  • Determination of Volatile Constituents,Total Flavonoids and Total Polysaccharides in the Roots of Artemisia argyi Levl. et Van. var.argyi cv. Qiai

    [Objectives] To analyze the volatile components,the contents of total flavonoids and total polysaccharides of the roots of Artemisia argyi Levl. et Van. var. argyi cv. Qiai,and to provide a scientific basis for the comprehensive development and utilization of the resources.[Methods]The volatile components of the roots were extracted by solid phase micro-extraction( SPME),and its volatile components were analyzed by gas chromatography-mass spectrometry( GC-MS); the contents of total flavonoids and total polysaccharides in the samples were measured by UV-spectrophotometry. [Results] 44 peaks were isolated from the roots and 30 components were identified,accounting for 80. 9% of the total volatile components; the content of total flavonoids in the roots was 9. 42%; the content of total polysaccharides in the roots was 11. 05%.[Conclusions] After a comprehensive investigation,the olefins in the roots generally have antibacterial activity,the contents of the total flavonoids and total polysaccharides contained in the roots were relatively high,the roots of Artemisia argyi do have broad prospects for the development.

  • Optimization of Extraction Process and Content Determination of Total Flavonoids from Sanguisorbae Radix and Carbonized Sanguisorba Root by Orthogonal Experimental Design

    [Objectives] To optimize the extraction process of total flavonoids from Sanguisorbae Radix and carbonized Sanguisorba root,compare quality of different batches of Sanguisorbae Radix,study the effects of processing on the content of flavonoids,and provide scientific basis for reasonable utilization of Sanguisorbae Radix. [Methods] Test samples were prepared by heating,refluxing,and extraction,the extraction process was optimized by orthogonal experiment design,color was developed by NaNO_2-Al( NO_3)3-NaOH,and total flavonoids were measured by UV method at the wavelength of 510 nm. [Results] The linear relationship of rutin was excellent in the concentration range of 0. 1248 mg/mL-0. 5712 mg/mL,R~2= 0. 9997; the average recovery was 99. 67% and the RSD was 0. 70%. The optimum extraction conditions were as follows: the volume fraction of ethanol was 50%,the extraction temperature was 90℃,the extraction time was 90 min,and the solid-to-liquid ratio was 1∶ 20( g/mL). [Conclusions] After optimization of the extraction process,the extraction rate of total flavonoids in samples of Sanguisorbae Radix was significantly increased; there was certain difference in the content of total flavonoids between different batches of Sanguisorbae Radix and processed products; the total flavonoids significantly declines in carbonized sanguisorba root,and the influence of processing on its curative effect was to be further studied.

  • GC-MS Analysis of Liposoluble Constituents from Calotropis gigantea Roots

    [Objectives] To effectively develop Calotropis gigantea resources and find the biological active constituents with medicinal value.[Methods]The liposoluble constituents from C. gigantea roots were studied by GC-MS for the first time. [Results] 62 constituents were separated and identified from C. gigantea roots,amounting to 60. 43% of the total constituents,α-amyrin( 4. 18%),ethyl palmitate( 3. 49%),1,7-dimethyl-naphthalene( 3. 06%) were the main liposoluble constituents. [Conclusions] Cardiac glycoside was mainly found in methanol extracts and not detected in liposoluble constituents by extraction of petroleum ether. However,the present study found another component of low toxicity—naphthalene and its derivatives.

  • Study on Endophytic Bacteria Diversity in the Root of Peperomia dindygulensis Miq. by Applying Clone Library Construction Method

    [Objectives] To study the endophytic bacteria diversity in the root of Peperomia dindygulensis Miq. [Methods] The root of P.dindygulensis Miq. was taken as materials,culture-independent method was applied to build 16 S rDNA clone library for endophytic bacteria of P. dindygulensis Miq. [Results] In the 16 S rDNA clone library for endophytic bacteria of P. dindygulensis Miq.,95 clones separately belonged to 46 OTUs,Phylogenetic analysis indicated that these cloning sequences separately belonged to phylum Proteobacteria,phylum Bacteroidetes,phylum Actinobacteria,phylum Nitrospirae,and phylum Gemmatimonadetes. In these,phylum Proteobacteria accounted for 70. 53% of total number of clones,so it was the most dominant group in this library. [Conclusions]The endophytic bacteria diversity in the root of P.dindygulensis Miq. is rich and this study is expected to provide scientific basis for protection,development and use of resources of P. dindygulensis Miq.

