  • Geographic Distribution and Application of Mucuna sempervirens Hems L.

    Mucuna sempervirens Hems L.belongs to genus Mucuna of family Leguminosae,which is an evergreen woody vine.By using the DivaGIS software,the geographic distribution maps of M.sempervirens were established based on the altitude layer and vegetation layer.The distribution rule and habitat characteristics of M.sempervirens were analyzed.Results showed that M.sempervirens was distributed below 34.5° N with the altitude of 300-3 000 m in China,such as the subtropical forests,bushes,streams and hillsides.It grew in Zhejiang,Guangdong,Guangxi,Sichuan,Hubei,Yunnan,Guizhou,Hunan,Jiangxi,Shaanxi,Gansu,Fujian and Anhui Provinces of China.Its geographical distribution and vegetation type were closely affected by the altitude.The vegetation types in distribution region were mainly subtropical evergreen broadleaved forest and tropical seasonal rain forest.As an all-purpose ornamental plant,M.sempervirens had relatively high medicinal value and ornamental value.

  • Effects of Different Seeds' Pretreatments on the Growth and Silymarin Content of Silybum marianum Gaertn.

    [Objective]To study the effects of different seeds' pretreatments on the growth and silymarin content of Silybum marianum Gaertn.and to determine the best seeds' pretreatment.[Method] Before sowing,S.marianum seeds were treated respectively by different low temperature such as refrigeration,freezing,and ultra-low temperature.Germination rate,plant height,plant width,budding time and yield per plant were studied,and silymarin content of fruits was compared in order to sieve the best seeds' pretreatment out.[Result]By comparison,seeds were treated by ultra-low temperature with lowest germination,growing badly and lowest silymarin content.Seeds were treated by natural overwintering(namely,autumn sowing) with growing well,highest yield and highest silymarin content.[Conclusion] S.marianum plants will grow well with high yield by autumn sowing.This is good for agricultural production and worthy of further studying.

  • Preliminary Study on the Optimal Pollination Period of Paris polyphylla var. yunnanensis

    [Objective]To select the optimal pollination period of Paris polyphylla var.yunnanensis.[Method] The pollen viability of P.polyphylla was evaluated by TTC test(2,3,5-triphenyl tetrazolium chloride) and its stigma receptivity was estimated by benzidine-H 2 O 2 method.[Result] Pollen viability was the highest at 68.2% when pollen sac cracked.It did not decrease significantly in the first 2 days.Since the fourth day,the pollen viability declined obviously,but still remained some activity.The stigma receptivity was in high activity within five days after pollen sac cracking and the pollination of P.polyphylla should be done in this period.[Conclusion]The optimal pollination period of P.polyphylla is selected in this experiment,which can provide basis for further development and utilization of P.polyphylla.

  • Survey on the Germplasm Resources of Cannabis sativa L.

    Cannabis sativa L.is the main original plant of medicinal material FRUCTUS CANNABIS that has a long cultivation history in China.Although numerous rare varieties have been formed,the planting area of C.sativa is sharply reduced due to the adjustment of industrial structure in recent years,which results in the great changes of resources distribution.We find,most of the wild resources in the main producing areas have been consumed,only a small part of which are restricted to Sinkiang Province,Yunnan Province and Shandong Province.Currently,C.sativa is cultivated extensively in Anhui,Yunnan,Gansu,Heilongjiang,Inner Mongolia,Guangxi,Shanxi,Sichuan,Henan and other provinces of China,sporadically cultivated in Ningxia,Shaanxi,Hebei,Hubei,Hunan and Shanxi,and seldom cultivated in the east coastal regions.Therefore,it is imperative to establish the germplasm resource library and nursery garden for special purpose in order to realize the protective development and utilization of C.sativa Furthermore,seeds of native varieties should be screened based upon the drug properties to ensure the effectiveness and safety of its medicinal material FRUCTUS CANNABIS and to guide its rational use.

