• The Simulation of Continuous Rectification Processing of Citral from Litsea cubeba (Lour.) Pers. Oil under Reduced Pressure

    Objective To simulate the reduced pressure continuous rectification processing of citral from Litsea cubeba (Lour.) Pers. oil by the use of Aspen Plus. Methods A technological process was first designed for separating citral from L.cubeba oil based on the properties of its components. Then DSTWU short-cut method was adopted to derive the number of stages, reflux ratio, quantity of distillate and other parameters for this rectification device. Finally, we used RadFrac rigorous model to verify the rectifying tower, analyzed the specificity of related technological parameters, and investigated the effects of number of stages, reflux ratio, quantity of distillate and other parameters on the continuous rectification of citral. Results Under the optimized conditions, the mass fraction of citral could be increased to 98.10%, with the total yield of 93.82%. Conclusions This study could provide an experimental basis for technological design and production of citral from L.cubeba oil using reduced pressure continuous rectification.

  • Effects of Ultrasonic Extraction on the Content of Total Flavonoids in the Superfine Powders of Viola tianshanica Maxim

    Objective To investigate the effects of ultrasonic extraction on the content of total flavonoids in the superfine powders of Viola tianshanica Maxim. Methods Both single factor test and L9(34) orthogonal test were used to investigate and detect the content of total flavonoids, the extraction rate of extracts and other indices. The score of comprehensive evaluation was calculated; and the optimal extraction technology was obtained. Results The optimal extraction technology for the superfine powders of V. tianshanica was as follows: 1∶12 solid-liquid ratio (W/V, g/mL), 70% ethanol concentration, 2 ultrasonic extraction times and 1.5 h extraction time. Under this condition, the content of total favonoids reached 19.19 mg/g; and the extraction rate of extracts was 15.3%. Conclusions This method was stable, feasible and reasonable.

  • Compression Characterization of the Mixture of Direct Compression Excipients and Traditional Chinese Medicine (TCM) Extract Powders

    Objective To evaluate the compression characteristics of the mixture of direct compression excipients and traditional Chinese medicine (TCM) extract powder. Method With the mixture of direct compression excipients and TCM powder as raw materials, the comprehensive characteristic of material was measured. The compression features of the mixture were evaluated from their flowability, compression and mechanical properties, and then analyzed by the mathematical models. Result TCM extracts show elastic characteristics, although they have better compressibility, but worse mobility, it needs to add some excipients with better mobility to improve the mobility of TCM. The mixture of TCM and excipients shows declined elasticity but stronger elastic recovery, and the mechanical properties of its compressed tablets are worse than that of direct compression excipients. Conclusion The related parameters or mathematical model can characterize the compressive characteristics of the mixture of TCM extracts and direct compression excipients. According to the actual situation, the critical values of powder compression properties could be measured and relatively objective criteria should be established to physicochemically analyze the effect of excipients on active pharmaceutical ingredient.

  • Optimization of Ultrasonic-assisted Extraction of Cultured Cordyceps militaris Polysaccharide by Response Surface Methodology

    Objective To optimize technique for extracting Cordyceps militaris Polysaccharide Method By adopting Response Surface Method and Center-rotated test, the duration of ultrasonic extraction, temperature and the ratio of liquid to solids effect on extraction rate was researched.Result The optimal extraction conditions were figured out: 80 ℃, the ratio of liquid to solid was 32 mL/g and 65 min of two-time extraction. Theoretically, the extraction rate can be 3.89%. Conclusion The Response Surface Method can optimize the techniques for ultrasonically extracting Cordyceps militaris polysaccharide, provide the optimum process parameters and lay the foundation for further research.

  • Optimization of Purification of Flavonoids from Lophatherum gracile Brongn.

    Objective To systematically study the factors which affect the purification of flavonoid from Lophatherum gracile Brongn.MethodHigh concentration of flavonoid extract from Lophatherum gracile Brongn.at pH 6.0 was eluted with water until the effluent was colorless,then 80% ethanol was used for elution at a rate of 2.0 mL /min until the effluent had no color reaction of flavonoids,then the eluent was collected.Result The average purity of flavonoids in solid product reached 57.5%.Conclusion This method is simple and feasible with good separation effect,which can meet the industrial requirement.

