• Effect of N Combined with K Fertilizers on the Quality of Platycodon grandiflorum(Jacq.) A. DC.

    [Objective] To study the effects of N combined with K fertilizers on the quality of Platycocon grandiflorus(Jacq.) A.DC.,so as to provide basic data for the cultivation of Platycodon grandiflorum(Jacq.)A.DC.[Method] The field experiment was carried out with randomized block design.[Result] When the N fertilization rate ranged from 0 to 210 kg/hm2 and the K application rate from 0 to 315 kg/hm2,the content of polysaccharide and total saponin in Platycodon grandiflorum(Jacq.)A.DC.showed rise trend with the increase of application amount,which reached the highest levels with 210 kg/hm2 N fertilizer combined with 315 kg/hm2 K fertilizer,being 22.08% and 8.73% respectively.[Conclusion] Reasonable application of N combined with K fertilizers is favorable for improving the quality of Platycodon grandiflorum(Jacq.)A.DC.,and this study would provide basis for the cultivation of Platycodon grandiflorum(Jacq.) A.DC..

  • Effects of Different K Fertilizer Application Rates on the Biomass,Yield and Quality of Biennial Notopterygium forbesii de Boiss.

    [Objective] To study the effects of K fertilizer on the biomass,yield and quality of Notopterygium forbesii de Boiss.,so as to provide scientific references for the reasonable fertilization.[Method] K fertilizer application test was carried in Duoba town of Xining by means of single factor and six levels randomized block design in the test,six application levels were set as 0,37.5,75.0,112.5,150.0 and 187.5 kg/hm2 respectively.[Result] K fertilizer had no significant impact on the plant height and number of compound leaves of Notopterygium forbesii de Boiss.;however the diameter of rhizome,the number of lateral roots,the dry weight of aboveground part,the root length,maximum leaf length,maximum leaf width and the dry weight of roots were improved by K fertilizer;the number of lateral roots was the largest when the application amount of K fertilizer was 75.0 kg/hm2;the diameter of roots and stem as well as the dry weight of the aboveground part were significantly promoted by 112.5 kg/hm2 K fertilizer;with the increase of K fertilizer,the number of roots,the content of volatile oil and alcohol extract in Notopterygium forbesii de Boiss.all improved gradually,which reached the maximum level under 150.0 kg/hm2 K fertilizer;with the applying amount of 150 kg/hm2,K fertilizer played remarkable role in improving the root length,maximum leaf length,maximum leaf width,dry weight of root,root yield,content of volatile oil and content of alcohol extract of Notopterygium forbesii de Boiss..[Conclusion] Taking into account the production costs,112.5 kg/hm2 K fertilizer was the best for the cultivation of Notopterygium forbesii de Boiss..

  • Effects of Fertilizing Amounts and Planting Densities on the Output of RADIX CAMPTAN DRAE YUNNANENSIS

    [Objective] To investigate the effects of different fertilizing amounts and planting densities on the output of RADIX CAMPTAN DRAE YUNNANENSIS.[Method] The artificial domesticated RADIX CAMPTAN DRAE YUNNANENSIS as tested material,different fertilizing amounts and planting densities were carried out.[Result] 11 250 kg/hm2 organic fertilization and 225 kg/hm2 compound fertilization were proved to be the suitable fertilizing conditions;and the reasonable planting density was 126 000 plants//hm2;the optimum combination of fertilizing amount and planting density was A2B2,under this condition the output of RADIX CAMPTAN DRAE YUNNANENSIS can be improved to 11 740.5 kg/hm2,7 times higher than the wild ones(1 500 kg/hm2).[Conclusion] This study would provide basis for the high efficiency and high yield cultivation of RADIX CAMPTAN DRAE YUNNANENSIS.

  • Study on Organochlorine Pesticides Residues in Soils from Genuine Producing Areas of RHIZOMA COPTIDIS

    [Objective] The research aimed to determine the contents of organochlorine pesticides residues in soils from genuine producing areas of RHIZOMA COPTIDIS.[Method] Gas chromatographic method was employed to determine the contents of organochlorine pesticides residues in soil samples from genuine producing areas of RHIZOMA COPTIDIS by reference to GB/T 14550-2003 standard.[Results] The results showed that the contents of organochlorine pesticides residues in the soil samples satisfied the standard.[Conclusion] Soil samples from most of the genuine producing areas of RHIZOMA COPTIDIS conformed to the GAP plantation requirement of traditional Chinese medicinal materials,which provided a foundation for guaranteeing the high-quality and security of RHIZOMA COPTIDIS medicinal materials.

