  • Extraction and Determination of Polysaccharides from FRUCTUS CHAENOMELIS

    [Objective] To establish a method for the extraction and determination of polysaccharides from FRUCTUS CHAENOMELIS. [Methods] Ethanol reflux method was adopted to extract polysaccharides from FRUCTUS CHAENOMELIS. Then, the content of polysaccharides was determined by the method of C6H5OH phenol-H2SO4. [Results] The linear regression equation was Y=0.008 8X-0.010 9, R2=0.992 3. There was good linearity between concentration and absorbance of polysaccharides in the range of 1.0 to 5.0 μg/ml. Through twice extraction, the content of polysaccharides was 13.21% in defatted sample and 9.26% in original sample. The average recovery was 97.04% with RSD of 3.52%. [Conclusion] This method is simple, stable, precise and reproducible, which is suitable for extraction and determination of polysaccharides from FRUCTUS CHAENOMELIS.

  • Extraction, Identification and Content Determination of Polysaccharides from the Leaves of Prunus pseudocerasus Lindl.

    [Objective] To study the extraction, qualitative identification and quantitative assay of polysaccharides from the leaves of Prunus pseudocerasus Lindl. (PPP), so as to provide reference for its comprehensive application. [Method] Polysaccharides were extracted with the method of water exaction and alcohol precipitation after the leaves were defatted with 95% EtOH and petroleum ether. Then, the protein in PPP was removed by Sevag method. A series of experiments were used to identify the PPP, including Molish test (α-naphthol test), anthrone-sulfuric acid method test, fehling test, fourier transform infrared spectrometry (FT-IR), and iodine test. The content of PPP was determined by anthrone-sulphuric acid colrimetry method. [Result] P. pseudocerasus polysaccharides were grey white in color. The content of polysaccharides in P. pseudocerasus leaves was 29.7 mg/g. [Conclusion] The method is simple, sensitive, reproducible, and reliable.

  • GC-MS Analysis of the Essential Oils from Fresh and Dry Leaves of Litsea glutinosa (Lour.) C. B. Rob.

    [Objective] To research the chemical constituents of essential oils from fresh and dry products of Litsea glutinosa (Lour.) C. B. Rob. [Method] The essential oils in L. glutinosa were obtained by using steam distillation, and identified by GC-MS. [Result] A total of 33 compounds were identified from the oils of fresh leaves, accounting for 91.01% of the total oils. Among them, the main components were β-caryophyllene(22.83%), β-ocimene(7.19%), phytol(6.90%), β-pinene(6.79%), α-pinene(5.97%), caryophyllene oxide(5.95%) and so on. A total of 42 compounds were identified from the essential oils of dry leaves, representing 93.89% of the total oils. They were mainly the β-caryophyllene (23.22%), β-ocimene (7.78%), bicyclogermacrene (7.24%), α-caryophyllene (5.89%), α-pinene(5.42%) and so on. Chemical composition analysis of the essential oils showed that there were only 30 common compounds in two kinds of essential oils. [Conclusion] This method was accurate and reliable. This research provided scientific basis for the establishment of quality standard, as well as the further development and utilization of L. glutinosa.

  • Determination of the Total Flavonoids in FLOS LONICERAE in Hainan Province

    [Objective] To detect the content of total flavonoids in flowers (FLOS LONICERAE) and leaves of Lonicera confusa. [Method] UV-Vis spectrophotometry was used to detect the total flavonoids contents in L. confusa leaves and flowers. With the extraction rate of total flavonoids as the index, conditions affecting the extraction rate of total flavonoids were preliminarily discussed. [Result] The optimal extraction condition for total flavonoids was as follows: 60 min extraction time, 1∶30 solid-liquid ratio, and 70% ethanol concentration. The contents of total flavonoids in L. confusa flowers and leaves were 5.655% and 3.459%, respectively. [Conclusion] L. confusa flowers and leaves were relatively rich in total flavonoids, which had great utilization value for Caprifoliaceae resources in southern China.

