• Inhibition Effects of Total Saponins from Panax japonicas Cultured Cells on Adjuvant Arthritis Rat

    [Objective]To investigate treatment effects of total Saponins from Panax japonicas cultured cells on the rat adjuvant inflammation.[Methods]Constructing the pathological model using male AA rats,by proinflammatory before and after oral administration,to study the total saponins of Panax japonicus in rat primary and secondary adjuvant arthritis prevention and treatment effect.[Results]The total Saponins isolated from Panax japonicas cultured cells can prevent and significantly reduce the AA rat adjuvant arthritis of primary lesions.Panax japonicus saponins can also well treat and alleviate adjuvant arthritis secondary lesions.The effective dose of total saponins of Panax japonicus was 35-140 mg/kg.[Conclusion]Total Saponins extracted from Panax japonicas cultured cells have good anti-inflammatory effects as nature Panax japonicus extract.

  • Shikonin Induced K562 Cell Apoptosis through Oxidative Stress

    [Objective]To study the role of shikonin in inducing K562 cell apoptosis and its inhibition mechanism.[Methods]Detected shikonin's anti-proliferative effect on K562 cell with thiazolyl blue staining.Observed morphology of apoptosis with Hoechst 33258 and acridine orange(AO)/ethidium bromide(EB)staining.With reverse transcription-polymerase chain reaction(RT-PCR)to detect apoptosis-related genes:the mRNA(Messenger RNA)expression changes of Bax(B-cell lymphoma gene 2 associated X protein),Bid(BH3 interacting domain death agonist),Bcl-xL(B-cell lymphoma gene xL),PUMAB(p53 up-regulated modulator of apoptosis),Bak(B-cell lymphoma gene2 homologous antagonist),Bcl-w(B-cell lymphoma-w),Bad(B-cell lymphoma gene 2 associated death promoter).Analyzed the changes of intracellular ROS(reactive oxygen species)with DCFHDA(oxalic acid-2',7'-dichlorofluorescein)staining.Analyzed content changes of intracellular GSH(reduced glutathione hormone)with CMFDA 5-chloromethylfluorescein diacetate staining.[Results]①0.2-1.6 mg/L shikonin had dose-dependent inhibition effect on K562 cell proliferation;②Cells treated with 0.2-0.5 mg/L shikonin had obvious apoptosis characteristics;③In groups treated with 0.2-0.5 mg/L shikonin,the mRNA expression of pro-apoptotic proteins(Bid,Bad,Bak,PUMA,Bax)increased significantly and the mRNA expression of anti-apoptotic proteins(Bcl-w,Bcl-xL)decreased significantly.④In 0.2-0.5mg/L shikonin treatment group,content of intracellular reactive oxygen increased and content of reduced glutathione decreased.[Conclusions]Shikonin had strong inhibition effect on K562 cell proliferation and could induce its apoptosis.The process was accompanied by the changes of intracellular redox state.K562 cell apoptosis induced by shikonin was related with intracellular oxidative stress.

  • Anti-inflammatory Effects of the Stems and Leaves of Platycodon randiflorum on Animal Models of Acute Inflammation

    [Objective]To study anti-inflammatory effects of water extract of the stems and leaves of Platycodon grandiflorum.[Methods]Observed anti-inflammatory effects of water extract of the stems and leaves of Platycodon grandiflorum using 4 kinds of animal models of acute inflammation which were consisted of rats'footpad swelling induced by egg white,ear swelling caused by xylene in mice,celiac capillary permeation in mice induced by 1%glacial acetic acid and the analgesia experiments of hot plate method.[Results]Water extract of the stems and leaves of Platycodon grandiflorum had a certain degree inhibition effect on ear swelling caused by xylene in mice,rats'footpad swelling induced by egg white and celiac capillary permeation in mice induced by acute inflammation.Compared with model group,low-dosage and highdosage groups of water extract of the stems and leaves of Platycodon grandiflorum were of significant differences(P<0.05 or P<0.01).[Conclusions]Water extract of the stems and leaves of Platycodon grandiflorum had strong anti-inflammatory effects.

