  • Effect of the Aqueous Extract of J. regia Leaves on the Seed Germination and Seedling Enzymatic Activity of S. baicalensis in Shangluo

    [Objective] To study the effect of different concentrations of aqueous extract from Juglans regia L. leaves on the seed germination and seedling enzymatic activity of Scutellaria baicalensis Georgi in Shangluo. [Method] The bioassay method was adopted to observe the seed germination and seedling enzymatic activity of S.baicalensis. [Result] Low concentration of aqueous extract promoted the seeds germination and seedling enzymatic activity (SOD, POD and CAT) of S. baicalensis; but with the increasing concentration of aqueous extract, the promoting role gradually decreased, disappeared and even changed into inhibition role. [Conclusion] The aqueous extract of J. regia could promote the seeds germination and seedling enzymatic activity of S. baicalensis to some extent, which provided theoretical support for rationally utilizing the allelopathy of J. regia and its intercropping.

  • Stereoscopic Planting Pattern of Kernel-used Apricot and Medicinal Plants in the Loess Drought Hilly Region in West Henan Province

    [Objective] To guarantee the high yield of quality kernel-used apricot and improve the economic benefits of farmers in the loess drought hilly region in West Henan Province. [Method] The stereoscopic planting pattern with eight cultivars of medicinal plants and three cultivars of kernel-used apricot was adopted to study the effect of different planting patterns on the growth, flowering, fruiting, disease and pest injury as well as the yield of the medicinal plants. [Result] To the same apricot species, Salvia miltiorrhiza Bge. and Pinellia ternata (Thunb.) Breit. produced weak impact on its growth, but Vaccaria segetalis (Neck.) Garcke and Malva rotundifolia L. had obvious influence on its growth and development. The same cultivars of medicinal plant had strong inference on the growths of kernel-apricot cultivars "Yiwofeng",less strong influence on "Longwangmao",and the weakest influence on "Chaoren"; the reproductive indexes of the three kernel-apricot cultivars were influenced by the same Chinese medicinal material followed the decrease scope of "Yiwofeng"<"Chaoren"<"Longwangmao". Under the stereoscopic planting pattern, as the canopy density of kernel-used apricot increased, the weight and medicinal yield of the herbal plants gradually decreased, most of the medicinal plant cultivarsgrew well under the canopy density of 0.5 or below, while the growth and yield of Pinellia ternata (Thunb.) Breit. were not influenced under the canopy density of 0.6. [Conclusion] The stereoscopic planting pattern of kernel-used apricot and traditional Chinese medicinal plants should select the apricot cultivars with strong growth vigor and those short herbal plants species.

  • Effects of NaCl Solution on the Physiological Indices of Polygonum lapathifolium L.

    [Objective] To study on the effects of NaCl solution on the physiological indices of Polygonum lapathifolium L. seedlings. [Method] Taking P. lapathifolium seedlings as the research materials, the contents of chlorophyll, soluble sugar, and MDA were detected under different treatments of NaCl solutions (0, 50, 75, 100, 125, 150 mmol/L), so as to determine the salt-resistant ability of P. lapathifolium. [Result] When NaCl concentration was less than 125 mmol/L, chlorophyll content increased as the NaCl concentration enhanced. But MDA content reduced significantly; and soluble sugar content firstly enhanced and then fell. When NaCl concentration was 150 mmol/L, chlorophyll content was significantly less than that in the CK; MDA content was significantly greater; and soluble sugar content was also greater than that in CK. [Conclusion] Within the range of 0-150 mmol/L, stress of NaCl on P. lapathifolium would start the self-defense mechanism of P. lapathifolium in order to adapt to the environmental stress. Thus, P. lapathifolium had certain salt-resistant ability.

  • Study on the Bud Inhibition Effect of Maleic Hydrazide against Atractylodes macrocephala Koidz.

    [Objective] To explore the chemical bud removal method of Atractylodes macrocephala Koidz., and determine the inhibition effects of 30.2% maleic hydrazide solution with different concentration at different periods. [Method] The chemical bud inhibition effects of maleic hydrazide with different concentrations against Atractylodes macrocephala Koidz. at different growth periods were systematically studied. [Result] The spraying of maleic hydrazide could completely replace bud-picking by hand and achieve the purpose of labor-saving, yield-increasing and efficiency-improving. The higher the concentration of maleic hydrazide and the earlier the spraying time, the better the bud inhibition effect; the plant height was related to the maleic hydrazide concentration and the spraying time, the higher the concentration, the shorter the plant; the later the spraying, the higher the plant; too early spraying and high concentration of maleic hydrazide would reduce the yield of Atractylodes macrocephala Koidz. [Conclusion] The spraying of maleic hydrazide in the range of 50.0 and 62.5-fold dilution during the budding period would increase the yield.