  • Separation and Purification of Polyphenols from Camellia semiserrata Seed Cake with Macroporous Resin and Their Antimicrobial Activity

    [Objective] To separate and purify polyphenols from Camellia semiserrata seed cake with macroporous resin and to study their antimicrobial activity against anthracnose. [Method] Six macroporous resins were used to separate and purify polyphenols from C. semiserrata seed cake using static and dynamic adsorption. [Result] The desorption rate of X-5 resin was higher than that of other types of macroporous resins. The polyphenols purity reached 44.9% after having been purified by X-5 resin. The polyphenols significantly inhibited the growth of plant pathogenic anthracnose hypha. [Conclusion] X-5 resin is superior to other types of macroporous resins in separating and purifying polyphenols from C. semiserrata seed cake. Polyphenols from C. semiserrata seed cake have significant antimicrobial activity against anthracnose.

  • Study on the Processing Technology of Fresh Tubers from C. yanhusuo in Production Place

    [Objective] To establish the processing technology of fresh tubers from Corydalis yanhusuo W.T.Wang in production place. [Method] Different boiled processing time test, water steam test, sliced sun-dried test, sulfur fumigation test or other processing test were compared with the traditional processing methods. The best processing method was determined based on the content of effective constituents, extract content and drying rate. [Result] The content of effective constituents in steam system processing samples and boiled samples presented the trend of decrease with the increasing of time, and both of them were higher than the traditional process. The content of water-extract in steam system processing was higher than that in boiled processing, but there was no remarkable difference between the boiling and the traditional processing. [Conclusion] Fresh tubers from C. yanhusuo can be processed by steaming or boiling, but steaming is better. And the steaming or boiling time should not last for long time, otherwise it will result in the loss of active ingredients, and non-white inner core is used as the gold standard in steaming or boiling processing.

  • Content Determination of β-elemene in Lantana camara Linn in Panxi Area of Sichuan Province of China

    [Objective] To establish HPLC analyses method for the β-elemene content in L. camara; and to detect the β-elemene content and distribution law of malignant weed L. camara in Panxi Area of Sichuan Province of China. [Method] HPLC method was used to detect the β-elemene content in L. camara. Eclipse XDB-C18 (4.6 mm × 150 mm, 5 μm) chromatographic column was adopted. The flow rate was 1.0 ml/min; detection wavelength was 210 nm; injection volume was 20 μl; column temperature was 30 ℃; and mobile phase was ethanol-acetonitrile-water (V/V/V, 70∶ 10∶ 20). Ultraviolet ultraviolet spectrophotometer and TLC were both used for the analyses of β-elemene in sample. [Result] β-elemene content in L. camara was 0.007 5-0.120 0 mg/ml by HPLC method, showing good linear relationship (r=0.999 5). And the average recovery rate was 99.3%.There was β-elemene in both the leaves and flowers of L. camara. And β-elemene content was the highest in leaves. β-elemene contents in fresh leaves and natural drying leaves were 0.025% and 0.083%, respectively. There were no significant differences between fresh leaves and natural drying leaves; but the two were significantly higher than that in 60 ℃ drying leaves. [Conclusion] This method was suitable for the content determination of β-elemene in L. camara. It also provided references for the development and utilization of L. camara, and offered new sources for the extraction of L. camara.

  • Effects of Microwave Processing on Ester Type Alkaloids from RADIX ACONITI LATERALIS PRAEPARATA

    [Objective] To discuss how the content of single and double ester type alkaloids was influenced by microwave processing. [Method] Effects of hot bleaching time (A), drying temperature (B) and microwave time (C) on the content of ester type alkaloids were discussed by orthogonal design, intuitive analysis, and variance analysis. [Result] Microwave time (C) had a very significant influence on the content of double ester type alkaloids, P<0.01. Microwave time (C) had a significant influence on the content of single ester type alkaloids, P<0.05. Bleaching time (A) and drying temperature (B) only had significant influences on the content of double ester type alkaloids, P<0.05. The optimal combination of factors was A3B3C3: hot bleaching time 3 min, drying temperature 60 ℃ and microwave time 5 min. Under above conditions, the content of single and double ester type alkaloids was 802.14 and 590.33 μg/g, respectively. [Conclusion] Microwave processing could reduce the content of double ester type alkaloid, and increase the content of single ester type alkaloid.

