  • Study on the HPLC Fingerprints of CORTEX MAGNOLIAE OFFICINALIS Extracts from Supercritical CO_2 Extraction

    [Objective] To study HPLC fingerprints of CORTEX MAGNOLIAE OFFICINALIS extracts from supercritical CO2 extraction, and to provide effective methods for quality control. [Method] A HPLC analysis was performed at 35 ℃ on a chromatographic column Phenomenex Luna  5u C18 (2) 100R (4.6 mm×250 mm, 5 μm) with the detection wavelength at 280 nm. The mobile phase consisted of chromatographic methanol and 0.1% acetic acid (V/V, 74∶26). Following the injection of sample solution, an elution was initiated at a flow rate of 1.0 ml/min. [Result] In repeatability test, the fingerprint similarity among samples was all greater than 0.99, so that HPLC-fingerprints could be use for quality control of CORTEX MAGNOLIAE OFFICINALIS extracts from supercritical CO2 extraction. The similarity among 10 batches of its extracts ranged from 0.9 to 1.0, which showed that SFE-CO2 extracts were uniform and stable. [Conclusion] CORTEX MAGNOLIAE OFFICINALIS extracts were proved to be high-purity products with very low impurity, in which main active ingredients magnolol and honokio could nearly occupy 100%.

  • Electrochemical Research on the Apigenin of Antitumor Chinese Medicinal Herb FLOS BUDDLEJAE

    [Objective] To study the electrochemical behavior of antitumor herbal drug apigenin and the interaction of apigenin with DNA at glassy carbon electrode. [Method] Cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were used in experiments. [Result] In CV, only one irreversible anodic peak with Ep = 0.580V was appeared in 0.1 mol/L B-R buffer solution (50% ethanol, pH 9.0). The peak currents of apigenin were linearly related to concentrations in a range from 5.0×10-6 to 9.0×10-5 mol/L. And the detection limit was 1.5×10-6 mol/L. A new electroanalytical method for this herbal drug was established according to this anodic peak. Using the established method without preseparation, apigenin in herbal drug was determined with satisfactory results. [Conclusion] This method is a new one to detect apigenin.

  • Determination of the Content of Total Flavonoids in Sophora alopecuroides L. Residues

    [Objective] To establish a method for determining the content of total flavonoids in residues, so as to provide basis for the reuse of Sophora alopecuroides L. [Method] The content of total flavonoids in S. alopecuroides residues was determined by NaNO2-Al(NO3)3-NaOH spectrophotometry. [Result] The regression equation was Y=11.403X+0.002 1, R=0.999 8, confirming a good linear relationship between the concentration and absorbance of rutin. The recovery of total flavonoids ranged from 98.67% to 100.33% with the RSD of 1.65% (n=9). In addition, the average content of total flavonoids in S. alopecuroides was determined as 3.90 mg/g. [Conclusion] The method is easy, convenient and low cost, which can be used for determining the content of total flavonoids in S. alopecuroides residues.

  • Determination of the Extracts Content from HERBA PLUBAGINIS ZEYLANICAE under Different Cultivation Conditions