  • Pharmacognostic Identification of Swertia nervosa(Wallich ex G.Don) C.B.Clarke

    [Objective]To engage in a systematic pharmacognostic identification study on Swertia nervosa(Wallich ex G.Don) C.B.Clarke.[Method]Characteristics and microscopic structure were studied by morphology and microanatomy method;Meanwhile.HPLC was used to measure the content of active components in S.nervosa [Result]Morphological and histological characters of S.nervosa were described and illustrated with line drawing.Chromatographic peaks of swertiamarin,gentiopicroside and mangiferin were well separated on the baseline.All calibration curves of the three active components showed good linearity within a certain concentration range.Of these,content of swertiamarin and gentiopicroside was 1.21 and 4.41 mg /g,while mangiferin was not detected.[Conclusion]These studies can provide evidence for further development,quality control,and identification of S.nervosa.

  • Microscopic Identification of Salvia deserta Schang

    [Objective]To identify the microscopic characteristics of Salvia deserta Schang.[Method]S.deserta was made into paraffin,surface,dissociation and powder slices.Their microscopic characteristics were observed,described,photographed,and developed by conventional methods of pharmacognostical identification.[Result]Microscopic and physicochemical identification were carried out on the characters of medicinal materials,cross section,dissociated tissue and powder of roots,dissociated tissue and powder of stems,blade surface,and pollen grain.[Conclusion] The method is proved to be simple,rapid and it can provide identification basis for searching and developing TCM resources in Xinjiang Province.

  • Effects of Polysaccharides Extracted from Tremella aurantialba Mycelium on Diabetic Mice

    [Objective]To study the effects of polysaccharides from Tremella aurantialba mycelium(TMP) on the blood glucose and glucose tolerance of diabetic mice induced by alloxan.[Method]Kunming female mice were selected.Diabetic mice induced by alloxan were randomly divided into four groups,which were low-dose group(50 mg /kg),middle-dose group(100 mg /kg),high-dose group(150 mg /kg) and diabetic model control group.And the normal control group was also established.After fed with polysaccharides in Tremella aurantialba mycelium(TMP) by intragastric administration for 28 d,effects of polysaccharides on the blood glucose and glucose tolerance of diabetic mice were studied.[Result]Compared with model group,the body weight of low-,middle-and high-dose groups increased significantly(P < 0.05).The serum glucose value of high-dose group decreased obviously by 15.4%(P <0.05).In glucose tolerance determination,compared with model control group,the values of serum glucose after fed with glucose for 0.5 and 2 h were reduced by 9.4%(P <0.05) and 12.8%(P <0.05) respectively.The area udner glucose curve declined by 10.3%(P <0.05).[Conclusion]Polysaccharides from Tremella aurantialba mycelium with the concentration of 150 mg /kg had obvious hypoglycemic activity and could enhance the glucose tolerance of diabetic mice.T Polysaccharides from Tremella aurantialba mycelium with the concentrations of 50,100 and 150 mg /kg could slow down the body weight decrease of diabetic mice.

  • Effects of Resveratrol on Membrane Potential and Calcium Ions of HA-VSMC

    [Objective]To study the effects of resveratrol on membrane potential and calcium ions of human vascular smooth muscle cell(T /G HAVSMC).[Method]VSMC was labeled with DiBAC4(3) and Fluo-3-AM,respectively;and then the changes of membrane potentials and the concentration of calcium ions were detected by flow cytometer.[Result] Resveratrol made the VSMC membrane potentials less negative or depolarized,and it increased the concentration of intracellular free calcium.[Conclusion] Resveratrol could depolarize the membrane potentials and increase the concentration of intracellular free calcium in HA-VSMC.