  • Study on the Extraction Process of Alkaloids from HERBA LOBELIAE CHINENSIS with Compound Enzymes under Ultrasonic Wave

    [Objective] To study the optimal technical conditions of extracting alkaloids from HERBA LOBELIAE CHINENSIS with complex enzymes under ultrasonic wave. [Method] With ethanol as the solvent, single factor experiment and orthogonal test method were employed to test the effects of five factors on extraction of alkaloids, including cellulase dose, pectinase dose, extraction time, pH value, and ethanol concentration. [Result] The optimal extraction procedure was as followed: cellulase 24 mg, pectinase 45 mg, extraction time 30 min, enzymatic pH 5.4, and ethanol concentration 70%. [Conclusion] High in extraction rate and good in stability, the method favored the determination of alkaloids from HERBA LOBELIAE CHINENSIS.

  • Study on the Extraction and Purification Process of Esculetin from CORTEX FRAXINI

    [Objective] To study the optimal extraction and purification process of esculetin from CORTEX FRAXINI. [Method] Using the content of esculetin measured by HPLC as the index, the optimal process of extracting esculetin was determined by L9(34) orthogonal test. With the isolation of esculetin as the index, the optimum isolation and purification process was determined by polyamide column. [Result] The optimal extraction process was extracting three times, each extraction 1.5 h, with eight-fold amount of 95% ethanol. Polyamide column could effectively isolate esculetin by following the steps below: the test solution was loaded on polyamide column at an adsorption flow rate of 8 BV/h and eluted with distilled water at an elution flow rate of 4 BV/h. The volume ratio of loading solution to resin was set at 8∶ 1 (g/ml) (W/V). Under the above conditions, the total elution rate and refining rate of esculetin were more than 90% and 50%, respectively. [Conclusion] The method used in the study is good for extracting esculetin from CORTEX FRAXINI.

  • Study on Microwave Extraction Technology of RADIX PAEONIAE RUBRA Coupled with RADIX SCUTELLARIAE

    [Objective] To discuss the microwave extraction technology of paeoniflorin and baicalin from RADIX PAEONIAE RUBRA coupled with RADIX SCUTELLARIAE. [Method] Using ethanol solution as solvent, microwave extraction technology was adopted to extract paeoniflorin and baicalin from RADIX PAEONIAE RUBRA coupled with RADIX SCUTELLARIAE. And the content of paeoniflorin and baicalin was tested by HPLC. Then the effects of microwave power, charging pump frequency, volume fraction of ethanol, solid-liquid ratio and soaking time on the yield of paeoniflorin and baicalin were investigated by means of orthogonal design and single factor test. [Result] The optimal microwave-assisted extraction technology of paeoniflorin and baicalin was soaking time 9 h, microwave power 36 kW, charging pump frequency 9 Hz, volume fraction of ethanol 50%, and solid-liquid ratio 1∶12 (g/ml). The yield of paeoniflorin and baicalin was 1.84% and 2.91% respectively under the above technology, which was higher than that of ultrasonic method and reflux method. [Conclusion] Microwave extraction method is superior to ultrasonic method and reflux method, which has widespread application prospects in extraction of active constituents from Chinese medicinal herbs.

  • Study on the Extraction Technology of Triterpenoids from Ganoderma applanatum (Pers.) Pat.

    [Objective] To study the extraction technology of triterpenoids from Ganoderma applanatum (Pers.) Pat. [Method] Microwave-assisted extraction (MAE), ordinary extraction, ultrasonic extraction and reflux extraction were used to study on triterpenoids from G. applanatum. By single factor test and central compose design, the extraction technology of triterpenoids was optimized. In addition, the effects of different methods on the extraction efficiency of triterpenoids were compared. [Result] The optimal extraction technology of triterpenoids was as follows: G. applanatum was extracted with 75% ethanol at the solid-liquid ratio of 1∶20 (g/ml) for 15 min at 75 ℃. Under these conditions, the average extraction rate of triterpenoids was 0.748%. It showed that the extraction efficiency of ordinary extraction, reflux extraction and ultrasonic extraction were respectively 74%, 80%, and 83% of microwave extraction by comparing the different methods. [Conclusion] MAE is a better extraction method for the content determination of triterpenoids in G. applanatum that has the advantages of fewer solvent, shorter time and higher extraction efficiency.