    <Abstract>[Objective] To determine the content of extracts from wild HERBA PLUBAGINIS ZEYLANICAE harvested at different time and HERBA PLUBAGINIS ZEYLANICAE cultivated under different conditions, so as to provide experimental basis for establishing the quality standards of HERBA PLUBAGINIS ZEYLANICAE. [Method] Samples were determined according to the methods recorded in the Appendix XA of 2010 edition Chinese Pharmacopoeia, a similarity evaluation was carried out on the contents of extracts from the stems, roots and leaves of wild and cultivated samples. [Result] Of all samples (apart from that cultivated under different light conditions), their roots, stems and leaves could be obviously distinguished, the similarity of roots and leaves was high. The total extracts contents in the roots, stems and leaves from HERBA PLUBAGINIS ZEYLANICAE harvested at different time were higher than that from wild samples; the total extracts contents were higher from January to March and from November to December, but due to the small plant size during January-March, the best harvesting period was determined to be from October to December, basically coinciding with local harvesting time; the total extracts of HERBA PLUBAGINIS ZEYLANICAE propagated with seeds and cutting seedlings were higher, but the yield of plants propagated with individual seedling was the highest. Due to the limited wild HERBA PLUBAGINIS ZEYLANICAE resources, it was suggested to use cutting seedling propagation; the HERBA PLUBAGINIS ZEYLANICAE cultivated in raw soil had the highest contents of extracts, but that cultivated in mellow soil had the highest yield, followed by that in clay. In view of the limited mellow soil resources, it was suggested to use clay; N, P and K fertilizers all could promote the plant growth. Appropriate amount of N, P and K fertilizers should be arranged; the HERBA PLUBAGINIS ZEYLANICAE cultivated under full sunlight had the highest contents of extracts; they were classified by cluster analysis into the first type, others belonged to the second type, it was suggested to adopt full light cultivation. [Conclusion] The method is simple and accurate with good reproducibility, which provides experimental data for the quality standards of HERBA PLUBAGINIS ZEYLANICAE.

  • Content Determination of Daucosterol in Roots,Stems and Leaves of Wild Plumbago zeylanica L. by RP-HPLC-ELSD

    <Abstract>[Objective] To establish a method for determining the content of daucosterol and compare its content from roots, stems and leaves of wild Plumbago zeylanica L. [Method] High performance liquid chromatography-evaporative light scattering detection (RP-HPLC-ELSD) was used to determine the content of daucosterol in wild P. zeylanica. Diamondsil C18 (250 mm×4.6 mm, 5 μm) was used with pure methanol as the mobile phase and the flow rate was 1.0 ml/min. The temperature of column was 25 ℃. The flow rate of nitrogen was at 2.5 L/min and the temperature of drift tube was 65 ℃. [Result] The calibration curve of daucosterol was linear with concentration in the range of 0.129 to 2.58 μg/ml with r=0.999 4 (n=7). The average recovery was 99.93% with RSD of 0.212% (n=9). The content of daucosterol was the highest in leaves and lowest in roots. For each part, the content of daucosterol in P. zeylanica was the highest in February, July, August, and the lowest in May and June. [Conclusion] The method is rapid, simple, stable. And the results provide data for establishing the cultivated technique of Plumbaginis Zeylanicae L.

  • Effects of Drying Methods on the Content of Ferulic Acid in Ligusticum chuanxiong Hort. Stem and Leaf

    [Objective] To study the effect of drying methods on the content of ferulic acid in different parts of Ligusticum chuanxiong Hort stem and leaf. [Method] The content of ferulic acid in L. chuanxiong stem and leaf was determined by HPLC. [Result]There was a good linear relationship between the concentrationand peak area of ferulic acid when the sample injection ranged from 0. 011 48 to 0. 114 8 μg ( r = 0. 999 9) . The average recovery was 100. 1% with the RSD of 1.18% ( n = 9) . The content of ferulic acid in naturally sun-dried L. chuanxiong leaf and stem was 0. 005 4% with the RSD of 1. 55% ,while that was 0. 008 0%with the RSD of 1. 80% in those dried naturally in the shade and that was 0. 010 5% with the RSD of 1. 07% in those dried naturally after sunshine. As to L. chuanxiong stem and leaf ,oven-drying after sunshine was superior to the above two methods. [Conclusion] Among the three drying methods,oven-drying after sunshine was the optimal one for L. chuanxiong stem and leaf .

  • Analysis on the Content of Reducing Sugar and Total Sugar in the Fruits of Capparis spinosa L.

