  • Pharmacognostical Identification of Achyranthes aspera L. Roots

    [Objective] To identify the roots of Achyranthes aspera L. [Method] A qualitative and quantitative study on the A. aspera roots was carried out through microscopic identification, TLC and HPLC. [Result] Obvious characteristics were found in the tissue structures of A. aspera roots. In addition, the average content of oleanolic acid and ecdysterone was 0.445 3% and 0.103 3%, respectively. [Conclusion] The result can be taken as the reference basis for the establishment of quality standards for A. aspera roots.

  • Effects of Storage Condition on Seed Vigor of Three Medicinal Plants

    [Objective] To study the effects of storage condition on seed vigour of three medicinal plants. [Method] With the seeds of Atractylodes macrocephala Koidz., Scutellaria baicalensia Georgi. and Polygala tenuifolia Willd. as the plant materials, seeds with water contents of 3%, 5% and 15% were stored at -10, 0, 20 and 36 ℃ for one year. Change laws of germination rate and vigour index of three medicinal plant seeds stored after one year were preliminarily researched. [Result] Among the two controlling factors, water content had more significant impact on seed germination rate and vigor index than temperature. Germination rate and vigor index of three medicinal plants seeds reduced with the decrease of storage temperature and seed water content. When the water content was controlled within 3%-5%, temperature had no significant impact on the seed germination rate and vigour index. However, seeds with 15% water content at 0 ℃ showed relatively low germination rate and vigour index. [Conclusion] Both seed germination rate and vigor index were affected by water content and storage temperature.

  • Selenium-accumulation Cultivation Technology of Ganoderma lucidum

    [Objective] To investigate the effects of Na2SeO3 concentration on the yield, quality and selenium-accumulation of Ganoderma lucidum. [Method] Plate natural breeding method was used to select the G. lucidum. Cultivation experiment of the strain selected was conducted. With biomass and mycelium polysaccharide yield as the indices, effects of Na2SeO3 concentration on the yield, quality and selenium-accumulation of G. lucidum were studied. [Result] Results of pate natural breeding showed that the stain Taizhi 1 had the greatest biomass and polysaccharide yield. Therefore, Taizhi 1 was selected as the test object for selenium-accumulation. Adding Na2SeO3 in the liquid medium could effectively promote the growth of thalli, but had no significant promotion on the synthesis of polysaccharides. When the Na2SeO3 concentration reached 0.08%, the biomass yield was the maximum. Adding Na2SeO3 in the cultivation medium had certain promotion on the growth of G. lucidum and the synthesis of polysaccharides. Selenium content in fruiting body of G. lucidum enhanced as the concentration of Na2SeO3 increased. [Conclusion] Adding Na2SeO3 in the cultivation medium could promote the synthesis of polysaccharides and the growth of fruiting body of G. lucidum. Selenium content in fruiting body of G. lucidum enhanced as the concentration of Na2SeO3 increased.

  • Pharmacognostical Study of Rhaphidophora hongkongensis Schott

    [Objective] To study the pharmacognostical characteristics of Rhaphidophora hongkongensis Schott. [Method] The pharmacognostical characteristics of R. hongkongensis stem, leaf and medicinal powder were studied through morphological, microscopic, physical and chemical identification. [Result] The cross sections of R. hongkongensis stem and leaf were characteristic of monocotyledons. That is, large secretory cavity was below the epidermis of stem, closed collateral vascular bundle scattered in the cortex and pith, calyptrate fiber bundles was on the outer surface of phloem, large xylem vessels were in the center of stem; endoderm was obvious; the main vein of leaf cross-section was mostly scattered in the closed collateral vascular bundle, most of the spongy tissue were scattered in the secretory cavities. Large stellate hairs could be seen in the whole plant powder; common druse was single or clustered in the parenchyma cells; number of calcium oxalate raphides was large; color reaction and TLC reactions were positive for steroids. [Conclusion] The experiment studies the pharmacognostical characteristics of R. hongkongensis, which provides scientific basis for the identification of medicines.

