• Isolation and Characterization of a New Diterpenoid Compound from Agastache rugosa

    [Objective] To extract and isolate terpenoids from the leaves and branches of wild Agastache rugosa in Qinba mountainous area,and to study on the structure of those terpenoids.[Method] The powders of A.rugosa leaves and branches were extracted with 95% ethanol,the concrete was dispersed in water and then extracted by petroleum ether,ethyl acetate,butanol in turn,the terpenoids in the ethyl acetate extracts were isolated with silica gel column chromatography,and then characterized by TLC,IR,UV,NMR and x-ray diffraction.Its crystal structure was also determined by single crystal x-ray diffraction.[Result] This diterpenoid was Longikaurin D,the crystal of Longikaurin D belonged to orthorhombic crystallographic system,space group P2(1)2(1)2(1),and cell parameters:a=0.845 1(4) nm,b=1.061 5(5) nm,c=2.237 1(10) nm,α = β = γ = 90.00°,Mr=404.43,V=2.007(16) nm3,Dc=1.352 mg/m3,Z=4,F(000)=872,μ=0.103/mm,R1=0.076 7,ωR2=0.146 8.[Conclusion] Longikaurin D was extracted from A.rugosa for the first time,which provided scientific basis for developing and utilizing the terpenoids in A.rugosa.

  • Isolation of Three Chemical Constituents from Alsophila spinulosa Stalks for the First Time

    [Objective] To study the chemical constituents from the stalks of Alsophila spinulosa(Hook.) Tryon.[Method] The chemical constituents isolated and purified from the crude powder of A.spinulosa stalks through silica column chromatography method were analyzed by TLC,1H-NMR,13C-NMR,etc.In comparison with related literatures,these compounds were further identified.[Result] Three chemical constituents were isolated from A.spinulosa stalks for the first time,including vitexin,isorientin and an organic acid(3,4,6-trihydroxy-1-ring vinyl carboxylic acid).And,this organic acids presented different degrees of antimicrobial activity,which provided support for the antimicrobial activity of A.spinulosa stalks from the angle of chemical constituents.[Conclusion] This method is simple,convenient and accurate to identify the chemical constituents that were isolated from A.spinulosa stalks.

  • Determination of Catechin in RHIZOMA BERGENIAE SCOPULOSAE by HPLC

    [Objective] To establish a method to determine the content of catechin in RHIZOMA BERGENIAE SCOPULOSAE by HPLC.[Method] Both ultrasonic extraction and HPLC method were adopted.Chromatographic column was Kromasil-C18 column(4.6 mm×150 mm,5 μm) with methanol-water-acetic acid(V/V/V,15:84:1) as mobile phase,flow rate at 1.0 ml/min,and detection wavelength at 278 nm.[Result] The linear range of catechin was 0.624-3.12 μg/ml(r= 0.999 3).The average recovery rate was 100.05%,with RSD=1.73%(n=6).[Conclusion] This method was simple,accurate and could be used for the content determination of catechin in RHIZOMA BERGENIAE SCOPULOSAE.

  • Extraction and Content Determination of Rhynchophylline from RAMULUS UNCARIAE CUM UNCIS

    [Objective] To study on the extraction and content determination of rhynchophylline from RAMULUS UNCARIAE CUM UNCIS.[Method] Extraction method of traditional alkaloids was adopted.And modern analysis method of HPLC was used to determine the content of rhynchophylline.Rhynchophylline content in RAMULUS UNCARIAE CUM UNCIS was calculated.[Result] Specialization investigation showed that the retention time of rhynchophylline was about 18.843 min,and that of isorhynchophylline was 22.323 min.The two peaks showed good resolution;and other components in RAMULUS UNCARIAE CUM UNCIS did not interfere with the assay.Results of content determination showed that the contents of rhynchophylline and isorhynchophylline were 23.23 and 129.17 μg/g,respectively.The average recovery rates of added recovery test were 99.12% and 101.33%,with RSD values being 1.84% and 1.56%,respectively.[Conclusion] This method was stable,effective,accurate,and could be used for the content determination of rhynchophylline in RAMULUS UNCARIAE CUM UNCIS.

