• Raising Seedlings from Wild Seeds of Disporum cantoniense (Lour.) Merr.by Cold Stratification at Different Time

    Objective To accelerate the artificial domestication of Disporum cantoniense (Lour.) Merr.wild variety.Methods Based on previous tests for different sowing time in 2006 and for different covers and different gibberellin in 2007,4 different stratification time were adopted to raise seedlings from D.cantoniense seeds in 2011,including A (90 d,starting from December 6,2010),B (60 d,starting from January 6,2011),C (30 d,starting from February 6,2011) and CK (0 d,no stratification).All seeds in treatment A,B,C and CK were planted in the soil on March 6,2011.Results Cold stratification could improve the emergence rate and emergence speed of seedlings,especially 90 days cold stratification,followed by 60 and 30 days.Conclusion Cold stratification could enhance the emergence rate of D.cantoniense wild seeds.

  • Study on the Biological Characteristics of Sophora flavescens Ait.Seed from Different Provenances

    The biological characteristics of Sophora flavescens Ait.seed from seven geographical provenance: Shanxi,Shaanxi,Gansu,Guizhou,Inner Mongolia,Liaoning and Henan were studied.The results showed that all indicators of Sophora flavescens Ait.seed from different provenances varied; there was a significant negative correlation between 1 000-grain weight and the germination rate.The vigor,the germination rate and the potentiality of the seed from Shanxi and Shaanxi were higher than those of other sources.

  • Investment of Traditional Chinese Medicine Resources and Their Utilization in Jinpenshan Forest Farm,Xinfeng County,Jiangxi Province

    Objective To conduct a survey of traditional Chinese medicine (TCM) resources in Jinpenshan Forest Farm,Xinfeng County,Jiangxi Province and a study of their sustainable utilization.Methods TCM resources of this region were preliminarily studied through filed survey,literature review,market survey and specimen identification.Result There are 132 species of medicinal plants belonging to 62 families in Jinpenshan Forest Farm,of which Compositae,Ranunculaceae and Rosaceae are the dominant families and genera of this region.Conclusion Based on the current situation of TCM resources,some suggestions are put forward on how to protect and utilize these medicinal plants in Jinpenshan Forest Farm.It is suggested that Jinpenshan Forest Farm should carry out basic research of medicinal plants,put base construction into practice,develop genuine medicinal materials,strengthen population conservation of medicinal plants,exploit unique health care medicine species,and develop scientific and technological industry of TCM modernization,all of which seeks to benefit the people.

  • The Virus Elimination Techniques of Lily

    Objective To research on the optimal approach for the production of virus-free bulbs of lily.Methods Researches on the shoot tip size of lily,heat treatment and ribavirin concentration were carried out.The elimination efficiencies of five lily verities to the viruses of LVX,CMV and LSV were researched by four different culture methods,which were the shoot tip culture,the shoot tip culture combined with heat treatment,the shoot tip culture combined with ribavirin,and the shoot tip culture combined with heat treatment and ribavirin.Results Considering the survival rate and virus-free rate,single shoot tip culture could hardly eliminate the viruses.And the method of shoot tip culture (0.5-0.8 mm) combined with heat treatment (changing temperature treatment) and ribavirin (10 mg/L) showed the optimal elimination efficiencies.Conclusions The optimal approach for the production of virus-free bulbs of lily was researched,which provided references for the further development and utilization of lily.

    Objective To carry out comprehensive evaluation on the soil fertility in cultivation base of Pithecellobium clypearia in Conghua of Guangdong Province.Methods Physico-chemical properties of soil samples at 0-20 and 20-40 cm layer at 4 slope directions were detected and analyzed.And comprehensive evaluation of soil fertility was carried out by improved Nemoro synthetical index method.Results pH value in soil was relatively low; organic matter content was lacked; total N,P and K contents were low; contents of available N and K were abundant; content of available P was relatively low; content of mineral element was slightly low.The contents of available N,available K and organic matter in west and east slopes were higher than those in north and south slopes.Conclusions Soil integrated fertility coefficients of all samples in P.clypearia base were lower than 0.9,showing that soil fertility was at poor level.