  • Study on the Optimum Harvest Time of FRUCTUS EVODIAE

    [Objective] To determine the optimum harvest time of FRUCTUS EVODIAE. [Method] The optimum harvest time of FRUCTUS EVODIAE was determined according to the apparent characteristics and the content of evodine, evodiamine and rutaecarpine in FRUCTUS EVODIAE by high-performance liquid chromatography (HPLC). [Result] In the investigated harvest time, the content change of evodine was insignificant, and the content of evodiamine and rutaecarpine first increased then slowly declined with the harvest time. [Conclusion] The proposed method was simple, rapid, accurate and reliable, and can be used as a tool for quality control of FRUCTUS EVODIAE. The optimum harvest time was during mid-to-late August.

  • Resource and Habitat Investigation of RADIX STEMONAE in Guangxi Zhuang Autonomous Region of China

    [Objective] To preliminarily investigate the species, resources and habitats of RADIX STEMONAE in Guangxi Zhuang Autonomous Region of China. [Method] Through literature consultation, specimen examination and filed investigation, preliminary investigation on the habitats, resources distribution and plant morphology of RADIX STEMONAE were conducted in Guangxi. [Result] RADIX STEMONAE in Guangxi belonges to Stemona tuberosa Lour., which is found mainly wild and is occasionally transplanted. RADIX STEMONAE usually grows in the stone caves, mixed forests and shrubs at the altitude of 30-1 600 m. The main production areas are Baise, Hechi, Guilin, Fangchenggang and Chongzuo cities. Due to the destructive collection and the damage of ecological environments, resources of RADIX STEMONAE have reduced sharply. Therefore, it is urgent to protect the RADIX STEMONAE. [Conclusion] The species, resources distribution and habitats of RADIX STEMONAE in Guangxi were investigated, in order to provide references for the sustainable development and normalized cultivation of RADIX STEMONAE.

  • Effects of Water and Fertilizer Coupling on the Yield and Quality of RADIX ANGELICAE SINENSIS

    Effects of water and fertilizer coupling on the quality and yield of RADIX ANGELICAE SINENSIS were studied, which provided theoretical basis for the water and fertilizer regulation of GAP normalized cultivation of RADIX ANGELICAE SINENSIS. Water content data and yield and quality data of RADIX ANGELICAE SINENSIS were collected by field experiment under different irrigation and fertilization conditions. According to the principle of quadratic general rotary unitized design, binary quadratic polynomial was adopted to research on the relationship among the irrigation amount, fertilization amount, ferulic acid content, and RADIX ANGELICAE SINENSIS yield. Relationship model of water and fertilizer coupling was established to carry out test analysis and correlation analysis. Results showed that under a given irrigation amount, RADIX ANGELICAE SINENSIS yield enhanced and then decreased as the fertilization amount increased; under a given fertilization amount, RADIX ANGELICAE SINENSIS yield also enhanced and then decreased as the irrigation amount increased; the maximum yield of RADIX ANGELICAE SINENSIS appeared in the plots with middle water and middle fertilizer. Under a given irrigation amount, ferulic acid content in RADIX ANGELICAE SINENSIS firstly enhanced and then fell as the fertilization amount increased; at a given fertilization amount, ferulic acid content also firstly enhanced and then fell as the irrigation amount increased; The maximum content of ferulic acid content in RADIX ANGELICAE SINENSIS appeared in the plots with middle-high water and middle-high fertilizer. Therefore, rational and effective irritation and fertilization could produce high-yield and high-quality RADIX ANGELICAE SINENSIS.

  • Study on the Numerical Taxonomy of the Genus Paris L. from Guangxi Province

    [Objective] To study the 36 characters of 9 species in the genus Paris L. from Guangxi Province by using numerical taxonomy. [Method] The principal components analysis (PCA) and UPGMA clustering analysis were performed to analyze the Euclidean distance. [Result] 9 species in Paris L. from Guangxi Province were divided clearly into four character groups. Paris polyphylla Smith and its variants were suggested to be treated as a new group, except for Paris polyphylla var. chinensis (Franch.) Hara, regarded as a variant in classical taxonomy. [Conclusion] It is significant to apply numerical taxonomy to solve the classification problems in the genus Paris L. or classification problems among species in Paris L. By combining numerical taxonomy, plant morphology and palynology, this study could draw a conclusion similar to classical taxonomy.