  • Protective Effect of Panax notoginseng Saponins on Experimental Liver Fibrosis in Rats and to Explore Its Possible Mechanisms

    [Objective]This article is written to observe the protective effect of Panax notoginseng saponins on experimental liver fibrosis in rats and to explore its possible mechanisms.[Methods]SD rats were divided at random into normal group,model group,Panax notoginseng saponins group(50,100 and 200 mg/kg)and colchicines group(Col,0.1 mg/kg).Rats in each group,except those in the normal group,were given subcutaneous injections of 50%CCl4twice per week in consecutive 18 weeks in order to abduct the liver fibrosis in rat models.From the 9th model building week on,the medicine-given groups were severally given intragastric administration of the corresponding drugs for testing while the normal and model groups were given equal amount of menstrua,once per day for continuous 10 weeks.After the experiment,the contents of hyaluronic acid(HA),laminin(LN),procollagen III N-terminal peptide(PIIINP),and Collagen IV(CIV)were detected through radio-immunoassay and the level of malondialdehyde(MDA)and superoxide dismutase(SOD)in liver homogenate through colorimetric method.Meanwhile,liver tissues from fixed position were fetched and the degree of liver fibrosis was observed through HE and Masson staining.The expressions of Bcl-2 and Bax protein in liver tissues were detected through immunohistochemical method.[Results]Comparing to the model group,Panax notoginseng saponins(100 and 200 mg/kg)have an impact not only on relieving liver fibrosis degree in rats,reducing HA,LN,PIIINP,CIV and MDA contents,raising SOD level in liver tissues,but also on inhibiting Bax expression and promoting Bcl-2 expression.[Conclusions]Panax notoginseng saponins have a certain influence on anti-fibrosis of rats'livers by an up-regulation of Bcl-2 expression and a down-regulation of Bax expression.It can be assumed that the inhibition of liver apoptosis is one of its possible mechanisms.

    [Objective]To study the analgesic and anti-inflammatory effects of the ethyl acetate extract of Cinnamomun longepaniculatum Folium.[Methods]Using the acetic acid writhing test and hot plate test to observe the analgesic effect of ethyl acetate extract of Cinnamomum longepaniculatum Folium on mice.By xylene induced mouse ear swelling model and acetic acid induced mouse capillary permeability increasing model to observe the anti-inflammatory effect of ethyl acetate extract of Cinnamomum longepaniculatum Folium.[Results]The ethyl acetate extract of Cinnamomun longepaniculatum Folium could significantly reduce the writhing times of mice induced by acetic acid with no significant analgesic effects on hot plate test model;could significantly inhibit abdomen capillary permeability increasing of mice induced by acetic acid,and significantly inhibit xylene induced mouse ear swelling in high dose.[Conclusions]The ethyl acetate extract of Cinnamomun longepaniculatum Folium has certain peripheral analgesic and anti-inflammatory effects.

  • Effect of Baicalin on Thermostable Nuclease Enzyme nuc Gene of Staphylococcus aureus and Its Activity

    [Objective]The effect of baicalin on Staphylococcus aureus thermostable nuclease enzyme nuc gene and its activity was analyzed.[Methods]The minimum inhibitory concentration of baicalin against Staphylococcus aureus was determined with the double dilution method,the impact of the drug on the activity of thermostable nuclease enzyme was determined with the method of Toluidine Blue DNAse Agar and the effect of baicalin on the expression of Staphylococcus aureus nuclease resistant bacteria nuc gene was detected with RT-PCR.[Results]The minimum inhibitory concentration of baicalin against Staphylococcus aureus was 0.156 mg/mL.The activity of Staphylococcus aureus thermostable nuclease enzyme could be completely inhibited the baicalin solution at the containing of 40.000-5.000 mg/mL and there was obvious inhibition to the activity of Staphylococcus aureus thermostable nuclease enzyme while the concentration of baicalin was diluted to 0.078mg/mL,which was significant difference with that of CK(P<0.01).The specific band of 867bp was successfully amplified with synthetic primers and its sequencing result was completely identical with the sequence of standard strain mu50.While the concentration of baicalin solution was 2.500 mg/mL the expression of Staphylococcus aureus thermostable nuclease enzyme nuc gene could be reduced.[Conclusions]The baicalin might inhibit the expression of Staphylococcus aureus thermostable nuclease enzyme nuc gene and reduce the activity of its heat-resisting.

  • Dosage-effect Relationship of Astragalus-Salvia's Effective Components in E-cadherin Expression of UUO Rat

    [Objective]By the observation of dosage-effect relationship of Astragalus and Salvia's effective components in E-cadherin Expression of UUO rat,an analysis of the effect on renal tubular epithelial cells was made and pathological changes of renal tubule interstitial fibrosis under different concentrations of Astragalus-Salvia drugs were observed and compared.[Methods]Forty SD rats were randomly classified into five groups:normal group,model group,Astragalus-Salvia drug granules compatibility(1∶1)low dosage group(hereinafter referred to as the low dosage group),middle dosage group and high dosage group.Double distilled water gavage was given to normal group and model group,rather 14 d therapy of relevant drug gavage to other groups to test serum creatinine(Scr),blood urea nitrogen(BUN)and 24 h urine protein content and to observe the renal pathology and collagen deposition with light microscope.The immunohistochemical method was used to detect E-cadherin changes in renal tissue.[Results]The renal function,pathological damage,collagen deposition and immunohistochemical results of different groups of kidney was the most obvious in module group and the least in high dosage group.[Conclusions]Astragalus-Salvia drugs have a certain protective effect on renal function of UUO rats and renal tubular function.Furthermore,with the dosage increased,the effect is more obvious,whose mechanism may be correlated with the gradual enhancement of E-cadherin expression;there exists dosage-effect relationship of Astragalus-Salvia's effective components in E-cadherin expression of UUO rats,with the most obvious effect of high dosage group.