  • Genetic Mode of Male Sterility in Platycodon grandiflorum(Jacq.) A.DC

    [Objective] To reveal the sterility genetic mode in Platycodon grandiflorum (Jacq.) A.DC, thus laying foundation for the utilization of male sterile materials. [Method] Hybridization, back-cross and selfing were performed between male sterile and normal germplasms for P. grandiflorum for observing the segregation of progenies. [Results] In F1 progenies of male sterile × normal fertile hybridization, there exist three combinations of fertility: (1) completely sterile, (2) completely fertile, and mostly (3) the segregating combination. The back-crossed progenies of completely sterile F1 combination still remained completely sterile. The selfed progenies of completely fertile F1 combination showed fertility segregation among individuals. Both the back-crossed and selfed progenies of the segregating F1 combination still presented fertility segregation among individuals. [Conclusion] According to the genetic features of nuclear and cytoplasmic nuclear infertility, the male sterile germplasms of P. grandiflorum detected in this study are infertile types of nuclear-cytoplasmic interaction, which can be used to produce three-line hybrid seeds using the conventional genetic breeding method.

  • Effects of Different Culture Media on Seed Germination of Pleione bulbocodioides (Franch.) Rolfe

    [Objective] To discover the optimum culture condition for seed germination of Pleione bulbocodioides (Franch.) Rolfe and provide technical support for the acquisition of test-tube plantlets. [Method] Effects of sucrose concentrations, active carbon concentrations, basal medium types, 4 hormones (6-BA, KT, NAA and IAA) concentration and 2 combinations of double-hormones on the germination of P. bulbocodioides seeds that were obtained through artificial pollination were investigated in immature embryos. [Result] The optimal culture condition for seed germination was a combination of 1/2 MS, 2% sucrose, 0.05% casein, 0.75% active carbon, 0.55% agar and 1.5 mg/L IAA (pH 5.8). The addictive IAA could cause a higher germination frequency, which induced more and larger protocorms whose average diameter reacheed 1-1.5 mm. [Conclusion] A good and effective seed germination system was established in this study, which would provide basis for acquisition of abundant test-tube plantlets of P. bulbocodioides.

  • Fructan Metabolism of Helianthus tuberosus L. at Different Altitudes

    [Objective] To study the fructan metabolism features of Helianthus tuberosus L. at different altitudes. [Method] " Qingyu 2" and "Qingyu 3" Helianthus tuberosus L. were used to study fructan metabolism in each organ of Helianthus tuberosus at different altitudes. [Result] As the altitude increased, the total content of fructan in each organ decreased, so did the polymerization degree (DP) of fructan in aboveground part, fructan accumulation in tuber and transferred fructan from aboveground part to tuber. At 2 600 m or below, with the increasing altitude, the DP of fructan in tuber decreased, so did the fructan accumulation in the aboveground part; above 2 900 m, both the DP of fructan in tuber and fructan accumulation in aboveground part were high. [Conclusion] This study could provide references for expanding planting area and promoting exploitation of Helianthus tuberosus L. resources.

  • Using an Aging Rat Model to Investigate the Antioxidative Ability of Artichoke (Cynara scolymus L.) Leaf Extract

    [Objective] To evaluate the antioxidative effects of artichoke (Cynara scolymus L.) leaf extract in D-galactose-induced aging rats. [Method] 50 Sprague-Dawley (SD) rats were divided into five groups, including a control group, an aging model group, and low-, medium-, and high-dosage artichoke treatment groups. Except for the control group, all other groups were injected with D-galactose solution daily for 36 consecutive days at the dosage of 400 mg/kg body weight. Rats in the three artichoke treatment groups of low-, medium- and high-dosage were orally administered artichoke at dosages of 20, 40 or 80 mg/kg body weight, respectively. Rats in control group and aging model group were orally administered an equal volume of distilled water. Along with body weight and hematological parameters, the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT), and the content of malondialdehyde (MDA) and lipofuscin (LF) was measured in the serum, liver, and brain. [Result] The results showed that SOD activity in the brain and liver, GSH-Px activity in the brain, and CAT activity in the liver of the artichoke treatment groups increased significantly compared with the aging model group, whereas the content of MDA in the serum and LF in the brain and liver decreased significantly. [Conclusion] Artichoke leaf extract showed good antioxidative ability in aging rats.

