  • Effects of Flavonoids from Stem and Leaf of Scutellaria baicalensis Georgi on Memory Impairment and Expression of ChAT and NOS in Chronic Cerebral Ischemia Rats

    [Objective]To investigate the effects of flavonoids from stem and leaf of Scutellaria baicalensis Georgi on memory impairment and expression of ChAT and NOS in chronic cerebral ischemia rats.[Methods]The model of chronic cerebral ischemia was established in female Sprague-Dawley (SD) rats by bilateral carotid artery occlusion for 2 months.By employing Morris water maze,expressions of chloine acetyl-transferase (ChAT) ,neuronal nitric oxide synthase (nNOS) ,inducible nitric oxide synthase (iNOS) and endothelial nitric oxide synthase (eNOS) were examined by immunohistochemical method to investigate the functions of learning and memory.[Results]Compared with the sham group,memory ability and the expressions of ChAT and eNOS,were significantly decreased,while the expressions of nNOS and iNOS were dramatically upregulated.However,the treatment of 17.5,35.0 and 70.0 mg/kg SSF for 38 d on ischemia rats showed significantly improved impairment of memory ability and the abnormal changes of ChAT,eNOS,nNOS and iNOS in the brain of ischemia rats.[Conclusion] Flavonoids from stem and leaf of Scutellaria baicalensis Georgi (SSF) can improve the memory impairment induced by chronic cerebral ischemia in rats,which may be related to regulate the abnormal changes of ChAT and NOS in the ischemic rat brains.

  • Isolation of the Endophyte from Alhagi pseudalhagi Desv and the Analysis of the Active Ingredient of Its Metabolite

    With the grinding method,20 endophyte strains were firstly isolated from the Alhagi pseudalhagi Desv stem and leaf,one species of herbal plant is grown in Xinjiang regoin,and a strain---XJAS-AB-11 with the strongest antagonistic activity to crop pathogen was selected. The XJAS-AB-11 was preliminary identified as one kind of Bacillus sp. based on the morphological observation and the analysis of physiological and biochemical characteristics and 16S rDNA sequence. After the expanding culture of the antagonistic strain,a preliminary study on its me-tabolites was conducted and the active monomer C2 and H2-2 were respectively isolated from its active site with the inhibitory effect by means of method of silica gel column chromatography and TLC. The compounds of C2 and H2-2 were proved to be flavonoids through the analysis of ultraviolet spectroscopy,NMR and mass spectrometry. The endophyte from Alhagi pseudalhagi Desv is rich in antimicrobial active substances and also is a potential resource for development of natural antibacterial drug.

  • The Effects of Total Flavonoids of Lespedeza virgata on Chronic Renal Failure Rats

    [Objective]To investigate the therapeutic function of Lespedeza virgata on chronic renal failure (CRF) rats and its effects on following indexes in rats: urine protein content,blood routine items,blood urea nitrogen (BUN) and serum creatinine (SCr).[Method]The tested animals were randomly divided into 4 groups: sham operation group,model group,group treated with low dose and group treated with high dose of total flavonoids of Lespedeza virgata.CRF rat models were constructed by removing 5/6 of the kidney and then treated with different dose of total flavonoids of Lespedeza virgata.After 35 and 70 days of treatment,the rat urine was collected and the urine protein content was determined; the rat blood was collected and the blood routine items were examined,the blood urea nitrogen and serum creatinine were tested; rats were sacrificed and the pathological and histomorphometric changes were observed,the pathological composite scores were evaluated.[Result]Treatment with low dose and high dose of total flavonoids of Lespedeza virgata decreased the urine protein content of chronic renal failure rats,but the difference is not statistically significant compared to model group; there is no obvious improvement of blood routine items in groups treated with low dose and high dose of total flavonoids of Lespedeza virgata; blood urea nitrogen (BUN) and serum creatinine (SCr) in CRF rats were reduced in groups treated with low dose and high dose of total flavonoids of Lespedeza virgata,the difference is statistically significant compared to model group (P<0.05); pathological and morphological examination shows that the pathological composite scores were obviously decreased in groups treated with low dose and high dose of total flavonoids of Lespedeza virgata,the difference is statistically significant compared to model group (P<0.05).[Conclusion]Lespedeza virgata has protective effects on the kidney of CRF rats,the mechanism of which is probably associated with an increase in renal blood flow,augmented glomerular filtration rate,promotion of metabolites excretion,reduction in the blood urea nitrogen and serum creatinine and other aspects.