  • Comparison of Extraction Methods of Total RNA from Eucommia Ulmoides Oliv

    [Objective] To extract high quality total RNA from the bark of Eucommia ulmoides Oliv and lay foundation for the isolation of mRNA,cloning of anti-fungi protein gene and library construction of cDNA.[Method] Total RNA from the bark of Eucommia ulmoides Oliv was extracted by modified CTAB-LiCl method,RNApure Plant Kit method and RNAiso Plus method respectively.The quality of total RNA was examined through ultraviolet spectrophotometer and formaldehyde denaturalization agarose gel electrophoresis.[Result] The results showed that total RNA extracted by modified CTAB-LiCl method and RNApure Plant Kit method had higher purity,integrity and yield rate,the ratio of A260/A280 was in the range of 1.8-2.0,the bands of 28S and 18SrRNA were clear;while the quality of total RNA obtained by RNAiso Plus method was poor,the ratio of A260/A280 was 1.652,and the band of RNA was diffusion,which suggested the total RNA was degraded during the extraction process.[Conclusion] The results showed that total RNA from the bark of Eucommia ulmoides Oliv extracted by modified CTAB-LiCl method and RNApure Plant Kit method was of high purity and quality,which could meet the requirement of RT-PCR and RACE etc.experiments as well as lay the foundation for cloning genes from Eucommia ulmoides Oliv.

  • Optimization of Proliferation Culture Medium of Atractylodes lancea(Thunb.) DC. subsp. luotianensis S. L. Hu et X. F. Feng

    [Objective] The research was aimed at identifying the optimum culture medium for the proliferation culture of Atractylodes lancea(Thunb.) DC.subsp.luotianensis S.L.Hu et X.F.Feng,so as to provide a technical support for the industrialized production of Atractylodes lancea(Thunb.) DC.subsp.luotianensis S.L.Hu et X.F.Feng seedlings.[Method] The optimum basic culture medium was screened out by employing tender branch as explants,and the proportion of 6-BA,KT,NAA and IBA was optimized by L9(34) orthogonal test.[Result] The optimum basic culture medium was N68,and the optimum combination of plant growth regulators was 1.0 mg/L 6-BA,1.0 mg/L KT,0.5 mg/L NAA and 0.2 mg/L IBA.[Conclusion] The optimum medium for the proliferation culture was obtained in this study,which provided a technical support for the industrialized production of Atractylodes lancea(Thunb.) DC.subsp.luotianensis S.L.Hu et X.F.Feng seedlings.

  • Effect of Different Cultivation Methods on Yield and Lodge Rate of Pinellia ternata(Thunb.) Breit.

    [Objective] To screen out a cultivation method that can lower the lodge rate of Pinellia ternata(Thunb.) Breit.and increase the yield of it.[Method] Cultured in perlite and soil respectively,nine treatments for each culture matrix were designed to cultivate Pinellia ternata(Thunb.) Breit.to analyze effect of cultivation method on yield and lodge rate of Pinellia ternata(Thunb.) Breit..[Result] The yield of Pinellia ternata(Thunb.) Breit.cultured in perlite was significantly higher than that cultivated in soil;effects of fertilizing amount on yield and lodge rate of Pinellia ternata(Thunb.) Breit.were significant.[Conclusion] The optimum artificial cultivation conditions for Pinellia ternata(Thunb.) Breit.are adding 1/5MS+100 mg/L KH2PO4 in perlite.

  • Preliminary Study on Transformation of Endophytic Fungus XT5 in T. brevifolia by Taxadiene Synthase Gene

    [Object] To transform endophytic fungus XT5 of T.brevifolia by taxadiene synthase gene.[Method] The taxadiene synthase gene was transferred into T.brevifolia endophytic fungus XT5 by soil agrobacterium-mediated method,and the fermentation yield of taxol was studied.[Result] 8 recombinants were obtained,and the taxol productivity of strain XT5-TS-2 reached as high as 423.983 4 μg/L,53.19% higher than the original strain,which suggested the output of taxol can be improved effectively by taxadiene synthase gene.[Conclusion] The study would provide theoretical foundation for improving the fermentation yield of taxol by T.brevifolia endophytic fungus.