    [Objective]To analyze the content of reducing sugar and total sugar in the fruits of Capparis spinosa L. [Method]3,5-dinitrosalicylic acid method waschosen to test the content of reducing sugar and total sugar in the fruits of Capparis spinosa L. [Result]Results showed that the content of reducing sugar extractedby hot-water ( 100 ℃ ) was about 11. 6% and temperature of water had obvious effect to the results. The content of total sugar was about 24. 5% . [Conclusion]The reducing sugar and total sugar were all relatively higher in the fruits of Capparis spinosa L. ,which indicated that they would have a good development perspective.

  • Analysis of the Flavonoids in the Leaves of Rhododendron pulchrum Sweet by HPLC-MS

    [Objective] To investigate the flavonoids in the leaves of Rhododendron pulchrum sweet by high performance liquid chromatograph - electrospray ionization tandem mass spectrometry. [Method]The chromatographic separation was performed on a C18column ( 4. 6 mm × 250 mm,5 μm) . The mobile phase A waswater containing 0. 1% formic acid and mobile phase B was anhydrous methanol. Gradient eluted program was: 0 - 24 min,31% - 42% B; 24 - 30 min,42% -50% B; 30 - 35 min,50% - 60% B; 35 - 40 min,60% B. Injection volume was 20 μl; flow rate was 0. 7 ml / min; column temperature was 30 ℃ ; and detectionwavelength was 356 nm. Mass spectrometer conditions were ESI,anion mode scanning,drying gas temperature 350 ℃ ,drying gas volume flow ( N2) 10 L/min,nebulizing gas pressure 275. 8 KPa,capillary voltage 3. 5 kV,capillary export voltage 100 V,and mass scan range m / z 200 - 700. Data were collected in anionmode. [Result] Compared with the standard references and literature data,five flavonoids compounds were identified as hyperoside,isoquercitrin,avicularin,quercitroside and quercetin. [Conclusion]This method was rapid,accurate and effective which could be applied for the constituent identification of R. pulchrum,and provided theoretical basis for the application of R. pulchrum.

  • Identification of RADIX NOTOGINSENG and Its Counterfeits by UASLG Method

    [Objective] To establish a method for distinguishing RADIX NOTOGINSENG from its counterfeits including RHIZOMA ALPINIAE OFFCINARUM,RHIZOMA BLETILLAE,RHIZOMA CURCUMAE LONGAE and RHIZOMA CURCUMAE by ultraviolet spectral line group ( UASLG) . [Method]The five kindsof medicines were extracted with 5 different solvents of water,ethanol,petroleum ether,ethyl acetate and chloroform. Then,the absorbencies in UV and first-orderderivative spectra were measured. [Result] There were significant differences in the peak position and absorbance inultraviolet absorption spectra among RADIXNOTOGINSENG,RHIZOMA ALPINIAE OFFCINARUM,RHIZOMA BLETILLAE,RHIZOMA CURCUMAE LONGAE and RHIZOMA CURCUMAE. [Conclusion] The method was simple and accurate,and could be used to distinguish RADIX NOTOGINSENG from its counterfeits.

  • Study on the Content of Forsythin,Total Flavonoids of Forsythia suspensa and Their Antioxidant Activity

    [Objective] To determine the content of forsythin and total flavonoids in different parts of Forsythia suspense,study their antioxidant activity,and analyze the relations of their content with antioxidant activity. [Method] Ultrasonic-assisted technology was applied to extract forsythin and total flavonoids from F.suspense. The content of forsythin and total flavonoids in F. suspense was respectively determined by means of HPLC and colorimetry. The antioxidant activity of theextracts from different parts of F. suspense towards free radical was evaluated by 1,1-diphenyl-2-icrylhydrazyl ( DPPH·) . [Result]The rank of different parts ofForsythia suspense in descending order was new leaves > old leaves > flowers > fruits according to the content of forsythin,while that was flowers > new leaves> old leaves > fruits according to the content of total flavonoids. Different parts of Forsythia suspensa had higher antioxidant activity,among which the antioxidantactivity of flower was the highest,followed by fruit,new leaf,and old leaf. [Conclusion]Extracts from F. suspensa flower,fruit and leaf all have higher antioxidant activity. Of these,total flavonoids are the primary active ingredients in flowers and leaves' extract of F. suspense,and fruits' extract is total flavonoids and forsythiaside.