  • Changes of the Ellagic Acid Content in the Granules of PERICARPIUM GRANATI Extracts with Different Storage Periods

    [Objective]To discuss the changes of ellagic acid content in the granules of PERICARPIUM GRANATI extracts with different storage periods,and to detect the stability of the granules.[Methods]High performance liquid chromatography was adopted.Chromatographic column was PT-230AColume heater Apollo C 18 column (150mm×4.6mm,5μm); column temperature was 30℃ ; mobile phase was aceto-nitrile-1.2% phosphoric acid solution (20∶80,V/V); detection wavelength was 254 nm and flow rate was 1.0 mL/min.[Results]The lin-ear range of the detection of ellagic acid content was between 0.52-5.20 μg/mL(R2 = 0.997 9); the average recovery rate was 97.8%; and RSD was 1.6%.The content of ellagic acids in the granules of PERICARPIUM GRANATI extracts was 0.495 mg/g on March 25,2008,and that was 0.439 mg/g on March 25,2011,which indicated that the granules of PERICARPIUM GRANATI extracts had good stability,and the period of validity could reach more than 3 years.[Conclusions]This method was simple and accurate and could be used for the quality control of PERICARPIUM GRANATI.

  • The Effects of Flavonoids from Scutellaria Stem and Leaf on the Expression of NMDAR and VEGF in Chronic Cerebral Ischemia Rats

    [Objective]To study the effects of flavonoids from Scutellaria stem and leaf (SSF) on the expression of NMDAR (N-methyl-D-as-partate receptor) and vascular endothelial growth factor (VEGF) in chronic cerebral ischemia rats.[Method]The model of chronic cerebral ischemia was established by bilateral carotid artery occlusion for 2 months with female SD rats.The effects of SSF were evaluated on mRNA expression in NMDAR of hippocampal cells and VEGF in cerebral cortex cells by the method of reverse transcription-polymerase chain reaction (RT-PCR).[Result]Compared with the sham group,the expression of NMDAR1,NMDAR2A and NMDAR2B in hippocampus and VEGF in cerebral cortex were significantly increased (P<0.01).However,The cerebral ischemia rats daily and orally administered SSF (17.5,35.0 and 70.0mg/kg) for 38 days appeared obviously reduced mRNA expressions of NMDAR1,NMDAR2A and NMDAR2B in hippocampus (P<0.05),and increased mRNA content of VEGF in the cortex (P<0.05).[Conclusion]SSF can decrease the expression of NMDAR in hippocampus,increase the expression of VEGF in cerebral cortex of cerebral ischemia rats,which suggested that the neuronal protection of SSF may be preformed by influencing the production of NMDAR and VEGF.

    [Objective]To provide a theoretical basis for the development of curcumin by structural modification.[Methods]Curcumin reac-ted with acetyl chloride to generate curcumin derivatives in ice-water bath.These synthetic products were separated by using silica gel chroma-tography.[Results] The synthetic products were identified as the derivatives of curcumin by digital melting point apparatus.[Conclusions] The reaction products were characterized of high purity and high yield.

  • Comparative Research on Antipyretic and Analgesic Effects of Different Extraction Parts of Rubus alceaefolius Poir.

    [Objective]To study the antipyretic and analgesic effects of water extracts,alcohol extracts and other solvent extraction parts of Rubus alceaefolius Poir.[Method]The alcohol extracts of Rubus alceaefolius Poir. were extracted with petroleum ether,chloroform,ethyl ace-tate,n-butyl alcohol according to their polarities. Their antipyretic effects were accessed by dried yeast-induced pyrexia method,and their an-algesic effects were accessed by HAc-induced writhing response and hot-plate test.[Results] The extraction parts of Rubus alceaefolius are provided with antipyretic effects on the pyrexia of rats induced by dried yeast-induced pyrexia method,and significant analgesic effects on HAc-induced writhing response and hot-plate test.[Conclusion] The antipyretic and analgesic effects of Rubus alceaefolius were good.