  • Extraction Technology of Total Flavonoids from the Roots of Ficus beecheyana

    ObjectiveTo select the optimum extraction technology of total flavonoids from the roots of Ficus erecta Thunb.var.beecheyana(Hook.et Arn.) King by ultrasonication-assisted extraction.MethodWith the content of total flavonoids as the index,single factor test was conducted to investigate the effects of ethanol concentration,extraction times,solid-liquid ratio and extraction time on the extraction yield of total flavonoids.And the technology of ultrasonication-assisted extraction was optimized by orthogonal test.ResultThe influencing degrees of four factors on the extraction yield of total flavonoids were in the order of solid-liquid ratio > ethanol concentration > extraction time > extraction times.The optimum extraction technology was 50% ethanol concentration,1:20 solid-liquid ratio(W /V,g /mL),2 extraction times and 60 min extraction time.Under this condition,the extraction yield of total flavonoids from the Ficus beecheyana roots was 27.83 mg /g.ConclusionThis extraction technology was reasonable,and the detection method was simple,rapid and accurate,which provided scientific references for the further development of Ficus beecheyana.

  • Optimization of the Ultrasonic Extraction Technology of Total Saponins from RHIZOMA DIOSCOREAE NIPPONICAE by Orthogonal Design

    ObjectiveTo select the optimal ultrasonic extraction technology of total saponins from RHIZOMA DIOSCOREAE NIPPONICAE. MethodsOrthogonal test was adopted. The content of total saponins in extracts was detected by spectrophotometry. ResultsThe detection wavelength was 410 nm; saponins showed good linear relationship ( r = 0. 999 8) within 0. 004-0. 020 mg / mL. the optimal extracting technology was as follows: 1∶ 30 solid-liquid ratio,180 W ultrasonic power and 70 ℃ extracting temperature,with n-butanol at water saturation as the extracting solvent. Conclusions This ultrasonic extraction technology was reasonable,economical and feasible.

  • Comparison of the Quercetin Contents in Young Buds and Old Leaves of HERBA TAXILLI

    ObjectiveTo detect the contents of quercetin in the young buds,old leaves and reference substances of HERBA TAXILLI,and to provide scientific basis for the material selection of HERBA TAXILLI tea. Methods With quercetin as the observation index,quercetin contents in the same host were detected by RP-HPLC. And the results were compared with those of reference substances of HERBA TAXILLI. Quercetin was extracted by the reflux extraction of anhydrous methanol hydrochloric acid ( 4∶1,V/V) solution. Chromatographic column was Welch Materials XB-C 18 ( 4.6 mm × 250 mm,5 μm) ; mobile phase was anhydrous methanol 0.05% phosphoric acid solution ( 47∶53,V/V) ; detection wavelength was 254 nm; flow rate was 1.0 mL/min; and column temperature was 30 ℃.Results Quercetin concentration had good linear relationship with the peak area between 0. 102 5 and 1. 640 0 ( R = 0.999 9) . The average recovery rate was 99. 25% . Quercetin content was 3.17 mg/g in young buds,6. 40 mg/g in old leaves,and 1.24 mg/g in reference substances of HERBA TAXILLI. Conclusions Both of the quercetin contents in young buds and old leaves of HERBA TAXILLI were higher than that in the reference substances of HERBA TAXILLI,so the young buds and old leaves could be used as the raw materials of HERBA TAXILLI tea.

  • Research on the HPLC Fingerprint of Zishen Tablet and the Content Detection of Five Chemical Components