  • Preparation and Quality Control of FOLIUM CRATAEGI Granules

    [Objective]To study the preparation technology of FOLIUM CRATAEGI granules and to establish methods for its quality control.[Method]FOLIUM CRATAEGI was extracted by 50% ethanol.With granules' angle of repose and hygroscopicity as the indices,the optimal auxiliary materials of prescription were selected.Total flavonoids and hyperin were detected by TLC.UV spectrophotometry was used to determine the content of total flavonoids.[Result]The optimal auxiliary material of prescription was 250 g extract powder,125 g powdered sugar and 125 g dextrin.Under this condition,the angle of repose and the hygroscopicity were both the minimum,which were 33° and 0.65%,respectively.Total flavonoids and hyperin were well isolated from the samples,the TLC spots were clear.The standard curve of total flavonoids was Y =25.455X +0.004 7 with R = 0.999 9.Linear range was between 0.016 and 0.024 mg /ml,the average recovery rate was 100.57%(RSD =0.13%).Moisture content,granularity and dissolvability of granules all met the requirements in Chinese Pharmacopeia.The average content of total flavonoids was 0.412 g /g(RSD = 0.38%).[Conclusion]The preparation process of FOLIUM CRATAEGI granules was simple,feasible and reasonable,and the quality was stable and controllable.

  • Extraction of Total Flavonoids from FLOS LONICERAE JAPONICAE and Its Antioxidant Activity

    [Objective]To study the antioxidant activity of total flavonoids from FLOS LONICERAE JAPONICAE.[Method] Total flavonoids were extracted from FLOS LONICERAE JAPONICAE by ethanol extraction.The antioxidant activity of total flavonoids was detected by oven method;the scavenging effect on hydroxyl free radical was detected by H 2 O 2 /Fe 2 + system method;the scavenging effect on superoxide anion was detected by pyrogallol method.[Result]Extracts from FLOS LONICERAE JAPONICAE showed significant inhibitory effects on the oxidation of lard;the scavenging rates on hydroxyl and free radical reached 52.59% and 17.12%,respectively.[Conclusion]Total flavonoids had significant antioxidant activity,and scavenging activities of hydroxyl free radical and superoxide anion.

  • Ultrasonic Extraction Technology for Total Flavonoids from the Roots of Hemerocallis fulva L.

    [Objective]To study the optimal condition for the ultrasonic extraction technology of total flavonoids from the roots of Hemerocallis fulva L.[Method]The optimal technology for the ultrasonic extraction of total flavonoids from H.fulva roots was obtained based single factor test and orthogonal test.[Result]Factors affecting the extraction rate of total flavonoids were in the order of ethanol concentration > solid-liquid ratio > extraction temperature > extraction time.The optimal extraction condition was 70% ethanol,1:15(g /ml) solid-liquid ratio,60 ℃ extraction temperature and 50 min extraction time.Under this condition,the content of total flavonoids in H.fulva roots reached 6.984 mg /g.[Conclusion]The extraction technology of total flavonoids from the roots of H.fulva was optimized,which provided theoretical basis for the further development and utilization of H.fulva roots.

  • Process Research of the Refined Tianma Gouteng Decoction by Macroporous Resin Method

    [Objective]To optimize the refined Tianma Gouteng Decoction by AB-8 macroporous resin.[Method]With gastrodin content and pasteforming rate as the indices,the maximum sample volume,sample injection flow,eluent concentration and other factors were optimized during the purification process by single factor test and orthogonal test.[Result] The average paste-forming rate was 21.252 7% before resin purification process and RSD was 1.009 6%;the average gastrodin content was 5.263 mg /g and RSD was 0.693%.The optimal process was as follows:30 ml maximum sample volume,2 BV /h sample injection flow,40% ethanol as eluent,165 ml eluent dosage,0.5 g /ml medical solution,and 1:10 diameter-height ratio.Under this condition,the dry extract rate was 9.98% and the average content of gastrodin was 13.364 mg /g.[Conclusion] This method was reasonable and feasible,could enhance the content of gastrodin,and reduce the extracts amount.

  • Extraction of Paeonol from CORTEX MOUTAN Based on Central Composite Design

    [Objective]To optimize the test parameters for the extraction of paeonol from CORTEX MOUTAN by distillation method.[Method]With paeonol yield as the index,the water addition amount,soaking time and the volume of distilled solution were taken as the investigated factors.Process parameters were optimized and analyzed by design expert7.05 data processing system.[Result]The optimal extraction condition was as follows:94 ml water addition amount,165 min soaking time,and 300 ml distilled solution.Under this condition,the yield of paeonol reached 16.42 mg /g.[Conclusion]Central composite design(CCD) was simple and predictable and could be used for the optimization of extraction parameters of paeonol from CORTEX MOUTAN.