  • Effects of Total Flavonoids Extracts from Apocynum venetum L. Leaves on the Content of Monoamine Neurotransmitters in Rat Brain Tissues

    [Objective] To study on the antidepressant effect and mechanism of total flavonoids extracts from Apocynum venetum leaves (AV-extract). [Method] The LC-MS/MS method was applied to investigate the contents of monoamine neurotransmitters, such as norepinephrine (NE), dopamine (DA), and serotonin (5-HT), in rats brain tissues by chronic depression stress. [Result] The concentration of NE, DA and 5-HT in chronic stress depression rats' brain decreased compared with the control group, while the contents of NE and DA in AV-extract group were higher than depression group, and the effect was more apparently with the increasing dosage of AV-extract. Treatment of 70 mg/kg AV-extract had the same effects with fluoxetine hydrochloride (3.5 mg/kg group). However, 5-HT content of the AV-extract group was close to the depression group, but the increased dosage of fluoxetine improved the content of DA, NE and 5-HT in the rat brain tissues of the depression group. [Conclusion] The study proved the neurotransmitter hypothesis of depression, and found out that AV-extract could reverse the decrease of NE and DA contents in rat brain tissues, which might be the antidepressant mechanism of A. venetum.

  • Callus Induction of Polygala tenuifolia Willd.

    [Objective] To study the factors influencing the callus induction of Polygala tenuifolia Willd., so as to establish callus culture method. [Method] The tissue culture method was adopted to compare the effects of different explants, pH, temperature, sunlight, hormone types and proportions on callus induction of Polygala tenuifolia Willd. [Result] The induction rate of cotyledon on MS was the highest at 88%; the callus induced from leaves and stems were few; the effect of different pH on the callus growth was not obvious; the optimum temperature for callus induction was 28 ℃; the induction rate under 66% light conditions was higher than that under 33% light conditions; the induction rate under the hormone combination of 1.0 mg/L 6-BA + 0.1 mg/L NAA was the highest. [Conclusion] The optimum culture conditions for the callus of Polygala tenuifolia Willd. were MS + 1.0 mg/L 6-BA + 0.1 mg/L NAA, pH 6.8, culture temperature 28 ℃, and the alternation between 66% light culture for 18 h and dark culture for 6 h.

  • GC-MS Analysis on Volatile Components in Mucilage from Christisonia hookeri

    [Objective] To analyze the volatile components in mucilage between calyx and corolla from Christisonia hookeri, and provide reference for the comprehensive development and utilization of Christisonia hookeri. [Method] The volatile components in mucilage were analyzed by GC-MS. [Result] There were two kinds of volatile components in the mucilage, i.e., 2-(2-butoxyethoxy) ethanol and methyl n-butyl sulfoxide. [Conclusion] The volatile constituents in mucilage from Christisonia hookeri could be used as raw materials for preparing some medicines and fine chemicals, while that were not the main compounds in mucilage, so the further research should focus on polysaccharides.

  • Analysis on X-ray Diffraction Fourier Fingerprint Pattern of RHIZOMA PINELLIAE

    [Objective] To develop a new identification and analytical method for RHIZOMA PINELLIAE. [Method] X-ray diffraction Fourier fingerprint pattern was adopted to obtain the micro-diffraction pattern and characteristic marked peaks of RHIZOMA PINELLIAE, based on which the identification of different samples, the difference between the original and processed RHIZOMA PINELLIAE products as well as the difference in the content of related components were analyzed, so as to realize the micro identification and analysis of common herbal medicines. [Result] According to their micro-diffraction patterns and characteristic marked peaks, 12 RHIZOMA PINELLIAE and their processed products could be divided into five groups of medicinal RHIZOMA PINELLIAE, RHIZOMA TYPHONII FLAGELLIFORMIS, and their processed products of RHIZOMA PINELLIAE PRAEPARATUM, RHIZOMA PINELLIAE PRAEPARATUM CUM ZINGIBERE ET ALUMINE and RHIZOMA PINELLIAE PRAEPARATUM CUM ALUMINE. The standardization of herbal medicinal quality was extended from the microscopic qualitative identification of macro traits to the repeatable micro information characterized by images and data. [Conclusion] The method of X-ray diffraction Fourier fingerprint pattern could be used for the identification of RHIZOMA PINELLIAE.