  • Effects of Calcium and Magnesium Deficiency on Growth of Polygonum multiflorum

    Objective To explore the effects of calcium and magnesium deficiency on the growth and development of annual Polygonum multiflorum.Method Using sand culture method,with the cuttings of Polygonum multiflorum as materials,the appearance,the content of chlorophyll,the quality of aboveand under-ground portion and their relationship were studied.Result After Polygonum multiflorum was treated with calcium and magnesium for one year,the color of its leaves became lighter and the chlorophyll content decreased,the above-ground portion was smaller,roots increased with less root tubers and more fibrous roots.There were three kinds of treatments: normal treatment,calcium deficiency treatment and magnesium deficiency treatment,the difference of root to shoot ratio among the three treatments was great and extremely significant,the root to shoot ratio of calcium deficiency treatment was the maximum,there was no obvious difference in the root proportion and drying rate.Conclusion In mineral element imbalance environment,the appearance,the quality of aboveand under-ground portion and the production of root tubers were significantly changed in annual Polygonum multiflorum,whether these changes have effects on the effective components of Polygonum multiflorum is unclear which needs further study.

  • Plumbagin Contents in the Above-ground Parts of Plumbago zeylanica L.at Different Harvest Periods

    Objective To analyze the plumbagin content in the above-ground parts of Plumbago zeylanica L.at different harvest periods.Methods The powder was soaked in 50 mL anhydrous methanol for 24 h.Then,ultrasonic extraction was carried out for 40 min,so as to obtain the extracting solution.Plumbagin contents in the branches of P.zeylanica at different harvest periods were detected by HPLC,so as to obtain the optimal collection period.Results The content of plumbagin in the branches of P.zeylanica enhanced from March to October,and reached the maximum at October (0.033%).Then,it decreased from October to the next February.The content of plumbagin was the minimum at February,which was only 0.009%.Conclusions Plumbagin content in the branches of P.zeylanica was the maximum in October,so the period from September to October was the optimal collection time.

  • Detection of Luteolin Content in CAULIS LONICERAEJAPONICAE by HPLC

    Objective To detect the content of luteolin in CAULIS LONICERAE JAPONICAE.Methods HPLC was adopted.Chromatographic column was Agilent ZORBAX SB-C18 (4.6 mm × 250 mm,5 μm); mobile phase was anhydrous methanol0.3% phosphoric acid solution (47∶53,V/V); flow rate was 1 mL/min; column temperature was 30 ℃; and detection wavelength was 350 nm.Results The linear range of luteolin was between 0.5 and 2.5 μg/mL.Correlation coefficient was R=0.999 6; average recovery rate was 100.71%,and RSD was 1.9%.The average content of luteolin in three batches of CAULIS LONICERAE JAPONICAE was 24.05 μg/g.Conclusions This method was simple,rapid,reliable and could be used for the quality control of CAULIS LONICERAE JAPONICAE.

  • Effects of FRUCTUS MUME on Curing Experimental Ulcerative Colitis in Mice

    Objective To discuss the effects of FRUCTUS MUME on SOD activity and MDA content in mice colon tissue with ulcerative colitis (UC).Methods Mice were given 3% DSS solution; UC model of mice was established.Mice in normal group and model group were given 0.33 mL normal saline by gavage; mice in low-,middleand high-dosage groups of FRUCTUS MUME were given the designed dosage.Administration was carried out once every day with continuous 10 d.During this period,occult blood test was carried out; and the fecal character was observed for DAI evaluation; morphological score of colon tissue was evaluated by both naked eye and microscope observation.The SOD activity and MDA content in mice colon tissue were detected.Results There was no gross blood stool in middleand high-dosage groups of FRUCTUS MUME,and loose stool could be observed.DAI in low-,middleand high-dosage groups reduced and had extremely significant differences with that in model group (P<0.01).In high-dosage group of FRUCTUS MUME,mucosal erosion was not serious with slight hyperaemia and edema.SOD activities in middleand high-dosage groups were both significantly higher than that in model group,having extremely significant differences (P<0.01).Middle-dosage group had significant differences with normal group (P<0.05); and high-dosage group showed no significant differences with normal group (P>0.05).MDA contents in middleand high-dosage groups were significantly lower than that in model group,having extremely significant differences (P<0.01).Conclusions Free radical had close relationship with UC in colic injury.According to the pathological process of UC,FRUCTUS MUME had significant curative effects on UC through reducing the MDA content and enhancing the SOD activity of lesion tissues.