  • Extraction of Anthraquinone Derivatives from RADIX ET RHIZOMA RHEI by Continuously Biphasic Hydrolysis Extraction

    [Objective] To optimize the extraction technology of anthraquinone derivatives from RADIX ET RHIZOMA RHEI by continuously biphasic hydrolysis extraction. [Method] Anthraquinone derivatives were extracted from RADIX ET RHIZOMA RHEI by continuously biphasic hydrolysis extraction. Content of anthraquinone derivatives was detected by UV Spectrophotometry. With the total content of anthraquinone derivatives as the index, L9(34) orthogonal test was conducted in order to select he optimal extraction technology. [Result] The optimal extraction technology was as follows: 1∶10 solid-liquid ratio (W/V, g/ml), 25 mol/ml sulfuric acid solution, and 5 h extraction time of biphasic hydrolysis. [Conclusion] This method was simple and repeatable.

  • ASS Determination of Iron Black in RADIX POLYGONI MULTIFLORI PREPARATA CUM SUCCO GLYCINES SOTAE

    [Objective] To establish an atomic absorption spectrophotometry (AAS) method for determination of iron black in RADIX POLYGONI MULTIFLORI PREPARATA CUM SUCCO GLYCINES SOTAE. [Method] Through ferric salt chemical reaction, the content of iron black was determined by AAS with standard solution of iron as control. Then experimental results were verified by ICP-MS. [Result] AAS method was specific for determining the content of iron black in RADIX POLYGONI MULTIFLORI PREPARATA CUM SUCCO GLYCINES SOTAE. The calibration curve of iron atoms showed good linearity in the range of 1.5 to 15 μg/ml. The average recovery rate was 96.21% with RSD of 1.3% (n=6). And the experimental data were similar to that determined by ICP-MS. [Conclusion] Some tested samples of RADIX POLYGONI MULTIFLORI PREPARATA CUM SUCCO GLYCINES SOTAE in the market contain chemical dye iron black. AAS is reliable, simple and easy, which can be used for determining the content of iron black.

  • Effects of Polygonum bistorla L.-413 on the Relaxant Mechanism of Isolated Thoracic Aortic Rings in Rats

    [Objective] To research the effects of Polygonum bistorla L.-413 on the relaxant mechanism of isolated thoracic aortic rings in rats. [Method] Effects of P. bistorla-413 on the contraction of vascular smooth muscle induced by norepinephrine (NA) were studied under the Ca2+-contained and Ca2+-free bath solutions. Time-dependent manner of the relaxing effects of P. bistorla-413 was investigated. Effects of P. bistorla-413 on the contraction of vascular smooth muscle induced by 40 and 80 mmol/L KCl solutions were researched. [Result] P. bistorla-413 had relaxing effects on the contraction of vascular smooth muscle induced by NA in a concentration-dependent manner. P. bistorla-413 (100 μmol/L) reached the maximum relaxing effects at 30 min; P. bistorla-413 in Ca2+-free group had greater inhibitory effects on the contraction of vascular smooth muscle induced by NA than that in Ca2+-contained group. P. bistorla-413 had inhibitory effects on the contraction of vascular smooth muscle induced by 40 and 80 mmol/L KCl solutions. And the dose-response curve of the two obviously moved up. [Conclusion] P. bistorla-413 had relaxing effects on the vascular smooth muscle. And the action mechanism might be that P. bistorla-413 could promote the synthesis and release of NO, open the calcium-activated potassium channels, and inhibit calcium influx and inner calcium release of smooth muscle cell.

  • Extraction and Content Determination of Flavonoids in Sonchus oleraceus L.

    [Objective] To study the extraction methods of flavonoids from Sonchus oleraceus L. and the variations of their contents with growth months and parts. [Method] The contents of flavonoids in S.oleraceus grown at different months and their distributions at different parts of S. oleraceus were detailed studied in this article. The extraction process of total flavonoids from S. oleraceus leaves was optimized through orthogonal design method. [Result] It was discerned from the experiment that the extraction solvent concentration> solid-liquid ratio>extraction time. The optimized extraction condition was that the solid-liquid ratio was 1∶20 (g/ml), extraction solvent was 75% alcohol and extraction time was 2 h. The flavonoids contents in different parts were flower>leaf>stem>root. Flavonoids in S. oleraceus L collected in April were higher than that of other months. [Conclusion] The result is valuable for further development and utilization of S. oleraceus resources.

