• Study on the Growth Rhythm of Aquilaria sinensis Seedlings at Seedling Stage

    [Objective] To better understand the growth rhythm of Aquilaria sinensis seedlings and culture high-quality seedlings for market demands.[Method] The growth rhythm of the container seedlings of semiannual Aquilaria sinensis was studied,the effect of meteorological factors on seedlings growth was analyzed,and the daily growth rate of the container seedlings was detected by ordered sample cluster analysis method.[Result] The growth period of the container seedlings of semiannual Aquilaria sinensis could be divided into four stages:early growth stage,rapid growth stage,slow growth stage and late growth stage.And the growth of basal diameter was divided into three phases of early growth stage,fast growth stage and late growth stage.Different seeding culture measures should be adopted according to the characteristics of different growth stages,at the early growth stage,more attention should be paid to such daily care as shading,weeding,chemical spraying and disease preventing and so on,so as to promote the growth of roots;at the rapid growth stage,the focus should be on the fertilization of high N,low P and K,the retention of soil moisture and comprehensive weeding;the slow growth stage should focus on decreasing the N fertilization proportion,improving the application proportion of P and K fertilizers,appropriately reducing the sprayed water amount,and controlling the pests and diseases;the emphasis of the late growth stage should be laid on stopping fertilization,decreasing water spraying,shading and preventing frost disasters,so as to promote the lignification of seedlings and improve the drought and cold-resistant capacity.[Conclusion] The study could provide guidance for the scientific and rational culturing of Aquilaria sinensis seedlings.

  • Investigation on the Medicinal Pteridophytes Resources in Yunnan Province

    According to the specimens kept in herbarium of Yunnan University and relevant literature data,the medicinal pteridophytes resources in Yunnan Province were statistically studied.Results showed that,there were totally 345 species in 111 genera of 49 families of medicinal pteridophytes in Yunnan Province,most of them belonged to Polypodiaceae,Dryopteridaceae,Aspleniaceae,Pteridaceae,Thelypteridaceae,Selaginellaceae,Athyriaceae,Sinopteridaceae,Adiantaceae,Huperziaceae and Davalliaceae.Those pteridophytes had diversified medical effects,most of them were used for clearing heat and detoxication,the medical parts of most medicinal pteridophytes were their whole plants and rhizomes.The habitats of medicinal pteridophytes differed largely with a wide vertical distribution.The distribution of medicinal pteridophytes in Yunnan Province concentrated in Jinping,Gongshan,Guangnan and other counties,where there were great development values of pteridophytes resources.But due to the immature studies on the development of local medicinal pteridophytes resources,most of the species were only used as folk medicinal herbs.Therefore,a lot of problems needed to be solved.A field investigation should be carried out on the medicinal pteridophytes resources in Yunnan Province,their medicinal functions and values should be evaluated,the pharmacognostic studies should be enhanced,species of pteridophytes with obvious efficacies were selected for industrialized production so as to achieve the marketization of medicinal pteridophytes resources development.

  • Content Determination of Rutin and Sophoricoside in FRUCTUS SOPHORAE at Different Harvesting Time

    [Objective] To investigate the relationship between the rutin,sophoricoside content in FRUCTUS SOPHORAE(originated from the dried fruits of S.japonica) and its growth time(harvesting time) in order to define the optimal harvesting time of FRUCTUS SOPHORAE for different applying purpose.[Method] The fruits of S.japonica were collected once a month from the mid-August to the next mid-January for three consecutive years between 2006 and 2008.The content of rutin and sophoricoside in the extracts obtained from FRUCTUS SOPHORAE by absolute methanol continuous extraction method was simultaneously determined by RP-HPLC.The separation of extracts was carried out on a Diamonsil C18 column(150 mm×4.6 mm,5 μm) with the detection wavelength at 260 nm.Following the injection of extracts,an elution with methanol-1% glacial acetic acid-acetonitrile(V/V/V,30.4:64.6:5) as mobile phase was initiated at a flow rate of 1.0 ml/min.[Result] Rutin peaked in the mid-August,with the level of(0.87±0.16)%,then declined gradually.Sophoricoside,the most predominant components of flavonoids,reached the highest between the mid-September and mid-October,which was(5.76±0.22)% and(5.33±0.39)%,respectively.The content of sophoricoside in FRUCTUS SOPHORAE harvested in autumn was much higher than that in the fruits collected in winter(P≤0.05).[Conclusion] The content of rutin and sophoricoside in FRUCTUS SOPHORAE at different development stages had the tendency of seasonal variation,which was the lowest in that harvested in winter.Being known the seasonal variation of the content of rutin and sophoricoside,the optimal harvesting time of S.japonica fruits for different applying purpose could be defined,aiming at providing certain theoretical basis for the scientific utilization of current resources.

