• Isolation and Purification Process of Total Flavonoids from Osmanthus fragrans Lour. with Macroporous Adsorption Resin

    [Objective] To study the isolation and purification process of total flavonoids(TF) from Osmanthus fragrans Lour.with macroporous adsorption resin.[Method] Taking the adsorption capacity and recovery rate as index,the optimal isolation and purification process of total flavonoids from Guizhou O.fragrans were respectively investigated by static test and dynamic test with AB-8 macroporous adsorption resin.[Result] pH,eluant,temperature,mass concentration of column-loading sample,diameter-height ratio,current velocity and total flavonoids-adsorption resins had great effects on the adsorption capacity and recovery rate of total flavonoids from O.fragrans.The optimal conditions for extracting and purifying total flavonoids in static test were:the AB-8 type macroporous resin was blended with 4 times of pH 4-5 sample solution(solid-liquid ratio of 1:4,(g:ml)) together and then stirred to make it reach the saturation adsorption;then,the isolated resin whose volume was 9.4 times as high as total flavonoids in sample solution was eluted with 70% ethanol.The optimum conditions in dynamic test were:17.86 mg/ml the extracts of total flavonoids from O.fragrans were loaded into the column with diameter-height ratio of 1.5:21.6 at a current velocity of 2-3 ml/min and then eluted with 2-3 BV of water to remove impurity at a current velocity of 3-4 ml/min.As the temperature increased,the adsorption capacity declined but the elution rate to enlarge.[Conclusion] The optimum conditions for extracting and purifying total flavonoids with AB-8 type macroporous resin are selected in this study,which could provide experimental basis for the industrial production of total flavonoids from O.fragrans.

  • Ultrasonic-assisted Extraction Technology of Total Flavonoids from the Leaves of Dioscorea alata Linn.

    [Objective] To study the ultrasonic-assisted extraction technology of total flavonoids from the leaves of Dioscorea alata Linn.[Method] With the leaves of D.alata as the raw materials,single factor test on the extraction temperature,extraction time,ethanol concentration and solid-liquid ratio was conducted.Based on this,L9(34) orthogonal test was conducted in order to determine the optimal ultrasonic-assisted extraction technology for the flavonoids from D.alata leaves.[Results] The optimal extraction technology was as follows:60% ethanol concentration,50 min extraction time,65 ℃ extraction temperature,and 1:30(g/ml)solid-liquid ratio.Under this condition,the extraction rate of total flavonoids from D.alata leaves was 5.17%.[Conclusion] The optimal ultrasonic-assisted extraction technology of flavonoids from D.alata leaves was selected,which provided new approach for the processing and utilization of D.alata leaves.

  • Ultrasonic Assisted Extraction Technology of Triterpenoid from Inonotus obliquus

    [Objective] To optimize the ultrasonic-assisted extraction technology of triterpenoid from Inonotus obliquus.[Method] With isopropanol as extracting solvent,the triterpenoid was extracted from I.obliquus by ultrasonic-assisted method.Taking the extraction rate of triterpenoid as an evaluation index,the extraction technology was optimized by means of single-factor experiment with treatment condition,ultrasonic power,ultrasonic frequency and ultrasonic processing time as experimental factors.[Result] The optimum ultrasonic-assisted extraction technology of triterpenoid from I.obliquus was determined as follows:ultrasonic power of 400 W,ultrasonic frequency of 30 times and ultrasonic processing time of 15 min.With the optimized technology,the extraction rate of triterpenoid was 1.06%.[Conclusion] The ultrasonic-assisted method was a kind of effective method to extract triterpenoid from I.obliquus.

  • Study on the Extraction Process of Polysaccharides from FRUCTUS CHAENOMELIS by Ultrasonic Technology

    [Objective] To optimize the extraction process of polysaccharides from FRUCTUS CHAENOELIS.[Method] The effects of solid-liquid ratio,ultrasonic temperature,ultrasonic power and ultrasonic action time on the extraction of polysaccharides from FRUCTUS CHAENOMELIS were studied through single-factor test and orthogonal test.[Result] The optimum ultrasonic conditions for extracting polysaccharides from FRUCTUS CHAENOMELIS were solid-liquid ratio 1:70 (g/ml),ultrasonic temperature 80 ℃,ultrasonic power 140 W and ultrasonic action time 50 min.Under these conditions,the yield of polysaccharides was 12.81%.[Conclusion] The ultrasonic technology is a simple,time-saving and high efficient method to extract polysaccharides from FRUCTUS CHAENOMELIS.

