• Scanning Electron Microscope Observation on Floral Organs and Leaf Epidermis of F. suspensa

    [Objective]To observe the morphological characteristics of floral organs and leaf epidermis of F.suspensa,so as to provide basic information for introduction,cultivation and taxonomy study of fine varieties.[Method]The pollen,stigma,style,upper and lower leaf epidermis of F.suspensa respectively adhered to the double faced adhesive tap of sample cup,observed,photographed and measured under scanning electron microscope(SEM).[Result]The pollen,whose extine surface is coated with netlike ornamentations and 3-colporate,is prolate in shape.Stigma is dichotomous branching and dry stigma.The stomatal apparatuses of style epidermis are anomocytic type.The upper and lower leaf epidermises have different cuticle ornamentations,both of which are covered by glands and without trichomes.In addition,the stomatal apparatus are restricted to the lower epidermis.[Conclusion]This method is accurate,reliable.It could provide theoretical basis for introduction,cultivation and taxonomy study of fine varieties of F.suspensa.

  • Research on Macroscopic and UV-Vis Spectrum Identification of Polygonum chinense L.

    [Objective]To study the pharmacognostic characteristics of Polygonum chinense L.[Method]A research was carried out on the morphological,macroscopic and UV-Vis spectrum identification of P.chinense.[Result]Viewed from cross section,the cortex is narrow,and phloem forms into a ring in the P.chinense root;the pericycle fibers are arranged in strips in stem;about 2 to 6 main vein vascular bundles are arranged in a ring,both sides of which have lignified fiber bundles;the leaves have round secretory cells and multicellular glandular hairs on upper epidermis; the multicellular glandular hairs,anisocytic stomata or anomocytic stomata are found on the lower epidermis of leaves;there are many glandular hairs,secretory cells,pollen grains,crystal fiber sheaths,brown compartmentalization,and starch grains in the crude powder of P.chinense;five kinds of P.chinense extracts all have absorption peaks at the wavelength of 200 to 800 nm in the UV-Vis spectrum.[Conclusion]Based upon morphological,macroscopic and UV-Vis spectrum identification,the pharmacognostic characteristics of P.chinense were researched.This study can provide a reference for identification,development,utilization and quality standard of P.chinense.

  • Changes of Nitrogen,Phosphorus and Potassium in Five-year-old Schisandra chinensis Leaves at Different Growth Stages

    [Objective]To study the changes of nitrogen(N),phosphorus(P)and potassium(K)contents in five-year-old Schisandra chinensis (Turcz.)Baill.leaves at different growth stages.[Method]Potassium bichromate titrimetric method-outside heating method was used to determine the soil organic matter.Alkaline hydrolyzation diffusion method was used to detect the available N.0.5 mol/L NaHCO3 extraction-Mo-Sb colorimetric method was used to detect rapidly available P;NH 4 OAC extraction-flame photometry was used to detect rapidly available K;H 2 SO4-H2 O2 method was used to digest the plant sample.Total N was detected by distillation titration method;total P was detected by vanadium-molybdenum-yellow colorimetry;total K was detected by flare photometer.Changes of N,P and K contents in five-year-old S.chinensis leaves at different growth stages were detected.[Result]S.chinensis leaves had the strongest absorption capacity to N in all growth period,followed with K.And P had the smallest absorption.N and K contents in S.chinensis leaves were relatively high in leaf-expansion stage;N and K contents fell smoothly from flowering stage to wilting stage.P content was relatively high in leaf-expansion stage and from late berry growth stage to early fruiting stage, and decreased greatly in late fruiting stage.[Conclusion]Artificial cultivation of S.chinensis needed enough base fertilizer and top-dressing of P was also needed at berry growth stage.