  • Analysis on the Chemical Components and Antioxidant Activity of Volatile Oil from FRUCTUS OCIMI BASILICI

    [Objective] To study the chemical components and antioxidant activity of volatile oil from FRUCTUS OCIMI BASILICI in Hetian of Xinjiang.[Method] The volatile oil was extracted by naphtha extractor and then determined by GC-MS,its chemical compositions were identified by computer retrieval technique,and the relative contents of those compositions were calculated by the method of peak area normalization.The antioxidant activities of those compositions were explored through their scavenging rate of DPPH·.[Result] A total of 65 compositions were isolated,among which 60 were identified,accounting for 92.59% of the total volatile oil components.The main compositions were linolenic acid(13.83%),n-Hexadecanoic acid(13.80%),pulegone(8.31%) and(-)-Spathulenol(6.72%).The volatile oil extracted by vapor distillation method could better scavenge DPPH·.[Conclusion] Among the chemical compositions of volatile oil extracted from FRUCTUS OCIMI BASILICI in Hetian of Xinjiang,the alcohols and ketons were the most,followed by acids and alkenes,and the volatile oil had DPPH· scavenging capacity.

  • Determination of Metal Elements Content in RHIZOMA IMPERATAE from Different Regions

    [Objective] To determine the content of metal elements in RHIZOMA IMPERATAE from different regions.[Method] The contents of metal elements Fe,Cu,Zn,Mn,Ca and Mg in RHIZOMA IMPERATAE were determined by flame atomic absorption spectrometry.[Result] The contents of metal elements in RHIZOMA IMPERATAE were different among varied regions.For example,the RHIZOMA IMPERATAE from Henan had high contents of Fe and Mn;while that from Anhui had high contents of Cu and Ca;the highest Mg content was seen in the RHIZOMA IMPERATAE from Shandong,while the highest Zn content was found in that from Inner Mongolia.The recovery of each metal element ranged from 92.8% to 104.6% with RSD between 0.9%-1.2%.[Conclusion] The method is simple with high precision and recoveries,therefore it can be adopted to determine the contents of metal elements in RHIZOMA IMPERATAE from varied regions.

  • Analysis of Flavonoids Constituents in HERB OF AIR-PLANT and Its Antioxidant Activity

    [Objective] To establish a qualitative and quantitative analysis method for the flavonoids in HERB OF AIR-PLANT and preliminarily evaluate its antioxidant activity.[Method] After hydrolysis,the total flavonoids were extracted and determined by UV spectrophotometry.TLC was applied to detect quercetin and kaempferol.Fention method was applied to determine the antioxidant activity.And the total quercetin and kaempferol content in the flavonoids was determined by HPLC.[Result] Rutin showed good linear relationship in the range of 11.0-110.0 μg/ml(r=0.999 8) with the average recovery of 99.3%(n=6,RSD=1.3%) by the UV spectrophotometry.Quercetin and kaempferol were detected by TLC.Quercetin showed good linear relationship in the range of 37.5 to 1 200.0 ng/ml(r=0.999 7) with the average recovery rate of 101.67%(n=6,RSD=1.92%);and kaempferol showed good linear relationship in the range of 62.5 to 2 000.0 ng/ml(r=0.999 4) with the average recovery rate of 98.68%(n=6,RSD=1.40%).The total flavonoids extracted from HERB OF AIR-PLANT showed high antioxidant activity,and antioxidant effect of total flavonoids from HERB OF AIR-PLANT increased as the concentration of total flavonoids enhanced.[Conclusion] This method is accurate and reliable,and could be used for the qualitative and quantitative determination of the flavonoids and for the quality control of HERB OF AIR-PLANT.