  • Extraction of Total Flavonoids from Ginkgo biloba by Decompressing Inner Ebullition

    [Objective] To study the extraction method of total flavonoids from Ginkgo biloba.[Method] By using the decompressing inner ebullition,the Ginkgo biloba powder was desorbed and extracted to investigate the effects of each factor on the extraction rate,the best desorption and extraction conditions were found out,and then compared with that of traditional extraction method.[Result] The optimal desorption conditions were:80% ethanol as the desorption solvent,20 min desorption time and 15 ml solvent dose;the best extraction technology was 0.067 MPa extraction pressure,60 ℃ extraction temperature,30% ethanol as extraction solvent,twice of extraction,5 min each time;under these conditions,the extraction rate of flavonoids could reach up to 2.55%.Compared with the traditional ethanol extraction method,the temperature was cut down by 20 ℃,the extraction was eleven times faster,ethanol dose was two times less,the extraction solution had little pigment and impurities,and could be directly purified with macroporous adsorption resin,the purity of total flavonoids from Ginkgo biloba was 29.8%,the advantages were obvious.[Conclusion] The method is economical,rapid and efficient,therefore could be applied to the extraction of flavonoids from Ginkgo biloba.

  • Study on the Extraction of Swertiamarin and Gentiopicroside from HERBA ARTEMISIAE SCOPARIAE by MAR

    [Objective] To extract the swertiamarin and gentiopicroside from HERBA ARTEMISIAE SCOPARIAE.[Method] The macroporous obsorbent resin No.9 developed by Zhejiang University was used to extract swertiamarin and gentiopicroside from HERBA ARTEMISIAE SCOPARIAE,based on the single factor test,orthogonal experiment and optimized parameters,the extraction system of swertiamarin and gentiopicroside was established with the absorbance as the response value and the contents of swertiamarin and gentiopicroside as the evaluation indices.[Result] Under 0.02 g/ml sample concentration,95% ethanol eluent concentration and 3.0 ml/min flow rate,the extraction of swertiamarin and gentiopicroside was the best.[Conclusion] The method was economical,practical and rapid,which could be used for the extraction of effective components from HERBA ARTEMISIAE SCOPARIAE.

  • Study on the Extraction Process of Total Flavonoids from Melilotus officinalis

    [Objective] To optimize the extraction process of total flavonoids from Melilotus officinalis.[Method] The effects of extraction solvent,extraction temperature,solid-liquid ratio,methanol concentration,extraction duration,and extraction times on the extraction rate of total flavonoids from Melilotus officinalis were investigated and the extraction process was optimized by using single-factor tests and the orthogonal test.[Result] The optimum extraction process was obtained as followed:solid-liquid ratio of 1:30 (g/ml),methanol concentration of 80%,extraction temperature of 60 ℃,extraction duration of 2 h and extraction times of 2.Under the above condition,the extraction rate of total flavonoids from Melilotus officinalis was approximately 1.42%.[Conclusion] The heat reflux method is stable,effective and suitable for industrialization,which will provide references for the development of total flavonoids from Melilotus officinalis.

  • Research on the Extraction Technology of Total Flavonoids from Broussonetia papyrifera by Microwave-assisted Method

    [Objective] To optimize the extraction process of total flavonoids from Broussonetia papyrifera leaves by Microwave-assisted Extraction.[Method] Single factor test was adopted to study the effects of microwave time,microwave temperature,ethanol concentration,microwave power,solid-liquid ratio on the extraction rate of total flavonoids from B.papyrifera leaves.[Result] The optimal extraction process was selected,which was 1:12 (g/ml) solid-liquid ratio,55% ethanol concentration,15 min microwave time,450 W microwave power,60 ℃ microwave temperature,and ethanol as the solvent.Under these conditions,the extraction rate of total flavonoids from B.papyrifera leaves was 79.63 mg/g.[Conclusion] This method was simple,repeatable and stable,and could be used for the extraction of total flavonoids from B.papyrifera leaves.