  • Study on Combined Hypoglycemic Effect of Wild Taraxacum mongolicum and Rhodiola sachalinensis in Diabetic Mice

    [Objective] To study the combined hypoglycemic effect of wild Taraxacum mongolicum and Rhodiola sachalinensis on diabetic mice.[Method] Thirty Kunming mice were selected(24 of them were diabetes model) and divided into the following five groups:blank control group,alloxan group,Rhodiola sachalinensis group,wild Taraxacum mongolicum group and combined dose group.The mice in Rhodiola sachalinensis group,wild Taraxacum mongolicum group and combined dose group received intragastric administration of self-made water decoction for five days,one time per day,and then their blood glucose was detected.[Result] The blood glucose of diabetic mice in intragastric administrated groups all lowered after five days,which had an extremely significant difference with the alloxan group(P<0.01).[Conclusion] Rhodiola sachalinensis,wild Taraxacum mongolicum as well as their combined treatment all had significant hypoglycemic effect on diabetic mice.

  • Effects of FRUCTUS SCHISANDRAE CHINENSIS on the Hepatic Antioxidant System and Trace Elements in Mice

    [Objective] To study the effects of FRUCTUS SCHISANDRAE CHINENSIS on the hepatic antioxidant system and trace elements in mice.[Methods] 40 male ICR mice were randomly divided into control group,low dose group,middle dose group and high dose group.Intragastric administration for 4 weeks,and then the content of reduced glutathione(GSH),malondialdehyde(MDA) and superoxide dismutase(SOD) in the hepatic tissue were determined.The hepatic tissue was digested by HNO3-HClO4 mixed acid,and the contents of Zn,Cu,Fe and Mn etc.four trace elements in it were determined by atomic absorption spectrometry.[Results] The contents of GSH and SOD in middle dose group were obvious higher than that in the control group(P<0.05).The contents of Cu,Fe and Mn in middle and high dose groups were obvious higher than that in the control group(P<0.05).[Conclusion] FRUCTUS SCHISANDRAE CHINENSIS could enhance the hepatic antioxidant ability of mice significantly through maintaining the balance of trace elements in liver.

  • Determination of Nutrient Contents in the Purple Cultivar from Dioscorea alata L. and in the Dioscorea opposite Thumb

    [Objective] To determine contents of nutritional compositions in the purple cultivar from Dioscovea alata Linn.originated from different habitats,and to compare with those in Dioscorea opposite Thumb.,so as to provide basis for the further development and application of the purple cultivar from Dioscovea alata Linn..[Method] The contents of nutrients in the purple cultivar from Dioscovea alata Linn.from different habitats like Zhejiang,Jiangxi and the farmstead in west suburb of Beijing were determined by conventional methods,and were comparatively analyzed with those of two varieties of Dioscorea opposite Thumb.from Henan Province(D.opposite Thumb.1 and D.opposite Thumb.2).[Result] D.opposite Thumb.2 was rich in crude protein,crude fiber,Fe and K;while D.opposite Thumb.1 was rich in fat,crude fiber,Cu,Ca,Mg and Se;the purple cultivar from D.alata L.from Zhejiang Province was abundant in Cu,Ca,Mn,K,glucose and sucrose;the purple D.alata L.in Beijing contained abundant soluble sugar,soluble acid,K and Se;that in Jiangxi was rich in Vc,P and Se.[Conclusion] The Chinese yam from different areas shows their own advantages in the nutrient content.

  • Effect of Mn~(2+) and Fe~(2+) Stress on the Seed Germination and Seedling Growth of Phytolacca acinosa Roxb

    [Objective] To discuss the effect of Mn2+ and Fe2+ stress on germination and seedling growth of Phytolacaa acinosa Roxb.seed.[Method] The germination rate,germination potential,germination index and seedling survival rate of Phytolacaa acinosa Roxb.seed under different Mn2+ and Fe2+ stress were calculated,investigating effect of Mn2+ and Fe2+ stress on germination and planting of Phytolacaa acinosa Roxb.seed.[Result] In comparison with control,when concentration of Mn2+ and Fe2+ were lower than 1 000 and 45 mg/L respectively,the germination of Phytolacaa acinosa Roxb.seed varied insignificantly;effect of Mn2+ and Fe2+ stress on seedling growth was stronger than seed germination with critical concentration of 800 and 45 mg/L,respectively.[Conclusion] This case provides reference for restoring plants polluted by Mn2+ and Fe2+ in soil and water and provides scientific evidence for the cultivation of Phytolacca acinosa Roxb..