  • Preparation and Pharmacodynamic Assay of Tongnao Oral Solution

    [Objective]To solve the severe precipitation of Tongnao oral solution and evaluate its pharmacodynamic effects.[Methods]The main components of Tongnao oral solution,including Red Ginseng,Notoginseng,Chuanxiong,Rhubarb and Leech,were extracted respectively for pair combination testing. Clotting time of normal mice and ischemic mice was employed as the indicator for evaluating the pharmacodynamics of final products.[Results]The pair combination testing showed that tannin in rhubarb would act on some components of other medicinal herbs and produce precipitation. After tannin was removed by using improved gelatin method,the Tongnao oral solution was clear and stable, and it could remarkably elongate the clotting time of normal mice and ischemic mice.[Conclusion]This technique can well solve the severe precipitation of Tongnao oral solution and ensure the effectiveness of the products.

  • The Effect of Matrine on Bone Marrow and Cell Apoptosis of Patients Radiated by ~(192)Ir

    [Objective]To observe the effect of Matrine on bone marrow and cell apoptosis of patients radiated by 192 Ir.[Method]54 patients who were radiated with 192 Ir (0.05-0.66 Gy) were taken for research objects and they were separately treated with Fufang ZaoFan Wan, Matrine and placebo.Double dyed with Annexin V-FITC and PI,FACScan for flow cytometry quantitative detection,in order to examine apoptosis rate of cells,aberration rate of lymphocyte.The bone marrow smear,aberration rate of lymphocyte and apoptosis rate of cells of patients were observed.[Result] Morphology abnormity was seen in all bone marrow samples and the total abnormity rate was 20.4% (reduction + extremely active); the apoptosis rate of medullary cells reduced with the pass of time.Among all abnormity of patients' chromosomes,the abnormity of instability showed higher rate for detection.The morphologies of groups treated with Matrine and Fufang Zaofan Wan for six months recovered to approximately normal level but with increased apoptosis rate of cells.The group treated with Matrine showed more significant effect (P<0.01) and in the following five years of reexamination,the difference between the patients of Matrine group and healthy people showed insignificance in statistics (P>0.05).Clinically,no tumor was found from the patients of Matrine group.[Conclusion]Matrine,as a anti-radiation agent,can improve bone marrow hematopoietic function of patients radiated by small amount of 192 Ir and induce the apoptosis rate of their medullary cells.

  • Research on the Hypoxia Resistant Ability of Vascular Endothelial Cell Improved by Tongxinluo via the PI-3K/Akt/Hif Pathway

    [Objective]To investigate the influence of PI-3K/Akt/Hif pathway on anti-hypoxia activity of Tongxinluo by studying the effect of Tongxinluo on hypoxia induced factor in vascular endothelial cell and its upstream signal transduction pathway,the PI-3K/Akt pathway,under the hypoxia condition.[Method]Human umbilical vein endothelial cells (HUVECs) were divided into the following treatment groups: control group,Tongxinluo group,hypoxia group,and hypoxia + Tongxinluo(100 μg/mL) group. The CCK-8 kits were used to detect viability and proliferation rate of cells in each group. The protein levels of Hif,Bcl 2 ,Mcl,Bax and phosphorylated Akt were studied by immunoblotting analysis. The HUVEC cells were transiently transfected with the dominant negative mutant of Hif-1α(DN-Hif) and apoptosis rates were analyzed by flow cytometry(FCM). The HUVEC cells were transiently transfected with the dominant negative mutant of PI-3K (ΔP85) or Akt (DN-Akt) to interrogate the role of PI-3K/Akt signaling pathway in the anti-hypoxia ability of Tongxinluo on endothelial cells. [Result]Though the proliferation rate increased significantly in Tongxinluo group under the hypoxia condition compared with hypoxia group,the degree weakened notably compared with the control group. The protein levels of Hif-1α,Bcl 2 ,Mcl and phosphorylated Akt were up-regulated by Tongxinluo. Meanwhile,the expression of Bax was down-regulated. Inhibition of Hif-1α activation by DN-Hif and inhibition of the PI-3k/Akt pathway by ΔP85 or DN-Akt attenuated the rise of Hif expression and HUVEC viability induced by Tongxinluo. But still,the percentage of apoptosis of HUVEC was down-regulated to a certain extent by Tongxinluo.[Conclusion] Tongxinluo could up-regulated the protein level of Hif,promote the expression of apoptosis related factors,improve the cell viability and eventually reduce the apoptosis rate,which may be one of the molecular mechanisms that Tongxinluo improved the anti-hypoxia ability of vascular endothelial cells. And futher study showed that Tongxinluo activated the PI-3K/Akt-dependent signaling pathway to up-regulate the expression of Hif.