  • Relationship between Main Compositions in Pine Needles of Pinus massoniana Lamb.and Harvest Periods

    Objective To investigate the effect of different harvest periods on main compositions in pine needles of Pinus massoniana Lamb.Methods Using volatile oils,caryophyllene,taxifolin and rutin as indicators,the pine needles of Pinus massoniana Lamb.sampled at different periods were compared for evaluating their medicinal values.In detail,volatile oils were extracted by vapor distill-extraction method,and caryophyllene was detected by GC-MS and the taxifolin and rutin by HPLC.Results The effective components in pine needles of Pinus massoniana Lamb.showed seasonal fluctuation.The highest content of volatile oils was 0.65% in samples collected in February; the content of caryophyllene reached 225.10 μg/g in samples collected in October; and the content of taxifolin and rutin respectively was as high as 35.41 and 13.41 μg/g in samples collected in June.Conclusion The effective components in pine needles of Pinus massoniana Lamb.vary among different harvest periods.Our results would be helpful for rational utilization and exploitation of new resources of Pinus massoniana Lamb.

  • Analysis of the Volatile Components of E.grandis × E.urophylla Leaves at Different Growth Years by GC-MS

    Objective To analyze the chemical components of volatile oils in E.grandis × E.urophylla leaves at different growth years,and to discuss the effects of growth years on the chemical components of volatile oils in E.grandis × E.urophylla leaves at different growth years.Methods The volatile components was extracted from the E.grandis × E.urophylla leaves by steam distillation method,which were planted in the years 2006,2007,2008,2009 and 2010.And their chemical components were identified by GC-MS.Results The yields of volatile oils in E.grandis × E.urophylla leaves at different growth years were between 0.55% and 1.35%; and the yields enhanced with the increase of growth years.A total of 63 chemical compounds were identified by GC-MS,accounting for 93.00%-97.07% of the total volatile oils.Among them,chemical compounds with high relative contents were 1,8-eucalyptol (55.72%-63.47%),α-terpineol (9.78%-14.15%),α-terpinyl acetate (4.96%-7.06%),α-pinene (0.99%-4.31%) and borneol (2.40%-4.89%).Conclusions There were certain differences in the relative contents of volatile components in E.grandis × E.urophylla leaves at different growth years.

  • The Co-toxicity of the Mixture of Cynanchum komarovii Al.and Sophora alopecuroides L.against Plutella xylostella L.

    Objective To investigate the common-toxicity of the mixture of Cynanchum komarovii Al.and Sophora alopecuroides L.against Plutella xylostella L.Method The dipping method was adopted to measure the activities of ethanol extracts from C.komarovii and S.alopecuroides and their mixture against Plutella xylostella L.,and the common-toxicity of the mixture was evaluated by the methods of Bliss and Sun,respectively.Result There is significant synergism between the ethanol extracts of C.komarovii and S.alopecuroides.The co-toxicity coefficient (CTC) is 248.30 under the dry powder mass ratio of 2∶ 1.The mixture exhibits significant contact toxicity against the 3rd instar larvae of P.xylostella with the mortality rate of 100% under the concentration of 20.00 mg/mL.The LC50 of the mixture is 2.074 7 mg/mL,which is much lower than that of C.komarovii (LC50=4.30 mg/mL) and S.alopecuroides (LC50=8.51 mg/mL).Conclusion There is synergism between C.komarovii and S.alopecuroides ethanol extracts.