    [Objective]To study the high and stable yield cultivation techniques of Cistanche tubulosa(Schenk)Wight.[Method]In order to screen the optimum cultivation technique,the effects of planting mode,inoculation method and inoculation techniques on the inoculation rate of C.tubulosa were analyzed,based upon the results of field test and experiment.[Result]The success rate of inoculation was relatively higher when the C.tubulosa seeds with the purity more than 70%and the maturity more than 90%were sowed into soils at the wide-narrow row spacing of 0.7 ×3.0(m×m)or 1.0×1.0×3.0(m×m×m)with the inoculating depth of 60 to 70 cm and horizontal distance of 20 to 30 cm from March to June every year.[Conclusion]The success rate of C.tubulosa inoculation was influenced by inoculation method,inoculation distance,inoculation time and planting mode,which was also closely related with the inoculation depth and irrigation time.

  • Effects of Plant Hormone on Seed Germination of Gentiana rigescens Franch

    [Objective]To explore the effects of plant hormone on seed germination of Gentiana rigescens Franch.[Method]With the matured seeds of G.rigescens as test materials,effects of GA 3 ,6-BA and IAA on the soaking time and germination status of G.rigescens were studied. [Result]Seeds soaked in 200 mg/L GA 3+5 mg/L 6-BA+5 mg/L IAA showed the best germination effects and the germination rate reached more than 93%,the germination starting day and germination lasting day were relatively short,germination rate and germination energy were the highest. [Conclusion]Based on orthogonal test,effects of factors on seed germination of G.rigescens were in the order of IAA﹥GA 3 ﹥soaking time﹥6-BA. Plant hormone treatment could enhance the germination rate of G.rigescens seeds.

  • Survey on Pinellia ternata (Thunb.) Breit.Resource in China

    Pinellia ternata(Thunb.)Breit.is extensively distributed all over the country except for several provinces such as Inner Mongolia,Jilin, Heilongjiang,Xinjiang,Qinghai and Tibet.In recent years,contradictions among the increasing market demand,the decline in wild resource,and the far lagged cultivation technique are increasingly aggravated.Through literature review coupled with visiting survey,we perform a comprehensive and systematic research on the sprouting resources and production status of P.ternata.As a result,the wild P.ternata mainly focus on Sichuan, Hubei,Anhui,Yunnan,Guizhou and other provinces of China.Most parts of cultivated resources are found in Gansu,Shanxi,Shandong, Guizhou,Yunnan,etc.Limited by natural environment and local human economic,the reserve and yield of P.ternata resource have already declined sharply in China.Now,to protect and study P.ternata has become an urgent problem.To guide the rational use of drugs,researchers should confirm the producing areas of P.ternata resource and ensure the availability and safety of medicinal materials,based on the collection and screening of P.ternata resource.These could contribute to the collection,protection,production,development and utilization of P.ternata resource.

  • Determination of Flavonoids Content in Basella rubra L.Fruits by HPLC

    [Objective]To establish the determination method for flavonoids content in Basella rubra L.fruits.[Method]With rutin as the reference substance,chromatographic column was Eclipse Zorbax XDB-C 18 (4.6 mm×150 mm,5μm);mobile phase was anhydrous methanol-water (V/V,55∶45),flow rate was 0.8 ml/min;detection wavelength was 358 nm;and column temperature was room temperature.[Result]Under the chromatographic conditions,regression equation of rutin standard curve was Y=20.889X-17.147(R2=0.998 2).Linear regression was good between the concentration of 2.00 and 40.0μg/ml.The average recovery rate was greater than 96%and RSD was smaller than 2.0%.Flavonoids content in B.rubra fruits was 179.0μg/g by this method.[Conclusion]This method was simple,rapid,efficient,repeatable,accurate and reliable, and was suitable for the content determination of flavonoids in B.rubra fruits.

    [Objective]To explore the PCR-DGGE reaction system on rhizosphere soil microorganism for Panax ginseng C.A.Mey.[Method] Some important factors for PCR-DGGE amplification system including the concentration of DNA template,Mg2+,dNTPs and primers were optimized with 1-year-old P.ginseng soil as materials.[Result]An optimal system(25μl)was obtained as 0.8μg/μl DNA template,0.2 mmol/L dNTPs, 0.4μmol/L primer,and 1.5 mmol/L Mg2+.[Conclusion]The method is simple and fast,which can lay basis for further analyzing the genetic diversity of rhizosphere soil microorganism for P.ginseng.