    [Objective]To study the contents and types of amino acids in Coreopsis tinctoria.[Method]Detection was conducted by the amino acid analyzer.[Result] There were 17 kinds of amino acids in C.tinctoria,accounting for 10.08% .Among them,8 were essential amino acids for human body,accounting for 40.3% of the total amount of amino acids.[Conclusion]As a genuine medicinal material in Xinjiang, C.tinctoria had relatively high health care value and good development prospect.

  • Study of Determination Methods of Polysaccharide Content in Poria cocos

    [Objective]To determine the content of Poria polysaccharide in Poria cocos from different places.[Method]The absorbance of pachyman was determined by phenol-sulfuric acid method.[Result]The optimal condition for the detection of Poria polysaccharide is when the wavelength detected was 490 nm,the dosage of 5% phenol was 1.2 mL; the dosage of concentrated sulfuric acid was 7.0 mL; the reaction time was 30 min and the temperature was 45℃ .At the same time,10 batches of samples from 8 different areas were determined by this method, result shows that the overall sample content in the products of Anhui Jinzhai is higher than that of Hubei Luotian and Yunnan Wuding,indicating the good quality of Poria cocos at Jinzhai.[Conclusion]This method can be used to monitor the quality of Poria polysaccharide in medicinal material market.

  • Antifungal Effect of Essential Oils from Five Medicinal Herbs on Candida albicans

    [Objective]To investigate the antifungal effect of essential oils from 5 medicinal herbs on Candida albicans.[Method]By adopting tube disk approach,the essential oils of cinnamon bark,palchouli,clove,fennel and Schisandra rosea were respectively diluted to 2.5-0.019 5 μL/mL by tiraes ratio dilution for MIC and MFC experiments on pathogenic Candida albicans strains (Monilia albican,Candida tropical,Candida krusei) from intestinal tract,lung and kidney.[Results]The MIC and MFC of cinnamon bark oil against Candida albicans were 0.039 and 0.078 μL/mL respectively; those of clove oil against Candida albicans were 0.156 and 0.312 5 μL/mL respectively; those of palchouli oil against Candida albicans were 0.313 and 0.625 μL/mL respectively; those of fennel oil oil against Candida albicans were 0.156 and 0.312 5 μL/mL respectively.The MIC of Schisandra rosea oil against Candida albicans (Monilia albican,Candida krusei) was 0.312 5 μL/mL,that against Candida tropical was 0.625 μL/mL,and the MFC of Schisandra rosea oil against Candida albicans was 0.312 5 μL/mL.For the control group (fluconazole) ,the MIC of Schisandra rosea oil against Candida albicans (Monilia albican,Candida tropical) was 0.312 5 μL/mL,and that against Candida krusei was 2.5 mg/mL.The essential oils from five medicinal herbs all showed strong antifungal effect against Candida albicans strains (Monilia albican,Candida tropical,Candida krusei).The antifungal activity was determined in turn as cinnamon bark oil > clove oil > palchouli oil > fennel oil > Schisandra rosea oil.Candida krusei showed drug resistance to fluconazole.[Conclusion]The essential oils extracted from natural medicinal herbs are effective in treating invasive fungal infection caused by Candida albicans,moreover,this does not bring drug resistance to Candida albicans.

