell. The mitochondrial transmembrane potential( △ψm) lowered and the number of cells in G0/ G1phase increased significantly which were detected by FCM. [Conclusions] The extracting solution of Euphorbia fischeriana Steud. could promote the apoptosis of K562 cell line and inhibit its growth by changing the cell cycle.

  • Experimental Study on the Antitussive,Apophlegmatic and Antiasthmatic Effect of Extract of Jianhua

    [Objective]To explore the antitussive,apophlegmatic and antiasthmatic effect of extract of Jianhua. [Methods]The antitussive, apophlegmatic and antiasthmatic effect of extract of Jianhua was studied by animal models. Control group,low-dosage group of extract of Jianhua,medium-dosage group of extract of Jianhua,high-dosage group of extract of Jianhua and positive medication group were included in the experiment. [Results] Compared with control group,low,medium and high-dosage of aqueous extract and alcohol extract of Jianhua could significantly inhibit cough induced by stronger ammonia water( P < 0. 05,P < 0. 01); medium and high-dosage of aqueous extract and alcohol extract of Jianhua could significantly increase phenol red excretion in the respiratory tract of mice( P < 0. 05,P < 0. 01); high-dosage of aqueous extract and alcohol extract of Jianhua could significantly prolong the incubation of asthma in guinea pigs induced by citric acid( P < 0. 05). [Conclusions]The extract of Jianhua had significant effect of relieving a cough,eliminating phlegm and relieving asthma.

  • Study on the Anti-inflammatory and Analgesic Effects of 12 Guangdong Native Heat-clearing Herbs

    [Objective]To explore the anti-inflammatory and analgesic effects of 12 Guangdong Native Heat-clearing Herbs with identical dose( 16 g/kg) and to provide reliable experiment basis for the clinical development and application of anti-inflammatory drugs. [Methods]To explore the anti-inflammatory and analgesic effects of 12 Guangdong Native Heat-clearing Herbs by acetic acid writhing,hot plate method,ear swelling caused by xylene in mice and celiac capillary permeation in mice induced by acetic acid. [Results]After intragastric administration of drugs( 16 g / kg) in mice for 7 days,the inhibiting effects of Radix Helicteris,Thinleaf Adina Fruit,Nervilia fordii,Radix Ilicis Asprellae and Trikeraia hookeri on ear swelling caused by xylene in mice were significant and the inhibiting effects of which on the increase of capillary permeability in mice induced by acetic acid were also significant( P < 0. 05). Radix Eupatorii Chinensis,Pronephrium triphyllum and Bidens biternata could inhibit ear swelling caused by xylene in mice significantly and Evodia lepta Merr,ovate leaf Holly Bark and Rubus parvifolius could inhibit the increase of capillary permeability in mice significantly( P < 0. 05). The effects of 12 Guangdong Native Heat-clearing Herbs on thymus and spleen were not significant( P > 0. 05); Trikeraia hookeri,Radix Ilicis Asprellae,ovate leaf Holly Bark,Radix Helicteris, Nervilia fordii and Herba Polygoni Chinensis could evidently reduce the times of writhing response in mice induced by acetic acid and increase the threshold of pain in mice induced by thermal stimulus; Evodia lepta Merr and Pronephrium triphyllum could evidently reduce the times of writhing response in mice induced by acetic acid,ovate leaf Holly Bark could significantly prolong the latent time of writhing response in mice induced by acetic acid( P < 0. 05) and Bidens biternata could significantly increase the threshold of pain in mice induced by thermal stimulus. [Conclusions]Radix Helicteris,Thinleaf Adina Fruit,Nervilia fordii,Herba Polygoni Chinensis,Radix Ilicis Asprellae,Trikeraia hookeri, Radix Eupatorii Chinensis,Pronephrium triphyllum,Bidens biternata,Evodia lepta Merr,ovate leaf Holly Bark and Rubus parvifolius had anti-inflammatory action; Radix Helicteris,Nervilia fordii,Herba Polygoni Chinensis,Radix Ilicis Asprellae,Trikeraia hookeri,Pronephrium triphyllum,Bidens biternata,Evodia lepta Merr and ovate leaf Holly Bark,etc. had analgesic effect.

  • Extraction of Polysaccharides from Trametes robiniophila Murr and Their Antioxidant Properties

    [Objective]To evaluate the antioxidant properties of polysaccharides from Trametes robiniophila Murr. [Methods]The polysaccharides of Trametes robiniophila Murr were separately extracted by cold caustic soda method,hot alkali method,conventional method,high temperature-ultrahigh pressure method,then analyzed via DPPH for evaluating their antioxidant properties in different groups. [Results]The deproteinization-treated grade I polysaccharides extracted by high temperature-ultrahigh pressure graded ethanol precipitation method yielded the highest content of polysaccharides( 41. 32%),and the grade II polysaccharides showed the highest antioxidant activities( 54. 73%). [Conclusions]Polysaccharides from Trametes robiniophila Murr are an effectively natural antioxidant that is endowed with high ability to scavenge free radicals.

  • Determination of Heavy Metals and Trace Elements from Rhizoma chuanxiong in Eleven Habitats by Microwave Digestion-ICP-OES

    [Objective]To measure the heavy metals and trace elements in 11 Rhizoma chuanxiong samples collected from 5 representative producing areas and perform correlation analysis. [Methods]The contents of the heavy metals including Hg,Cd,Pb,As,Cu,Zn,Se,Mn and Fe and the trace elements in the Rhizoma chuanxiong samples tested were measured by inductively coupled plasma optical emission spectrometer( ICP-OES),then the correlation between heavy metals,trace elements and producing region was analyzed by partial least squares using software simca- P +. [Results] The regression equations for each element detected by ICP-OES all showed a good linear relationship, with the correlation coefficient between 0. 999 7- 1,and recovery of 90. 02%- 108. 02%. [Conclusions]The microwave digestion-ICP-OES established in this study is simple,efficient and accurate,and can be used for the determination of heavy metals and trace elements in Rhizoma chuanxiong. The contents of heavy metals and trace elements are to some extent correlated with producing regions,in detail,the content of Fe, Mn,Zn and Cu showed an ascending trend in traditional Rhizoma chuanxiong producing regions. To conclude,it is suggested to periodically monitor the soil condition in traditional Rhizoma chuanxiong producing regions,and the pollution of heavy metals caused by climate.