  • Comparative Study on Curcumin of Curcuma Species from Nanpanjiang and Beipanjiang District, Guizhou Province

    [Objective]To study the content of curcumin in Curcuma species sampled from Nanpanjiang and Beipanjiang District,Guizhou Province,so as to provide scientific basis for quality evaluation of related medicinal materials.[Method]The content of curcumin,demethoxycurcumin and bisdemethoxycurcumin in the root and/or rhizome of 7 Curcuma plants was detected by HPLC.HPLC analysis was performed on an Inertsil ODS-3 column with detection wavelength at 420 nm.The mobile phase consisted of anhydrous methanol,isopropyl alcohol,water and acetic acid (V/V/V/V,20∶27∶48∶5),and the flow rate was 0.5 ml/min.[Result]The content of curcumin came from Anlong was higher than others,and the one came from Xingyi and Zhenfeng was lowest.Different original plants of RADIX CURCUMAE sampled from Xingyi varied greatly in the content of curcumin.Of these,the content of curcumin was the highest in RHIZOMA CURCUMAE LONGAE(3.27%),followed by RADIX CURCUMAE (0.005 4%)and RHIZOMA CURCUMAE(0.001 5%).[Conclusion]This method is simple and fast.The experimental result can provide scientific reference for quality evaluation of Curcuma medicinal materials from Nanpanjiang and Beipanjiang District,Guizhou Province.

  • Simultaneous Determination of Multi-components in RHIZOMA POLYGONI CUSPIDATI by HPLC

    [Objective]To develop a method for simultaneously determining multi-components(i.e.,polydatin,resverotol,emodin-8-O-β-D-glucopyranosid,emodin,chrysophanol and physcion)in RHIZOMA POLYGONI CUSPIDATI and to assess the quality of RHIZOMA POLYGONI CUSPIDATI growing in different locations.[Method]The HPLC separation was performed on a reversed-phase C 18 Hyperrsil column(250 mm×4.6 mm, 5μm)with a gradient solvent system of acetonitrile-1%aqueous phosphoric acid as mobile phase(20%-38%of acetonitrile at 0-30 min and 38%-100%at 30-70 min).The flow rate was 1.0 ml/min,and the detection wavelength was 284 nm.[Result]All calibration curves of the six components showed good linearity within a certain concentration range.The recoveries(n=5)were in the range of 95.00%to 105.00%.[Conclusion]The method is simple,accurate and reliable and not only enables the simultaneous determination of the content of the stilbenes and anthraquinones in RHIZOMA POLYGONI CUSPIDATI,but also provides a simple,feasible and characteristic quality evaluation method for the Chinese herb RHIZOMA POLYGONI CUSPIDATI.

  • GC-MS Analysis of Volatile Oils Extracted from Lysimachia christinae Hance by Different Methods

    [Objective]To study volatile oil extracted from the fresh and dry leaves of Lysimachia christinae hance by using steam distillation and blended solvent extraction.[Method]The chemical compositions of volatile oil were evaluated by GC-MS and the relative content of every compound was determined by peak area normalization method.[Result]The results showed that 19 and 14 kinds of compounds were identified from airdried and fresh leaves of L.christinae by using steam distillation,respectively.Whereas 20 and 11 kinds of compounds were identified from air-drie and fresh leaves of L.christinae by using blended solvent extraction,respectively.Compared with two methods,the highest relative content of volatile oil was β-myrcene using by steam distillation,which was 39.67%in the fresh leaves and 42.80%in air-drie leaves,respectively.However,the highest relative content of volatile oil was diethyl phthalate using by blended solvent extraction,which was 18.05%in the fresh leaves and 28.64% in air-dried leaves,respectively.[Conclusion]This result indicated that the compositions of volatile oil depended on not only different extraction,but also different processing methods.