  • Electrophysiology Effects of Cestrum nocturum n-Butyl Alcohol Extract on Isolated Guinea Pig Atria

    [Objective]To study the electrophysiology effects of Cestrum nocturumn n-Butyl Alcohol Extract (CNAE) on isolated guinea pig atria.[Methods] The conventional experimental methods for electrophysiology of isolated guinea pig left and right atria were adopted,such as CNAE on contractility,spontaneous beat of right atrium,function refractory,post-rest potentiation and positive staircase phenomenon of left atrium were observed in isolated guinea pig.[Result] CNAE (0.1,0.1 × 3,1,1 × 3,10.0,10 × 3,100 mg/mL) prominently decreased spontaneous beat of right atrium.As for the contractibility of right atrium,it had a effect of biphasic manner,CNAE at a lower concentration (0.1,0.1 × 3,1 mg/mL) enhanced the contraction of left atria in a concentration-dependent manner,but at a higher concentration (1 × 3, 10.0,10 × 3,100 mg/mL) it exerted an inhibitory effect.And CNAE possessed the effect of depressing the excitability via increasing the threshold of excitability.And CNAE at a higher concentration (50.0,100.0 mg/mL) could shorten the function refractory period and restrain the positive staircase phenomenon of isolated guinea pig left atrium,but it had no effect on the post-rest potentiation phenomenon of myocardial contractility in left atrium.[Conclusion]The pharmacological effect of CNAE on isolated guinea pig atria may be related to the mechanism by restraining the voltage-dependent calcium channel (VDC) and natrium channel,or promoting the Potassium channel.

  • Studies on Inhibitory Effect of Eighteen Kinds of Chinese Herbal Medicines against Escherichia coli

    [Objective]The purpose was to investigate the bacteriostatic effects of eighteen kinds of Chinese herbal medicines including HER-BA PYROLAE,MAHONIA BEALEI CARR,FRUCTUS ROSAE LAEVIGATAE,and RADIX RHIZOMA RHEI.[Method]Water decoction method was adopted to extract the active ingredients from Chinese herbal medicine,whose bacteriostatic effect was then determined by filter pa-per tablet method with Escherichia coli as the indicator bacteria.Double dilution method was used to determine the minimum inhibition concen-trations of four kinds of Chinese herbal medicine.[Result] Among the eighteen kinds of Chinese herbal medicine,the extracts from fifteen kinds of medicine had the inhibitory effect on Escherichia coli.The minimum inhibition concentration of HERBA PYROLAE was 31.25 mg/mL, while that of MAHONIA BEALEI CARR,FRUCTUS ROSAE LAEVIGATAE,and RADIX ET RHIZOMA RHEI was all 62.50 mg/mL.[Conclusion]The HERBA PYROLAE showed a good inhibition effect against Escherichia coli.

  • The "Dose-Time-Toxicity " Relationship of Mice Hepatotoxicity Caused by the Administration of Water-extract Component of FRUCTUS EVODIAE after Multiple Doses

    [Objective]To research the " dose-time-toxicity" relationship of mice hepatotoxicity caused by the administration of water-extract component of FRUCTUS EVODIAE after multiple doses.[Methods]Mice were given water-extract component of FRUCTUS EVODIAE by gavage at different time points or in different doses. The situation of death and toxic reaction of mice were observed. The serum ALT and AST activities at the same time point after administration were examined. The liver,renal and viscera indexes were calculated. And the morphological changes of liver tissue were observed under optical microscope.[Results]ALT and AST levels in mice serum and liver tissue enhanced at 1 d after administration; hepatotoxicity became significant at 3 d after administration,and it remained at 7 d. Compared with normal group, 0.63 5.0g/kg dose range of the water-extract component of FRUCTUS EVODIAE could cause significant hepatotoxicity injury within 7 d of administration; ALT,AST,AKP and TBI enhanced; ALB reduced; liver weight/body weight enhanced; edema of liver cells in different degrees was detected,as well as the fatty degeneration and hyperaemia among tissues,which showed certain dose-toxicity and time-toxicity relationships. These changes became serious with the increase of dose. And different dose groups had significant differences with normal control group.[Conclusion] Administration of water-extract component of FRUCTUS EVODIAE after multiple doses would cause significant mice hepatotoxicity,showing certain " dose-time-toxicity" relationship of mice hepatotoxicity.