  • Effect of Volatile Oil from Bupleurum chinense DC. on Energy Metabolism of the Liver in Rats

    [Objective] To explore the energy mechanism of volatile oil from Bupleurum chinense DC. for hepatotoxic injury. [Method] Volatile oil from B. chinense at the dose of 0.19, 0.28 and 0.42 ml/kg was given to rats by intragastric administration for 15 d continually. The changes in respiratory function, ATP content and ATPases activity of hepatic mitochondrion were detected. [Result] Compared with normal control group, the respiratory control rate (RCR) and phosphorus/oxygen rate (P/O) reduced from (1.76±0.28)% and (1.28±0.38) to (0.76±0.12)% and (0.71±0.21) in volatile oil from B. chinense 0.42 ml/kg group respectively, and the respiratory oxygen uptake were reduced from (2. 82±0. 64) to (1.04±0.23) mol/min·g. ATP content was reduced from (1.00±0.18) to (0.72±0.13) μmol per gram of protein, and activities of Na+-K+-ATPase, Mg2+-ATPase and Ca2+-ATPase from (0.94±0.14), (0.95±0.14) and (0.97±0.16) mmol/h·g protein to (0.53±0.08), (0.67±0.10) and (0. 65±0.11) mmol/h·g protein, respectively. Compared with normal control group, RCR, P/O, respiratory oxygen uptake, ATP content and ATPases activities in volatile oil from B. chinense 0.28 and 0.19 ml/kg groups were also significantly reduced.[Conclusion] Volatile oil from B. chinense can cause hepatotoxicity in rats by inhibiting function of hepatic mitochondrion and influencing the energy metabolism of the liver.

  • Isolation of Endophytic Fungi with Acetylcholinesterase Inhibitory Activities from Huperzia serrata

    [Objective] To isolate endophytic fungi with acetylcholinesterase (AChE) inhibitory activities from Huperzia serrata(Thunb.)Trev. [Method] Stems, leaves and roots of H. serrata were collected from the open field. Then, they were inoculated on PDA culture medium after the surface sterilization. Based on the isolation of endophytic fungi, shaking flask fermentation was conducted; and fermentation products were extracted by ultrasound. Inhibitory effects in vitro of fermentation products on AChE were detected by DTNB method. [Result] A total of 194 endophytic fungi were isolated from the roots, stems and leaves. After the fermentation in potato glucose liquid medium (PDB), in vitro AChE inhibitory activity was detected in endophytic fungi fermentation extracting liquid. 31 endophytic fungi showed in vitro AChE inhibitory activity. Among them, 13 strains had relatively strong inhibitory activity, with the inhibitory rate being 50.2%-80.5%. 9 strains were isolated from the H. serrata stems; and 4 strains were isolated from leaves. [Conclusion] Endophytic fungi could be isolated from the H. serrata. The fermentation products of the endophytic fungi showed relatively strong in vitro inhibitory activity to AChE, which might be one of the effective approaches to find the inhibitory drugs of AChE.

  • Study on the Seed Germination Characteristics of the Medicinal Plant Bergenia purpurascens (Hook. F. et Thoms.) Engl.

    [Objective] To study the effects of light, temperature and pH on the germination of Bergenia purpurascens (Hook. F. et Thoms.) Engl. seeds. [Method] The purity, thousand seed weight, after-ripening, and germination characteristics of B. purpurascens seeds were tested by conventional determination methods for crops. [Result] No after-ripening dormant characteristics were found in B. purpurascens seeds. The most suitable temperature was 25 ℃ for germination. They had wide adaptability to the solution pH from 5 to 11. And the germination rate gradually increased as the pH rose. [Conclusion] In natural state, B. purpurascens is mainly propagated by the clustered underground rhizomes along surface soil layer but not by seeds due to inappropriate external natural environment. In suitable environment of cultivation, the germination rate of seeds could reach 86.67%. It is feasible to domesticate B. purpurascens.

    [Objective] To survey the optimal harvesting period of Pinellia ternata (Thunb.) Breit. and to establish the matching filed planting and management technique system for tissue culture and rapid propagation of P.ternata. [Method] The optimal harvesting period was discussed by comparing drying rate and tuber of P.ternata from different harvesting periods. [Result] The optimal harvesting period of tissue culture P.ternata was in mid- to late-September. [Conclusion] This study can provide theoretical basis for cultivation and harvesting techniques of tissue culture seedlings and lay foundation for protection and utilization of P.ternata resource.