  • Trace Analysis on Aristolochic acid A in RADIX ET RHIZOMA ASARI from Different Production Areas

    [Objective] To determine and compare the content of aristolochic acid A in RADIX ET RHIZOMA ASARI from 12 different production areas by establishing Reversed-Phase High Performance Liquid Chromatography(RP-HPLC).[Method] The content of aristolochic acid A in the extract of RADIX ET RHIZOMA ASARI prepared by ultrasonic extraction was determined by RP-HPLC.RP-HPLC used the chromatographic column Zorbax SB-C18(4.6 mm ×150 mm,5 μm) with the detection wavelength at 316 nm.Following the injection of 5 μl of sample solution at room temperature,an elution with acetonitrile-water solution(V/V,50:50) as mobile phase was initiated at a flow rate of 1.0 ml/min.[Result] Except the RADIX ET RHIZOMA ASARI in Fengcheng,no aristolochic acid A was detected.The content of aristolochic acid A in RADIX ET RHIZOMA ASARI produced in Fengcheng showed good linear relationship within the range of 2.04 to 20.4 ng/ml.[Conclusion] This method is rapid and simple with high recovery rate and good linear relationship,sensitivity and repeatability.Thus,it could be used for the determination of aristolochic acid A content in RADIX ET RHIZOMA ASARI.And the RADIX ET RHIZOMA ASARI used in this research had relatively high quality.

  • Analysis of Polysaccharides Compositions in Ripe Peel of Citrus reticulate 'Chachi'

    [Objective] To study the polysaccharides compositions in ripe peel of Citrus reticulate 'Chachi'.[Method] Employing HPLC method,PMP pre-column derivatization was performed to measure the monosaccharide composition of pectic polysaccharides of Citrus reticulate 'Chachi',under the chromatographic conditions as follows:chromatographic columns of the Phenomenex Luna C18(250 mm×4.60 mm,5 μm),with the mobile phases of 0.05 mol/L mixture solution of phosphates-acetonitrile(80:20,V/V,pH=7.1),flow rate of 1.0ml/min;detection wavelength of 245 nm,column temperature of 30 ℃ and injection volume of 20 μl.[Result] The pectic polysaccharides of Citrus reticulate 'Chachi' contained mannose,ribose,rhamnose,galacturonic acid,glucose,galactose,xylose,arabinose and so on.The mole ratios of each monosacchavide was correspondingly 0.08:0.002:0.05:4.80:1:1.52:0.11:3.82.[Conclusion] This method has the advantages of simple operation,shortened period,repeatability and yield,and could be used for the measurement of polysaccharides of Citrus reticulate 'Chachi'.

  • Identification of Volatile Components from Shells of Juglans sigillata D.by GC-MS

    [Objective] To study the volatile components from shells of Juglans sigillata D.by GC-MS.[Method] The volatile components were extracted from J.sigillata shells by steam distillation,and then identified by GC-MS.[Result] A total of 20 compounds were identified from J.sigillata shells,among which fatty acid and aliphatic hydrocarbon were the main volatile components,accounting for 50.41% of the total volatile oil.The content of hexadecanoic acid occupying 25.46% of the total volatile oil was the highest in J.sigillata shells,followed by anthracene(4.41%),linoleic acid(4.37%),diphenylene-oxide(2.50%) and heptacosane(2.27%).[Conclusion] The major volatile components in the J.sigillata shells were fatty acid and aliphatic hydrocarbon.

  • Effect of Different Processing Methods on the Content of Geniposide in FRUCTUS GARDENIAE

    [Objective] To compare the content of geniposide in FRUCTUS GARDENIAE before and after being processed.[Methods] The content of geniposide in FRUCTUS GARDENIAE was determined by HPLC.A HPLC analysis was carried out on a Inertsil ODS-2 C18 chromatographic column(250 mm × 4.6 mm,5 μm) with the detection wavelength at 430 nm and column temperature at 40 ℃.Following the injection of 20 μl of sample solution,an elution with acetonitrile-water(V/V,15:85) as mobile phase was initiated at a flow rate of 1.0 ml/min.[Result] Different processed products of FRUCTUS GARDENIAE varied in the content of curcumin and listed in a decreasing order as follows:raw materials(3.60%) > carbonized sample(3.36%)> parched sample(3.22%) > slightly brown-parched sample(3.02%) > yellow-parched sample(2.77%).[Conclusion] Different processed methods may have some effects on the content of geniposide in FRUCTUS GARDENIAE.