  • Determination and Analysis of Gross Protein in Wild Pteridium aquilinum var.latiusculum

    Kjeldahl determination was adopted to determine the gross protein content in the wild Pteridium aquilinum var.latiusculum grown in Yuping,Tongren,Guizhou.The test results indicated that the gross protein content in the Rhizomes,Petioles and Blades of bracken were 6.89% ,28.19% and 33.01%.It is suggested that rational exploration and utilization be enhanced based on the protection of wild bracken.

    [Objective]To study the effect of curcumin on the proteome of HepG2 cells by using two-dimensional electrophoresis and mass spectrum.[Methods] The proteins from HepG2 cells and HepG2 cells treated with 30 μmol/L curcumin were separated with two-dimensional electrophoresis and the gels were scanned after silver staining.All images were analyzed by Image Master 6.0 software,and the different pro-teins were identified by mass spectrum.[Result] The expression of protein disulfide isomerase,replication protein A2,carnitine deficiency-associated gene and cyclin-dependent kinase inhibitor 2A increased in the HepG2 cell treated with 30 μmol/L curcumin,while the expression of PCNA,ER-60 protease,Ran binding protein 1,Stathmin 1/oncoprotein 18,Nm23 protein decreased.[Conclusion] Curcumin could in-hibit tumor cell growth,interfere cell cycle,promote apoptosis,inhibit tumor invasion and metastasis,and promote cell repair.In addition,the hypolipidemic and anti-atherosclerosis effects of curcumin may correlate with promotion of the transmembrane transport of fatty acids and maintain of cell membrane stability.

  • Study on Establishment of the Clone of Centipeda minima

    [Objective]To study the tissue culture of the tender stem of Centipeda minima and provide an effective means to protect the wild C.minima resources.[Method]The tender stems of C.minima as samples were sampled for the induction and differentiation of adventitious buds,as well as the rooting,successive transfer culture,transplanting and plantation of tube seedlings.[Result]MS+0.4 mg/L 6-BA+0.1 mg/L NAA was the best medium for the direct induction and differentiation of adventitious buds from tender stems; MS+0.1 mg/L NAA+0.5 mg/L GA 3+0.6 mg/L 6-BA was the ideal medium for the proliferation of adventitious buds; the best method for the rooting of adventitious buds was to immerse the bottom incision of adventitious buds into 40 mg/L IAA solution for 6 min,and then inoculate it into the N 6 +0.2 mg/L NAA medium for rooting.The survival rate of transplanted and planted tube seedlings maintained at high level,and the planted tube seedlings maintained all botanical traits of wild C.minima resources.[Conclusion]An asexual clone system of C.minima is established for its artificial cultivation,which provides an effective means to satisfy the market demands and the protection of wild C.minima resources.

  • Effects of Dry Methods on the Content of Butylphthalide in Ligusticum chuanxiong Hort. Stems and Leaves

    [Objective]To study the effect of dry methods on the content of butylphthalide in Ligusticum chuanxiong Hort. stems and leaves (Miwu). [Methods]The content of butylphthalide in Miwu from different dry methods was determinated by HPLC. [Results]There was a good linear relationship between butylphthalide concentration and peak area when the sample injection was from 0.123 1 to 1.231 0 μg/mL (R=0.999 9),the quantification limit was 0.012 31 mg/mL,the average recovery was 100.1%,and RSD was 0.93% (n=9). Different dry methods had some influence on the butylphthalide content in Miwu. The content of butylphthalide was 0.024 84%,0.051 78% and 0.064 68% in tested samples of Miwu that were respectively obtained by sun-drying,shade drying and oven-drying after sunshine. As to Miwu,oven-drying after sunshine was superior to the above two methods. [Conclusions] Among the three drying methods,oven-drying after sunshine was the optimal one for Miwu.