  • Study on Infrared Spectrum of Lonicera hypoglauca Miq. and Lonicera macranthoides Hand.-Mazz. from Guangxi Province

    [Objective] To develop a rapid method for identifying and evaluating Lonicera hypoglauca Miq. and Lonicera macranthoides Hand.-Mazz. species with IR spectrums. [Method] IR spectrums were applied to research Lonicera hypoglauca Miq., samples collected from 10 different producing areas and Lonicera macranthoides Hand.-Mazz. samples collected from 7 different producing areas in Guangxi Province. Then, the baseline of infrared spectrums was smoothed and corrected using Omnic 7.0 software, and each samples similarity was calculated using their average spectrum as the standard spectrum. Based upon theirs infrared spectrums characteristic absorption peaks and the similarity, these samples were identified and evaluated. [Result] The characteristic absorption peaks of Lonicera hypoglauca Miq. and Lonicera macranthoides Hand.-Mazz. from Guangxi Province were obtained. The similarity of Lonicera hypoglauca Miq. samples ranged from 0.976 7 to 0.993 4 with the RSD of 0.58% (n=10). Lonicera macranthoides Hand.-Mazz. samples similarity was in the range of 0.685 4 to 0.823 8 with the RSD of 6.92% (n=6). [Conclusion] This method is a simple, fast and stable one that can be used to identify and evaluate the species of Lonicera hypoglauca Miq. and Lonicera macranthoides Hand.-Mazz.

  • Effects of Formula Fertilizer of N, P and K on the Growth of Annual Scutellaria baicalensis Georgi

    [Objective] To explore the effects of formula fertilizer of N, P and K on the growth of annual Scutellaria baicalensis Georgi. [Method] Three factors secondary D-saturation optimal design was employed to discuss the effects of N-P-K ratio on the growth of annual Scutellaria baicalensis Georgi, thereby establishing an optimal fertilizing method. [Result] Growth indexes of Scutellaria baicalensis Georgi in fertilization treatments were higher than that in blank control group, which pointed out that the combined application of N, P and K was conductive to the growth of Scutellaria baicalensis Georgi. On the whole, the combined application of N, P and K was superior to the combination of two elements and single element. The optimal fertilization condition for Scutellaria baicalensis Georgi was 79.41 kg/hm2 N, 150.00 kg/hm2 P2O5, and 150.00 kg/ hm2 K2O. Under this condition, the content of polysaccharides in the roots of Scutellaria baicalensis Georgi was 7.4%. [Conclusion] The optimal fertilizing method was selected, which provided basis for the production of Scutellaria baicalensis Georgi.

  • Study on the Liquid Suspension Culture of Dendrobium candidum Wall.ex Lindl. Seeds

    [Objective] To study the liquid suspension culture of Dendrobium candidum Wall.ex Lindl. seeds. [Method] 15 formulas of culture liquids were designed with 5 influencing factors for the liquid suspension culture of D. candidum seeds, including MS basal medium, hormone, natural additives, pH and sucrose. Of these, the optimal condition for the liquid suspension culture of D. candidum seeds was screened. [Result] Moderate salt concentration, NAA, low pH value, higher sucrose concentration and moderate potato juice were conducive to the germination of D. candidum seeds. The culture medium suitable for seeds germination was 1/2 MS + 0.5 mg/L NAA + 80 g/L potato juice + 45 g/L sucrose. [Conclusion] The medium formula suitable for liquid suspension culture was selected, which provided theoretical basis for the cultivation of D. candidum.