    ObjectiveTo establish the analysis method for the HPLC fingerprint of Zishen tablet,to detect the contents of ligustilide,n-butylene( n-butene) phthalide,tanshinoneⅠ,cryptotanshinone and tanshinoneⅡA in Zishen tablet,and to provide references for the quality control of Zishen tablet. MethodsHPLC was adopted; chromatographic column was Ultimate XB C 18 column( 250 mm × 4. 6 mm,5 μm) ; mobile phase was acetonitrile 0. 3% phosphoric acid solution; and detection wavelength was 260 nm. According to the HPLC of Zishen tablets,tanshinone ⅡA was taken as the control substance. The RSD values of the relative retention time and relative peak area of the chromatographic peak of tanshinoneⅡA were analyzed. Similarity analysis of the 10 batches of Zishen tablets was carried out based on the similarity evaluation system for chromatographic fingerprint of TCM ( Version 2004A) . By using the external standard method,contents of ligustilide,n-butylene( n-butene) phthalide,tanshinoneⅠ,cryptotanshinone and tanshinoneⅡA were detected in 10 batches of Zishen tablets. ResultsUnder the selected conditions,32 peaks together constituted the fingerprint characteristics of Zishen tablets; and sample in each batch had the same characteristics; the relative contents of characteristic peaks had basically the same distribution. The corresponding retention time of the chromatographic peak of tanshinone ⅡA was the same in different batches; RSD values of the relative retention time and the relative peak area of the chromatographic peaks of tanshinone ⅡA in different batches all met the requirements of fingerprint detection. The similarity degrees of the fingerprints of 10 batches of samples were all greater than 0. 940; and 5 chemical components had basically the same contents in the 10 batches. ConclusionsHPLC fingerprint and the detection method of 5 components in Zishen tablet were accurate,stable and simple,and the two methods could be used for the quality control of Zishen tablet.

  • Comparative Analysis of Polydatin and Resveratrol in Polygonum cuspidatum from 5 Regions in Southwest China

    ObjectiveThe technical system of the simultaneous determination of polydatin and resveratrol in Polygonum cuspidatum was established and the content of the two ingredients in the Polygonum cuspidatum from 5 producing areas in southwest China was compared. MethodThe two ingredients could be determined with HPLC method at once. The column was Agilent C 18 ( 150 mm × 4.6 mm,5 μm) ; the mobile phase,acetonitrile water ( 23∶77,V/V) and the detection wavelength,306 nm. ResultsThe injection concentration of polydatin and resveratrol was in the range of 48 480 μg/mL ( r = 0. 999 0) or 7. 2 72. 0 μg/mL ( r = 0. 999 2) ,respectively,which integral value of each peak area showed a good linear relationship and the average recovery rate of two components was 98. 69% and 99. 42% ,RSD was 1. 68% and 3. 39% ( n = 6) . The order of the content of polydatin in Polygonum cuspidatum from five producing areas was Leshan in Sichuan > Dujiangyan in Sichuan,Yongchuan in Chongqing > Zunyi and Bijie in Guizhou; the content of resveratrol,Zunyi and Bijie in Guizhou > Dujiangyan and Leshan in Sichuan,Yongchuan in Chongqing. Conclusion This test method can be used for the quality control of Polygonum cuspidatum,and the test results would provide a reference for the research on the medicinal function of Polygonum cuspidatum.

  • Analysis on Chemical Constituents of Raw and Processed Radix Bupleuri

    ObjectiveTo investigate the changes in chemical constituents of Radix Bupleuri before and after processing. Methods The chemical constituents of Radix Bupleuri were detected before and after processing with UV,HPLC and TLC methods. ResultsThe chromatographic conditions were determined as follows: chromatographic column of Kromasil C 18 ( 4.6 mm ×250 mm,5 μm) ,column temperature of 25 ℃ ,mobile phase of acetonitrile ( A) water ( B) ,gradient dilution of phase A at 10% within 0 3 min,phase A at 10% 15% within 3 5 min,phase A at 15% 25% within 5 20 min,phase A at 25% 40% within 20 30 min,phase A at 40% 55% within 30 40 min,phase A at 55% 75% within 40 45 min,phase A 75% 90% within 45 60 min. Under these parameters,the chemical constituents could be well separated. Comparison of HPLC fingerprints showed that chemical constituents in raw and processed Radix Bupleuri were changed remarkably,and some new constituents produced after processing. Thin layer chromatography conditions were determined as follows: expansion system of chloroform methanol water ( 100∶20∶1,V/V/V) ,chromogenic agent of 5% vanillin-sulfuric acid to heat to 105 ℃ till the appearance of clear spots. The spot location of TLC fingerprints of chemical constituents differed between raw and processed Radix Bupleuri. ConclusionUV,HPLC and TLC all confirmed the changes of chemical constituents of Radix Bupleuri before and after processing.