  • Study on Immunoregulation Effects of Different Extracts from RADIX ASTRAGALI and RADIX HEDYSARI

    [Objective] The research aimed to comparatively study the immunoregulation effects of different extracts from RADIX ASTRAGALI and RADIX HEDYSARI using macrophages as target cells.[Method] Lipopolysaccharide(LPS) and dexamethasone(DEX) were used to treat cells as stimulant and immunodepressant,respectively.Taking NO as index reflecting functional changes of macrophages,effects of RADIX ASTRAGALI and RADIX HEDYSARI extracts with different concentration on NO secretion of macrophages under 3 states including activated,suppressed and normal were compared.[Results] Water extracts from RADIX ASTRAGALI and RADIX HEDYSARI could intensify either the normal cells function or those cells treated with 10 ng/ml LPS,and the former was better.Ethanol extracts of the two were provided with little effects on macrophages under suppressed state,but water extracts from residues of ethanol extracts could also activate the function of macrophages.[Conclusion] The active components that could activate macrophages were contained in water extracts from RADIX ASTRAGALI and RADIX HEDYSARI,of which the former effect was more obvious.

  • Determination of Zn in FOLIUM NELUMBINIS from Funiushan by UV Spectrophotometry

    [Objective]To determine the Zn content in FOLIUM NELUMBINIS,so as to provide scientific basis for the deep development and utilization of FOLIUM NELUMBINIS in pharmaceutical and food industries.[Method]Content of Zn in FOLIUM NELUMBINIS was determined by direct UV spectrophotometry using PAN as the color-developing agent and with standard zinc solution as control at the wavelength of 562 nm.[Result]Zn(Ⅱ) and PAN could form red complex,and the absorbance presented good linear relationship with concentration of standard zinc solution in the range of 0.48-5.03 μg/ml(r=0.999 9).The mean content of Zn in FOLIUM NELUMBINIS was 87.44 μg/g with average recovery of 99.8% and RSD value of 0.34%.[Conclusion]Simple in operation,accurate in determining result and good in repeatability and stability,the spectrophotometry established in this case can be used for determining the content of zinc in FOLIUM NELUMBINIS.

  • Effect of RADIX et RHIZOMA RHODIOLAE CRENULATAE on the Immunologic Function of Mice

    [Object] To investigate the effect of RADIX et RHIZOMA RHODIOLAE CRENULATAE on the immunity of mice.[Method] 192 BALB/C mice were randomly divided into 4 groups,and each group was divided into 4 treatments,i.e.,low dose group,middle dose group,high dose group and control group,then they were given medicine(20 ml/kg·weight) for 30 d,the immune indexes were determined.[Result] The carbon clearance ability of mice in high dose group was heightened(P<0.05);the delayed-type hypersensitivity(DTH) induced by sheep red blood cell(SRBC)(P<0.05),the activity of NK cell(P<0.05),the synthesis of serum prothrombin(P<0.05)and antibody cell(P<0.05),the phagocytosis of peritoneal macrophage(P<0.05) were all improved in middle and high dose groups,while the immune organs-body weight ratio and the transformation ability of mice spleen lymphocytes induced by ConA in middle and high dose groups were not promoted significantly.[Conclusion] RADIX et RHIZOMA RHODIOLAE CRENULATAE is endowed with the function of enhancing the immunity of mice,and this case would provide experimental experience for the development of valuable health food.

  • Effect of Stratification and GA_3 Joint Treatment on the Germination of Mosla chinensis Maxim. Seed

    [Objective]To study the effect of stratification and gibberellin(GA3) joint treatment on the germination of Mosla chinensis Maxim.seeds,so as to provide basis for the artificial cultivation of Mosla chinensis Maxim.in large scale.[Method] Treated at 5-10,15,20 ℃ and laminated with river sand,sawdust and filter paper respectively,then the seeds of Mosla chinensis Maxim.were soaked in 400,600,800 mg/L GA3 solution for 18,24,30 h respectively and then subjected to germination test in 25 ℃ germinator.[Result] The optimal germination conditions were determined to be germination temperature of 25 ℃,stratification treated under 5-10 ℃,GA3 concentration of 800 mg/L,soaking duration of 30 h.Under the above conditions,the germination rate of Mosla chinensis Maxim.seeds reached as high as 96.25%.[Conclusion] The germination rate of Mosla chinensis Maxim.seeds can be promoted significantly by stratification and GA3 joint treatment,which would lay foundation for the artificial cultivation of Mosla chinensis Maxim.in large scale.