  • Effect of Polygonum Perfoliatum On the Expression of HBsAg and HBeAg in HepG2.2.15 Cells

    [Objective]To study the effect of alcohol extraction of Polygonum Perfoliatum on the expression of HBsAg and HBeAg in HepG2.2.15cells.[Methods]The half toxic concentration(TC50)and the maximum non-toxic concentration(TC0)of Polygonum Perfoliatum extracts to HepG2.2.15 cells can be detected via methyl thiazolyl tetrazolium(MMT)method.With enzyme-linked immunosorbent assay(ELISA)method,the secretion level of HBsAg and HBeAg in cell supernates could be measured after a 72-hour action of drugs with four different concentrations on the HepG2.2.15 cells.[Results]Liquids with concentration of 0.4,0.2 and 0.1 mg/mL have an inhibitory effect on the secretion of HBsAg and HBeAg after being cultured for 72 h in the cell strain of HepG2.2.15 and there is a dose-response relationship between the concentration and the effect.Therapeutic indexes of the liquids were all greater than 1 but lesser than 2,which showed that alcohol extract of Polygonum Perfoliatum was an effective therapeutic drug of low toxicity.[Conclusions]Alcohol extraction of Polygonum Perfoliatum L.has an anti-HBV effect in vitro and certain toxicity as well.

  • Investigation and Utilization of Anti-cancer Medicinal Plant Resources in Tangshan

    Anti-cancer medicinal plant resources are rich and of a great variety in Tangshan.On the base of related articles,we authenticated medicinal plant specimens collected from various representative sample areas.By investigating resources of medicinal plants,we discovered that there are 20 families,31 genera,35 species of anti-cancer medicinal plants in Tangshan,there are 3 families,5 genera,5 species of monocotyledon,and there are 17 families,26 genera,30 species of dicotyledon.We suggested that more anti-cancer researches of medicinal plant resources in Tangshan should be performed,so that they can provide the basis for the development and utilization of anti-cancer drugs.

  • Effect of Soil pH on Cadmium Concentration Accumulated in Genuine Medicinal Plant Species RHIZOMA CHUANXIONG Produced in Sichuan Province

    [Objective]To investigate the accumulation characteristics of Cadmium(Cd)in RHIZOMA CHUANXIONG cultivated in different producing areas.[Methods]The soil sample and paired RHIZOMA CHUANXIONG materials was randomly collected from the main producing areas and used as experimental materials.The Soil Physi-chemical Detection Industry Standard issued by Ministry of Agriculture was used to analyze soil samples,and AAS was employed to determine the Cd content accumulated in RHIZOMA CHUANXIONG materials.Based on the physi-chemical characteristics determined above,the dynamic change of Cd content in RHIZOMA CHUANXIONG materials and of active state of Cd in typical soil samples.[Results]The results showed that there was no remarkable difference in pH value between fresh and dried soil samples;the soil accumulating higher Cd content in RHIZOMA CHUANXIONG materials had a relatively lower pH value,while that with lower Cd content had a relatively higher pH value.The proportion of active state Cd in soil with lower pH value was relatively higher.[Conclusions]The Cd content in RHIZOMA CHUANXIONG materials is negatively correlated with soil pH value at the significant level.

  • Effects of Different Fertilities on the Antioxidant Enzyme Activities and Chlorophyll Content of Radix pseudostellariae

    [Objective]To explore a suitable fertility level for the growth of Radix pseudostellariae.[Methods]Employing antioxidant enzyme activities and chlorophyll content as the indicators,an orthogonal experiment L9(33)was designed to analyze the field plantation and chemical properties of R.pseudostellariae by potted cultivation collected from the GAP production base in Shibing County of Guizhou Province.[Results]At the fertility of 3 g urea,7.5 g calcium superphosphate and 1 g potassium sulfate for each pot,R.pseudostellariae plants showed the minimum antioxidant enzyme activities and the maximum chlorophyll content.In addition,chlorophyll content,CAT and SOD activity reached the highest significant correlation under different level of potassium,while no statistical significance was detected in the correlation between antioxidant enzyme activities and chlorophyll content under other treatments.[Conclusions]Under lower fertilities,the antioxidant enzyme activities of R.pseudostellariae plants were relatively high,while chlorophyll content was lower.With the increase of fertility,the antioxidant enzyme activities descended gradually and the chlorophyll content showed an ascending trend,but excessive fertigation would affect the production of R.pseudostellariae.