  • Determination of Trace Elements in Poria cocos

    [Objective] To measure the contents of trace elements in Poria cocos. [Method] Contents of trace elements including Mg, Fe, Mn, Zn, K and Na in P. cocos samples, collected from three different drugstores, were measured using HNO3-HClO4 system digestion with flame atomic absorption spectrometry. [Results] P. cocos was found abundant in K, Mg, Na, Fe, Mn and Zn, among which the contents of K and Mg are much higher. [Conclusion] P. cocos has higher content of K and Mg, which could promote it to expel dampness and diereses. This study provided reference to reveal the medication mechanism of P. cocos.

  • Content Determination of Methyl Salicylate from Volatile Oil of Gaultheria yunnanensis Leaves in Different Production Areas

    [Objective] To determine the contents of methyl salicylate from volatile oil of Gaultheria yunnanensis leaves in different production areas. [Method] Gas chromatography was adopted to determine the methyl salicylate content in volatile oil of Gaultheria yunnanensis leaves in different production areas. [Result] Content of methyl salicylate in volatile oil varied in different production areas. And the Gaultheria yunnanensis in Shuitian County, Wudang District, Guiyang City was the best in quality; while that in Gaopo Village, Huaxi District, Guiyang City was the poorest. [Conclusion] This detection method was quick and mature, with high accuracy and good repeatability, which provided basis for the establishment of quality standard and content determination method of Gaultheria yunnanensis.

  • Determination of the Contents of Seven Active Components in FRUCTUS EVODIAE

    [Objective] To establish a HPLC/DAD (photodiode-array detection) method for simultaneous determination of synephrine, chlorogenic acid, rutin, hyperoside, quercetin, evodiamine, and rutaecarpine in FRUCTUS EVODIAE. [Method] The samples were prepared with 50% methanol aqueous solution. The chromatographic separation was carried out on a C18 column (250 mm×4.6 mm, 5 μm) and the acetonitrile-1% acetic acid was mobile phase with gradient dilution. The flow rate was 1.0 ml/min, and the detection wavelength was 254 nm for rutin, hyperoside and quercetin, 270 nm for synephrine and rutaecarpine, and 340 nm for chlorogenic acid and evodiamine, respectively. [Result] The separation of synephrine, chlorogenic acid, rutin, hyperoside, quercetin, evodiamine, and rutaecarpine was relatively ideal.Their average recovery rates approached 100%, and RSD values were all below 3.1%. [Conclusion] The method was suitable for the simultaneous determination of synephrine, chlorogenic acid, rutin, hyperoside, quercetin, evodiamine and rutaecarpine, and it provided better technical support for the quality evaluation of FRUCTUS EVODIAE.

  • Comparison of the Phillyrin Content in FRUCTUS FORSYTHIAE from Different Producing Areas

    [Objective] To compare the phillyrin content in FRUCTUS FORSYTHIAE from different producing areas. [Method] The main components of FRUCTUS FORSYTHIAE were qualitatively analyzed by thin layer chromatography (TCL) method and the content of phillyrin in FRUCTUS FORSYTHIAE from different producing areas was assayed by HPLC. [Result] One compound was separated from FRUCTUS FORSYTHIAE and identified to be phillyrin. Phillyrin content in FRUCTUS FORSYTHIAE from Shanxi was twice as much as that from Henan, while the content of phillyrin from Yunnan was slightly lower than that from Henan, showing that there was a significant difference among the content of phillyrin in FRUCTUS FORSYTHIAE from different areas which might be caused by the ecosystem environment of producing areas and different cultivation management measures. [Conclusion] The experimental results were of great significance to expanding the producing areas, ensuring the quality of FRUCTUS FORSYTHIAE and improving its clinical application and so on.

  • Effects of Several Auxiliary Materials on the Particle Size and Zeta Potential of Paclitaxel Liposome

    [Objective] To identify the effects of several auxiliary materials on the particle size and Zeta potential of Tax liposome. [Method] Tax liposome was prepared with film dispersed-ultrasonic method, the orthogonal test was adopted to select the optimal formulation of Tax liposome, and to detect the particle size and Zeta potential of Tax liposome. [Result] The particle size and stability of liposome were significantly influenced by the materials such as eunatrol. And with the increasing amount of those materials, the particle size of liposome became small, Zeta potential value decreased, while the stability improved. [Conclusion] Materials like eunatrol could significantly decrease the particle size and Zeta potential of Tax liposome, the Tax liposome prepared with the optimal technology had high entrapment efficiency, even particle size and good stability.

  • Study on the Chemical Constituents of Chlorogenic Acids from the Dried Leaves of Lonicera similis Hemsl.

