• Exploring Study on the Scavenging Activity on O_2~(·-) and Correlative Chemical Characteristic Fingerprint of Crataegus Leaves

    [Objective] To study the relationship between HPLC fingerprint and scavenging activity on O·-2 of Crataegus leaves.[Method] Using O·-2 scavenging activity as evaluation index,the content of active ingredients in 5 species of Crataegus leaves was determined by HPLC for establishing mathematic models about the relationship between peak area and O·-2 scavenging activity(also named peak-efficacy fingerprint) .The HPLC fingerprint of Crataegus leaves was translated into bioactivity fingerprint including efficacy fingerprint and potency fingerprint with the helps of mathematics models,chemometrics,bioinformatics and computer programming and simulation etc.[Result] A new evaluative pattern,whose correlation coefficient between predicted value and practical value was 0.846(P<0.05) ,was successfully established for the quality of traditional Chinese medicine(TCM) .Besides,the predicted values of five species were all more than 70% of those in practical value.[Conclusion] This evaluative pattern is an all-around evaluative system,which includes not only chemical identification but also effective evaluation for TCM.This study that has made up for the deficiencies of current evaluative pattern for the quality of TCM would provide a new idea for modern study on TCM.

  • Determination of Protocatechuic Acid in CORTEX ACAN THOPANACIS by High Performance Liquid Chromatography-Four Channel UV-Vis Detection Method

    [Objective] To establish a method to determine the content of protocatechuic acid in CORTEX ACAN THOPANACIS.[Method] Content of protocatechuic acid in CORTEX ACAN THOPANACIS was determined by high performance liquid chromatography-four channel UV-Vis detection method.Chromatographic conditions were as follows:ODSC18 chromatographic column(250 mm×4.6 mm,5 μm) ,and buffer solution of methanol and sodium dihydrogen phosphate as the mobile phase(0.1 mol/L,15∶85,V/V,pH=2.5) .4 detection wavelengths were 245,260,275 and 290 nm with the quantitative detection wavelength of 260 nm.[Result] Protocatechuic acid had good linear relationship within the range of 0.5-6.0 mg/L(r=0.999 9) .The average recovery rate was 99.87% with RSD=2.11%.[Conclusion] The method was reliable,stable,and simple,and could be used for the content determination of protocatechuic acid in CORTEX ACAN THOPANACIS.

  • Study on the Synthesis and Antifungal Activity of Chalcone Analogues

    [Objective] To synthesize chalcone analogues and study their antifungal activity so as to find high-efficient and safe compounds with antifungal activity.[Method] With benzoyl chloride compounds and amines as the materials,16 chalcone analogues were designed and synthesized through the aminolysis reaction of acyl chloride,and their molecular structures were determined by IR and 1H NMR.[Result] The chalcone analogues had certain antifungal activity.The antifungal rate of 100 mg/L compound A2(2-chloro-N-phenyl benzamide) against Rhizoctonia solani and Sclerotiua sclerotiorum reached 90.27% and 92.56%,respectively.[Conclusion] Product A2 was an analogue of the natural product chalcone that could naturally degrade without causing environmental pollution.What's more,its chemical structure was simple and easy to prepare.Using A2 as a lead compound,new high-efficient and low-toxic antifungal agents which were safe to the non-target organisms could be developed through optimizing its chemical structure and studying the relationship between structure and activity.

  • Study on the Quality Standard of ALLIGATOR ALTERNANTHERA HERB

    [Objective] To provide basis for quality control of ALLIGATOR ALTERNANTHERA HERB and comprehensive analysis of its pharmacognosy features.[Method] The pharmacognosy features of ALLIGATOR ALTERNANTHERA HERB were comprehensively studied from the aspects of morphological,microscopic,physical and chemical identification,conventional ingredients check,TLC identification and content determination.[Result] According to the experimental results,a quality standard of ALLIGATOR ALTERNANTHERA HERB was established:contents of moisture,total ash and acid-insoluble ash were less than or equal to 12%,26%,6.5%,respectively;contents of water-soluble extract,ethanol-soluble extract and total flavonoids were respectively higher than or equal to 40%,25% and 2.0%.[Conclusion] The study could provide scientific basis for quality control of ALLIGATOR ALTERNANTHERA HERB.