  • The Protective Effect of CoQ10 on ox-LDL——induced HUVEC Injury by AP-1 and PON2

    [Objectives] To explore the protective effect and possible mechanisms of the coenzyme Q10( CoQ 10) on the human umbilical vein endothelial cell( HUVEC) injury induced by the oxidized low-density lipoprotein( ox-LDL). [Methods]With the human umbilical vein endothelial cells( HUVECs) cultured in vitro as the test target,the HUVECs were randomly divided into 5 groups: normal control group; model group; low concentration CoQ10 group( 12. 5); medium concentration CoQ10 group( 25); high concentration CoQ10 group( 50). The CCK-8 method was used to test the cell viability,and the drug concentration was screened in 60 μM of CoQ 10 toxic concentration; the total protein was extracted and Western blot was used to detect the protein expression of c-fos,c-jun and PON2; the RT-PCR method was used for determination of the content of c-fos,c-jun and PON2 mRNA. [Results]Compared with the normal group,the cell viability was significantly reduced in the 35 μg/m L high ox-LDL model group,and the cell injury was induced; compared with model group,after 12 h pre-protection,12. 5 μM,25 μM,50 μM CoQ10 could all significantly improve the survival of injured cells( P < 0. 05); compared with the normal group;the protein and mRNA levels of c-fos,c-jun,PON2 in the model group all declined; compared with the model group,the protein level and mRNA expression of c-fos,c-jun,PON2 in different CoQ10 groups increased to varying degrees( P < 0. 05). [Conclusions] CoQ 10 could reduce the HUVEC injury induced by high ox-LDL,and possible mechanism was achieved by upregulating the expression of AP-1 and PON2.

  • In vitro Antioxidant Activity of Extracts from Eleutherine plicata Herb by Different Extraction Methods

    [Objectives] To study in vitro antioxidant activity of extracts from Eleutherine plicata Herb by different extraction methods.[Methods]1,1-diphenyl-2-picrylhydrazyl( DPPH) free radical scavenging method,2,2'-Azinobis( 3-ehtylbenzothiazolin-6-sulfnic acid) diammonium salt( ABTS) free radical scavenging method,and iron ion reduction ability determination method were applied to measure the extracts of Eleutherine plicata Herb by ethanol extraction,water extraction,and ethanol extraction after water extraction,and comparison was made with positive control Vc( vitamin C). [Results]The ethanol extract,water extract,and ethanol extract after water extraction of Eleutherine plicata Herb had a high DPPH free radical,ABTS free radical scavenging activity,and reducing activity. [Conclusions]E. plicata Herb is a natural antioxidant with high potential.

Studies have shown that the effects of anti-hepatitis B virus,hepatoprotective,anti-tumor,antipathogenic microorganism,anti-oxidation,and anti-thrombosis were good and worthy of further research and development. The research progress on the pharmacological effects of P. urinaria was reviewed in order to provide a reference for the rational development of this product.

  • Extraction and Separation of Oxysophocarpine from Sophora alopecuroides by Reverse Three-phase Membrane Cycle Method

    [Objectives] To establish the high performance liquid chromatography( HPLC) determination method for oxysophocarpine and optimize the extraction and purification technology of oxysophocarpine from Sophora alopecuroides by inverse three-phase membrane cycle.[Methods]Based on the single-factor experiment,the effects of aqueous phase and organic phase volume ratio,the concentration of sodium hydroxide,concentration of hydrochloric acid and extraction cycle time on extraction process of oxysophocarpine were investigated using orthogonal design method,to determine the optimal extraction process. [Results]The oxysophocarpine was determined by Shim-pack VP-ODS chromatographic column( 4. 6 mm × 250 mm,5 μm),mobile phase was methanol-0. 2% phosphoric acid aqueous solution( 7∶ 93),gradient elution,flow rate was 1 m L/min,the sample size was 5 μm,column temperature was 30℃,detection wavelength was 221 nm. Aqueous phase and organic phase volume ratio was 1∶ 1,hydrochloric acid concentration was 0. 3 mol/L,sodium hydroxide concentration was 0. 75 mol/L,water pump flow rate was 6 m L/min,60 min cycle time. The extraction rate of oxysophocarpine 98. 21 % within 60 min under the best experimental conditions. Oxysophocarpine has good linearity relationship within the range of 0. 01-0. 7 mg/m L( r~2= 0. 9978,n = 6),the respective average recovery rate was 97. 47%( RSD = 1. 95%). [Conclusions] This extraction process is simple in operation,the organic solvent has low consumption,and can be used for extraction of alkaloid.