  • Extraction Process of Total Flavonoids and Polysaccharides from Leaves of Diospyros kaki

    Objective To explore the best extraction conditions of total flavonoids and polysaccharides from Diospyros kaki.Method By using the single factor test and orthogonal experiment,the influence of extraction time,solid-liquid ratio,extraction solvent and extraction frequency on the extraction rate of total flavonoids and polysaccharides was studied,and the best extraction conditions were determined.Result The best extraction conditions for total flavonoids were 60% ethanol,2 h of extraction time,twice of extraction frequency,and 1∶30(g/mL) of solid-liquid ratio; while the best extraction conditions for polysaccharides were 60% ethanol,1 h of extraction time,one time of extraction frequency,and 1∶20(g/mL) of solid-liquid ratio.Conclusion The study provides references for improving the extraction process,reducing the energy consumption and realizing the industrialized production of flavonoids and polysaccharides,and lays a theoretical foundation for their future development and utilization.

  • Ultrasonic Extraction Process of Total Saponin in Codonpsis pilosula

    Objective To optimize the experiment conditions for ultrasonic extraction of total saponin from Codonpsis pilosula.Method The orthogonal test design was adopted to optimize the factors of the extraction process of total saponin in Codonpsis pilousla,and the colorimetric method was used to determine the content of total saponin.The new method was compared with the traditional extraction methods.Result The optimal ultrasonic extraction conditions were as follows: methanol was used as the extracting solvent,the ratio of solid to liquid was 1∶20 (W/V,g/mL) and ultrasonic extraction was conducted three times with 30 minutes each time.Under these conditions,the total saponin extracted from Codonpsis pilosula was 65.48 mg/g.Conclusion This method is simple with stable conditions,and it improves utilization of medical material,which can be used to extract total saponin from Codonpsis pilosula.

  • Determination of Total Flavonoids in Different Plant Parts of Five Species of HERBA MOGHANIAE in Different Grown Areas

    Objective To detect the content of total flavonoids in different plant parts of five species of HERBA MOGHANIAE in different grown areas; to evaluate the qualities of different species of medicinal materials in different grown areas; and to find out new medicinal resources and medicinal plant parts.Methods The content of total flavonoids was detected at 510 nm by UV-Vis spectrophotometry.Results Rutin showed good linear relationship between 9.17 and 64.19 μg/mL; the average recovery rate was 99.63% and RSD was 1.36%(n=9).Among the different species,contents of total flavonoids in the roots of Moghania strobilifera (Linn.)Ait.and M.latifolia Benth.were lower than those of M.philippinensis (Merr.et Rolfe)Li and M.macrophylla (Willd.) O.Kuntze.Total flavonoids content in the roots of M.glutiaosa (Prain)Y.T.Wei was approximated to those in the roots of M.philippinensis (Merr.et Rolfe)Li and M.macrophylla (Willd.) O.Kuntze.Total flavonoids contents in the leaves of different species were relatively high,but those in the stems were relatively low.As for the plants in different grown areas,M.macrophylla (Willd.) O.Kuntze purchased from Chengdu City of Sichuan Province (grown in Yunnan Province) had the maximum content; M.philippinensis (Merr.et Rolfe) Li grown in Yiling District of Yichang City of Hubei Province had the maximum content.Total flavonoids contents in M.strobilifera (Linn.)Ait.in different grown areas were all relatively low.Conclusions This method was simple,rapid and accurate,and could be used for the quality control of HERBA MOGHANIAE.There were great differences in the total flavonoids contents in HERBA MOGHANIAE in different grown areas,indicating that altitude,climate,soil condition,growth years and storage years had significant impacts on the total flavonoids content in HERBA MOGHANIAE.With respect to the total flavonoids,the effective and sustainable utilization of resources,it was suggested that the leaves and stems of different species of HERBA MOGHANIAE could be used as medicines together with roots; and the stems could also be used.M.glutiaosa (Prain)Y.T.Wei could also be used as a alternative medicinal resource.