  • Differences of Total Flavone Content in Xiangxi Drynaria fortunei among Different Germplasm Resources

    [Objective] To study the differences of total flavone content in Xiangxi Drynaria fortunei among different germplasm resources.[Method] The total flavones and total naringin were extracted from D.fortunei by microwave method.With rutin as standard substance,the content of total flavones in leaf,petiole and rhizome of 8 D.fortunei germplasm resources harvested in Xiangxi was determined by colorimetry.In addition,with naringin as standard substance,the content of total naringin in different parts of D.fortunei was determined by colorimetry.[Result] There was significant difference about the content of total flavones in Xiangxi D.fortunei among different germplasm resources.More specifically,the content of total flavones in Longshan samples was the highest at 13.5%,followed by Wangcun samples at 1.8%,and the least in Baojing samples.Moreover,the content of total naringin in different parts of D.fortunei followed a trend of leaf>rhizome>petiole.[Conclusion] D.fortunei harvested from Longshan County could be used as the original species for artificial cultivation because of the highest content of total flavones.Besides,D.fortunei leaves could be utilized and developed for the highest content of total naringin,aiming at providing experimental foundation for the research on germplasm resources screen and artificial cultivation of D.fortunei.

  • Comparison Study on Extraction Methods of Volatile Components from Schizonepeta tenuifolia Briq.

    [Objective] To study the methods for extracting volatile components from Schizonepeta tenuifolia Briq.[Method] The volatile components were extracted from the S.tenuifolia by simultaneous distillation extraction,backward flow,program raise of temperature microwave extraction and headspace.Then,GC-TOFMS was used to analyse their components and relative content.Besides,effects of different extracting technologies on volatile components were also compared.[Result] 60,27,38 and 34 volatile components were identified from S.tenuifolia extracts prepared by simultaneous distillation,backward flow,program raise of temperature microwave extraction,and headspace,respectively.Both components and relative content of volatile components were notably different in extracts prepared by different method.Moreover,estragole was the main component of the above S.tenuifolia extracts that accounted for 34.90%,39.90%,39.90%,and 84.82%,respectively.[Conclusion] The simultaneous distillation extraction is better than others,which could be used for analysing volatile components in S.tenuifolia.

  • Determination of Cu,Pb and Cd Content in PORIA by Atomic Absorption Spectrometry

    [Objective] To determine the content of Cu,Pb and Cd in PORIA by atomic absorption spectrometry(AAS).[Method] Different PORIA samples were collected,and then digested in the mixed solution of concentrated HNO3 and HClO4(4:1),the content of Cu in the sample solution was determined by FAAS,while that of Pb and Cd was determined by GFAAS.[Result] The content of Cu,Pb and Cd in the six PORIA samples all met the limit control of heavy metals in importing and exporting medicinal plants in China.[Conclusion] The method is rapid,sensitive and stable,which could be applied to the quality control of the PORIA market.

  • Study on the Anticomplement Constituents from FLOS ROSAE RUGOSAE

    [Objective] To isolate and identify the anticomplement constituents from FLOS ROSAE RUGOSAE,the flower buds of Rosa rugosa Thunb.[Method] Diverse chromatographic methods were adopted to isolate and purify the anticomplement constituents,and then chemical structures of the compounds were compared and identified by using nuclear magnetic resonance,mass spectrum data and literature data.[Result] 3 flavonoids were isolated from the main active part of FLOS ROSAE RUGOSAE,including luteolin(1),quercetin(2) and kaempferol(3).[Conclusion] The FLOS ROSAE RUGOSAE extracts had in vitro anticomplement effect due to the flavonoids in the extracts.