  • Preparation and Purification of Protoplast from Nervilia fordii (Hance) Schltr. Leaves

    [Objective] To set up protoplast preparation and purification method from Nervilia fordii (Hance) Schltr. leaves. [Method] Taking the yield and survival rate of protoplast as indices, the effects of different factors, including pretreatment, enzymolysis method, enzymolysis temperature, centrifugation speed, illumination, and sieve mesh number were studied for the separation and purification of protoplast from N. fordii leaves. [Result] The optimum process for protoplast isolation was obtained as follows: N. fordii leaves were first pretreated for 1 h in CPW solution containing 13% mannitol, and then incubated at (25±1)℃ in enzyme solution under dark conditions; after the enzymolysis, the enzyme solution was filtrated through 200 mesh screen and centrifuged at 800 r/min for 8 min. The filtration and centrifugation were repeated once before microscopic examination. Large amount of intact and clean protoplast with very few cell debris was obtained. [Conclusion] The preparation system has been successfully established, which lays foundation for establishing protoplast regeneration system from N. fordii leaves.

  • Study on Rapid Propagation System of Rehmannia glutinosa f. hueichingensis (Chan et Sehih) Hsiao Test-tube Plantlet

    [Objective] To establish the rapid propagation system of Rehmannia glutinosa f. hueichingensis (Chan et Sehih) Hsiao test-tube plantlet. [Method] In rapid propagation test, the stem tip of R.glutinosa test-tube plantlet was respectively cultured in medium containing NAA, 6-BA and KT at different concentrations. [Result] At low and medium concentration, NAA and 6-BA could promote the rapid propagation of R. glutinosa test-tube plantlet to some extent. On the contrary, high concentration of NAA and 6-BA would inhibit the rapid propagation of test-tube plantlet. The optimal medium for test-tube plantlet was MS+0.002 mg/L NAA+0.2 mg/L 6-BA. [Conclusion] This study could provide theoretical basis for the development and utilization of R. glutinosa.

  • Screening of Differentiation Medium for Apocynum venetum L. Seeds

    [Objective] To screen out an optimal medium for induction and differentiation of Apocynum venetum L. seeds. [Method] To obtain the optimal medium, effects of NAA concentration on the differentiation of A. venetum seeds were investigated using MS as the basic medium and pollution-free bulbils as explants. [Result] Different concentration of NAA flavored the survival of A. venetum seeds, among which there was no significant difference. The optimal medium for differentiation of A. venetum seeds was MS+0.02 mg/L NAA+1.00 mg/L IBA +10 g/L agar+30 g/L sucrose. [Conclusion] This study could provide basis for the industrialized production of A. venetum.

  • Pilot Study on Serum Pharmacochemistry of Extract of Citrus reticulata 'Chachi' Peel

    [Objective] To study the serum pharmacochemistry of extract from Citrus reticulata 'Chachi' Peel. [Method] Based on the establishment of HPLC fingerprints of extract of C. reticulata peel, the constituents absorbed into blood were determined by comparing HPLC fingerprints of the methanol extract of C. reticulata peel, medicated plasma with extract of C. reticulata peel and blank serum. [Result] Seventeen compounds absorbed into blood were found in medicated serum, among which five were original constituents of extract from C. reticulata peel and the other might be metabolites of the original constituents. [Conclusion] Polymethoxylated flavones, nobiletin and hesperetin which are absorbed into blood might be bioactive components of extract of C. reticulata peel. Further studies on them will help clarify the bioactive constituents and functional food factors of extract from C. reticulata peel.

  • Effects of Polysaccharides from Potentilla anserine on the Levels of Cytokines in Tumor Mice

    [Objective] To study the effects of polysaccharides from Potentilla anserine (PAP) on the levels of interleukin-2 (IL-2) and tumor necrosis factor-α (TNF-α) from peripheral blood. [Method] The mice models with tumor S180 were established and treated with 50, 100 and 200 mg/kg PAP by gastric perfusion, respectively. The inhibition rate of S180 tumor was measured to investigate the antitumor activity in vivo of PAP. The levels of IL-2 and TNF-α in mice peripheral blood were determined with ELISA. [Result] The tumor inhibition rates were 46.41%, 52.29% and 59.48% in different groups treated with 50,100 and 200 mg/kg PAP in vivo, respectively. Of these, the inhibition rate of CY and low-dose PAP was equal to that of medium- and high-dose PAP, significantly higher than that of cyclophosphamide. Three dose groups of PAP could significantly increase the content of IL-2 and TNF-α spleen index. [Conclusion] The compound PAP could improve the antitumor activity and immunological function of S180 tumor-bearing mice.