  • Study on Germination Characteristic of Peucedanum praeruptorum Dunn. Seeds

    [Objective] To study the breeding characteristics of Peucedanum praeruptorum Dunn., the original plant of Chinese medical herb RADIX PEUCEDANI, so as to provide reference for domestication and large area industrialized cultivation of wild P. praeruptorum. [Method] Four different storage methods were used to treat P. praeruptorum seeds. In group A, the seeds in a kraft bag were stored under natural conditions. In group B, the seeds in a kraft bag were stored in a dryer at room temperature. In group C, the seeds sealed in many layers of plastic bag were stored at 4-5 ℃ in a refrigerator. In group D, the seeds sealed in many layers of plastic bag were stored at -10 ℃ in a refrigerator. 240 and 460 days later, the germination rate of seeds in each group was measured. Meanwhile, seeds stored for more than 15 months were soaked in different plant growth regulators (A. Soaked for 8 h in 200 mg/L GA at 25 ℃; B. Soaked for 8 h in warm water at 25 ℃; C. No soaking treatment). [Result] Within 240 days, the germination rate of P. praeruptorum seeds stored by four methods ranged from 63%±5% to 77%±5%. Among them, the seeds which were stored at 4-5 ℃ in a refrigerator had the highest germination rate. Within 460 days, the germination rate of seeds was between 0 and 62%±1%, but those stored under natural conditions had been entirely inactive. And the germination rate of refrigerated seeds was the highest at 62%±1%. From soaking tests of seeds, the germination rate of seeds treated by GA was 70%±2%, while that of untreated seeds was only 35%±88%. [Conclusion] Storage methods affect the germination rate of P. praeruptorum seeds directly. If the interval between sowing and harvesting is less than half a year, the seeds will not need special storage treatment, otherwise, they should be stored in a refrigerator. It is better to soak the refrigerated seeds prior to germination tests.

  • Effects of Different Hormone Pretreatments on the Germination of Mentha arvensis (Peppermint) Seeds

    [Objective] To study the effects of different hormone pretreatments on the germination of Mentha arvensis (peppermint) seeds. [Method] Effects of different hormones on the germination of peppermint seeds were studied, including GA3, ethephon, NAA, and 2, 4-D. [Result] The germination rate and germination potential of peppermint seeds were gradually increased with GA3 concentration increasing from 40 to 160 mg/ml, but from then on, they began to decline apparently. And its optimum concentration was between 160 and 200 mg/ml. Ethephon could effectively promote the germination of peppermint seeds and improve the germination rate when being used in concentrations between 40 and 100 mg/ml, but that used in concentrations above 100 mg/ml evidently inhibit germination. When the NAA concentration was increased from 0.08 to 0.1 mg/ml, the germination rate and germination potential were gradually increased, that is to say, NAA in appropriate concentrations could significantly promote the germination of peppermint seeds and improve the germination rate. If NAA concentration continued to rise, there would be a significant reduction in germination rate and germination potential (P<0.05), confirming the presence of inhibitory effects for NAA in higher concentration above 0.5 mg/ml. Similarly, 2, 4-D used in concentrations between 10 and 30 mg/ml could promote the seeds germination and improve the germination rate, while that in higher concentration above 50 mg/ml inhibited germination. The germination rate and germination potential were gradually increased with 2, 4-D concentration increasing from 10 to 30 mg/ml, but they began to significantly decline if concentration continued to rise (P<0.05). [Conclusion] GA3, ethephon, NAA, and 2,4-D have different effects on the germination of peppermint seeds.

  • Study on Optimal Fertilizer Techniques for Standardized Cultivation of Plantago asiatica L. in Jiangxi Province

    [Objective] To study the optimal fertilizer techniques for standardized cultivation of Plantago asiatica L. [Method] Through the combination design of quadric rotation, the regression model of its production and the four elements like N, P, K and organic fertilizers was set up, the optimization of the mixed fertilization was determined by the application of DPS data base. [Result] The regression model was founded as: =2 937.44+105.46X1+17.06X2 +62.69X3 +265.18X4 -251.17X12 -80.01X22 -150.62X32 -55.67X42 +22.75X1X2 +22.75X3X4. Using 3 450 kg/hm2 as the critical value of system optimization scheme, the optimal mixed fertilization was the following: N fertilizer (urea) 247.65-314.81 kg/hm2, P fertilizer (calcium superphosphate) 346.05-553.95 kg/hm2, K fertilizer (potassium sulfate) 213.3-340.425 kg/hm2, organic fertilizer (digest pig manure) 32 737.5-35 782.5 kg/hm2. [Conclusion] The fitting degree of this formula is good enough to reflect the relationship between its yield and N, P, K and organic fertilizers.