  • Study on Extraction Process of Triterpene Acids from Rosemary

    Objective To explore the optimal extraction conditions of rosemary triterpene acid.Method Using cellulase method,by the single factor experiment and orthogonal design to study the extraction process of rosemary triterpene acid.Results The optimal enzymatic extraction conditions of rosemary triterpene acid were as follows: the enzyme dosage was 6 mg/g,and the enzymolysis time was 3.0 h,and the enzymolysis temperature was 55 ℃,and the pH value was 6.5.Under this condition,the rosemary triterpene acid extraction rate was 9.16%.Conclusion The cellulase can effectively destroy the cell wall,accelerate releasing of effective substance.Cellulase enzymolysis is provided with advantages as low price,mild enzymolysis conditions,simple process and good safety,and is worthy of popularization and application.

  • Antioxidant Activities of 3 Parts of Danshen Preparations

    Objective To study the antioxidant activities of 3 parts of Danshen preparations: aqueous extract,lipophilic extract and hydrolysate of aqueous extract.Method The antioxidant activities of 3 parts were determined by DPPH(1,1-Diphenyl-2-picrylhydrazyl) and TEAC(Trolox equivalent antioxidant capacity) method.The antioxidant activities of these 3 parts were analyzed by UV-Vis spectrophotometry,and then compared.Result Hydrolysate showed better antioxidant activity than aqueous extract,while lipophilic components had extremely poor antioxidant activity in the two systems.ConclusionAqueous extract and hydrolysate of aqueous extract of Danshen preparations were provided with strong antioxidant activities,and the activity of hydrolysate of aqueous extract was even better than aqueous extract.

  • Antioxidant Activity of the Extracts from Dalbergia odorifera T.Chen Leaves in Vitro

    ObjectiveTo evaluate the antioxidant activity of different extracts from Dalbergia odorifera T.Chen leaves,and their correlations with the contents of total flavonoids.MethodsThe aqueous extract from D.odorifera leaves was fractionated into four extracts using 3 different organic solvents of petroleum ether,chloroform,ethyl acetate and water.The DPPH radical scavenging activity assay,superoxide radical scavenging activity assay,hydroxyl radical scavenging activity assay,peroxide scavenging activity assay,and nitric oxide scavenging activity assay were employed to evaluate the antioxidant activities.The contents of total flavones were measured by Aluminum salt complex method,and ascorbic acid as the positive control.HPLC method was adopted to analyze their fingerprints.Results The four extracts from D.odorifera leaves have varying degrees of in vitro antioxidant activity,which were ranked in descending order: ethyl acetate extract(EAE),chloroform extract(CE),water extract(WE) and petroleum ether extract(PE).The antioxidant activities of these extracts were positively correlated with total flavonoids contents.Conclusions Antioxidant components could mainly exist in the ethyl acetate extract,and the antioxidant capacity is positively associated with the content of total flavonoids.

  • Study on Anti-hepatic Fibrosis Effect of Polysaccharides from Dicliptera chinensis (L.) Nees

    Objective To study the antagonism of polysaccharides from Dicliptera chinensis (L.) Nees towards hepatic fibrosis induced by dimethylnitrosamine (DMN) in Wistar rats.Methods The antagonism of polysaccharides from D.chinensis towards hepatic fibrosis was studied by exploring their effects on ALT,AST,TBIL and ALB contents of blood serum,liver wet weight and spleen wet weight in DMN-induced Wistar rats.The formation of hepatic fibrosis (HF) was observed with the help of Masson dyeing.Results The ALT,AST,TBIL and ALB contents of blood serum were significantly lower in highand low-dose groups of polysaccharides from D.chinensis than in model group,but by contrast the ALB content of blood serum in highand low-dose groups was higher than that in model group.The liver wet weight and spleen wet weight of rats in medication groups were obviously declined compared with model group.The polysaccharides from D.chinensis could significantly alleviate the inflammation,necrosis and fibrosis degree of liver.Conclusion The polysaccharides from D.chinensis have an antagonism towards hepatic fibrosis induced by DMN in Wistar rats,which may be associated with protecting liver and lowering transaminase.