    [Objective] To study the chemical constituents in the dried leaves of Lonicera similis Hemsl., so as to provide guidance for its further utilization and finding out new active substance. [Method] Chlorogenic acids were isolated and purified by solvent extraction and column chromatographic methods, and then identified by various spectroscopic methods. [Result] Six compounds were isolated from the dried leaves of Lonicera similis Hemsl., including chlorogenic acid (I), chlorogenic acid methyl ester (II), chlorogenic acid ethyl ester (III), 5-caffeoyl-1, 3-quinide (IV), methyl 3, 4-di-O-caffeoyl quinate (V) and methyl 3, 5-di-O-caffeoyl quinate (VI). [Conclusion] Apart from the chlorogenic acids, other compounds were extracted from this plant for the first time.

  • Microwave-assisted Extraction of Essential Oil from the Stem of Paederia scandens and Analysis of Its Compositions

    [Objective] To study the microwave-assisted cyclohexane optimum extraction conditions of essential oils from the stem of Paederia scandens and analyze their content by GC-MS, so as to provide references for the scientific utilization of P. scandens. [Method] The essential oils were first extracted from the stem of P. scandens by microwave-assisted cyclohexane extraction and then analyzed by GC-MS. The optimum conditions were screened by orthogonal experiment. [Result] The optimum extraction conditions were as follows: MP. scandens stem∶ Vcyclohexane=2∶ 60 (g/ml), irradiation time 10 min, microwave power 800 W and irradiation temperature 50 ℃, under which the yield of essential oils reached 1.32%. 10 compositions were isolated from the stem extracts of P. scandens by GC-MS, of which seven compositions accounting for 90.690% of the total peak area were identified, mainly including 18.424% n-palmitic acid, 24.243% phthalic acid isobutyl nonyl ester and 15.131% oleic acid. [Conclusion] The microwave-assisted cyclohexane extraction method was rapid and efficient to extract essential oils from the stem of P.scandens, but that remain needed to be further improved because of the too few isolated components.

  • Extraction Process and Content Determination of Total Flavonoids from GINKGO FOLIUM

    [Objective] To study on the extraction and content determination methods for total flavonoids from GINKGO FOLIUM. [Method] Both the ultrasonic extraction and Soxhlet extraction methods were studied and improved. Content of total flavonoids in extraction solution was determined by coloration method. The optimal extraction condition for the extraction of total flavonoids from GINKGO FOLIUM was determined by the single factor test. [Result] The extraction rates of ultrasonic extraction and Soxhlet extraction methods were 0.83% and 1.10%, respectively. And the extraction rate of the combination of ultrasonic extraction and Soxhlet extraction was 1.37%. The optimum extraction condition was 20 min ultrasonic time, 60% ethanol concentration, 1∶50 solid-liquid ratio, and heated refluxing for 2 h under 85 ℃. [Conclusion] The optimum extraction process for total flavonoids from GINKGO FOLIUM provided references for its further research and application.

  • Microwave Extraction Technology of Polysaccharides from Sargassum thunbergii

    [Obejective] To study on the microwave extraction technology of polysaccharides from Sargassum thunbergii. [Method] Single factor test was used to determine the effects of various factors on extraction rate. And orthogonal test was used to select the optimum technology. After deproteinization and decoloration of polysaccharides in S. thunbergii, the main components were detected by infrared spectrum. [Result] The optimized process conditions were 8 min microwave time, 540 W microwave power, 1∶30 (g/ml) solid-liquid ratio, and 3 min ultrasonic time. And the extraction rate of polysaccharides by microwave extraction was 6.5%. According to the results of infrared spectrum, pyranose was the main component of polysaccharides in S. thunbergii. [Conclusion] The microwave extraction method of polysaccharides from S. thunbergii was feasible and simple with an ideal extraction rate.

    [Objective] To optimize the extraction process of oil from Agriophyllum squarrosum seeds in Xinjiang and analyze its ingredients. [Method] Taking the yield of the seeds oil as index, the effects of extraction pressure, extraction temperature and extraction time on the extraction of oil from Agriophyllum squarrosum seeds by SFE CO2 method were investigated in single-factor tests, based on which the optimum extraction conditions were obtained through orthogonal test. Ingredients of A. squarrosum seeds oil were analyzed by GC-MS. [Result] The optimum extraction process was as follows:extraction pressure of 30 MPa, extraction temperature of 40 ℃ and extraction time of 3 h. Under above conditions, the yield of A. squarrosum seeds oil reached as high as 13.62%, the principle component of which was linoleic acid. [Conclusion] Linoleic acid is one of the essential fatty acids that can reduce serum cholesterol and control arterialthrombus. SFE CO2 method is stable, efficient and of high application value.