  • Analysis of β-elemene Contents in Eupatorium adenophorum Spreng and Lantana camara L. by HPLC

    ObjectiveTo find out new and low-cost resources of β-elemene and new approach of utilizing two malignant invasive plants ( Eupatorium adenophorum Spreng and Lantana camara Linn. ) . Method The content of β-elemene in different parts of E. adenophorum and L. camara from Panxi Area were analyzed by HPLC on Eclipse XDB-C 18 ( 4.6 mm ×150 mm,5 μm) . The mobile phase was ethanol∶acetonitrile∶water ( 70∶10∶20,V / V) ; flow rate was 1. 0 mL / min; detection wavelength was 210 nm; injection volume was 20 μL; and column temperature was 30 ℃ . Result Content of β-elemene in naturally dried leaves of L. camara was 0. 126% ,which was close to that in RHIZOMA CURCUMAE. Content of β-elemene in naturally dried leaves of E. adenophorum was 0. 383% ,which was 2 times of that in RHIZOMA CURCUMAE. Conclusion Extracting β-elemene from E. adenophorum and L. camara could greatly reduce the material cost of β-elemene,and it provided new approach for the control and development of malignant weeds.

  • Research and Comparison of Anti-oxidation of Polyphenols of Chinese Dates from North of Shaanxi

    ObjectiveThis research aimed to study the antioxidant activity of polyphenols of Chinese Dates from different counties of north of shaanxi province and compared in four antioxidant systems separately. Methods The antioxidant activities of each polyphenols of Chinese Dates were tested in vitro by scavenging different free radicals and H2O2 tests: ①tested the capability of scavenging·OH; ② tested the capability of scavenging O-2 ·; ③ tested the capability of scavenging DPPH·; ④tested the capability of scavenging H 2 O 2 . ResultsAll the polyphenols of Chinese Dates from different counties of north of shaanxi had certain antioxidant capacity,but different polyphenols from different parts had different capacity in clearing away free radicals or H2O2 . ConclusionsPolyphenols of Chinese Dates from north of Shaanxi has stong antioxidant capacity and it is a good natural antioxidant.

  • Effects of Growth Years on the Polysaccharide Content in RADIX OPHIOPOGONIS

    Objective To study the correlation rules of polysaccharide content in RADIX OPHIOPOGONIS with different growth periods,to determine the characteristic quality,and to provide scientific basis for GAP quality control and for resources rational exploitation and utilization of RADIX OPHIOPOGONIS.Method In situ dynamic method was used for the main production area for RADIX OPHIOPOGONIS ( 3 years old) . Root tuber samples with 0. 5,1. 0,2. 0 and 3. 0 years old were randomly collected. The washed fresh root tubers were dried at-50 ℃ . Polysaccharide content in each treatment was detected by phenol-sulfuric acid colorimetry. Correlation rules of polysaccharide content in RA-DIX OPHIOPOGONIS with different growth periods were analyzed by SPSS17. 0 software. Result The average content of polysaccharide in RADIX OPHIOPOGONIS with different growth periods was between 17. 68% and 34. 86% . With the growth of root tuber,polysaccharide content showed extremely significant positive correlation ( P < 0. 001) . The regression equations were as follows: y 1 = 9. 126ln( x) + 24. 58(r2 = 0. 980) and y2 = 9. 065ln( x)-28. 85(r2 = 0. 981) . ConclusionThe content of polysaccharide which was a secondary metabolite was closely related to the growth period. There was scientific basis for harvesting at 3 years old. It was proper to collect one year after plantation,because in the summer and fall of the second year,a large amount of root tubers became rotten due to the high temperature and humidity.