  • The Optimization of Ultrasonic Extraction Process of Total Flavonoids from Rhodiola Crenulata by Response Surface Method

    [Objectives]To study the optimal extraction process of total flavonoids in Rhodiola rosea L.,and further facilitate the development and utilization of Rhodiola rosea L. [Methods]With Rhodiola crenulata as raw material,ethanol as extractant,ultrasonic extraction as the extraction method,the single factor method was first used for preliminary investigation of effect of ethanol volume fraction,solid-liquid ratio,extraction temperature and extraction time on the flavonoids extraction rate,and then the Box-Behnken response surface method was used to optimize the extraction process of total flavonoids in R. crenulata. [Results] The optimal extraction conditions: ethanol concentration of 72%;solid-liquid ratio of 1∶ 43; extraction temperature of 66℃; extraction time of 50 min. Under these conditions,the extraction rate of total flavonoids from R. crenulata was 2. 591%. [Conclusions] The results showed that the method of response surface was reasonable and feasible for the optimization of ultrasonic extraction of total flavonoids from R. crenulata.

  • HPLC Finger-print Study of Marchantia convoluta in Guangxi

    [Objectives]To establish HPLC finger-print of Marchantia convolute Gao et Chang,and to control the quality of medicinal materials. [Methods] Agilent ZORBAX SB-C18 chromatographic column( 4. 6 mm × 250 mm,5μm) was adopted,acetonitrile( B)-0. 1% phosphoric solution( D) was used as the mobile phase( gradient elution),and the detection was conducted at the wavelength of 230 nm and at the column temperature of 25℃. HPLC finger-print of herb materials in the upland-field was established,and 8 lots of medicinal materials were used for the similarity assessment to establish the finger-print. [Results] Theprecision,repeatability and stability RSD of HPLC finger-prints of the 8 lots of Marchantia convoluta Gao et Chang were all less than 3%,and their similarities were all greater than 0. 75; 6 common peaks were observed,and the control finger-print was also established. [Conclusions] With reliable data and simple and efficient manipulations,HPLC finger-print analysis can provide references for the quality assessment of Marchantia convoluta Gao et Chang.

  • Preparation Process of Liposome of Polysaccharides of Dicliptera chinensis

    [Objectives] The research aimed to study preparation process of liposome of polysaccharides of Dicliptera chinensis( DCP) and investigate its quality.[Methods] Thin film ultrasonic technology and uniform design were used to prepare DCP liposome,and the process was optimized. [Results] DCP liposome obtained by the optimized process had below parameters: entrapment efficiency( 54. 79 ± 1. 30) %,average drug loading rate( 5. 48 ± 0. 12) %,average particle size( 354. 63 ± 7. 04) nm,Zeta potential(-22. 51 ± 1. 13) m V. [Conclusions]DCP liposome obtained by the optimized process had higher entrapment efficiency and drug loading rate.

  • Identification of Lablab Semen Album by DNA Barcode Technology

    [Objectives] To identify ITS2 barcode of Lablab Semen Album and its adulterants,and provide a new method for the identification of Lablab Semen Album. [Methods] The ITS2 sequence was amplified by PCR and sequenced bi-directionally. After splicing by Codon Code Aligner,the data were processed with the aid MEGA software to construct the cluster dendrogram( neighbor-joining,NJ tree). [Results]The ITS2 sequence of Lablab Semen Album had length of 218 bp; the constructed cluster dendrogram indicated that all species were monophyletic and could be distinguished from other species. [Conclusions] The ITS2 barcode can be used for rapid identification of Lablab Semen Album and its adulterants and this experiment further verified that DNA barcode technology is effective in identification of traditional Chinese medicines.