  • Ultrasonic-assisted Extraction of Flavonoids from HERBA TARAXACI

    [Objective] To select the ultrasonic-assisted extraction conditions of flavonoids from HERBA TARAXACI.[Method] With the extraction rate of total flavonoids as the investigation index,the single factor test and orthogonal experiment were adopted to study the effects of ethanol concentration,solid-liquid ratio,extraction time and other factors on the ultrasonic extraction of flavonoids from HERBA TARAXACI,and the best extraction conditions were selected.[Result] Under the best conditions of 120 W ultrasonic power,60% ethanol as solvent,1:30 (g/ml) solid-liquid ratio,30 min extraction time and 70 ℃ ultrasonic temperature,the extraction rate of total flavonoids could reach 4.02%.[Conclusion] The method selects the optimal extraction conditions,which provides theoretical basis for the industrialized production of flavonoids from HERBA TARAXACI.

  • Determination of Total Flavonoids in the Fruits of Elaeagnus mooraroftii Wall and Elaeagnus oxcarpa Schlechtend

    [Objective] To detect the water contents,total ash contents and total flavonoids contents of Elaeagnus mooraroftii Wall and Elaeagnus oxcarpa Schlechtend in Xinjiang.[Method] According to the 2010 Chinese Pharmacopeia,water content and total ash content were detected.With rutin as the reference substance,NaNO2-Al(NO3)3-NaOH color system was taken as chromogenic reagent.Total flavonoids content was detected by UV spectrophotometry.[Result] Water content was less than 13.75% and total ash content was less than 2.91% in both E.oxcarpa and E.mooraroftii fruits.The total flavonoids contents of E.oxcarpa and E.mooraroftii in Xinjiang were (8.75±0.101 5) and (2.15±0.065 6) mg/g (n=3),respectively.[Conclusion] This method was simple accurate and repeatable and could be used for the content determination of total flavonoids in Elaeagnus angustifolia fruits.

  • Monosaccharide Composition in RADIX ASTRAGALI Polysaccharides by Gas Chromatography

    [Objective] To research on the composition of monosaccharide from RADIX ASTRAGALI polysaccharides.[Method] RADIX ASTRAGALI polysaccharides sample was hydrolyzed with 2 mol/L sulfuric acid.Then,the dried hydrolysate was dissolved in pyridine,and derivatization was conducted by trimethylsilyl ether.Finally,the composition of monosaccharide from RADIX ASTRAGALI polysaccharides was detected by Capillary Gas Chromatography.[Result] Arabinose,fructose,glucose and mannose were identified from the RADIX ASTRAGALI polysaccharides.And their molar ratio calculated according to the equation was 1:10.309:24.667:0.462.Besides,three unknown matters were detected,with the retention time being 10.627,14.783 and 18.249 min.[Conclusion] This method was simple and sensitive,and could be used for the component analysis of monosaccharide in RADIX ASTRAGALI polysaccharides.

  • Determination of Polysaccharide Contents in the Different Base Resources of RADIX CURCUMAE

    [Objective] To study the polysaccharide contents in different base recourses of RADIX CURCUMAE,and to provide references for the quality control of RADIX CURCUMAE.[Method] The crude polysaccharide was extracted by hot water extraction;after using the spectrophotometry and phenol-sulfuric method,the contents of total sugar,reducing sugar and polysaccharide in the purified polysaccharide were detected at 489 nm.[Result] Polysaccharide content varied in different base recourses of RADIX CURCUMAE.Among the 3 different base resources of RADIX CURCUMAE,Curcuma phaeocau lis Val.had the maximum polysaccharide content (25.775%),followed with Curcuma kwangsiensis S.G.Lee et C.F.Liang (3.955%);and Curcuma longa L.had the minimum polysaccharide content (2.695%).[Conclusion] This method was simple and stable and could be used for the content determination of polysaccharide in RADIX CURCUMAE.