  • Preliminary Research on the Inducing Effects of Wogonin on the Apoptosis of Lung Cancer Cells and Its Mechanism

    [Objective]To investigate the effects of wogonin on the proliferation and apoptosis of lung cancer cell lines A549 and explore its molecular mechanism.[Methods]MTT method was used to evaluate the effects of wogonin on the proliferation of A549;Hochest staining method and flow cytometer were used to evaluate the effects of wogonin on the apoptosis of cells;Western-blotting detector was used to evaluate the effects of wogonin on the apoptosis of related proteins.[Results]Wogonin can inhibit the growth of A549 cells,and its inhibitory rate of cell proliferation was dependent on the drug concentration and the reaction time.According to the results of morphological observation,wogonin can obviously lead to the apoptosis of lung cancer cells A549,and inhibit the expression of apoptosis-related proteins Bcl-2 and survivin,as well as improve the expression of Bax protein.[Conclusions]Wogonin can apparently induce the apoptosis of lung cancer cells A549,and its molecular mechanism of inducing apoptosis was likely to be associated with the down-regulation of Bcl-2 and the expression of survivin protein.

  • Optimizing Technical Parameters for the Extraction of Total Flavonoids in Different Hypecoum leptocarpum Hook.f.et Thoms

    [Objective]To find out the optimal extraction process for separating total flavonoids from Hypecoum leptocarpum Hook.f.et Thoms.and its application for determination.[Methods]Using total flavonoids as indicator,an orthogonal experiment was performed to optimize the extraction process by investigating the methanol concentration,material-liquid ratio and extraction time.[Results]The absorbance for total flavonoids was found peaked at 510 nm.The orthogonal experiment results indicated that the optimal extraction condition was A1B2C2,i.e.,methanol concentration of 50%,material-liquid ratio:1∶15(W/V,g/mL),ultrasonic extraction twice and 20 min for each extraction.Given these conditions,the total flavonoids contained in the Hypecoum leptocarpum Hook.f.et Thoms was about 48.63 mg/g.[Conclusions]The method is efficient,easy to operate,and yields accurate results.

  • Study on the Infrared Spectra of Stemona tuberosa Lour.from Different Producing Areas in Guangxi

    [Objective]To explore the infrared spectra characteristics of the medicine Stemona tuberosa Lour.and lay a theoretical foundation for its medicine quality assessment.[Methods]A test of Stemona tuberosa produced in 10 different places of Guangxi Province was conducted by means of infrared spectrometry,and the similarity of the samples were detected by comparing them with a standard spectrogram(this standard spectrogram was set to be 1),which was actually the average spectrogram of each sample.[Results]The infrared characteristic absorption peaks of Stemona tuberosa were acquired.It was shown from the results that the similarity of the 10 samples varied in a range of 0.925 2 to0.980 4,thus the threshold value was settled as 0.90.[Conclusions]This method has a good repeatability and stability.Since this method is of convenience,celerity and dexterity,it can be used as a qualitative testing method for Stemona tuberosa.

    As a main producing country of onion(Allium cepa L.),China accounts for 30%of total cultivation acreage and yield of onion in the world.However,the novel onion varieties are seldom released by Chinese breeders,most of them were the hybrids introduced from other countries,directly causing the higher production cost that reduces the economic benefit of farmers.Intensive study on the genetic diversity of onion resources would facilitate the breeding process of onion in China,improving both breeding efficiency and quality.Here we review the research advances in onion genetic diversity from origin and classification,morphological level,cellular level,biochemical level and molecular level,and prospect the breeding of elite novel onion varieties as well.

  • Infrared Fingerprint Analysis of Gentianae macrophyllae Radix Coupled with Sequential Analysis of Dual-indexes and Cluster Analysis

    [Objective]To find a method for rapid and effective identification of Gentiana macrophyllae habitats and varieties.[Methods]Using dual-index sequence analysis method(common peak ratio and variant peak ratio)and cluster analysis,infrared spectroscopy fingerprint of Gentiana macrophyllae of different habitats and varieties were analyzed.[Results]The identification results were consistent with the habitats and varieties of Gentiana macrophyllae;4 species could be divided into 3 categories,Gentiana crassicaulis and Gentiana straminea as one class,the others as another class.[Conclusions]Dual-index sequence analysis method and cluster analysis method combined with infrared spectroscopy fingerprint,can identify reliably Gentiana macrophylla samples of 2 or more different habitats and varieties.