  • Study on the Antimicrobial Activity of Liposoluble Compounds from Sargassum pallidum (tvrn) Ag against Food Spoilage Bacteria

    [Objective] To study the antimicrobial activity of liposoluble compounds from Sargassum pallidum (tvrn) Ag against food spoilage bacteria. [Method] Using the circle agar disc diffusion method, the antimicrobial activity of liposoluble compounds and fractions isolated by column chromatography from S. pallidum was assayed against food spoilage bacteria. [Result] The inhibition zone of S. pallidum crude grease against Escherichia coli and Pseudomonas stutzeri was 1.0 and 1.5 mm. Bacillus subtilis and Staphylococcus aureus were not sensitive to crude grease. Benzene fraction had no antimicrobial activities against Escherichia coli and Pseudomonas stutzeri, and its inhibition zone against Staphylococcus aureus and Bacillus subtilis was both 1.0 mm. Petroleum ether fraction had no antimicrobial activity against 4 food spoilage bacteria. Ethanol fraction showed strong antimicrobial activities against four food spoilage bacteria and the inhibition zone against Bacillus subtilis, Escherichia coli, Staphylococcus aureus and Pseudomonas stutzeri could reach 3.0, 4.5, 4.0, and 2.5 mm. In addition, the blank control group had no antimicrobial activities. [Conclusion] Ethanol fraction of S. pallidum showed strong antimicrobial activities against four food spoilage bacteria. The antimicrobial activities of liposoluble compounds were attributed to their strongly polar lipids.

  • Study on Acute Toxicity and Analgesic Effect of Total Flavonoids from FRUCTUS CANARII

    [Objective] To observe the acute toxicity and analgesic effect of total flavonoids from FRUCTUS CANARII (TFFC). [Method] Mice in experimental medicine group were intragastrically administered TFFC, with the thickest concentration and the largest volume permitted. Mice in control group were administered the equal volume of solvent. The acute toxicity reaction of mice was observed in the successive 7 days. The analgesic effect of TFFC was observed by using the writhing assay and the hot-plate method in mice. [Result] After 7 days, all mice in the experimental medicine and control groups survived. The weight of mice in the two groups respectively increased by 28.0% and 25.0% (P>0.05), no acute toxicity was observed in both groups. The maximal tolerant dose (MTD) of TFFC by ig in mice was 11.46 g/kg, which was about 115 times of adult clinical daily dose. TFFC reduced the times of writhing induced by intraperitoneal injection of acetic acid in mice, improved pain threshold in hot-plate. [Conclusion] TFFC has little acute toxicity and good analgesic effect in mice.

  • Extraction and Free Radical Scavenging Activity of Polysaccharides from HERBA DESMODII STYRACIFOLII

    [Objective] To discuss the methods for extraction and content determination of polysaccharides from HERBA DESMODII STYRACIFOLII and to study their scavenging activity to hydroxyl radical. [Method] Polysaccharides were extracted from HERBA DESMODII STYRACIFOLII with hot water method and then preliminarily purified, the content of which was measured by means of phenol-sulfuric acid method. In addition, ·OH scavenging activity of polysaccharides was tested. [Result] The content of polysaccharides in tested sample was 5.11%. The recovery rate was 99.46% with the RSD of 1.38% (n=5). HERBA DESMODII STYRACIFOLII polysaccharides could effectively scavenge ·OH generated from Fenton system. Moreover, the ·OH scavenging activity of polysaccharides was improved along with the increase of concentration. [Conclusion] HERBA DESMODII STYRACIFOLII polysaccharides have stronger scavenging activity to ·OH.