  • Determination of the Inorganic Elements Contents in Several Wild Traditional Chinese Medicines in Tianjin City,China

    [Objective] To analyze the contents of 9 inorganic elements contents in wild Salsola collina Pall.,Polygonum aviculare L.,and Artemisia argyi in Tianjin City,so as to provide scientific basis for the research on their efficacy.[Method] Salsola collina Pall.,Polygonum aviculare L.,and Artemisia argyi were treated by wet method.And 9 inorganic elements(Ca,Mg,Fe,Cu,Zn,K,Pb,Na and Mn) were determined by FAAS.[Result] In the 3 traditional Chinese medicines,Ca content ranged from 6 440.49 to 9 364.01 μg/g,Cu from 13.19 to 31.14 μg/g,Mg from 5 156.66 to 10 423.99 μg/g,Mn from 63.71 to 108.03 μg/g,K from 18 262.22 to 24 836.81 μg/g,Na from 109.92 to 6 329.75 μg/g,Zn from 67.07 to 90.94 μg/g,and Fe from 969.32 to 2 147.52 μg/g.A certain amount of Pb was detected in all the three medicines.Polygonum aviculare L.had relatively low contents of inorganic elements,while Artemisia argyi had relatively high contents.[Conclusion] The method was accurate,stable,easy and rapid.

  • Determination of Bergenin in the Rhizoma Bergeniae Scopulosae by HPLC

    [Objective] To establish a method to determine the content of bergenin in RHIZOMA BERGENIAE SCOPULOSAE by HPLC.[Method] The research was carried out on Kromasil-C18 column(4.6 mm×150 mm,5 μm) .And the mobile phase was acetonitrile-0.2% phosphoric acid(10∶90) with a flow rate of 1.0 ml/min.The column temperature was 35 ℃ and the detection wavelength was 275 nm.Thus,the content of bergenin in RHIZOMA BERGENIAE SCOPULOSAE was detected.[Result] Bergenin showed good linear relationship between the range of 0.248 and 1.240 μg/ml(r=0.997 9) ;and the average recovery was 100.95% with RSD=2.47%(n=6) .[Conclusion] This method was simple,repeatable and accurate and could be used for the quality control of RHIZOMA BERGENIAE SCOPULOSAE.

  • Effects of Aloe on the Lifespan and SOD Activity of Drosophila melanogaster

    [Objective] To study on the effects of aloe on the lifespan and life cycle of Drosophila melanogaster,so as to provide theoretical basis for further developing the medical and health care effects of aloe.[Method] D.melanogaster was added with an appropriate amount of aloe leaf homogenate to keep the final concentration at 10,25,50,75 and 100 g/L respectively.Then,the effects of aloe on the lifespan and SOD activity of D.melanogaster were studied.[Result] Different dosages of aloe could prolong the average lifespan and longest lifespan of D.melanogaster to different extents.As the feeding time and concentration increased,the SOD activity in D.melanogaster improved significantly.[Conclusion] With a variety of ingredients to eliminate oxygen radicals,aloe had high medical and edible values and could extend the lifespan of D.melanogaster.

  • Study on the Antimicrobial Activity of Flavonoids in Clausena indica(Datz). Oliv. Fruits

    [Objective] To extract flavonoids from Clausena indica(Datz) .Oliv.fruits and study their antimicrobial activities.[Method] Microwave-assisted method was used to extract flavonoids,whose antimicrobial activities were determined by filter paper diffusion method and LB medium double dilution method.[Result] The concentration of concentrated flavonoids extracted from Clausena indica(Datz) .Oliv.fruits was 162.298 5 mg/ml,and flavonoids presented good antimicrobial activities against Staphylococcu saureus,Bacillus subtilis and Escherichia coli with the MIC of 20.29,40.58 and 81.15 mg/ml,but the antimicrobial activity against Mucor and Saccharomyces cerevisiae was not obvious.[Conclusion] The study could provide theoretical basis for further exploring and developing Clausena indica(Datz) .Oliv.resources and producing new,safe and high-efficient biopreservatives.

  • Study on Control Efficiency of Several Fungicides on Spot Blight in Scrophularia ningpoensis Hemsl.