  • Study on the Preparation Process of PERICARPIUM GRANATI Effervescent Granules

    [Objective]To explore the preparation process of PERICARPIUM GRANATI effervescent granules.[Method]Taking the content of total flavonoids as an index,the ultrasonic extraction process of PERICARPIUM GRANATI was optimized by means of L 9(3 4) orthogonal test with ethanol concentration,extraction times,solid-liquid ratio,extraction temperature as experimental factors.Refined extract and adjuvants were mixed to prepare effervescent granules.The angle of repose,moisture content,dissolubility,and particle size of effervescent particles were detected.[Result]The optimum ultrasonic extracting conditions of PERICARPIUM GRANATI were as follows:solvent of 70% ethanol,extraction times of 1,extraction time of 30 min and solid-liquid ratio of 1:25(g /ml).The formula of effervescent granules was established as follows:PERICARPIUM GRANATI thick paste 10 g,lactose 74 g,cane sugar 4 g,citric acid 3 g,sodium bicarbonate 25 g,polyethylene glycol 6000 14 g,70% ethanol.The angle of repose of effervescent granules was 29.6° and showed good liquidity.The moisture content,the particle size and the dissolubility of granules conformed to the pharmacopoeia stipulation.[Conclusion]The preparation process is simple,and the quality is stable and controllable,so it is fit for industrialized production of PERICARPIUM GRANATI effervescent granules.

  • Extraction Process of FRUCTUS SCHISANDRAE CHINENSIS in Zhenzaoanshen Capsule

    [Objective]To study the optimum extraction process of FRUCTUS SCHISANDRAE CHINENSIS in Zhenzaoanshen capsule.[Method] Taking the yield of schizandrin as index,L 9(3 4) orthogonal test designed with four factors like ethanol concentration,extraction time,solid-liquid ratio and extraction times was conducted to select the optimum extraction process for FRUCTUS SCHISANDRAE CHINENSIS.[Result]The optimum extracting process for FRUCTUS SCHISANDRAE CHINENSIS was boiled in 10 times water [solid-liquid ratio = 1:10(g /ml) ] and refluxed for 3 times,1.5 h every time.[Conclusion] This extraction process was a simple and feasible method that was suitable for extracting FRUCTUS SCHISANDRAE CHINENSIS in Zhenzaoanshen capsule.

  • Optimization of Extraction Process for Polysaccharides from Dendrobium huoshanense by Response Surface Methodology

    [Objective]To optimize the extraction process of polysaccharides from Dendrobium huoshanense.[Method]Effects of extraction time,solid-liquid ratio and ethanol concentration on the extraction rate of polysaccharides from D.huoshanense were studied by single factor test.Relationship between the process parameters and response value was studied by response surface methodology.And the optimal extraction process was forecasted.[Result]The optimal extraction process of polysaccharides from D.huoshanense was 1:340(g /ml) solid-liquid ratio,3.25 h extraction time and 86% ethanol concentration.Under this condition,the theoretical extraction rate of polysaccharides from D.huoshanense reached 39.10%.And the measured value was 38.78%,which was close to the predictive value.[Conclusion] Extraction process optimized by response surface methodology was accurate and reliable with high extraction rate,which provided references for the production of preparations.

  • Study on Backflow Extraction Technology of Total Saponins from Stem Base of Panax notoginseng(Burk) F.H.

    [Objective]To optimize the methods for content determination and backflow extraction technology of total saponins from stem base of Panax notoginseng(Burk) F.H.[Method]The content of total saponins was determined by the methods of vanillin-perchloric acid and the extraction technology was optimized by the content of total saponins.[Result]Panax saponin Rg 1 presented good linear relationship over the range of 0.008 3 to 0.049 6 mg /ml(R =0.999 4).The average recovery was 98.6% with the RSD of 3.54%(n =4).The optimal solvent backflow extraction conditions were as follows:P.notoginseng stem base was soaked for 12 h in 60% ethanol at the solid-liquid ratio of 1:10(g /ml) and extracted for 3 times for 2 h each time.[Conclusion]This method is simple and feasible,highly accurate and precise.The extraction process is economical,reasonable and deserved popularizing all the more.