  • Effect of Two Chinese Herbal Compounds on the Protection of Gastric Mucosal Lesion in Rat and Its Blood Index

    [Objective] To observe the effects of two Chinese herbal compounds ( LWZY) on the protection of gastric mucosal lesion in rat and its blood index.[Method]Three models were set up including the gastric mucosal lesion caused by anhydrous alcohol,gastric mucosal lesion caused by aspirin and gastric mucosallesion caused by hydrochloric acid,and the medicine was intragastrically administrated,the indices of gastric mucosa damage index,prothrombin time ( PT) ,fibrinogen ( FIB) ,activate part blood coagulation time ( APTT) and thrombin time ( TT) were selected to evaluate the anti-gastric mucosal lesion performance of twoChinese herbal compounds and its impact on the blood index. [Result]Cimetidine and LWZY groups had significant protective effects on rats in model of gastricmucosal lesion by anhydrous alcohol,the model of gastric mucosal lesion by aspirin and the model of gastric mucosal lesion by hydrochloric acid,the gastric mucosadamage index of all treatment groups significantly reduced compared with that of model group ( P < 0. 05; P < 0. 01) ,the blood in the anhydrous alcohol model withLWZY reduced significantly in PT,FIB,APTT and TT compared to the normal group ( P < 0. 05; P < 0. 01) with a statistic significance; the blood in the hydrochloric acid model increased slightly in PT,FIB,APTT and TT than the normal group; PT and FIB of LWZY group significantly reduced compared with modelgroup,but without statistic significance,APTT and TT were reduced significantly( P < 0. 05; P < 0. 01) ,and had a statistic significance,the indices of blood PT,FIB,APTT and TT in aspirin model were obviously higher than that in the normal group,while the LWZY group was reduced significantly in PT,FIB,APTT andTT compared to model group ( P < 0. 05; P < 0. 01) ,and had statistic significance. [Conclusion]LWZY could significantly protect the rat gastric mucosa damagecaused by anhydrous alcohol,aspirin and hydrochloric acid,and promote blood clotting in vivo,the action mechanism might be related to the blood coagulation factor and hormone level.

  • High-quality and High-yield Cultivation Technique of Two Segments Method for Crocus sativus L. in Zhejiang Province,China

    Based on the cultivation test and demonstration,the characteristics,medicinal value,cultivation technique,field management,pest control,harvest andprocess of Crocus sativus L. were analyzed systemically. Two Segments Method for the cultivation of C. sativus in Zhejiang Province was put forward. Selecting bulbsabove 8 g was the key for high-yield cultivation of C. sativus in Zhejiang Province. In early November,C. sativus was cultivated in the open field,and grew overwinter under scientific application of fertilizer and water. In the following May,bulbs were collected in ventilating,shady and humid room to accelerate germination. Flowers were collected from late October to November. Popularization of this matching technique provided scientific guidance for the industrial developmentand cultivation management of C. sativus.

    [Objective] To identify the anamorph of Cordyceps taishanensis.[Method] The rDNA ITS fragment was amplified,cloned and sequenced using total DNA extracted from three strains of COA1,COA2 and COA3.[Result] COA1 was Paecilomyces militaris,COA2 was Beauveria bassina and COA3 was Isaria tenuipes.[Conclusion] The findings indicated that Cordyceps taishanensis was named based on the geographical position.