  • Determination of Resveratrol Content in RHIZOMA ET RADIX POLYGONI CUSPIDATI by HPLC

    [Objective] Based on optimizing a variety of experimental conditions,a method for the determination of resveratrol in RHIZOMA ET RADIX POLYGONI CUSPIDATI by HPLC was established by using ethanol as extractant.[Method] Phenomenex C18(250 mm×4.6 mm,5 μm) was selected as column,with methanol-0.25% acetic acid(V/V,65:35) as mobile phase,0.8 ml/min mobile speed and 306 nm detection wavelength.Taking the extraction rate of resveratrol as the evaluation index,single factor test and L25(54) orthogonal test were used to optimize the experimental conditions.[Result] The extraction conditions for resveratrol were 60% ethanol,1:15(g/ml) solid-liquid ratio,55 ℃ extraction temperature and 15 min extraction time.Under these conditions,the extraction rate of resveratrol reached 0.826 μg/g.[Conclusion] This method is proved to be simple,rapid,accurate and could be used for the determination of resveratrol content in RHIZOMA ET RADIX POLYGONI CUSPIDATI.

  • Effect of Different Extraction Methods on the Chemical Constituents of Essential Oils from FOLIUM ARTEMISIAE ARGYI

    [Objective] To study the effect of different extraction methods on the chemical constituents of essential oils from FOLIUM ARTEMISIAE ARGYI.[Method] The essential oils were first extracted from FOLIUM ARTEMISIAE ARGYI by steam distillation and SFE-CO2,and then qualitatively identified by GC-MS.Next,the relative content of each component was calculated by normalization method.[Result] There were significant differences in the chemical constituents of essential oils that were extracted from FOLIUM ARTEMISIAE ARGYI by different methods like steam distillation and SFE-CO2.The essential oils extracted by SFE-CO2 mainly contained 1,2-benzenedicarboxylic acid-2-ethylhexyl mono ester(17.81%) and 1,2,3,4,4a,5,6,8a-octahydro-7-methyl-4-methylene-1-(1-methylethyl)-(1a.alpha.,4a.alpha.,8a.alpha.)-naphthalene(11.13%),while that extracted by steam distillation mainly contained 4,6,6-trimethyl-[1S-(1a,2β,5a)]-bicyclo [3.1.1] hept-3-en-2-ol(22.05%),4-methyl-1-(1-methylethyl)-[1S-(1α,4β,5α)]-bicyclo [3.1.0] hexan-3-one(10.69%) and caryophyllene oxide(10.62%).[Conclusion] The essential oils extracted from FOLIUM ARTEMISIAE ARGYI by SFE-CO2 contained more chemical constituents,especially low-polarity substance.However,the chemical constituents of essential oils extracted by steam distillation focused on higher-polarity alcohols.It was difficult to extract low-polarity volatile components by steam distillation.

  • Content Determination of Vitexin and Apigenin-7-O-β-D-Pyranglycuronide in Broussonetia papyrifera Leaves by HPLC

    [Objective] To determine the content of vitexin and apigenin-7-O-β-D-pyranglycuronide in Broussonetia papyrifera leaves.[Method] Using absolute methanol-1% acetic acid glacial(V/V,30:70) as mobile phase,the content of vitexin in B.papyrifera leaves was measured by HPLC method at the detection wavelength of 359 nm.Using absolute methanol-0.05% phosphoric acid(V/V,40:60) as mobile phase,the content of apigenin-7-O-β-D-pyranglycuronide was measured by HPLC method at the detection wavelength of 324 nm.[Result] There was good linearity in the concentration range of 0.056 to 0.488 μg/ml for vitexin(r=0.999 2).The average recovery rate of vitexin was 99.2% with the RSD of 1.42%.Apigenin-7-O-β-D-pyranglycuronide presented good linearity in the concentration range of 1.808 to 4.068 μg/ml(r=0.999 8).The average recovery rate of apigenin-7-O-β-D-pyranglycuronide was 100.9% with the RSD of 1.58%.[Conclusion] This method that takes advantages of simplicity and good separating effect could be used for establishing the quality standard of B.papyrifera leaves.

  • Determination on the Main Chemical Components and Antibacterial Activities of Volatile Oil from Datura stramonium L.