  • Research on the Isolation and Purification Technology of Gardenia Yellow Pigment by HPD-400 Macroporous Resin

    [Objective]To study the technological conditions for isolation and purification of gardenia yellow pigment by macro-porous resin.[Method] The macroporous resin suitable for adsorption and separation of gardenia yellow pigment was chosen from 9 types of ones by investigating their adsorbing and eluting characteristics.To screen out the optimal technology,the effects of influence factors such as temperature,acidity and ethanol concentration on the adsorption capacity of HPD-400 macroporous resin were investigated.[Result]In the nine types of macroporous resin which were tested,the adsorption capacity of HPD-400 macroporous resin was the strongest.The adsorption performance of HPD-400 macroporous resin was the best when the temperature was 25 ℃,pH was 3.0 and concentration of crude extract A 440 nm was 0.590.The elution of gardenia yellow pigment performed best with 70% ethanol at 25 ℃.[Conclusion] HPD-400 macroporous resin can effectively isolate and purify gardenia yellow pigment,which can be used in industry.

  • Optimization of Microwave-assisted Extraction Technology of Total Flavonoids from FRUCTUS LYCII

    [Objective]To optimize the microwave-assisted extraction technology of total flavonoids from FRUCTUS LYCII.[Method]Using rutin as reference substance,the factors that influenced microwave-assisted extraction of total flavonoids were optimized by means of single-factor test and orthogonal design test.[Result]The optimal extraction process of total flavonoids was as follows:70% aqueous ethanol solution,solid-liquid ratio 1:30(g /ml),microwave radiation time 8 min,microwave power 400 W,radiation temperature 120 ℃.In this condition,the yield of total flavonoids was 18.3 mg /g.[Conclusion]Compared with the traditional technology,microwave-assisted extraction is more efficient and easy-operating in extracting total flavonoids from FRUCTUS LYCII.

  • Preparative Separation of Safflower Yellow from FLOS CARTHAMI by Medium Pressure Column Chromatography

    [Objective]To isolate and purify safflower yellow from FLOS CARTHAMI by medium pressure column chromatography with macroporous resin.[Method]The crude extracts of FLOS CARTHAMI were prepared using water extraction method and column chromatography with macroporous resin.Next,highly pure safflower yellow was separated from crude extracts using medium pressure column chromatography,and its content was measured by means of ultraviolet spectrophotoetry.[Result]Among 5 main chromatographic peaks,the peak area of HSYA was the largest.The 4 chromatographic peaks at the retention time of 30.930(Peak No.1),32.803(Peak No.2),36.981(Peak No.3) and 40.316 min(Peak No.4) showed the maximum absorption at the wavelength of 240 and 403 nm.According to related literatures,the chromatographic peak A may be safflower yellow for the ultraviolet spectrograms of 4 absorption peaks were consistent with that of HSYA.The purity of safflower yellow prepared through MPCC with macroporous resin was more than 79% and its content was 75.14%.[Conclusion]Medium pressure column chromatography is first introduced to separate safflower yellow from crude extracts of FLOS CARTHAMI.The method is simple,time-saving,effective and worthy of popularization.

  • Variation Characteristics of Total Saponins Content in Panpax notoginseng at Different Growth Ages

    [Objective]To determine the contents of total saponins in Panpax notoginseng(Burk.) F.H.chen stems,leaves and roots at different growth ages in different production areas,and to analyze the effects of growth age and production area on the total saponins of P.notoginseng.[Method]Contents of total saponins in P.notoginseng stems,leaves and roots were detected by UV spectrophotometry.And the detection wavelength was 560 nm.[Result] Contents of total saponins in P.notoginseng stems,leaves and roots enhanced with the increase of growth age.Among them,total saponins content in roots enhanced by 46.5% each year,which was the most significant increase.Fluctuation was the most greatest in two-year-old P.notoginseng roots.At the same time,contents of total saponins in P.notoginseng stems and leaves enhanced by 23.9% and 33.9% each year,respectively.And total saponins content in one-year-old P.notoginseng leaves showed relatively great fluctuation;the total saponins content in P.notoginseng roots was higher than those in P.notoginseng leaves and stems.[Conclusion]There were differences in the total saponins contents in different parts of P.notoginseng at different growth ages.