  • Determination of Metal Elements in the Leaves of Magnolia officinalis by Flame Atomic Absorption Spectrometry

    [Objective] To determine the content of six metal elements like Ca,Mg,Fe,Zn,Cu and Mn in M.officinalis leaves.[Method] Digested with HNO3-HCIO4 mixed acid(V/V,5:1),the content of Ca,Mg,Fe,Zn,Cu and Mn in M.officinalis leaves was determined by flame atomic absorption spectrometry(FAAS).[Result] The content of Ca,Mg,Fe,Zn,Cu and Mn in M.officinalis leaves was 580.57,278.63,177.21,16.70,1.02 and 8.25 mg/kg,respectively.[Conclusion] The FAAS method is simple,rapid and accurate to determine the content of six metal elements in M.officinalis leaves.The study can provide useful data for exploring the relationship between the content of metal elements and pharmacological actions in M.officinalis leaves.

  • First Isolation of 4 New Compounds from C. robustum Root

    [Objective] To study the chemical constitutes of C.robustum root,thus to supply evidence for its pharmacological research and therapeutic mechanism.[Method] The chemical constitutes were isolated from C.robustum by silica gel column chromatography,SephadexLH-20 and recrystallization,and their structures were elucidated on the basis of spectral analysis and physicochemical property.[Result] Nine compounds were isolated from the ethyl acetate extract of C.robustum root,and their structures were identified as palmitic acid(I),α-spinasterol(II),α-spinasteryl-3-O-β-D-glucoside(III),β-stigmasterol(IV),16α,23,28-trihydroxy-olean-12-en-3-O-α-L-arabinopyranoside(V),16α-hydroxy-olean-12-en-28 oic acid-3-O-α-L-arabinopyranoside(VI),hederagenin-3-O-α-L-arabinopyranoside(VII),caulophyllogenin-3-O-α-L-arabinopyranoside(VIII),hederagenin-3-O-β-D-glucopyranosyl-(1→2)-α-L-arabinopyranoside(IX).Of them,compound I,II and III were isolated from this plant for the first time.[Conclusion] This method could be used for studying the chemical constitutes of C.robustum root,and the study provides basis for the further development and utilization of this resource.

  • Study on the GC Fingerprints of Volatile Oil Extracted from ASARI RADIX ET RHIZOMA

    [Objective] To establish a determination method for GC fingerprints of volatile oil in ASARI RADIX ET RHIZOMA from different habitats of Liaoning,so as to provide basis for evaluation,development and quality control of ASARI RADIX ET RHIZOMA medicinal resources.[Method] The volatile oil was first extracted from 10 batches of ASARI RADIX ET RHIZOMA by steam distillation.Then,comparative analysis was performed on GC fingerprints of volatile oil in ASARI RADIX ET RHIZOMA from different habitats of Liaoning by GC through optimizing the temperature programming.[Result] A total of ten common peaks were identified from the GC fingerprints of volatile oil in ASARI RADIX ET RHIZOMA,and the established GC fingerprints of ten batches of samples had good similarities.Moreover,the precision,repeatability and stability could meet the requirement.[Conclusion] The established method is reliable and feasible,which could provide reliable basis for evaluation,development and quality control of ASARI RADIX ET RHIZOMA medicinal resources.

  • Induction of Callus from P.cuspidatum Leaves and Determination of Resveratrol

    [Objective] To induce the callus from P.cuspidatum leaves and to determine the content of resveratrol.[Method] P.cuspidatum leaves as tissue culture materials were induced on MS induction medium containing 0.2 mg/L 2,4-dichlorophenoxyacetic acid(2,4-D) and 1 mg/L 6-amino-benzyl adenine(6-BA) at 28 ℃ and dark.15 d later,the induced callus was transferred to the same culture medium for 15 d,which were transferred 3 times.Subsequently,the callus was cut and then cultured in the same medium for three times.The content of resveratrol in the callus induced from P.cuspidatum leaves was determined by HPLC.[Result] The callus induced from P.cuspidatum leaves grew well,and its resveratrol content was determined at 0.24 mg/g.[Conclusion] The HPLC method was simple.Convenient,quick and accurate to determine the content of resveratrol in P.cuspidatum leaves.