    [Objective] To study the control efficiency of several fungicides on spot blight in Scrophularia ningpoensis Hemsl.[Method] Scrophularia ningpoensis Hemsl.son shoots were respectively soaked in 4 fungicides before planting,and then their control efficiencies on field diseases were investigated after onset;field control experiment against disease was carried out to screen the effectual chemicals for the control of this disease.[Result] By soaking son shoots,4 fungicides could effectively control spot blight and significantly improve survival rate of Scrophularia ningpoensis Hemsl.seedling.The control efficiency of 80% carbendazim at 1∶800 concentrations against spot blight was the highest(67.1%) ,followed by 70% thiophanate-methyl at 1∶1 000 concentrations(65.6%) .All fungicides could achieve better control efficiency in the early stage of spot blight.[Conclusion] Treatment of son shoots with fungicides combined with field spraying control could effectively control spot blight in Scrophularia ningpoensis Hemsl.and minimize losses.

  • Effects of Total Flavonoids from EUCOMMIAE CORTEX on the Expression of Osteoprotegerin by Rat Osteoblasts

    [Objective] To study the effects of total flavonoids from EUCOMMIAE CORTEX on the expression of osteoprotegerin mRNA and protein by osteoblast in vitro cultured from the cranium of newly-born rat.[Method] 10,100 and 200 μg/ml total flavonoids from EUCOMMIAE CORTEX were respectively added into osteoblast isolated from the cranium of SD rats.Then,effects of total flavonoids from EUCOMMIAE CORTEX on the expression of osteoprotegerin mRNA and protein were respectively detected by Realtime-PCR and immunofluorescence technique.[Result] Compared with control group,total flavonoids from EUCOMMIAE CORTEX could obviously promote the expression of osteoprotegerin mRNA and protein by osteoblast.[Conclusion] Total flavonoids from EUCOMMIAE CORTEX could directly promote the expression of osteoprotegerin by osteoblast at the RNA and protein level.

  • Differences of the Contents of Berberine Hydrochloride by Three Preparation Methods in Sanhuang Xiexin Decoction by HPCE Internal Standard Method

    [Objective] To establish a method to determine the berberine hydrochloride content in Sanhuang Xiexin decoction by HPCE(high performance capillary electrophoresis) Internal Standard Method,and to compare the differences in berberine hydrochloride contents by three different peroration methods.[Method] Diphenyl guanidine was used as the internal standard with a fused silica capillary(50 μm×80 cm,effective length of 71.7 cm) .Other conditions were DAD detector,detection wavelength at 275 nm,25 mmol/L borax-acetonitrile(3∶1,V/V,pH=9.12) solution as the electrophoretic medium,30 kV working voltage,50 kPa sampling pressure for 15 s,5 kPa electrophoretic medium sealing for 5 s,and 15 ℃ column temperature.Thus,the content of berberine hydrochloride in sample was determined.[Result] When the internal standard of diphenyl guanidine was 60.10 μg/ml,berberine hydrochloride showed good linear relationship within the range of 6.40-97.28 μg/ml,with r = 0.999 57,recovery rate=100.17%,and RSD=1.75%.The content of berberine hydrochloride in recipe extracts was significantly higher than in others by this method.[Conclusion] This method was simple,rapid,accurate,good in repeatability,and could be used as the quality detection means for Sanhuang Xiexin decoction.

  • Study on the Diurnal Variation of Photosynthesis of Platycodon grandiflorum A.DC in the Hilly Region of Shangluo

    [Objective] To study the diurnal variation of photosynthesis of Platycodon grandiflorum A.DC,in order to provide references for its cultivation.[Method] By using Li-6400 portable photosynthesis system,the photosynthetic rate,transpiration rate,stomatal conductance,intercellular CO2 concentration and photosynthetic active radiation of Platycodon grandiflorum A.DC at different time periods was detected,and its photoresponse curve was fitted.[Result] The diurnal variation of photosynthetic rate showed a two-peaked curve and an obvious "noon-break".As indicated by the correlation analysis result,the noon decrease of photosynthetic rate was caused by the stomata factors.The light saturation point,light compensation point and apparent quantum yield were 1 418 μmol/(m2·s) ,68 μmol/(m2·s) and 0.067 7,respectively.[Conclusion] Platycodon grandiflorum A.DC was a shade plant that had strong ability to utilize low light.