    [Objective] To determine the main chemical components and antibacterial activities of volatile oil from Datura stramonium L.[Method] Volatile oil was extracted from Datura stramonium L.by steam distillation.Chemical components of volatile oil were analyzed by GC-MS.Alternaria alternate,Botryosphae riadothidea,Rhizoctonia solani Kühn,Fusarium solani(Mart.),and Fusarium avenaceum were taken as the target bacteria to carry out in vitro antibacterial activity test.[Result] Ten components were detected,which accounted for 92.66% of the total volatile oil.In the volatile oil of Datura stramonium L.,content of 5,6-dihydro-6-pentyl-2H-pyran-2-one was the highest,accounting for 44.28%.Volatile oil from Datura stramonium L.had different inhibitory effects on 5 plant pathogens,which were in the order of Botryosphae riadothidea>Fusarium avenaceum>Fusarium solani(Mart.)>Rhizoctonia solani Kühn>Alternaria alternate.[Conclusion] This method was simple and rapid,and could be used for the determination of main chemical components and antibacterial activities of volatile oil from Datura stramonium L.

  • Effect of Four Taste Tibet Inula Root Soft Capsule on Immunodeficiency Mice Model Induced by Cyclophosphamide

    [Objective] To study the effect of four-taste Tibet inula root soft capsule on the immunologic function of immunodeficiency mice model induced by cyclophosphamide.[Method] Kunming mice were divided into blank group,model group,four taste Tibet inula root soft capsule group(low-dose and high-dose) and positive medicine group(levamisole hydrochloride tablets).After having been treated with the corresponding dose of drug,the phagocytic percentage and phagocytic index of peritoneal macrophage in immunodeficiency mice model induced by cyclophosphamide were observed.Effects of generated hemolysin antibody and drug on the mice delayed-type hypersensitivity induced by DNFB were analyzed by hemolysin antibody measurement method.[Result] Compared with model group,four taste Tibet inula root soft capsule could significantly enhance the phagocytic percentage and phagocytic index of peritoneal macrophage in immunodeficiency mice,and promote the generation of hemolysin antibody from body.In the test,the difference in the weight between left and right ears was significant in mice with delayed-type hypersensitivity,which was significantly higher than that of mice in model group and control group,having statistical difference.[Conclusion] Four taste Tibet inula root soft capsule could significantly enhance the immune function of immunodeficiency mice induced by cyclophosphamide.

  • Research on the Effect of FRUCTUS GARDENIAE Extract on Reducing Uric Acid in Hyperuricemia Mice and Its Action Mechanism

    [Objective] To study the effect of FRUCTUS GARDENIAE extract on the level of uric acid in hyperuricemia mice and investigate the mechanism.[Method] After intragastric administration of FRUCTUS GARDENIAE extract at different doses,the level of uric acid and the activity of xanthine oxidase(XOD) in hyperuricemia mice induced by potassium oxonate were respectively observed.[Result] Different doses of FRUCTUS GARDENIAE extract could obviously reduce the level of serum uric acid and inhibit the activity of XOD in hyperuricemia mice.However,extract had no significant effect on the level of serum uric acid in normal mice.[Conclusion] FRUCTUS GARDENIAE extract could significantly reduce the level of serum uric acid in hyperuricemia mice,which is related to its inhibitory effect on XOD.

  • Ultrasonic Ethanol Extraction Technology of Polyphenol from FRUCTUS CANARII

    [Objective] To study the extraction technology of polyphenol from FRUCTUS CANARII by ultrasonic ethanol method.[Methods] The ultrasonic ethanol extraction conditions of polyphenol from FRUCTUS CANARII were optimized by single factor test and orthogonal test.[Results] The optimal ultrasonic ethanol extraction conditions were as following:40% ethanol,ultrasonic power of 40 W,solid-liquid ratio of 1:20 g/ml and extraction time of 45 min.Factors which affected the extraction of polyphenol from FRUCTUS CANARII were in order of ethanol concentration > solid-liquid ratio> ultrasonic power>extraction time.[Conclusion] This research provided scientific basis for the further exploitation of medicinal value of FRUCTUS CANARII.