  • Study on Ultrasonic Extraction Technology of Sterol from the Rhizome of Begonia grandis Dry. subsp. sinensis (A. DC.)Irmsch

    [Objective] To study the ultrasonic extraction technology of sterol from the rhizome of Begonia grandis Dry. subsp. sinensis ( A. DC. ) Irmsch. .[Method]In this study,the sterol was first extracted from B. grandis rhizome using ultrasonic technology,and then quantitatively analyzed using the advancedLiebennann-Bur chard colorimetric method. The single factor experiment and orthogonal experiment were carried out to study the effects of factors including extraction temperature,extraction time and solid-liquid ratio on the yield of sterol. The regression equations to three factors were established by RSREG software in SAS8.1. In addition,the types of sterols were analyzed using HPLC. [Result]Results show that the regression equation was significant,the optimal technological conditions of sterol extraction were as follows: extracting temperature was 58 ℃ ,extracting time was 57 min,solid-liquid ratio was 1: 44 ( g / ml) . Under these conditions,the content of sterol was 1. 80 mg / g in the rhizome of B. sinensis. [Conclusion]Ultrasonic-assisted technology is an efficient,energy-saving and time-savingmethod that can be used to extract sterol from the rhizome of B. grandis. In addition,the analysis of HPLC spectrum reveals that sterol composition in the rhizomeof B. sinensis is complex,including β-sitosterol and other sterols.

  • Changes of Phenolics Content and Related Enzymatic Activities in Fritillaria thunbergii Miq and Their Relationships with Dormancy Release

    [Objective] To discuss the changes of phenolics content and related enzymatic activities in Fritillaria thunbergii Miq.bulbs and their relationship with dormancy release.[Method] F.thunbergii bulbs were stored at 4 and 10 ℃ for 75 d,respectively.Sampling was conducted regularly and the phenolics content,PPO activity and PAL activity in bulbs were detected.At the same time,phenolics content,PPO activity and PAL activity in germinated and dormant bulbs were compared.[Result] During low temperature treatment,phenolics contents in bulbs and buds both showed downward trend.Treatment at 10 ℃ for 30-45 d and treatment at 4 ℃ for 45-60 d were the two key periods for phenolics reduction and dormancy release.There was significant negative correlation between phenolics content and PPO activity in F.thunbergii.Phenolics content in terminal buds had significant positive correlation with PAL activity;and phenolics content in scales showed no had significant correlation with PAL activity.[Conclusion]Phenolics content in F.thunbergii bulbs had certain correlation with its dormancy;reduction of phenolics content was one of the physiological mechanisms for dormancy release of F.thunbergii bulbs by low temperature treatment.

  • Extraction Technology of Total Alkaloids from RADIX ACONITI LATERALIS PRAEPARATA

    [Objective] To determine the optimal extraction technology of total alkaloids from RADIX ACONITI LATERALIS PRAEPARATA.[Method] With dry extracts rate,and the purity and extraction rate of total alkaloids as the indices,effects of extraction solvent,extraction method,extraction temperature,extraction time,grind degree,solid-liquid ratio and extraction times on the extraction rate of total alkaloids were studied.And the optimal extraction technology was selected.[Conclusion] The optimal extraction technology was as follows:RADIX ACONITI LATERALIS PRAEPARATA crude powder was added into anhydrous ethanol with the ratio of 1:10(g/ml)under 15 ℃.After cold leaching for 3 times with each time for 1 d,the dry extracts rate,extraction rate of total alkaloids,and purity of total alkaloids were 3.15%,0.64% and 20.32%,respectively.[Conclusion] This method was efficient,low toxic,economical,convenient and can be used for industrial production.

  • Extraction Technology of Serotonin Derivatives from Seed Meal of Hehuang Carthamus tinctorius L. by Water Reflux Method

    [Objective] To optimize the extraction technology of serotonin derivatives from seed meal of Hehuang Carthamus tinctorius L. by water reflux method.[Method]Taking the content of serotonin derivatives as index,the extraction technology of serotonin derivatives was optimized by means of orthogonal test with extraction temperature,extraction time,extraction times and solid-liquid ratio as experimental factors. [Result]The extraction of serotonin derivatives from seed mealof Hehuang C. tinctorius was impacted mostly by extraction temperature,followed by extraction time,extraction times and solid-liquid ratio. The optimal technologywas extraction temperature of 80 ℃ ,extraction time of 1. 5 h,extraction times of 3 and solid-liquid ratio of 1: 20 ( g / ml) ,with which the content of serotonin deriv-atives was 0. 66% . [Conclusion] This optimal extraction technology could provide basis for the development and production of Hehuang C. tinctorius seed meal.