  • Study on Chemical Constituents from the Ancient Ephedra Species Found in Yanghai,Xinjiang

    [Objective]To separate and identify the chemical constituents of the ancient Ephedra species and investigate their stability.[Method] The chemical constituents were extracted from the ancient Ephedra species by ethanol reflux method,further separated by column chromatography, and their structures were elucidated by spectral data analyses.[Result]Eight compounds were isolated from the ethanol extract of Ephedra species and identified as kaempferol-3-O-α-L-ramnopyranoside(compound 1),quercetin-3-O-β-L-rhamnopyranoside(compound 2),kaempferol-3-O-α-L-ramnopyranosyL-4″-O-E-(4-hyd-roxy)-cinnamoyl(compound 3),ciwujiatone(compound 4),tetradecanoyl-cholest-5en-3β-ol(compound 5),β-sitosterol(compound 6),octacosanol(compound 7),and hexacosanoic acid(compound 8).[Conclusion]Flavonoids from the ancient Ephedra species could stably preserve and indicate some level of phytochemical fidelity.

  • Determination of Puerarin and Paeoniflorin in Yanbao I Capsule by HPLC

    [Objective]To establish the method for determining puerarin and paeoniflorin in Yanbao I capsule.[Method]The HPLC analysis used chromatography glass column Diamonsil C18(2)(4.6 mm×250 mm,5μm)with the detection wavelength at 250 nm for puerarin and 230 nm for paeoniflorin and the column temperature was set at 25℃.Following the injection of sample solution,an elution was initiated at a flow rate of 1 ml/min with anhydrous methanol-water(V/V,28∶72)for puerarin and acetonitrile-0.1%phosphoric acid(V/V,14∶86)for paeoniflorin as mobile phase.[Result]The linear range of puerarin was from 0.001 18 to 0.007 08μg/ml(r=0.999 9).The average recovery of sample was 102.4%with RSD of 1.17%(n=6).The linear range of paeoniflorin was from 0.044 to 0.261μg/ml(r=0.999 9).The average recovery of sample was 97.8%with RSD of 1.71%(n=6).In Yanbao I capsule,the content of puerarin and paeoniflorin could reach 0.055 and 0.168 mg/g,respectively.[Conclusion]The method is simple,accurate and repeatable,which is suitable for determining the content of puerarin and paeoniflorin in Yanbao I capsule.

  • Bacteriostasis in Vitro of the Extracts from Rosa laxa Ratz.Fruits

    [Objective]To study the bacteriostasis in vitro of different solvent extracts from the fruits of Rosa laxa Ratz.and to find the antibacterial components.[Method]Effects of R.laxa fruit extracts by different polar solvents on the growth and biofilm formation of Streptococcus mutans and Escherichia coli were determined by microporosity plate method and dilution method.[Result]The ethyl acetate extracts from R.laxa fruits had certain inhibitory effects on the growth of Streptococcus mutans.Extracts from R.laxa fruits by five polar solvents could obviously inhibit the formation of biofilm of Streptococcus mutans Extracts from R.laxa fruits by five polar solvents had inhibitory effects on the biofilm formation of Escherichia coli,among which,chloroform and ethyl acetate extracts showed the strongest antibacterial effects.[Conclusion]Extracts from R.laxa fruits had relatively good inhibitory effects on Streptococcus mutans and Escherichia coli;and further research was needed.

  • Analysis of Chemical Constituents of Essential Oils from Stems,Leaves and Flowers (Fruits) of Fatsia japonica

    [Objective]To compare the chemical constituents of the essential oils from different parts of Fatsia japonica.[Method]The essential oils were extracted from stems,leaves,flowers(fruits)of F.japonica by steam distillation,and GC-MS was used to analyze the chemical constituents.The relative percentage of each chemical constituent was calculated using peak areas normalization.[Result]A total of 97 compounds were identified from three essential oils of F.japonica,and the main component was sesquiterpene,while following was aliphatic compounds.There were 30 kinds of the same composition,accounting for 64.55%in stems,77.90%in leaves,and 69.40%in flowers(fruits).[Conclusion]The essential oils from different parts of F.japonica have some differences in composition and content.