  • Technology of Isoflavone Antialcoholism Tablets from RADIX PUERARIAE and Its Properties

    [Objective]To study the tabletting technology of isoflavone antialcoholism tablets from RADIX PUERARIAE and its properties.[Method] With wild RADIX PUERARIAE in Jurong as the raw materials,isoflavone from RADIX PUERARIAE was extracted and purified.With isoflavone of RADIX PUERARIAE as the major component,xylitol,maltodextrin and citric acid were taken as the auxiliary materials.Pressed technology of isoflavone antialcoholism tablets from RADIX PUERARIAE was studied;and the solubility and dispersibility were discussed.[Result]Optimized technology of isoflavone antialcoholism tablets from RADIX PUERARIAE was as follows:1:2(W /W) condiment-main material ratio,4% moisture content and 60 ℃ baking temperature.Among them,the ratio of maltodextrin to RADIX PUERARIAE isoflavone in main material was 1:2(W /W) and the xylitol-citric acid ratio in condiment was 1:1(g /g).Isoflavone antialcoholism tablets from RADIX PUERARIAE had relatively high water holding capacity and water solubility;temperature had relatively significant effects on solubility and dispersibility.[Conclusion]Isoflavone antialcoholism tablets from RADIX PUERARIAE had good chew sense,appropriate hardness,suitable acid and sweet flavor,which suited the public taste.

  • Research Advances in the Detoxificating Preparation of Swainsonine in " Locoweed" and Its Application to Antitumor

    Swainsonine is the main toxic component of " locoweed" that belongs to polyhydroxyl-substituted indolizidine alkaloid.Swainsonine can inhibit α-mannosidase in animal body and poison animals;it also has strong anti-tumor activity which is beneficial to human beings.The paper introduces the research advances in the biochemical characteristics,poisoning mechanism,detoxification preparation and anti-tumor function of swainsonine,aiming to provide references for preventing and curing swainsonine poisoning and rationally developing swainsonine.

  • Effects of Different Bacteriocides on the Tissue Culture of Hypoestes sanguinolenta

    [Objective]To study the effects of different bacteriocides on the tissue culture of Hypoestes sanguinolenta.[Method] Explants were sterilized by different bacteriocides.After that,the inoculated materials were cultured at 23 ℃ with 12 h /d illumination time and 2 000 lx illumination intensity.The pollution and growth situation of materials were observed.[Result]H.sanguinolenta explants treated by 75% ethanol + 0.1% HgCl 2 showed significant pollution control effects with the pollution rate being only 13.33%,but the growth situation was poor.H.sanguinolenta explants treated by 75% ethanol + 0.2% NaClO showed relatively low pollution rate and the growth situation was relatively good.[Conclusion]75% ethanol + 0.2% NaClO was the optimal bacteriocide for tissue culture of H.sanguinolenta explants.

  • Screening of an Endophyte Producing Ginkgo biloba L. Flavonoids

    [Objective]To protect the Ginkgo biloba L.resources,and to find the endophyte producing G.biloba flavonoids.[Method]With G.biloba leaves and roots as the plant materials,tissue block isolation method and streaking cultivation method were adopted for the isolation and purification of endophytes.With extracting solution of G.biloba flavonoids as the reference,TLC analysis on fermentation liquids was conducted.Al(NO 3) 3-NaNO 2 was used as the specific chromogenic agent of flavonoids.[Result] A total of eight kinds of endophytes were screened.TLC analysis showed that the color band or spot of 07-Y8 fermentation liquid had the same color and similar mobility as the standard extracting solution of G.biloba flavonoids.[Conclusion] Strain 07-Y8 might have the capability of being fermented into G.biloba flavonoids or other similar compounds,which provided new approach for the development and utilization of G.biloba flavonoids.