  • Morphology Research on the Individual Growth of Silybum marianum

    [Objective] To study the morphology characteristics of the individual growth of Silybum marianum (L.) Gaertn. [Method] Seedling materials were cultivated in laboratory; young plants were planted in open filed. The external morphology and growth rhythm of S. marianum were observed and recorded. Then, hand-held slicing method and temporary slice method were used to observe the structures of fresh leaves and roots. Morphological characteristics of seedling, roots, stems and leaves were researched by conventional paraffin section technique. [Result] As for the seed germination method, cotyledon was epigeal and shallow sowing should be adopted. Structure of nutritive organs of seedlings and plants was characterized by typical herbaceous dicotyledon structure. Synantherous stamen was a little shorter than the corolla lobe. And the growth period was 125-140 d. [Conclusion] The morphology characteristics of the individual growth of S. marianum were researched, which provided references for the standard cultivation of S. marianum.

  • Induction of Hairy Root in Scutellaria baicalensis Georgi and Optimization of Culture Conditions by Response Surface Methodology

    [Objective] To induce roots of Scutellaria baicalensis Georgia by Agrobaaerium rhizogenes strains 1.2556 and optimize culture conditions of hairy root using response surface methodology. [Method] Hairy roots were induced by coculture. Based on single factor experiments, culture time, temperature, shaking speed, inoculation amount were analyzed by Box-Benhnken design and response surface methodology. [Result] Hairy root free grew on hormone MS medium after infection by Agrobaaerium rhizogenes strains 1.2556, which showed the representative characteristic of hairy root. The optimal culture conditions of hairy root were as follows: culture time 24.95 d, temperature 27.4 ℃, shaking speed 123.69 r/mim, inoculation amount 0.500 g. The average yield of fresh weight of hairy root (fm) reached 9.271 28 g per bottle under optimal conditions. [Conclusion] This study optimizes the culture conditions of hairy root, which can provide basis for further development and utilization of S. baicalensis.

  • Simultaneous Determination of Eight Lignans in FRUCTUS SCHISANDRAE CHINENSIS from Different Habitats by HPLC

    [Objective] To determine the schisandrin, gomisin J, schisandrol B, gomisin G, schisantherin A, deoxyschisandrin, schisandrin B and schisandrin C of FRUCTUS SCHISANDRAE CHINENSIS from different habitats, including Liaoning Province, Hebei Province and Heilongjiang Province, and to supply a detection means for quality control of FRUCTUS SCHISANDRAE CHINENSIS. [Method] Methanolic extract of FRUCTUS SCHISANDRAE CHINENSIS was separated on the chromatographic column Elite ODS-C18 column (250 mm×4. 6 mm, 5 μm) with gradient mobile phase of acetonitrile (A)-water ( B) (50% of A at 0-17 min; 50%-55% of A at 17-25 min; 55%-75% of A at 25-30 min; 75% of A at 30-35 min; 75%-65% of A at 35-40 min; 65%-50% of A at 40-45 min) at the flow rate of 1.0 ml/min. The column temperature was maintained at 30 ℃ and the detection wavelength was set at 217 nm.[Result] Excellent chromatographic separation was achieved for eight studied lignans with good linearity (r>0.999 5) over the studied concentration ranges. The average recovery rates (RSD) (n = 9 ) were 99.75% (0.38%) for schisandrin, 100. 69% (2.31%) for gomisin J, 100.46% (1.39%) for schisandrol B, 99.87% (1.15%) for gomisin G, 100.22% (1.45%) for schisantherin A, 100.15% (0.99%) for deoxyschisandrin, 100.61% (0.25%) for schisandrin B and 101.31% (1.03%) for schisandrin C, respectively. [Conclusion] The HPLC method developed for simultaneous determination of eight lignans is simple and feasible, which can be used for simultaneous quantitative analysis of lignans in FRUCTUS SCHISANDRAE CHINENSIS.