    [Objective]To detect the copper,lead,chromium,arsenic and mercury contents in RADIX ARDISIAE CRENATAE in different production areas of Guizhou Province.[Method]RADIX ARDISIAE CRENATAE was treated by microwave digestion;the contents of copper,lead,chromium,arsenic and mercury were detected by atomic absorption spectrometry.[Result]Except the chromium content exceeded the standard,contents of the rest metals were all lower than the limited amount in Green Standards of Medicinal Plants and Preparations for Foreign Trade and Economy.The over standard rate was 37.5%.[Conclusion]This method was simple,rapid,accurate and could effectively detect the contents of copper,lead,chromium,arsenic and mercury in RADIX ARDISIAE CRENATAE.

    [Objective]To study the analgesic effects and acute toxicity of essential oil from Helleborus thibetanus Franch.[Method]The analgesic effects of essential oil were tested by hot plate test and twisting body test;the modified Karber's method was used to evaluate LD 50 .[Result]The essential oil from H.thibetanus could significantly reduce the mice writhing response induced by acetic acid and enhance the mice pain threshold of hot plate stimulation.LD50 of essential oil from H.thibetanus was 8.50 ml/kg;and 95%confidence interval was 7.20-10.00 ml/kg.[Conclusion] The essential oil from H.thibetanus showed significant analgesic effects and had relatively low toxicity.

  • Acute Toxicity of Reinwardtia trigyna Water Extracts to Mice Liver and Kidney Functions and Blood Cell Count

    [Objective]To detect the acute toxicity of R.trigyna water extracts to mice,and to detect the toxicity of R.trigyna water extracts in short term.[Method]After given the R.trigyna by gavage,mice were observed for 10 d and the physical signs and activity situation were recorded.Liver and kidney functions and blood routine were detected.[Result]Effects of R.trigyna water extracts on the mice leukocyte were the greatest,but the two showed no linear relationship.The low and high doses had little effects but the middle dose had significant effects.R.trigyna water extracts had few effects on lymphocyt and monocyte,had great effects on neutrophil,had relatively small effects on erythrocyte except group D, and had no significant effects on hemoglobin.R.trigyna had no significant effects on the physical signs and activity condition of mice,which reduced the leukocyte and neutrophil in blood and led to the decrease of immune function.High dose caused the liver toxicity and had no significant effects on kidney.[Conclusion]R.trigyna water extracts had certain toxicological effects on mice,and high dose showed relatively great toxicity.

  • The Effect of Compound Danshen Dripping Pills on the Pharmacokinetics of Steady-state Warfarin in Rats

    [Objective]To determine the potential interaction between the Chinese herb medicine,Compound Danshen Dripping Pills(CDDP)and a widely used oral anticoagulant warfarin in steady-state in vivo when administered concomitantly.[Method]Each day for 4 days the warfarin control group and the warfarin plus single-dose of CDDP group of rats were treated orally with warfarin(0.2 mg/kg,once per day at 8 a.m.),and the warfarin and CDDP combined treatment group of rats were treated orally with warfarin(0.2 mg/kg,once per day at 8 a.m.)and CDDP(500 mg/kg, once per day at 8 a.m.),the warfarin steady-state was achieved after 4 days of treatment,blood samples were collected each day(at 8 a.m.)for HPLC analysis.Until the 5 day the warfarin control group of rats were still treated orally with warfarin(0.2 mg/kg,once at 8 a.m.),the warfarin plus single-dose of CDDP group of rats were treated orally with warfarin(0.2 mg/kg,once at 8 a.m.)and CDDP(500 mg/kg,once at 8 a.m.), and the warfarin and CDDP combined treatment group of rats were still treated orally with warfarin(0.2 mg/kg,once at 8 a.m.)and CDDP(500 mg/kg,once at 8 a.m.),and blood samples were collected at 0,0.5,1,2,4,8,12,24,36,48 h after warfarin-treatment.The concentration of warfarin in blood plasma was determined by high performance liquid chromatography(HPLC).[Result]The linear calibration curve ranged from 0.05 to 10μg/ml,the limit of quantitation(LLOQ)for warfarin was 0.05 ng/ml(r>0.999),moreover,all the validation data including accuracy, precision(intra-and inter-day),sensitivity,recovery,stability,were within the required limits.CDDP did not alter the pharmacokinetic(PK)behavior of warfarin in steady-state in vivo in rats after simultaneous and concomitant treatment.[Conclusion]The simultaneous and concomitant treatment of rats with CDDP did not give rise to significant effects on the pharmacokinetics in steady-state of warfarin.