  • Determination of Paeoniflorin and Albiflorin Contents in Different Germplasms and Parts of Paeonia lactiflora Pall.

    [Objective]To compare the content differences and distribution situation of paeoniflorin and albiflorin contents in different germplasms and parts of Paeonia lactiflora Pall. by using the same analysis method.[Method]HPLC method was adopted. Chromatographic column was LichroCART RP-C 18 (4 mm × 250 mm,5 μm);mobile phase was acetonitrile 0.1% phosphoric acid (14∶86,V/V); flow rate was 1 mL/min;detection wavelength was 230 nm; column temperature was 30 ℃ and injection volume was 10 μL.[Result]There were great differences in the paeoniflorin and albiflorin contents of different germplasms of P.lactiflora.The paeoniflorin contents in different underground parts of P. lactiflora were in the order of rhizome>fibrous root>main root; and the albiflorin contents were in the order of fibrous root>rhizome>main root. [Conclusion]The main factor affecting the contents of paeoniflorin and albiflorin was germplasm.And the paeoniflorin and albiflorin contents in RADIX PAEONIAE ALBA had small variation in authentic producing areas.

  • Optimization of Explants and Hormone Combinations for Callus Induction of Apocynum venetum L.

    [Objective]To investigate the effects of different media and hormone combinations on callus formation of apocynum.[Method] Using bud-containing stems,stems and roots of Apocynum venetum L as explants,6.5 g/L agar,27 g/L sucrose and four hormones (6-BA,KT,NAA,2,4-D) were added to three media (MS,B5,KC) separately,totally nine schemes designed finally.The induction and browning rate,growth potentials and inducement value of callus were adopted as investigation indices for optimizing the callus induction conditions.[Result]The parameters for callus induction of A.venetum were optimized as follows: bud-containing stems as materials,medium for callus induction as MS + 6-BA 0.6 mg/L + NAA 0.5 mg/L.Under the optimized conditions,the callus induction rate reached 100% .[Conclusion] This study provided reference for establishing the A.venetum L suspension cell lines and researching the secondary metabolites.

  • Determine Equilibrium Solubility and Oil-water Partition Coefficient of Dauricine by HPLC

    [Objective]To determine equilibrium solubility under different pH value and the oil-water partition coefficient of dauricine in Octanol buffer system.[Methods] To determine the concentration of dauricine by HPLC and the equilibrium solubility by saturation method;to determine dauricine's concentration in octanol and water phase so that its oil-water partition coefficient can be calculated.[Results]The concentration of dauricine increased with the addition of pH value at 37℃;the oil-water partition also increased with the addition of pH value; the logP value of dauricine was around 0.5 when pH value remained in the range of 6.8-8.0(equavelent to the pH condition inside small intestine),which indicated it has higher lipid solubility.[Conclusion] The equilibrium solubility and oil-water partition coefficient of dauricine were both under the influence of pH value;the methods established in this experiment was able to accurately determine the equilibrium solubility and oi-water partition coefficient of dauricine and to procast its interrior process.

  • Screening of the Effective Components from RHIZOMA COPTIDIS by Dialysis-HPLC

    [Objective]To screen the effective components from RHIZOMA COPTIDIS by dialysis-HPLC.[Method]Taking liposome as the simulated biomembrane,components in RHIZOMA COPTIDIS interacted with liposome was screened by dialysis-HPLC.Effects of liposome concentration and buffer pH value on the interaction between RHIZOMA COPTIDIS and liposome were investigated.[Result] There are 6 components in RHIZOMA COPTIDIS having significant interaction with liposome.Compared with the reference substances,the 6 components were columbamine,jatrorrhizine,epiberberine,coptisine,palmatine and berberine hydrochloride.[Conclusion] Dialysis-HPLC could effectively and rapidly predict the absorption situation of multi-components in RHIZOMA COPTIDIS,and could select the potential active components,aiming at providing references for the further research on the effective components in RHIZOMA COPTIDIS.