Chemical Composition of Natural Medicine

  • HPLC Fingerprint of Pyrrosia calvata (Bak.) Ching

    [Objective]To establish the fingerprint of Pyrrosia calvata( Bak.) Ching for evaluating its quality by HPLC. [Methods] The HPLC analysis was carried out on SinoChrom ODS-BP( 5 μm,4. 6 mm × 250 mm) column with mobile phase of acetonitrile- 0. 2% phosphoric acid solution( gradient elution) at the flow of 1. 0 mL / min,and the detection wavelength was 205 nm. [Results]The established fingerprint of P. calvata was used to determine 10 batches of sample from different sources. 12 common peaks were confirmed as the character fingerprints through similarity analysis on the fingerprint of the P. calvata. [Conclusions]The method had good precision,stability and reproducibility, providing a scientific base for controlling the quality of P. calvata by establishing fingerprint of the P. calvata.

  • Contents of Forsythin and Forsythiaside A in Different Parts of Forsythia suspensa and Its Medicinal Value

    [Objective]To compare the contents of forsythin and forsythiaside A in different parts of Forsythia suspensa( Thunb.) Vahl.,and to provide scientific references for the development and utilization of F. suspensa resources. [Methods] Contents of forsythin in the fruits, leaves,branches and fruit stems of F. suspensa were detected by HPLC. Chromatographic column was Luna5uC18( 2) 100A( 250 mm ×4.6 mm, 5 μm). As for forsythin,the mobile phase was acetonitrile- water( 25∶ 75,V / V); detection wavelength was 277 nm; column temperature was room temperature and flow rate was 1. 0 mL / min. As for forsythiaside A,the mobile phase was acetonitrile- 0. 4% glacial acetic acid solution( 15∶85,V/V); detection wavelength was 330 nm; column temperature was room temperature and flow rate was 1.0 mL/min. [Results]Forsythin had good linear relationship within 3. 33- 13. 32 μg / mL( R2= 0. 999 7); the average recovery rate was 99. 30% and the RSD was 1. 4%. Forsythiaside A had good linear relationship within 4. 17- 6. 69 μg / mL( R2= 0. 999 7); the average recovery rate was 98. 26% and the RSD was 1. 8%. [Conclusions]Forsythin content was the maximum in leaves,followed with fruits and branches; and its content was the minimum in fruit stems. Forsythiaside A content was also the maximum in leaves,followed with fruits and fruit stems; and branches had the minimum content.

  • Optimization of the Ultrasonic Extraction Technology of Pigments from the Leaves of Coix lacryma-jobi

    [Objective]To research the extraction technology of pigments from the leaves of Coix lacryma-jobi L. by ultrasonic wave,and to provide theoretical basis for the utilization of C. lacryma-jobi resources. [Methods]The optimal extraction technology was obtained based on single factor test and orthogonal test. [Results] The optimal extraction technology for pigments from C. lacryma-jobi leaves was as follows: 1∶20 solid-liquid ratio,120 W extraction power and 9 min extraction time. [Conclusions] Raw materials and the reagent were low-cost and easy to get; the extraction method was simple,repeatable and time saving.

  • Determination of Rutin and Quercetin Content in Yantai Bupleurum by HPLC

    [Objective]To determine the content of rutin and quercetin in the whole plant of Yantai Bupleurum by HPLC. [Methods] An Agilent C18column( 250 mm × 4. 6 mm,5 μm) was applied with a mobile phase of acetonitrile- 0. 1% phosphoric acid solution,gradient elution was selected with the flow rate of 1. 0 mL / min,the UV detector was set at 360 nm. [Results]The average recovery rates of rutin and quercetin were 100. 47% and 99. 50%,the RSD were 1. 34% and 0. 41%. The average content of rutin and quercetin in the whole plant of Yantai Bupleurum reached 83. 00 and 102. 68 μg / g. [Conclusions]The method is fast,accurate and reliable. It is suitable for content determination of rutin and quercetin in the whole plant of Yantai Bupleurum.

  • Contents of Inorganic Elements in the Stems of Wild Plumbago zeylanica Linn. and Their Seasonal Dynamic Changes

    [Objective]To determine the contents of inorganic elements in the stems of wild Plumbago zeylanica Linn. in different harvest periods( from January to December),and to discuss the effects of harvest period on the content of inorganic elements. [Methods]Microwave digestion and Inductively Coupled Plasma Optical Emission Spectrometer( ICP-OES) were both used to determine the inorganic elements in the stems of wild P. zeylanica. The detection results were researched by correlation analysis,principal component analysis and cluster analysis. [Results]The contents of inorganic elements in the stems of P. zeylanica varied in different harvest periods. Al,Ca,Fe,Sn,Ti,As,V, Zn,Mn,Mg and Ni were the eigenelements of P. zeylanica stems. Cluster analysis showed that the 12 samples of P. zeylanica stems could be divided into two categories: September was the first major category,and the others were the second category. [Conclusions]The detection results offered useful data for the synergic relationship among elements,and provided theoretical basis for the better utilization and development of P. zeylanica resources.