  • Effect of Water Stress on the Structure and Chlorophyll Content of Sedum tatarinowii Maxim Leaves

    [Objective] To study the effect of water stress on the structure and chlorophyll content of Sedum tatarinowii Maxim leaves,so as to provide references for its wide application.[Method] Wild S.tatarinowii sampled from Qiliyu National Forestry Park in Huoshan of Shanxi Province were cultivated and then treated by water stress.Four treatments were set up with the absolute water content in soil of 17.50%,13.60%,9.60% and 6.20% respectively,among which 17.50% was the control.The absolute water content of soil was determined by dry weighting method;while the chlorophyll content in the leaves was detected by acetone extraction method;the well-grown and clean leaves were sampled to prepare paraffin section and epidermis section so as to observe the type and distribution features of stomatal apparatus as well as the stomatal density.[Result] Under normal conditions,the leaves of S.tatarinowii were coated with wax on the surface,and had developed water storage tissue and idioblast with high water absorption capacity inside;the stomata closed during the daytime,while opened at night,the stomatal apparatus subsided;under water stress,the immature stomata appeared on the upper epidermis,and the density of mature stomata showed declining tendency,while the change of stomatal density on the lower epidermis was not obvious.The reaction of chlorophyll a and b to active oxygen was not significantly different,and the content of chlorophyll increased under certain degree of drought conditions;when the absolute water content in soil was 9.60%,S.tatarinowii could maintain its normal photosynthesis;while when the absolute water content in soil decreased to 6.20%,the chlorophyll content declined.[Conclusion] S.tatarinowii had typical features of xerophyte,when the absolute water content in soil maintained at a low level,the chlorophyll content remained unchanged or stable increasing trend,and the normal photosynthesis of plants could be maintained.

  • Extraction and Determination of Emodin in Polygonum perfoliatum L.

    [Objective] To extract and determine the content of emodin in Polygonum perfoliatum L.[Method] The content of emodin was determined by reverse phase HPLC.A column of Kromasil C18 was used and a mixture of methanol-water-glacial acetic acid(87∶13∶215) was used as the mobile phase at a flow rate of 110 ml/min.The UV detection wavelength was 254 nm and the column temperature was room temperature.[Result] The emodin showed a good linear relationship within the range of 1-40 μg/ml(r=0.999 9) ;the linear regression equation was Y=112.89X-8.912(r=0.999 9) .The optimum extraction technology was 80% ethanol concentration,35 ml ethanol dosage and 2 h extraction time.The average recovery rate was 99.16%(RSD=1.49%) .[Conclusion] The method was simple,rapid and accurate.It could be used for the extraction and content determination of emodin from Polygonum perfoliatum L.

  • Optimization of the Extraction Technology of Flavonoids from HERBA TARAXACI

    [Objective] To optimize the extraction technology of flavonoids from HERBA TARAXACI.[Method] By adopting orthogonal experiment and artificial neutral network,the content of total flavonoids was determined,the effects of extracting time,extracting temperature,solid-liquid ratio and ethanol concentration on the extraction process were studied,based on which the extraction technology was optimized.[Result] The effects of factors on the extraction decreased as solid-liquid ratio>ethanol concentration>extracting time>extracting temperature,and the optimum conditions simulated by artificial neural network were extracting temperature 75 ℃,ethanol concentration 65%,solid-liquid ratio 1∶30 g/ml and extracting time 3 h.[Conclusion] The artificial neural network based on orthogonal test could be used in optimizing the extracting technology of flavonoids from HERBA TARAXACI.

    [Objective] To establish a method for extracting protocatechuic acid from Syringa oblata Lindl.leaves.[Method] With Syringa oblata leaves as raw material,absolute ethanol as solvent,the content of protocatechuic acid as the inspection index,an orthogonal test was carried out by selecting the material-liquid ratio,extraction times,extraction temperature and extraction time to optimize the extraction technology of protocatechuic acid from Syringa oblata leaves.[Result] The optimum extraction technology was as follows:material-liquid ratio of 1∶30,extraction time of 4 h,and extraction temperature of 65 ℃.[Conclusion] It was feasible to extract protocatechuic acid from Syringa oblata leaves by solvent-extraction method.

  • Extraction Process of Oleanolic Acid in LIGUSTRI LUCIDI FRUCTUS

    [Objective] To study on the effects of different extraction methods on the extraction rate of oleanolic acid from LIGUSTRI LUCIDI FRUCTUS.[Method] Taking LIGUSTRI LUCIDI FRUCTUS as the material,the three extraction methods were compared according to the extraction rate of oleanolic acid,which were the microwave-assisted extraction,the ultrasonic-assisted extraction,and the supercritical fluid extraction.[Result] Content of oleanolic acid was 1.100% by the microwave-assisted extraction(retention time of 4.116 min) ,1.107% by supercritical fluid extraction(retention time of 4.115 min) ,and 1.291% by ultrasonic-assisted extraction(retention time of 4.107 min) .[Conclusion] Compared with the conventional solvent extraction method,the three methods had the characteristics of short extraction time,high extraction yield,simple operating method and so on.The research results provided theoretical basis for the different extraction requirements of oleanolic acid from LIGUSTRI LUCIDI FRUCTUS.