  • Regulating Effect of Sargassum fusiforme (Harv.) Setch. on the Level of Blood Lipid in Experimental Rats with Hyperlipidemia

    Objective To discuss the regulating effect of Sargassum fusiforme (Harv. ) Setch. on the level of blood lipid in experimental rats with hyperlipidemia. Methods To establish animal models with hyperlipidemia,high fat forage was fed to 40 healthy female Wistar rats. Then they were fed S. fusiforme powder with the purpose of intervention treatment. Biochemical techniques were adopted to detect the levels of triglyceride (TG) ,total cholesterol (TC) ,low density lipoprotein (LDL) ,high density lipoprotein (HDL) ,and the activities of lipoprotein lipase (LPL) and hepatic lipase (HL) . Enzyme linked immunosorbent assay was used to measure the level of hydroxy methylglutaryl coenzyme A (HMG-CoA) and the activity of hydroxy methylglutaryl coenzyme A reductase (HMG-CR) . Results TG,TC and LDL levels of blood serum in treatment group were significantly lower than those in model group (P < 0. 05) ,but HDL level was significantly increased (P < 0. 05) . LPL and HL levels of blood serum and liver tissue were significantly higher than those in model group (P < 0. 05) ,HMG-CoA level was signif-icantly higher in liver tissue than in model group,but HMG-CR activity was significantly lower in liver tissue than in model group (P < 0. 05) . Conclusions S. fusiforme could affect the metabolism of blood fat components and regulate the level of blood fat via enhancement of LPL. HL and HMG-CR activities.

  • Effects of Flavonoids from Scutellaria Stem and Leaf on Memory Impairment and Nerve Inflammation in Rats with Chronic Cerebral Ischemia

    Objective To study the effects of flavonoids from Scutellaria stem and leaf ( SSF) on memory impairment and nerve inflammation in rats with chronic cerebral ischemia. Methods The model of chronic cerebral ischemia was established by permanent ligation of bilateral common carotid artery of female Sprague-Dawley ( SD) rats for 2 months. The functions of learning and memory were detected by Morris water maze,the expression of glial fibrillary acidic protein ( GFAP) in astrocytes of hippocampus and cortex was observed by immunohistochemical method and the levels of IL-1β,TNF-α,IL-8 and IL-10 were determined with ELISA method. Results Compared with the sham group, learning ability and retention ability significantly decreased,the GFAP expression remarkably increased,the IL-1β,TNF-α and IL-8 levels no-ticeably heightened and the IL-10 content notably lowered in cerebral ischemia rats,respectively. However,the treatment of 17. 5,35. 0 and 70. 0 mg / kg SSF for 38 d showed significant improvements in the impairment of learning ability and retention ability,embodied by the effectively suppressed GFAP expression ( P < 0. 05) ,lowered IL-1β,TNF-α and IL-8 levels and markedly enhanced IL-10 content with cerebral ischemia rats ( P < 0. 01) . Conclusion SSF can obviously improve the memory impairment and nerve inflammation in rats with chronic cerebral ischemia,probably due to inhibition of astrocyte proliferation and lowering of the inflammatory cytokine production in the brain.

  • Content Determination of Flavonoids and Chlorogenic Acid in Different Organs of Sweet Potatoes

    Objective To fully utilize the stems and leaves of sweet potatoes,and to provide references for the extraction of effective components from plants. Method Chlorogenic acid content was detected by high performance liquid chromatograph; and flavonoids content was detected by UV-Vis spectrophotometry. Result Contents of flavonoids and chlorogenic acid varied greatly in different organs of sweet potatoes. Among them,leaves had the maximum contents of flavonoids and chlorogenic acid,which were 9. 46% and 4. 45% . Young stems had the minimum content of chlorogenic acid,which was 0. 72% ; and old stems had the minimum content of flavonoids,which was 2. 20% . Conclusion Contents of flavonoids and chlorogenic acid were very high in the leaves and stems of sweet potatoes,which could be developed into natural medicines.

    Objective To evaluate the inhibitory effect of Ilexgenin A ( IA) on the proliferation of cervical cancer cell line Hela. Method Using the methods of determination of CCK-8,LDH and BrdU,the proliferation inhibitory rate and the 50% inhibitory concentration of IA on the human cervical cancer cell line Hela were examined. Result The inhibition of IA on Hela cell line was dose-and time-dependent.Conclusion IA has an inhibitory effect on the proliferation of Hela cells.