  • Determination of Gastrodin Contents in Wild and Cultivated Gastrodia elata from Different Regions of Chongqing by RP-HPLC

    [Objective] To compare the gastrodin contents in wild and cultivated Gastrodia elata from different regions of Chongqing.[Method] After ultrasonic extraction,gastrodin content in G.elata was detected by RP-HPLC.The mobile phase was acetonitrile-0.05% phosphate solution (V/V,3:97);chromatographic column was DiamonsilC18 column (250 mm × 4.6 mm,5 μm);detection wavelength was 220 nm;flow rate was 1 ml/min;and column temperature was 30 ℃.[Result] Gastrodin content varied greatly in different production areas,and there were relatively great differences between cultivated and wild G.elata.Gastrodin content in wild G.elata was relatively higher than that in cultivated G.elata.Among them,cultivated G.elata in Chengkou and wild G.elata in Yunyang had relatively high content of gastrodin;while cultivated G.elata in Wushan showed relatively low content of gastrodin.[Conclusion] The gastrodin contents in cultivated and wild G.elata in Chengkou were far higher than the national standard.Therefore,it was completely feasible to establish planting base for G.elata in Chengkou that complies with the requirements of the national GAP.

  • Study on the Application of HPLC Fingerprint in Genuineness Identification of RADIX ACONITI LATERALIS PRAEPARATA

    [Objective] To establish the high performance liquid chromatogram (HPLC) fingerprint chromatogram of RADIX ACONITI LATERALIS PRAEPARATA and apply it to identify the genuine regional and non-genuine regional medicinal herb.[Method] The HPLC analysis was performed on a Waters XB-C18 (250 mm×4.6 mm,5 μm) with the detection wavelength at 240 nm and column temperature at 30 ℃.Following the injection of 20 μl of sample solution,an elution with the mixture of 40 mmol/L ammonium acetate (adjusted to pH 10.0) and acetonitrile as mobile phase was initiated at a flow rate of 1.0 ml/min,about lasted 70 min.[Result] A total of 22 common peaks were identified from the fingerprints of 6 bathes of genuine regional medicinal herbs,and their similarity was all above 0.926%.From the perspective of peak area ratio of common peaks,there were certain differences in the chemical constituents among different genuine regional medicinal herbs.The 6 batches of non-genuine regional medicinal herbs had 15 common peaks with the similarity all less than 0.879,which were significantly different in chemical constituents.In addition,the chemical constituents were more complicated in genuine regional than non-genuine regional medicinal herb.[Conclusion] The established HPLC fingerprint could distinguish the genuine regional and non-genuine regional medicinal herb,which could provide scientific basis to better control the inner quality of RADIX ACONITI LATERALIS PRAEPARATA.

  • Determination of Mineral Elements in Lycium barbarum L. from Ningxia by Sealed Microwave Digestion ICP-AES

    [Objective] To determine the content of mineral elements in Lycium barbarum L.from Ningxia.[Methods] Taking HNO3-H2O2 as the acid-soluble system,the sample was digested by microwave-assisted closed digestion.Inorganic elements in L.barbarum from Ningxia,including K,Ca,Na,Mg,Fe,Cu,Mn,Zn,Pb and Cr,were also detected.Based on the test on recovery rate,the reliability of data was verified.[Results] Relative standard deviations obtained by this method were all smaller than 5%.The recovery rate was between 94.40% and 107.00%.The high accuracy indicated that the data was reliable.[Conclusion] This method was rapid,simple,accurate and could satisfied the sample examination request with more elements together,which provided a scientific method for the research on mineral content and products quality of L.barbarum.

  • Effects of Different Digestion Methods on the Determination of Copper Content in RADIX ET RHIZOMA NOTOGINSENG by FAAS

    [Objective] To investigate the effects of different digestion methods on the determination of copper (Cu) content in RADIX ET RHIZOMA NOTOGINSENG by Flame Atomic Absorption Spectrometry (FAAS).[Method] Using RADIX ET RHIZOMA NOTOGINSENG sampled from Wenshan City of Yunnan Province as experimental materials,the effects of different digestion methods of dry ashing and wet digestion on the determination of Cu content were investigated by atomic absorption spectrometry.[Result] The optimal recovery reached 3.221 μg/g at 600 ℃ for 4 h using dry ashing method,and 3.721 μg/g under HNO3-HClO4-H2O2 system using wet digestion method.[Conclusion] AAS could be used to determine the Cu content in Panax pseudo-ginseng var.Notoginseng simply and accurately,and the content of heavy metal Cu in Panax pseudo-ginseng var.Notoginseng produced in Wenshan,Yunnan Province is in accordance with Medicinal Plants and Preparations for the Import and Export of Green Industry Standards of China.