  • Determination of Naringin in Citrus wilsonii Tanaka Leaf by HPLC

    [Objective] To establish a method for the determination of naringin in the leaf of Citrus wilsonii Tanaka by HPLC.[Method] The content of naringin in the leaf of C.wilsonii was determined by HPLC.The separation was performed on a Kromasil column C18(250 mm×4.6 mm,5 μm) with the detection wavelength at 286 nm.Following the injection of sample solution,an elution with methanol-water-glacial acetic acid(V/V/V,30:68:2) as mobile phase was initiated at the flow rate of 1.1 ml/min.[Result] There was a good linearity between the naringin concentration of 0.192 6 and 1.152 μg/ml.The addition recovery rate was 100.6% with the RSD of 1.5%(n=6).[Conclusion] The established HPLC method for determining the content of naringin in C.wilsonii leaf takes advantages of simplicity,accuracy,good repeatability,high precision and strong specificity,which could provide scientific basis for quality control specification of C.wilsonii leaves,variety identification of FRUCTUS CITRI and development of different medicinal parts.

  • Study on the Dynamic Variation of the Main Constituents of P.japonicas Rhizome at Different Cultivating Years

    [Objective] To establish HPLC method that could determine simultaneously the content of three main chemical constituents like P.japonicus saponin V,P.japonicus saponin IVa and P.japonicus saponin IV in P.japonicas medicinal materials,and to study the dynamic variation of three chemical constituents of Hubei Enshi P.japonicus rhizome at different cultivating years.[Method] The content of P.japonicus saponin V,P.japonicus saponin IVa and P.japonicus saponin IV in each sample of P.japonicas rhizome was determined by HPLC.The HPLC analysis was performed on a Diamonsil C18 column(4.6 mm×250 mm,5 μm) with the detection wavelength at 203 nm.Following the injection of sample solution,an elution with acetonitrile-0.2% ammonium acetate(V/V,32:68) as mobile phase was initiated at a flow rate of 1.0 ml/min.In addition,the column temperature was 30 ℃.[Result] The calibration curve of P.japonicus saponin V,P.japonicus saponin IVa and P.japonicus saponin IV was linear in the range of 0.06-4.00 μg(r1=0.999 6),0.04-3.00 μg(r2=0.999 4),0.04-3.00 μg(r3=0.999 5),respectively.The average recovery of the above 3 chemical constituents was 100.61%(RSD=1.28%),99.06%(RSD=1.63%),101.12%(RSD=0.89%),respectively.The content of the main constituents in P.japonicus rhizome differed greatly at different cultivating years.[Conclusion] The method is simple,reliable,and can be used for determining the content of the main constituents in P.japonicus rhizome.It could provide reference to formulate standardized cultivation measures for P.japonicus by monitoring the accumulation of its main constituents.

  • Preliminary Research on the Tissue Culture of Asclepsus curassavica

    [Objective] To establish a rapid propagation system for the tissue culture seedlings of Asclepsus curassavica,in order to provide theoretical basis for the fine variety breeding.[Method] Taking the seeds of wild A.curassavica as research materials,induction effects of different concentrations of hormone combinations on the germination,differentiation,and rooting of A.curassavica were compared,based on the aseptic germination.[Result] A.curassavica seeds showed good germination on MS culture medium.On the culture medium(4) of MS+0.2 mg/L 6-BA+0.5 mg/L NAA,bud showed good proliferation.The culture medium(7) of MS+0.5 mg/L NAA had good induction effect on the root.[Conclusion] Technique of tissue culture and rapid propagation were suitable for the seedling cultivation of A.curassavica.