  • Protective Effect of Swertiamarin on H_2O_2-Induced Apoptosis of Mice Cardiomyocytes

    [Objective] To observe the protective effect of swertiamarin on H2O2-induced apoptosis of mice cardiomyocytes and explore its protective mechanism.[Methods] Cultured with selective plating method,the cell viability,SOD and MDA contents of SD mice cardiomyocyte were determined;the expression rates of apoptosis proteins bcl-2 and Bax were detected with immunocytochemical technique,and the cell apoptosis was detected by acridine orange fluorescent staining.[Results] The viability and SOD activity of cardiomyocytes in medicine groups with different concentrations were all improved,and the content of MDA was lowered as well,which had significant differences with that in H2O2-induced group(P<0.05;P<0.01).[Conclusion] Swertiamarin can protect the H2O2-induced cardiomyocytes effectively,and the protective mechanism may be related to the oxidation system and the regulation of specific proteins.

  • Study on the Anti-hypoxia and Anti-exercise Fatigue Effects of Oplopanax elatus Nakai

    [Objective] To observe the anti-fatigue and anti-hypoxia effects of Oplopanax elatus Nakai on mice,so as to provide reference for the development and utilization of O.elatus.[Methods] Given different doses of O.elatus water suspension for 14 days,the burden swimming endurance,anti-hypoxia duration,the content of blood lactic acid of mice were determined.[Results] The mice in 5.2,2.6 and 1.3 g/kg O.elatus water suspension groups showed stronger anti-hypoxia effect and burden swimming endurance,while with lower blood lactic acid.[Conclusion] O.elatus could improve the anti-fatigue and anti-hypoxia ability of mice.

  • Study on Hypoglycemic Effect of Dahongpao Tea

    [Objective] To explore the hypoglycemic effect of Dahongpao tea.[Method] 50 mice were carried intraperitoneal injection with 200 mg/kg alloxan,repeated in the next day,then 24 mice with the blood glucose above 9.0 mmol/L were obtained after 4 days.Those hyperglycemic mice were divided into the control group and tested group according to the similar blood glucose level of(15.11±6.47) mmol/L and(14.77±5.40) mmol/L respectively,and the difference between the groups was not significant(P>0.05).The mice in the control group were fed with basal feed and cold water,while those in the tested group ate basal feed with 3% of Dahongpao tea and drank 0.5% Dahongpao tea.The blood glucose of the mice in the above two groups was tested again after 7 days and the variation was studied.[Result] The blood glucose of the mice in the control group was(12.23±5.11) mmol/L and the variation was not notable(P>0.05);while the blood glucose of the mice in the test group dropped to(7.93±1.88) mmol/L,reduced by 46.3% compared with that before treated(P<0.001).[Conclusion] Dahongpao tea could effectively reduce the blood glucose of the hyperglycemic mice.

  • Optimization of Volatile Oil Extraction from Asparagus officinalis L. Powder and Its Components Analysis

    [Objective] To optimize the technology of extracting volatile oil from powder of Asparagus officinalis L.by using SC-CO2,lay a foundation for further researches and wider application of the powder of Asparagus officinalis L..[Method] With the extraction amount of volatile oil as the evaluation index,an orthogonal test of 4 factors and 3 levels was designed to optimize the extraction technology,and GC-MS was used to analyze the chemical components of extractives.[Result] The optimal conditions of extracting volatile oil from the powder of Asparagus officinalis L.by SC-CO2 were to extract for 2h at 25 MPa,35 ℃ and CO2 flow 20 L/h.The extraction yield of volatile oil from the powder of Asparagus officinalis L.was up to 98%;37 kinds of compounds were included in the extractives,among which alkanes account for the most,then aldehydes and esters.[Conclusion] Under the optimal extraction conditions,the extraction yield of volatile oil from the powder of Asparagus officinalis L.by using SC-CO2 was high.The work lays a solid foundation for the further researches and wider application of the powder of Asparagus officinalis L..