  • A Study on the Establishment of Rapid Propagation System and Propagation Techniques of High-quality Seedling of Wolfberry (Lycium barbarum L.)

    [Objective] To study various factors that affect the rapid propagation of wolfberry (Lycium barbarum L.) in vitro and to establish the rapid propagation system of wolfberry superior strains with the purpose of breeding a large number of high-quality seedlings. [Method] Using Ningqi No.1 twigs as explants, the sterilization methods, lateral bud induction, subculture, rooting culture mediums of wolfberry were optimized. Meanwhile, the transplanting medium of test-tube plantlet were optimized. [Result] Explants were first soaked for 5-8 min in 500 mg/L penicillin injecta then sterilized. The optimized culture mediums were MS+ 0.5 mg/L 6-BA+ 0.1 mg/L IAA for lateral bud induction, MS+ 0. 5 mg/L 6-BA+1.0 mg/L IAA for subculture, and 1/2 MS+ 0.3 mg/L IBA for rooting culture. The transplanting medium of test-tube plantlet was the mixture of perlite and peat, in which the survival rate was 96%. [Conclusion] The establishment of rapid propagation system of high-quality wolfberry lays good technological base for accelerating breeding progress, breeding and promoting new improved varieties. It is feasible to breed high-quality seedlings of wolfberry through choosing superior strains.

  • Analysis on Chromosome Karyotype and C-banding Pattern of Eulaliopsis binata

    [Objective] To explore the chromosome karyotype and C-banding pattern of Eulaliopsis binata, so as to provide theoretical basis for studies of plant chromosome karyotype and origin of E. binata species. [Method] Fresh root tip of E. binata was treated by F-BSG method, and microscopy method was used to observe and record its chromosomes. All experimental data were analyzed statistically using the method proposed by Li Mao-xue. [Result] The total number of E. binata chromosomes was 2n=40, and the karyoutype formula was 2n=40=36m+4sm. The karyoutype type was 1B. The absolute length of chromosomes was between 2.37 and 6.34 μm with an average of 4.52 μm per chromosome. Study on C-banding of Eulaliopsis binata chromosomes was not reported as yet. F-BSG was used to show C-banding, and firstly acquire karyotype formula, which was 2n=40=22C/C+10W/C+4W/W+2IC/CI+2S/C. The absolute length of C-banding chromosomes was between 3.49-11.82 μm with an average of 7.46 μm per C-banding chromosome. [Conclusion] The study can provide theory basis for selecting new high-quality breeding and for studies of plant chromosome karyotype and E. binata origin.

  • Inhibiting Effects of Ilexgenin A from Ilex hainanensis Merr. on Tumor Cell HepG2

    [Objective] To investigate the proliferation inhibition and apoptosis in human hepatic cancer cell strain HepG2 induced by ilexgenin A (3α, 19β, 20β-trihydroxyurs-12-ene-24, 28-dioic acid) and its mechanism. [Method] HepG2 were treated by ilexgenin A with different concentration (20, 40, 60, 80 mg/ml) and time (24, 36, 48 h). Methylthiazolyl tetrazolium bromide (MTT assay), acridine orange (AO) fluorescence staining method, SP method of immunocytechemistry and flow cytometry (FCM) were used to determine the inhibiting effects on HepG2 cells, the cell cycle and apoptosis morphological changes and the expressions of bcl-2, Bax. [Result] Ilexgenin A inhibited the proliferation of HepG2 cells in dosage and time dependent manner. The HepG2 cells that were treated by ilexgenin A for 24 h. Ilexgenin A induced a G1 cell cycle arrest and reduced the cell population in S phase. The apoptosis rate of cells treated by 40, 60 and 80 mg/ml ilexgenin A were 7.89%, 8.37%, 9.93% respectively. With the increasing of the medicine density, the expression of bcl-2 was down-regulated, while the expression of Bax up-regulated in the apoptosis. [Conclusion] Ilexgenin A can inhibit the proliferation of HepG2 ceils and induced a G1 cell cycle arrest and reduced the cell population in S phase. It also can induce apoptosis of human prostatic carcinoma through down-regulating the expressions of bcl-2 and up-regulating the expression of Bax.