  • Analysis of the Factors Affecting the Quality of RADIX CODONOPSIS

    In order to study the factors affecting the quality of RADIX CODONOPSIS,the literatures were studied from the aspects of germplasm,growth environment,cultivation technology,processing method,drying method,storage condition and so on. The qualities of different verities of RADIX CODONOPSIS were compared,as well as the effects of their environment,cultivation and processing on the effective component and efficacy of RADIX CODONOPSIS. The contents of lobetyolin,polysaccharides,and atractylenolide Ⅲ in RADIX CODONOPSIS were taken as the evaluation indices. It was found out that the germplasm,growth environment,cultivation technology,processing method and storage condition affected the quality of RADIX CODONOPSIS in different degrees. Among them,germplasm had the maximum influencing degree; and the influence degrees of the rest factors still needed further study. Technical measures were used to enhance andstabilize the quality of RADIX CODONOPSIS: pay attention to the germ plasm and select varieties with high quality and active ingredient in order to enhance thequality of RADIX CODONOPSIS; since environmental conditions affecting the quality and active ingredient,genuine environment for medicinal materials should selected in order to ensure the quality stability of RADIX CODONOPSIS; effects of agronomic measures on the quality and output should not be ignored,and the optimal agronomic measures could further enhance the output and active ingredient content in RADIX CODONOPSIS; processing method not only affected the content ofeffective component,but also influenced the storage; processing method affected the chemical composition and efficacy of RADIX CODONOPSIS,and research onthe optimal processing method could ensure the clinical efficacy; the optimal storage method could effectively control the loss of effective component and enhance thequality of RADIX CODONOPSIS.

  • Process Research on the Reflux Extraction of Flavonoids from Rubus crataegifolius Leaves

    [Objective] To study the optimum extraction condition of flavonoids from Rubus crataegifolius leaves by using reflux extraction method.[Method] Taking ethanol as the solvent,effects of ethanol concentration,solid-liquid ratio,extraction temperature,and extraction time on the extraction rate of flavonoids were studied.Based on the results of single factor test,orthogonal test was designed and the optimal technology condition was selected.[Result] Factors affecting the extraction rate of flavonoids were in the order of solid-liquid ratio > ethanol concentration > extraction temperature > extraction time.The optimal extraction condition was as follows:70% ethanol,1:20(g/ml)solid-liquid ratio,85 ℃ extraction temperature and 1.5 h extraction time.Under this condition,the extraction rate of flavonoids was 9.51%.[Conclusion] Reflux extraction of flavonoids from R.crataegifolius leaves was simple and had relatively high extraction rate.

  • Culture Condition and Nutritional Components of Spirulina platensis in Spring under Industrialized Condition

    [Objective] To optimize the culture condition of Spirulina platensis under industrialized condition and to compare the contents of nutritional componentsin spring. [Method] In the enclosed workshop with collecting light panel as the ceiling,S. platensis was cultured by photo bioreactors with four different opticalpaths. The maximum cell densities of photo bioreactors with different optical paths were compared as well as the contents of different nutritional components. [Result] The maximal cell density was 1. 52 g / L in 20 cm bioreactor. The contents of protein and phycocyanin were the highest in 20 cm light path,which were 61.2% and 10. 9% ,respectively. The contents of polysaccharides,crude fat and chlorophyll a were the maximum in 10 cm bioreactor,the content of crude fat reached 7. 48% ,the content of polysaccharides reached 8. 2% ,and the content of chlorophyll a reached 1. 3 mg / g. [Conclusion]This method optimized the culture condition of S. platensis under industrialized condition,which provided theoretical references for the development and utilization of S. platensis.