  • Determination of the Effective Components in Geranium sibiricum

    [Objective] To develop an HPLC method for the simultaneous determination of free and hydrolyzed protocatechuic acid and gallic acid in Geranium sibiricum.[Method] Agilent SBC18 column(4.6 mm ×250.0 mm,5 μm),mobile phase of acetonitrile-0.2% H3PO4(4:96),the flow rate of 0.8 ml/min,the detection wavelength of 253 nm and the room temperature as column temperature.[Result] The average recoveries of free protocatechuic acid and gallic acid were 98.42% and 97.36% respectively with RSD of 1.47% and 1.56%;while the average recoveries of hydrolyzed protocatechuic acid and gallic acid were 99.58% and 99.43% with RSD of 1.79% and 1.44%.The average contents of free protocatechuic acid and gallic acid in the samples were 0.001 2% and 0.041% respectively;the average contents of hydrolyzed protocatechuic acid and gallic acid were 0.013% and 1.387% respectively.The linear relationships of protocatechuic acid and gallic acid were in the range of 0.204-1.02 μg(r=0.999 3) and 0.078-0.39 μg(r=0.999 4) respectively.[Conclusion] The method described in this paper was simple and accurate with good reproducibility,which could be adopted for the quality control of Geranium sibiricum.

    [Objective] To identify the structure of flavone aglycones in Momordica grosvenori flower,so as to provide basis for the development of Momordica grosvenori flower.[Method] The flavone aglycones were subjected to melting point determination,TLC analysis,HPLC analysis,infrared spectroscopic analysis and NMR analysis for characterizing the structure.[Result] Only kaempferol was observed in the flavone aglycones from Momordica grosvenori flower.[Conclusion] The structure of flavone aglycones were identified in this study,which would provide basis for the development and utilization of Momordica grosvenori flower.

  • Components Analysis of Volatile Oil from Different Tissues of Aconitum carmichaeli Debx

    [Objective] To analyze the compositions of volatile oil from different parts of Aconitum carmichaeli Debx.,so as to provide scientific evidence for the comprehensive application of Aconitum carmichaeli Debx..[Method] By employing GC-MS,the components of volatile oil were analyzed.[Result] Thirty-five,forty,thirty-two and twenty-one peaks were isolated from the mother root,stem and leaf,daughter root and fibrous root respectively,and eight,nine,nine and six compounds were identified from them separately,taking account for 8.98%,19.53%,7.61% and 9.84% of total volatile oil,respectively.[Conclusion] Different tissues of Aconitum carmichaeli Debx.differ from each other in volatile oil with low content.

  • Determination of Trace Elements in Traditional Chinese Medicine HERBA ERIGERONTIS

    [Objective] The research aimed to determine the contents of trace elements in traditional Chinese medicine HERBA ERIGERONTIS,so as to provide a basis for the development and application of HERBA ERIGERONTIS.[Method] Pretreated by HNO3-HClO4 mixed acid in 5:1 volume ratio as digestive juice,the contents of 10 trace elements including K,Ca,Cu,Na,Mg,Mn,Fe,Zn,Pb and Cd in HERBA ERIGERONTIS were determined by inductively coupled plasma atomic emission spectrometry(ICP-AES).[Result] HERBA ERIGERONTIS was richest in K with the content of 1 128.40(md) μg/g.Cd and Pb were not detected.The other 7 trace elements decreased according to content in the following order Na,Ca,Mg,Zn,Fe,Cu and Mn,which were 947.30,779.20,557.10,109.80,66.40,33.30 and 7.27(md) μg/g respectively.[Conclusion] The method used in this study was accurate and rapid,which could provide a theoretical basis for the study on internal relations between trace elements in HERBA ERIGERONTIS and its efficacy,in addition to for better development and utilization of HERBA ERIGERONTIS.

  • Extraction Technology and Stability of Flavonoids from Pteridum aquilinum var. latiusculum

    [Objective] The research aimed to extract and purify flavonoids from Pteridium aquilinum var.latiusculum,and to study their stability.[Method] The flavonoids from Pteridium aquilinum var.latiusculum were extracted with ethanol at the concentration of 70%,and the content was determined by UV spectrophotometry.Effects of temperature,illumination,pH value,stabilizing agent,carbohydrate and metallic ion on the stability of flavonoids were also investigated.[Result] The flavonoids content in the root reached as high as 4.264 0%.Except for carbohydrate and pH value within the range of 4 to 6,effects of other factors such as natural light,strong light,reducing agent and Mg2+,Al3+ and Fe3+ on the stability of flavonoids from Pteridium aquilinum var.latiusculum were distinct.[Conclusion] This study provided a theoretical basis for further exploitation and utilization of the Pteridium aquilinum var.latiusculum.