  • Expression of Recombinant Curcin in Jatropha curcas L.and Its Anti-tumor Activity in Vitro

    [Objective]To study the expression of recombinant curcin in Jatropha curcas L.and its anti-tumor activity in vitro.[Method]Curcin was cloned from the leaves of J.curcas,and the recombinant vector pET-22b was established.The recombinant protein(29 KD)was expressed in E.coli.[Result]Induced by 0.5 mmol/L IPTG for 6 h at 30℃,expressing quantity of recombinant protein was about 20%of the total protein.Recombinant curcin was expressed in the form of inclusion body.After dissolution,renaturation and purification,purity of recombinant curcin reached 95.6%.[Conclusion]In vitro research showed that recombinant protein had significant proliferative inhibition effects on the A549 and HepG2 tumor cells.And their IC50 were 92.246 and 691.42μg/ml,respectively.But the recombinant protein had no significant inhibitory effects on Hela cell.

    [Objective]To study the extraction conditions for procyanidins assited by ultrasonic wave from Rhodiola rosea.[Method]Effects of solid-liquid ratio,ethanol concentration,extraction time and extraction temperature on the extraction rate of procyanidins were studied by single factor test.Based on the single factor test,the optimal extraction condition for procyanidins from R.rosea was obtained by L 9 (34)orthogonal test.[Result]The influence degree of factors on the extraction rate of procyanidins was in the order of ethanol concentration(B)>solid-liquid ratio(A)> extraction temperature(C)>extraction time(D).The optimal extraction condition was as follows:1∶40(g/ml)solid-liquid ratio,60%ethanol concentration,50℃extraction temperature and 30 min extraction time.Under this condition,the extraction rate of procyanidins from R.rosea was 11.26%.[Conclusion]The optimal condition was obtained for the ultrasonic assisted extraction of procyanidins from R.rosea,which provided references for the development and utilization of R.rosea.

  • Optimization of the Preparation Technology of Chrysanthemum Oral Solution by Orthogonal Design

    [Objective]To discuss the preparation technology of chrysanthemum oral solution.[Method]Taking the content of total flavonoids as index,the extraction technology of chrysanthemum was optimized by means of L 9 (34)orthogonal experiment with ethanol concentration,solid-liquid ratio,reflux time,and reflux times as investigated factors.The content of total flavonoids was determined by UV spectrophotometry.The chrysanthemum extract was filtered,concentrated,diluted with water,filtered with the aid of talcum powder,and finally blended with other auxiliary materials to prepare chrysanthemum oral solution.[Result]The optimal extraction technology was:ethanol concentration of 60%,solid-liquid ratio of 1∶20 (g/ml),reflux time of 1 h,and reflux times of 3.Under this condition,the content of total flavonoids could reach 3.108 6%.The regression equation of total flavonoids was Y=25.455X+0.004 7,R2=0.999 8.The prepared oral solution was a kind of brown,transparent and sweet liquid, in which there would be little sediment over a long time.[Conclusion]This is a simple technology,by which stable finished products are prepared.

  • Optimization of the Ultrasonic Assisted Extraction of Polysaccharides from Dendrobium huoshanense by Response Surface Method

    [Objective]To optimize the extract technology of polysaccharides from Dendrobium huoshanense.[Method]Effects of solid-liquid ratio, extraction time and ultrasonic power on the extraction rate of polysaccharides from D.huoshanense were studied by response surface method.[Result]The optimal extraction condition was as follows:1∶10(g/ml)solid-liquid ratio,30 min extraction time,and 250 W ultrasonic power.Under this condition,the extraction rate of polysaccharides from D.huoshanense reached 28.02%.[Conclusion]This method optimized the ultrasonic assisted extraction of polysaccharides from D.huoshanense,which provided references for the development and utilization of polysaccharides in D.huoshanense.