    Objective To measure the content of oleanolic acid in Acanthopanax giraldii Harms from different producing areas and different collecting time,A. leucorrhizus var. fulvescens Harms and A. setchuenensis Harms ex Diels from different medicinal parts. Method The HPLC-ELSD method for determination of oleanolic acid was performed on the Kromasil C18 column ( 4. 6 mm × 250 mm,5 μm) with the mixture of acetonitrile,water,glacial acetic acid and triethylamine ( 85∶15∶0. 03∶0. 05) as the mobile phase at a flow rate of 0. 8 mL/min. The column temperature was set at 30 ℃,the ELSD drift tube temperature was set at 78 ℃,and the flow rate of atomization gas high-purity nitrogen was set at 2. 0 L/min. ResultThe linear range of oleanolic acid was 2.96-118. 50 μg/mL,the regression equation was lgA =1. 247 3lgC +3. 006 9 ( r =0.999 1,n =6) ,the average recovery was 97.52%,and the RSD was 1.75%. These experimental materials,arranged in descending order of oleanolic acid content,were as follows: A. giraldii Harms stem bark ( 375. 00 μg/g) > A. leucorrhizus var. fulvescens Harms stem bark ( 216.26 μg/g) > A. giraldii Harms root ( 215.63 μg/g) > A. giraldii Harms root bark ( 133.78 μg/g) > A. leucorrhizus var. fulvescens Harms leaf ( 35. 53 μg/g) > A. setchuenensis Harms ex Diels stem bark ( 35. 46 μg/g) . No oleanolic acid was found in A. giraldii Harms peeled stem,A. giraldii Harms leaf and A. setchuenensis Harms ex Diels leaf. The content of oleanolic acid was relatively higher in A. giraldii Harms stem bark collected from March to July. As with A. giraldii Harms stem bark,the content of oleanolic acid was the highest in that collected from Majia Ditch,Hongyuan County,Aba Prefecture,Sichuan Province. Conclusion June and July are regarded as the optimal time for harvesting A. giraldii Harms stem bark when they are used as medicine in Sichuan Tibetan and Qiang folk. Majia Ditch,Hongyuan County, Aba Prefecture,Sichuan Province is considered the genuine producing area of A. giraldii Harms,by which the medicinal material containing the highest content of oleanolic acid is produced. It is suggested that A. leucorrhizus var. fulvescens Harms stem bark could serve as the new resource plant for oleanolic acid. It is necessary to carry out in-depth studies about the effects of harvesting time and altitude on the content of oleanolic acid.

  • Comparative Analysis of Essential Constituents in Herbal Pair Atractylodes macrocephala-Ramulus cinnamomi and Its Single Herbs by GC-MS with Chemometric Resolution Method

    Objective To investigate the changes in main essential constituents in Atractylodes macrocephala and Ramulus cinnamomi and their herbal pair,further providing basis for getting the pairing rules of Traditional Chinese Medicine and unveiling novel bioactive constituents. Methods The Essential constituents were extracted from Atractylodes macrocephala and Ramulus cinnamomi macrocephala and their herbal pair by employing the steam distillation,measured by GC-MS and the overlapped spectral peaks were analyzed by chemometric resolution method (CRM) ; temperature-programmed retention indices were employed for qualitative analysis. Results Totally 29,66 and 55 Essential constituents were identified from Atractylodes macrocephala and Ramulus cinnamomi and their herbal pair respectively,accounting for 95. 93% , 94. 43% and 96. 35% of total essential oils correspondingly. Single herbs and their pair had four common constituents,accounting for 13. 14% of total essential oils. After processed in the herbal pair,four new constituents were identified from the herbal pair; 33 constituents were not identified from Ramulus cinnamomi and 7 were not found from Atractylodes macrocephala. The main Essential constituents in herbal pair were 2-( 2-Methoxyphenoxy) phenol,γ-selinene and cinnamic aldehyde,which mainly from Atractylodes macrocephala. Conclusion The chemical constituents in herbal pair are not the simple totaling of single herb,and the mechanism still remains to be studied further.