  • Study on the Antimicrobial Activities of Ceratostigma minus Stapf ex Prain

    [Objective] To determine the antimicrobial activities of different extracts from Ceratostigma minus Stapf ex Prain,so as to provide scientific evidence for its further development.[Method] The extracts of Ceratostigma minus Stapf ex Prain were subjected to bacteriostatic tests in vitro by agar spread method.MIC and MBC value were determined by micro-dilution method.[Result] The extracts were endowed with inhibitory effect against Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa and Candida albicans in different degrees,among which,the petroleum ether layer of Ceratostigma minus Stapf ex Prain showed strong inhibition activity against the above 4 bacteria with the minimum inhibitory concentrations(MIC) of 0.005,0.078,0.156 and 0.005 mg/ml respectively.[Conclusion] The extracts of Ceratostigma minus Stapf ex Prain have broad spectrum of antimicrobial activity.And this study would provide scientific evidence for further development of Ceratostigma minus Stapf ex Prain.

  • Effects of Low-temperature Swimming Combined with GBE on the Blood Lipid Metabolism of HLP Rats

    [Object] To observe the effects of low-temperature swimming and Ginkgo biloba extract(GBE) joint treatment on the blood lipid metabolism of HLP rats.[Method]Fed with high-fat forage for 42 d,the hyperlipidemia rats were obtained and randomly divided into 4 groups(N=3),i.e.,control group,GBE group,low-temperature swimming group,low-temperature swimming combined with GBE group.The contents of cholesterol(TC),triglyceride(TG),high density lipoprotein-TC(HDL-C),MDA and superoxide dismutase(SOD) in serum,liver and heart were determined.[Result] The hepatic mass index and SOD activity were decreased in each group;TC and TG of rat serum were remarkably reduced in low-temperature swimming and GBE joint treatment group.[Conclusions] Low-temperature swimming combined with GBE method can regulate the blood lipid metabolism of HLP rats,and lower TC and TG significantly,which is helpful for improving hyperlipidemia.

  • Effect of Different Drying Methods on the Properties of Pectin from Premna phylla Turcz. Leaves

    [Objective] To study the effects of different drying methods on the rehydration of dried Premna microphylla Turcz.leaves and pectin strength.[Method] Hot-air drying,microwave drying and natural drying methods were adopted to process the leaves of Premna microphylla Turcz.respectively.[Result] The leaves dried with microwave method had the best rehydration effect and the highest pectin strength,while natural drying method was the most inefficient one.[Conclusion] The optimum technology for drying the leaves of Premna microphylla Turcz.was obtained in this study,which would provide reference for the annual production of Premna microphylla Turcz.foods.

  • Comparison of Extraction Methods for Total Flavonoids from RADIX RHIZOMA GLYCYRRHIZAE and Optimization of the Extraction Process

    [Objective] To comparatively study the extraction methods for total flavonoids from RADIX RHIZOMA GLYCYRRHIZAE,and to optimize the extraction conditions of the optimum extraction method.[Method] The influences of accelerated solvent extraction,ultrasonic extraction,heat reflux extraction and warm soaking extraction on yield of total flavonoids from RADIX RHIZOMA GLYCYRRHIZAE were investigated,and the extraction conditions of accelerated solvent extraction were optimized.[Result] The accelerated solvent extraction was highest in extraction efficiency in comparison with ultrasonic extraction,reflux extraction and warm immersion extraction,and its optimum extraction processes were:7 min of extraction duration,110 ℃ of extraction temperature,one time of extraction times and extraction solvent of 70% ethanol.[Conclusion]The optimal extraction method with optimum extraction conditions of flavonoids from RADIX RHIZOMA GLYCYRRHIZAE was obtained in this study,which provided an effective and rapid method for the extraction of flavonoids from RADIX RHIZOMA GLYCYRRHIZAE.

  • Purification of High-voltage Electric Field Transformed Ginsenoside Rg_2

    [Objective] To prepare and purify the conversion product of ginsenoside [Method] The ginsenoside was transformed by high-voltage electric field,and the conversion product was purified by resins of HPD100,HPD200A,HPD200B,HPD300,D101 and AB-8 respectively,and determined by RP-HPLC.[Result] The yield rate of ginsenoside Rg2 reached as high as 90% under the appropriate high voltage electric field conditions;AB-8 resin was endowed with the best purification effect,with which,the purity of Rg2 was above 70%.[Conclusion]The highly purified ginsenoside Rg2 was obtained in this study,which would provide evidence for the industrialized production of ginsenoside Rg2.