  • Optimization of the Production Process of Yam and Honey Jelly

    [Objective] To optimize the production process of yam and honey jelly.[Method] Using yam and honey as raw materials,the single factor test and orthogonal experiment were adopted to optimize the production technology of jelly.[Result] The optimum conditions for producing yam and honey jelly were:20.00% of yam juice,15.00% of mixed syrups(honey∶sugar=1∶1) ,0.20% of citric acid,and 1.00% of gelling agent(xanthan gum∶sodium alginate=1∶3) .[Conclusion] The study provided experimental basis for producing the yam and honey jelly with nice flavor and health-care function.

  • Extraction Technology of Total Alkaloids from RADIX ACONITI LATERALIS PREPARATA by the Uniform Design Method

    [Objective] To screen the optimal extraction technology of total alkaloids from RADIX ACONITI LATERALIS PREPARATA,so as to find out the technology applicable to the industrial production.[Method] The total alkaloids was extracted from RADIX ACONITI LATERALIS PREPARATA by the uniform design method,and then the effects of ethanol concentration,dosage and extraction time on the extraction rate were discussed.The optimal extraction technology was selected and then compared with the methods used in other literatures,so as to determine its applicability to industrial production.The total alkaloids was firstly extracted from RADIX ACONITI LATERALIS PREPARATA by TS-NS extraction and concentration unit,and then determined by titration method after being freeze-dried and dissolved.[Result] The optimal extraction technology was:85% ethanol as solvent,60-mesh size,1∶9 solid-liquid ratio,and 2 h extraction time.Under those conditions,the extraction rate of total alkaloids could reach 0.312%.[Conclusion] The method had good extraction effect,and it was applicable to the industrial production.

  • Extraction Processes of Total Sterol from LONGAN SEEDS

    [Objective] To optimize the extraction process of total sterol from LONGAN SEEDS.[Method] Total sterol was extracted from LONGAN SEEDS by using reflux method and ultrasonic method.As for the reflux method,effects of reflux time,solid-liquid ratio and extraction times were studied by single factor test.As for the ultrasonic method,effects of ultrasonic time,solid-liquid ratio and extraction times were studied by L9(33) orthogonal test.Taking the extraction rate of total sterol as the index,total sterol content in LONGAN SEEDS using two different extraction processes was determined by UV-Vis spectrophotometer.[Result] The optimal extraction conditions by ultrasonic method were 1∶8 solid-liquid ratio,2 times of extraction and 20 min for each extraction.Under these conditions,the extraction rate of total sterol from LONGAN SEEDS was 4.662 mg/g.At the same time,the optimal extraction conditions by reflux method were 3 h extraction time,1∶8 solid-liquid ratio,and 3 times of extraction.Under these conditions,the extraction rate of total sterol from LONGAN SEEDS was 4.383 mg/g.[Conclusion] Ultrasonic method was selected to extract the total sterol from LONGAN SEEDS.

  • Application Research of Complex Enzyme on the Extraction of Effective Components from HERBA PHYLLANTHI URINARIAE

    [Objective] To optimize the complex enzyme-assisted extraction process of effective components from HERBA PHYLLANTHI URINARIAE,so as to provide references for its high-efficient utilization.[Method] The flavonoids were extracted from HERBA PHYLLANTHI URINARIAE by using water as the extracting solvent.Using the yield of total flavonoids as index,the extraction conditions were optimized by the orthogonal test with 4 levels and 5 factors,namely,material-liquid ratio,extraction temperature,extraction time,cellulase concentration and pectase concentration.[Result] The yield of total flavonoids in extracts was the highest when fine powder was refluxed for 2 h at 40 ℃ with 25 times of water as extracting solvent in the presence of 1.4 g/L cellulase and 2.0 g/L pectase.Under the optimized conditions,the yield of total flavonoids was 1.589%.The influence of five inspected factors on the yield of total flavonoids decreased in the order of extraction temperature > pectase concentration > material-liquid ratio > extraction time > cellulase concentration.[Conclusion] Complex enzyme-assisted extraction method was rapid,efficient and environmental-friendly with low carbon consumption and high extraction yield.