  • Study on Anti-inflammatory Effect of Total Flavonoids from FRUCTUS CANARII

    [Objective] To observe anti-inflammatory effects of total flavonoids from FRUCTUS CANARII. [Method] Using the inhibition rate as index, three conventional models including dimethylbenzene-induced ear swelling, egg white-induced paw edema and acetic acid-induced celiac capillary permeability addition in mice were applied to evaluate the anti-inflammatory effect of total flavonoids from FRUCTUS CANARII. For each model, mice were divided into 5 groups, which were respectively injected with 80, 40, 20 mg/kg total flavonoids from FRUCTUS CANARII, 10 mg/kg dexamethasone and normal saline containing 5% Tween-80 (as blank control). [Result] Total flavonoids from FRUCTUS CANARII could inhibit the mouse ear swelling induced by dimethylbenzene, inhibit the mouse paw edema swelling induced by egg white and inhibit the increase of the mouse celiac capillary permeability induced by acetic acid. [Conclusion] Total flavonoids from FRUCTUS CANARII have certain anti-inflammatory effect.

  • Proliferation Inhibition and Induced Differentiation Effects of Carthamin on Leukemic Cells

    [Objective] To research the proliferation inhibition and induced differentiation effects of carthamin on K562 leukemic cells. [Method] With the cultured K562 leukemic cells as the research materials, the proliferation inhibition and induced differentiation effects of carthamin were observed by the XTT test, colony formation test, hemoglobin benzidine staining test and morphological experiment. [Result] The activity and colony formation of leukemic cells were dose-dependently suppressed by carthamin (P<0.05); the hemoglobin benzidine staining test and morphological experiment also proved that the carthamin dose dependently induced the K562 leukemic cells to the hemoglobin end cells. [Conclusion] Carthamin had significant effects of proliferation inhibition and induced differentiation on K562 leukemic cells.

  • Study on the Antioxidant Activity of Ellagic Acid from Rubuso suavissimus S. Lee

    [Objective] To study the antioxidant activity of ellagic acid from Rubuso suavissimus S. Lee. [Method] The ellagic acid was extracted and purified from R. suavissimus by solvent extraction and recrystallization, and its DPPH· scavenging ability and reducing capacity were studied by in vitro test. In addition, the total antioxidant activity of ellagic acid was determined by phosphomolybdate method. [Result] The ellagic acid could effectively eliminate DPPH· free radicals, which was characterized by fast speed, low dosage and strong antioxidant activity. When the concentration of ellagic acid was 0.10 mg/ml, the DPPH· scavenging rate reached more than 92.5%. The reducing capacity of ellagic acid was improved by raising concentration, which was lower than that of BHT at the concentration below 0.5 mg/ml and higher than that of BHT at the concentration above 0.5 mg/ml. Furthermore, the total antioxidant activity increased with the increase of concentration and with the extension of action time. [Conclusion] It is significant to develop and utilize ellagic acid with strong free radical scavenging capacity.

  • Preliminary Study on the Antibacterial Activity of RADIX ET CAULIS SOLANI TORVI Extract in Vitro

    [Objective] To study the antibacterial activity of RADIX ET CAULIS SOLANI TORVI extract against tested strains in vitro. [Method] The antibacterial activity of RADIX ET CAULIS SOLANI TORVI extract against 10 kinds of bacteria such as Staphylococcus aureus was tested by drug-sensitive disk diffusion method and tube double dilution method. [Result] The water extract from RADIX ET CAULIS SOLANI TORVI ① presented antibacterial activity against Staphylococcus aureus, Staphylococcus citreus, Micrococcus, Bacillus subtilis, Shigella and Staphylococcus albus. Ethanol extract ② and petroleum ether extract ③ had strong inhibitory effect on methicillin-resistant Staphylococcus aureus, Staphylococcus aureus, Staphylococcus citreus, Micrococcus, Bacillus subtilis and Shigella. Ethanol extract ② and chloroform extract ④ had weak inhibitory effects on Staphylococcus albus. [Conclusion] Different extracts from RADIX ET CAULIS SOLANI TORVI presented varying antibacterial activity against tested strains. The antibacterial activity of ethanol extract ② was higher than that of water extract ①, but petroleum ether extract ③ and chloroform extract ④ were the main active ingredients that contributed to antibacterial activity.