  • Induction of Leaf Callus and Detection of Stilbene Glucoside Content in RADIX POLYGONI MULTIFLORI

    [Objective] To carry out the induction of leaf callus and the detection of stilbene glucoside content in RADIX POLYGONI MULTIFLORI.[Method] Under dark culture conditions at 25 ℃,leaves of RADIX POLYGONI MULTIFLORI were cultured on MS medium(1 mg/L 2,4-D + 0.4 mg/L NAA) for 15 d.Then,the calluses induced were transferred to the same MS medium for 3 times,with 15 d for each subculture.Content of stilbene glucoside in leaf callus was detected by HPLC.[Result] Calluses induced grew well,and the content of stilbene glucoside in leaf callus was 0.37 mg/g.[Conclusion] There was stilbene glucoside in leaf callus of RADIX POLYGONI MULTIFLORI.And the leaf callus could be used for the further physiological and biochemical studies.

  • Study on the Preparation Technique on Synthetic Seeds of Atractylodes macrocephala Koidz.

    [Objective] To study the best synthetic endosperm combination of Atractylodes macrocephala Koidz.seeds and to provide the theoretical basis for development and large-scale application of A.macrocephala synthetic seeds.[Method] Factors to affect germination rate and seedling conversion rate,such as encapsulated propagule and synthetic endosperm components were studied with germfree tube plantlet system of A.macrocephala.[Result] The germination rate and seedling conversion rate of synthetic seeds could reach 100% and 92% with auxilizry buds,2.5% sodium alginate + 2% CaCl2 as synthetic endosperm encapsulation,and 0.5 mg/L IAA and 30 g/L sucrose as synthetic endosperm components.[Conclusion] The technique of rapid multiplication of synthetic seeds on the basis of tissue culture of A.macrocephala has definite superiority.

  • Research on the Microwave-ultrasonic Synergistic Extraction of Total Flavones from Dendroalamus latiftorus Munro Leaves

    [Objective] To obtain the optimum extraction conditions of total flavones from Dendroalamus latiftorus Munro leaves.[Method] Taking the content of total flavones as the index,the extraction conditions of total flavones from D.latiftorus leaves were studied by uniform design based on extraction rate.[Result] The optimum extraction conditions were as follows:extraction solvent was 54.73% ethanol solution,extraction temperature was 76 ℃,extraction time was 5 min,and the solid-liquid ratio was 1:8(g/ml).[Conclusion] Microwave-ultrasonic extraction had high yield,short extraction time,small ethanol consumption and low ultrasonic power,which was an effective method for the extraction of total flavones from D.latiftorus leaves.

  • Extraction and Purification of Dihydromyricetin in Ampelopsis grossedentata

    [Objective] To optimize the extraction and purification technology of dihydromyricetin from Ampelopsis grossedentata.[Method] Dihydromyricetin was extracted from Ampelopsis grossedentata with water as extracting agent,then purified and determined,the extraction conditions were optimized through single factor test and orthogonal experiment.[Result] The optimum extraction conditions were:extracting temperature 90 ℃,solid-liquid ratio 1:10(g:ml) and extracting time 60 min,under which the yield of dihydromyricetin was the largest,the content of total flavonoids increased from 68.51% to 91.0% after 5 times re-crystallization.[Conclusion] The extraction and purification technology was simple and affordable with convenient source of materials,therefore,it had broad prospect in producing dihydromyricetin as antioxidant.

  • Extraction and Quantitative Determination of Chlorogenic Acid from FLOS LONICERAE JAPONICAE

    [Objective] To determine the content of chlorogenic acid in extracts of FLOS LONICERAE JAPONICAE at different flowering periods that were prepared by different method.[Method] The chlorogenic acid(CA) in FLOS LONICERAE JAPONICAE was determined and identified by UV spectrophotometric method after extraction with ethanol reflux method or ultrasonic method,aiming at discussing the difference of chlorogenic acid at different flowering periods.[Result] The concentration and content of chlorogenic acid in original extract of FLOS LONICERAE JAPONICAE prepared by ethanol reflux method were 1.905 mg/ml and 3.81%,while those in original extract prepared by supersonic method were 1.306 mg/ml and 2.61%.Compared with supersonic method,ethanol reflux method was optimized and validated.Moreover,the concentration and content of chlorogenic acid in original extract of L.japonica full-flowering flowers prepared by ethanol reflux were 0.167 7 mg/ml and 0.34%,while whose in original extract of L.japonica buds were 0.260 1 mg/ml and 0.52%.Hence,the bud contained more chlorogenic acid.[Conclusion] The method is easy,simple and rapid to extract chlorogenic acid and determine its content in FLOS LONICERAE JAPONICAE.