  • Measurement of Hederagenin and Oleanolic Acid Contents in Clematidis Radix et Rhizoma from Different Areas

    Objective To establish an HPLC method for the determination of hederagenin and oleanolic acid in Clematidis Radix et Rhizoma.Method Using Waters XbridgeTM-C18 column (250 mm×4.6 mm,5 μm) at 25 ℃,acetonitrile-water (90∶ 10) as the mobile phase,the HPLC analysis was performed with flow rate of 1.0 mL/min.and PDA detection wavelength at 205 nm.Result The hederagenin showed a good linear relationship between 2.008-25.100 ng/mL (r2=0.999 8),and the oleanolic acid assumed a good linear relationship between 2.238-22.380 ng/mL (r2=0.999 1).The content of hederagenin in Clematidis Radix et Rhizoma from different areas could satisfy the quantitative specification of Chinese Pharmacopoeia,while that of oleanolic acid does not.Conclusion The contents of bioactive compositions of Clematidis Radix et Rhizoma vary significantly among different producing areas,on the aspect of quantitation of the chemical markers.

  • Detection of the Purity of Tartary Buckwheat Powder Based on Infrared Spectrum

    Objective To establish the infrared spectrum identification system for Tartary Buckwheat Powder.Methods Edible starches including Barley,Wheat and Oat Powders were added into Tartary Buckwheat Powder according to the proportions of 5%,10%,20% and 30%,respectively.After direct tabletting of powder and tabletting of the dry products of the concentrate of water extracts,detection of infrared spectra was carried out.The characteristic fingerprint chromatogram and that of comparative analysis was carried out.Results After the direct tabletting of powder,the contour features of infrared spectra of the different cultivars of Tartary Buckwheat Powder were accurately matched,having great similarities.The absorption peaks of the infrared spectra between Tartary Buckwheat Powder and Adulterated Samples,which were adulterated with different proportions of Barley,Wheat and Oat Powders,had relatively similarities in peak shapes and peak positions.However there were significant differences in infrared absorption peak at 2 300-2 400 cm-1,such as the absorption peak of Tartary Buckwheat Powder sample was relatively low,while that of the adulterated samples was relatively high.Moreover with the increase of the adulterated proportion,the intensity of absorption peak enhanced.At the same time,it was found out that the intensities of the absorption peaks of three adulterated samples were in the order of Oat > Barley > Wheat.There were relatively similarities in peak shapes and peak positions between tabletting of the dry products of the concentrate of water extracts and the dry products of water extracts concentrate of adulterated samples in different proportions.However,there were significant differences in infrared absorption of the stretching vibration area of C=O or C=C at about 1 650 cm-1,such as Tartary Buckwheat Powder had two absorption peaks,while the adulterated sample had only one peak.Moreover with the increase of the adulterated proportion,the intensity of absorption peak enhanced.At the same time,it was found out that there were obvious differences in the absorption peaks of three adulterated samples.For example,Barley and Oat had one weak absorption peak at 2 300-2 400 cm-1,respectively; and the intensities of the absorption peaks were in the order of Oat > Barley.However,that of Wheat showed no absorption peak.Conclusions This method was simple,easy,repeatable,and could rapidly and effectively identify whether the edible starches including barley,wheat and oat powders were added into Tartary Buckwheat Powder.

    Based on the related literatures and reference books,the antitumor function as well as its action mechanism and clinical application progress of HERBA SOLANI NIGRI in the latest ten years were reviewed.As is indicated by the results,HERBA SOLANI NIGRI shows significant antitumor functions by inhibiting the proliferation of tumor cells,inducing their apotosis and cytotoxicity,and enhancing the body immunity.Thus,HERBA SOLANI NIGRI has better efficacies and wide prospect in treating the cancer in liver,lung,bladder and nose.