  • Optimization of the Extraction Technology of Flavonoids from Chrysanthemum coronarium L.by Microwave Method

    [Objective] To optimize the extraction conditions of flavonoids from Chrysanthemum coronarium L.[Method] Single factor test was adopted to inspect the effects of ethanol concentration,solid-liquid ratio,microwave power and extraction time on the extraction rate of flavonoids from Chrysanthemum coronarium L.,and the extraction technology was optimized by orthogonal experiment.[Result] The optimum conditions for extracting flavonoids by microwave method were:60% ethanol,1:20 solid-liquid ratio,microwave thawing and 120 s extraction time.Under those conditions,the yield of flavonoids could reach 3.19%.[Conclusion] The microwave extraction of flavonoids from Chrysanthemum coronarium L.is simple and practical with the advantages of time-saving,energy-conserving and high efficient.

    [Objective] To investigate the ultrasonic-assisted extraction technology of total polyphenol from persistent calyx of PHYSALIS CALYX SEU FRUCTUS.[Method] Regarding the total polyphenol yield as the indicator,the ultrasonic-assisted extraction technology of total polyphenol from persistent calyx of PHYSALIS CALYX SEU FRUCTUS was optimized by firstly single factor test and then an orthogonal design.[Result] The optimized results were as follows:50% ethanol as the extraction solvent,solid-liquid ratio of 1:20(g/ml),extraction time of 30 min,and 2 extraction times.[Conclusion] The ultrasonic method developed in this study is simple,reliable and time-saving and efficient,which provided theoretical basis for further study on the extraction of total polyphenol from the persistent calyx of PHYSALIS CALYX SEU FRUCTUS.

  • Microwave-assisted Extraction of Melanin from the Seed Coat of Mucuna prurients(L.)

    [Objective] To research the extraction technology of melanin from the episperm of Mucuna prurients(L.) by microwave-assisted alkaline extraction and acid precipitation.[Method] Melanin was extracted from the seed coat of M.prurients by the method of microwave-assisted alkaline extraction-acid precipitation and the method of alkaline extraction-acid precipitation.Content of melanin in M.prurients was detected by UV spectrophotometry.[Result] The content of melanin in M.prurients seed coat was 2.41%.The optimal conditions for alkaline extraction-acid precipitation were 1.0 mol/L NaOH solution,1:20 g/ml solid-liquid ratio,4-6 h extraction time,and pH=2 for the precipitation of melanin.Under these conditions,the yield of melanin crude extracts was(91.2±2.0)%,and its purity reached(36.5±2.1)%.In addition,the optimal technology of microwave-assisted alkaline extraction-acid precipitation was as follows:in accordance with the ratio of solid to liquid 1:20 g/ml,the seed coat and 1.0 mol/L NaOH solution were together exposed to high power ultrasonic for 25 min,with which the maximal yield of melanin was obtained,that is to say,the yield of melanin crude extracts was(95.3±1.5)%,and its purity reached(37.4±1.5)%.[Conclusion] The seed coat of M.prurients was rich in melanin,which deserved further research and development.Microwave assistance could significantly enhance the extraction rate of the crude extracts of melanin and greatly shortened extraction time remarkably.

  • Research on Preparation Process of Dropping Pills from Pueraria lobata(Willd.) Ohwi Bud and Content Determination of Puerarin

    [Objective] To optimize the preparation process of dropping pills from Pueraria lobata(Willd.)Ohwi bud and determine the content of puerarin.[Method] Taking the percent rate of roundness,weight difference rate and dissolving time duration of dropping pills as indexes,the PEG6000-S-40 ratio,matrix-drug ratio,refrigerant temperature and dropping rate were optimized by orthogonal test with table L9(34) in the course of preparing dropping pills from P.lobata bud.And the content of puerarin in dropping pills was determined by HPLC.[Result] The optimal conditions for preparing dropping pills from P.lobata bud were as follows:PEG6000-S-40 ratio 3:1(g/g),matrix-drug ratio 4:1(g/g),refrigerant temperature 35-40 ℃,dropping rate 50 drops/min.And the average content of puerarin in three batches of dropping pills were(52.26 ±1.33) mg/g.[Conclusion] The preparation process of dropping pills from P.lobata bud is reasonable and repetitive.The dropping pills have good quality of appearance and meet the requirements of China Pharmacopoeia.The HPLC method is advanced,accurate and reliable,which can be used to control the quality of dropping pills from P.lobata bud.