  • Process Research of the Polysaccharide Water Extraction from RADIX CHANGll

    [Objective] To study on the optimum extraction process for the polysaccharides extraction from RADIX CHANGll.[Method] L9(34) orthogonal test was used to select the optimum extraction condition for polysaccharides from RADIX CHANGll with polysaccharides content as the index.[Result] The optimum process for polysaccharides extraction from RADIX CHANGll was 1:30 solid-liquid ratio,75 ℃ extraction temperature,three times of extraction,and 2 h extraction time.And the yield rate of polysaccharides was as high as 21.73%.[Conclusion] This method was reasonable,steady and feasible and could be used as an extraction method for polysaccharides extraction from RADIX CHANGll.

  • Extraction Technology of Prim-O-glucosylcimifugin and 5-O-methylvisammioside by Microwave from Saposhnikovia divaricata

    [Objective] To optimize the extraction conditions of chromones from Saposhnikovia divaricata(Turcz.) Schischk.by microwave.[Method] The contents of prim-O-glucosylcimifugin and 5-O-methylvisammioside were taken as indicating indices;and the effects of solid-liquid ratio,ethanol concentration,extraction time and temperature on chromones content were studied.[Result] The optimal microwave condition was as follows:1:30(g/ml) solid-liquid ratio,70% ethanol concentration,90 ℃ extraction temperature and 3 min extraction time.[Conclusion] The extraction of chromones from S.divaricata by microwave was found to be a rapid and efficient extraction method with short time and high extraction rate.

    [Objective] To select the optimal extraction process for shikimic acid from the PINECONE of Pinus yunnanensis Franch.[Method]On the basis of single factor test,the extraction process of shikimic acid in the PINECONE of Pinus yunnanensis Franch was optimized by L9(34)orthogonal test.[Result]The optimal extraction process was as follows:80% methanol,1:40 solid-liquid ratio,2 extraction times and 20 min for each extraction.[Conclusion] The extraction process was simple,stable and efficient.

    [Objective] To review the research advances on the chemical constituents and pharmacological activities of Ilex cornuta Lindl.et Paxt.,and study the future research orientation.[Method] The existing researches on the chemical consitutent and pharmacological activities of cornuta are collected and arranged.[Result] 40 compounds have been seperated from Ilex cornuta Lindl.et Paxt.,including 18 triterpenoid saponins and its aglycon,5 flavonoids and its glycoside,5 polyphenols and its derivatives,7 fatty acids and 5 others(chain sesquiterpene,tanacetene,daucosterol,cornutaside A and B).The extract had the hypolipidemic,antimicrobial and antifertility effects.Monomeric compound like ursolic acid has the sedative,anti-imflammatory,antimicrobial,antibiabetic,antiulcer and hypolipidemic efficacy,and improve the immune function,anti-carcinogeneic and anti-angiogenic effect,and induce differentiation of F9 teratoma cells;quercetin and hyperin have strong sedative and anti-cerebral ischemic injury effect,and could enhance cerebral blood flow;cornuta glycosides could protect the myocardial ischemia of rats induced by pituitrin and significantly reduce myocardial contractility;adenosine increase coronary blood flow,3,4-Dicaffeoyl-quinic acid significantly improve the release of PGI2.[Conclusion] The study could provide references for better developing and utilizing the Ilex cornuta Lindl.et Paxt.resources.