  • Change Rule of Polysaccharides in 3 Huoshan Species of Dendrobiumn

    [Objective] To study the content of polysaccharides in 3 Huoshan species of Dendrobiumn with the variation of time.[Method] The content of polysaccharides in 3 species of Dendrobiumn including D.officinale,D.huoshanense and D.moniliforme at 8 growth stages was determined by sulfuric acid-phenol method.And,the changing trend and impacting factors were also analyzed.[Result] The content of polysaccharides was increasing steadily based on a low level at the stage of tissue culture,and it decreased in the following order,D.officinale,D.huoshanense,D.moniliforme.The highest content of polysaccharides in those Dendrobium was pursued after two years culture,the order was D.huoshanense > D.officinale >D.moniliforme,among which the content of polysaccharides in D.huoshanense was as high as 35.74%.[Conclusion] Those results lay a foundation for the breeding,cultivation and application of those three kinds of Dendrobium.

    [Objective] To study the components of haematochrome in wild Zingiber strioatum from Mount Fanjing.[Method] The haematochrome solution was prepared by ultrasonic-assisted extraction method,and then the components of haematochrome were preliminarily identified by spectroscopic and chromatographic methods.[Result] The main components in haematochrome of wild Z.strioatum were flavonoids (No.1),paeonidin-3-rhamnoside (No.2) and malvidin-3-galactoside (No.3).[Conclusion] The experiment identified the components of Z.strioatum haematochrome in Mount Fanjing,which provided references for the follow-up research.

  • Determination of Total Flavonoids in Mukdenia rossii (Oliv.) Koidz. by Spectrophotometry

    [Objective] To establish a method for determining the content of total flavonoids in Mukdenia rossii (Oliv.) Koidz.[Method] By comparison of two color development methods,AlCl3 was chosen as the color-developing agent.With the detection wavelength at 270 nm,the ultraviolet-visible spectrophotometry was adopted to determine the content of total flavonoids in M.rossii.[Result] The mass concentration of total flavonoids in M.rossii was linearly related to absorbance in the range of 0 to 0.024 mg/ml with the correlation coefficient of 0.999 8.The content of total flavonoids in M.rossii was 11.099 mg/g.[Conclusion] The ultraviolet-visible spectrophotometry is a simple and fast method with high precision,reproducibility and stability that is fitter to determine the content of total flavonoids in M.rossii.

    [Objective] To study the scavenging activity of FRUCTUS MORI polysaccharides to free radical in vitro.[Method] The ability of FRUCTUS MORI polysaccharides to scavenge DPPH·,·OH and O-2· was respectively assayed by DPPH,salicylic acid and pyrogallol auto-oxidation methods.[Result] FRUCTUS MORI polysaccharides had strong scavenging activity to DPPH·,·OH and O-2·,the highest scavenging rate of which to the above 3 free radicals was 90.01%,76.40% and 49.85% respectively.[Conclusion] FRUCTUS MORI polysaccharides had strong scavenging activity to free radicals in vitro.

  • Advances in the Cultivation Techniques of Dendrobium officinale

    Dendrobium officinale is a kind of traditional Chinese medicine that belongs to national second class protected medicinal plants.It has unique health-care and medicine values for many people due to the abundant polysaccharides and mannose.This paper reviews briefly the general development situation,industrial policy,species collection,and tissue culture production of seedlings.Moreover,this paper focused on the key points of cultivation techniques,regional properties and security of planting base,developing perspective and problems about D.officinale,as well as investigate the effects of variety breeding,cultivation model,fertilization technology,disease and insect pests on the medicinal value of D.officinale.As a result,the success to expand distribution range on condition of ensuring the security of planting base could improve the quality of medicinal materials.Industrial production of tube seedlings could rapidly breed high-quality varieties.Besides,other factors like breeding,light,substrate,fertilization,and harvesting could influence the quality of cultivars to some extent.Consequently,to design a whole set of efficient and safe cultivation model based on these theoretical bases could improve the quality of D.officinale,promoting the sustainable development of the entire industry.