  • Standardized Culativation Technique for Saussurea lappa by Covering Plastic Mulch in Yunnan Lijiang Prefecure of Yunnan Province

    Objective To develop standardized cultivation technique for Saussurea lappa by covering plastic mulch in Lijiang Prefecture of Yunnan Provience. Method The standardized cultivation technique for S. lappa by covering plastic mulch in whole growth period was summarized on the basis of several years trial research. The reasons for increase in yied of S.lappa by covering plastic mulch were also discussed simultaneously. Results Through the cultivation technique for S.lappa by covering plastic mulch,the germination percentage was more than 90% ,the length of biennial taproot was 20.36-42. 71 cm and the maximum diameter was 3.37-5. 40 cm,the drying rate was 31.60%-34.74% and the production peak was 10 032 kg/hm 2 ; the sulphated ash weight was less than 4%; the acid insoluble ash weight was less than 2%. Conclusions The standardized cultivation for S.lappa on the production in Yunnan could be realized by this technique,the industrial development of S.lappa in Yunnan would also be promoted in Lijiang and its circumjacent areas.

  • The Geochemical Features of Genuine Eucommia ulmoides Oliv. Producing Area in Longmen Mountains

    Through field investiagation,sample collection and testing analysis of genuine Eucommia ulmoides Oliv. in Longmen mountain section,the research indicates that the geochemical backgroud of Eucommia ulmoides Oliv. producing area is similar to the Chinese national value,but different from the Clarke value. The content of Zn,B and Pb in this area is above while that of Mo,Mg,K and Ca is below the national background value and the Clarke value. The content of soil elements changes vertically as follows: the content of N,Org·C,P,CaO,As, Hg,Cd and Pb decreases while that of Mn increases from the soil surface layer to the parent material layer; the content of B,Mo,K2O,MgO, Zn and Al2O3 increases from Layer A to Layer B to Layer C,but decreases in Layer D. The evaluation of soil quality indicates that Cd and Zn have obvious influence on soil quality of Eucommia ulmoides Oliv. Therefore,the two elements should get more management.

  • Application of Self-made Frozen Section Embedding Medium for Extracting Total RNA from the Seeds of Viola yedoensis

    Objective To extract the high-quality total RNA from the seeds of Viola yedoensis. Method The seeds of V. yedoensis were embedded in both self-made embedding medium and synthetic glue,then sectioned with frozen section machine for extracting RNA; the concentration,purity and integrity of yielded RNA were detected for comparing two embedding methods. Results The concentrations RNA extracted by self-made embedding medium and synthetic glue were determined 3. 42 and 1. 04 μg/μL respectively. Agarose gel electrophoresis apparently detected two distinct bands of 28s rRNA and 18s rRNA,with brightness ratio of 28S rRNA to 18S rRNA higher than 1. However, the total RNA extracted by the synthetic glue embedding medium were smeared. Conclusion Using self-made frozen section embedding medium method to extract total RNA from seeds of V. yedoensis method can well meet the requirements of most molecular biological experiments. It is a cost-saving,effective and simple method for high-quality RNA extraction.

  • Research Advances in the Development and Utilization of Pueraria peduncularis (Grah. ex Benth)

    Pueraria peduncularis ( Grah. ex Benth) ,a species of perennial herb in genus Pueraria of family Leguminosae,is abundant in resources. In this paper,the research advances in the development and utilization of Pueraria peduncularis in both the medical and agricultural fields were briefly introduced. It was found out that the Pueraria peduncularis had the efficacies of heat-clearing,detoxifying,lung-nourishing and insect-killing and it also showed strong biological activities towards phytopathogen and molluscs. Thus,a deep research into Pueraria pedu-ncularis is of great importance to the comprehensive development and utilization of its resources.

  • Advances in the Treatment of Rapid Arrhythmia Using Corydaline

    Corydaline has the pharmacological efficacies of analgesia,calmness,myocardial ischemia and antiarrhythmic effect. Its anti-ar-rhythmic mechanism is to act on the potassium and calcium channels,inhibit potassium and calcium currents,reduce automatic rhythmicity and calcium overload,thus extending the effective refractory period and action potential duration. Corydaline has been found clinically effective in atrial premature beats,premature ventricualr contraction,atrial fibrillation and anomalous atrioventricular excitation. This paper reviewed the advances in treatment of rapid arrhythmia using corydaline both clinical study and basic electrophysiology,aiming at providing reference for further exploitation and application of corydaline.