  • Components Analysis of Amino Acids and Mineral Elements in Rhizoma Dioscorea cirrhosae

    [Object] To analyze and determine the types and contents of amino acids and mineral elements in Rhizoma Dioscorea cirrhosae,so as to provide theoretical evidence for the development and utilization of Rhizoma Dioscorea cirrhosae.[Method] Contents of amino acids and mineral elements were determined by HPLC and inductively coupled plasma mass spectrometry respectively.[Result] 16 amino acids were obtained and the essential amino acids made up 37.98%,of which aspartic acid,glutamic acid,leucine ranked the first,second and third respectively,while the content of methionine was lowest;Rhizoma Dioscorea cirrhosae was rich in K,Ca,Na,P and Mg etc.five major elements as well as B,Mn,Mo,Co,Ni,Cr,Cu,Se,Fe,Sr,Zn,Ba,Cd and Li etc.fourteen trace elements.[Conclusion] There are multiple nutrients and active ingredients in Rhizoma Dioscorea cirrhosae,and this case would provide theoretical evidence for the further development and utilization of Rhizoma Dioscorea cirrhosae.

  • Optimization of Ultrasonic Extraction Process of Tannins from Rhodiola crenulata(Hook.f.et Thoms.)H. Ohba

    [Objective] To optimize the ultrasonic extraction process of tannins from Rhodiola crenulata(Hook.f.et Thoms.)H.Ohba.[Method] Based on single factor experiment,the extraction conditions were optimized by orthogonal test,and the content of tannins was determined by casein method.[Result] The optimal extraction conditions were as follows:70% acetone as extraction solvent,solid-liquid ratio of 1:10,ultrasonic power of 450 W and extraction duration of 15 min.Under the above conditions,the content of tannins from Rhodiola crenulata(Hook.f.et Thoms.)H.Ohba reached as high as 5.17 mg/ml.[Conclusion] The study provides theoretical evidence for the extraction of tannins from Rhodiola crenulata(Hook.f.et Thoms.)H.Ohba.

  • Study on the Extraction Process of Active Ingredients from Fritillaria unibracteata

    [Object] To optimize the extraction process of total alkaloids from Fritillaria unibracteata.[Method] Based on the single factor test,the extraction process of total alkaloids from Fritillaria unibracteata has been optimized through orthogonal design L36(42×32).[Result] The optimum extraction process was as follows:particle size of 200 meshes,alcohol concentration of 75%,extraction duration of 30 min,extraction time of one,among which,extraction time had extremely significant effect on the result(P<0.01﹚,and the particle size had significant effect on the extraction result(P<0.05﹚,while extraction duration and alcohol fraction had no significant influence on the result.[Conclusion] The study would provide scientific references for the production,quality control and reasonable utilization of Fritillaria unibracteata resources.

  • Optimization of Microwave Extraction Process of Total Alkaloid from Aconitum flavum

    [Objective] To optimize the microwave extraction process of total alkaloid from Aconitum flavum,so as to lay the foundation for the development and application of total alkaloid from Aconitum flavum.[Method] The extraction process of total alkaloid from Aconitum flavum was optimized by orthogonal test based on single factor test.[Result] The optimal extraction conditions were as following:8 min of extraction duration,1:10 of solid to liquid ratio,40 ℃ of extraction temperature and extraction solvent pH value of 3.Under the optimum extraction process,yield of total alkaloid from Aconitum flavum reached as high as 0.59%.[Conclusion] The optimum extraction conditions are obtained in this study,which will provide basis for the development and application of total alkaloid from Aconitum flavum.

  • Ultrasonic Extraction of Proanthocyanidins from Litchi chinensis Sonn

    [Object] To optimize the extraction process of proanthocyanidins from Litchi chinensis Sonn.with ultrasonic extraction method.[Method] Litchi chinensis Sonn.as tested material,effects of extractant,volumetric fraction,pH,solid-liquid ratio,ultrasonic power,extracting duration,extraction times and the partical size of Litchi chinensis Sonn.on the yield rate of proanthocyanidins were investigated in this study,and the optimum extraction process of proanthocyanidins was obtained through orthogonal test.[Result] The optimum extraction process of proanthocyanidins was as follows:100 mesh of Litchi chinensis Sonn.powder,solid-liquid ratio of 1:8,ethanol fraction of 70%,pH of 5.0,ultrasonic powder of 600 W,extraction duration of 30 min for one time,and the influence factors were in the order as:extraction duration>partical size>solid-liquid ratio>ultrasonic power.[Conclusions] The optimum extraction process of proanthocyanidins from Litchi chinensis Sonn.was obtained in this study,and the yield rate was 13.11%,which would provide reference for the effective utilization of proanthocyanidins and food colors from Litchi chinensis Sonn..