  • Effect of Seed Preservation Technique on the Germination of Artemisia annua Seeds

    [Objectives] To explore the techniques of long-term preservation of Artemisia annua. [Methods] Using different storage methods,germination test and statistical analysis,the effect on seed germination of Artemisia annua was studied. [Results] In cold storage,the envelope preservation had the best effect,while the self-sealing bag had the worst effect; under freezing at low temperatures,the hop-pocket preservation had the best effect,while the self-sealing bag effect had the worst effect. In short,under different storage methods,the self-sealing bag had the worst packaging effect,indicating that the ventilation property of self-sealing bag was poor,affecting the seed breathing,thereby affecting the vitality of the seed,and shortening the life of the seed. [Conclusions]The cold storage envelope packaging was the most appropriate way of storage for Artemisia annua seed,and the storage time should be less than 270 d.

  • Study on Underground Stem Tissue Structure and Agronomic Traits of Different Populations of Wild Houttuynia cordata Thumb

    [Objectives] To study the underground stem tissue structure and agronomic traits of different populations of wild Houttuynia cordata Thumb. [Methods] 8 different populations of wild Houttuynia cordata Thumb. in southern China were selected,the preliminary study was carried out about underground stem tissue structure( stem wall thickness,underground stem cross section diameter,medullary cavity,large vascular bundle area,small vascular bundle area,and number of vascular bundles),and main agronomic traits of aboveground and underground parts. [Results]There were significant differences in the underground stem tissue structure and agronomic traits of some populations of wild Houttuynia cordata Thumb. [Conclusions] From comparison,it reached the conclusion that the populations of wild Houttuynia cordata Thumb. in Kunming of Yunnan,Hechuan of Chongqing,and Nanning of Guangxi showed better underground stem tissue structure and agronomic traits. The results are expected to provide certain guidance for cultivation,reasonable development,and utilization of wild populations of Houttuynia cordata Thumb.

  • Effect of Panax Notoginseng Saponins in the Treatment of Traumatic Spinal-cord Injury in Rats: A Meta-Analysis

    [Objectives] To systemically review the efficacy and safety of Panax Notoginseng Saponins( PNS) in the treatment of traumatic spinal-cord injury( TSCI) in rat models. [Methods] Randomized controlled trials( RCT) about the effects of PNS for TSCI in rats were searched in Pub Med,EMbase,The Cochrane Library,CBM,CNKI,Wan Fang Data from inception to November 2016. Two reviewers independently screened literature according to the inclusion and exclusion criteria,extracted data,and assessed the risk of bias of included studies.Then Meta-analysis was performed using Rev Man 5. 3 software. [Results] A total of 6 RCTs involving 264 rats were included. The results of Meta-analysis showed that: compared with the control group,the Basso,Beattie and Bresnahan( BBB) scores in the PNS group were superior in 7 days( SMD = 1. 81,95% CI( 1. 18,2. 43),P < 0. 00001),14 days( SMD = 1. 92,95% CI( 1. 51,2. 33),P < 0. 00001),21 days( SMD =2. 33,95% CI( 0. 98,3. 68),P <0. 0007) and 28 days( SMD =2. 72,95% CI( 1. 78,3. 67),P <0. 00001) after surgery. The results of Meta-analysis showed that: compared with the control group,ramp test in the PNS group were superior in 7 days [SMD = 1. 55,95% CI( 0. 78,2. 31),P <0. 0001]and 14 days[SMD =1. 83,95% CI( 1. 46,2. 20),P < 0. 00001]. [Conclusions] Current evidence indicates that PNS can promote the recovery of motor function in the rats with TSCI. Due to limited quality of the included studies,the above conclusion should be verified by conducting more large-scale,high quality RCTs.

  • Clinical Effect of Dendrobium officinale Kimura et Migo from Longhushan on Treatment of Type 2 Diabetes

    [Objectives] To observe and compare the clinical effect and safety of Dendrobium officinale Kimura et Migo from Longhushan on treatment of type 2 diabetes. [Methods] 90 patients with type 2 diabetes were randomly divided into group A,group B and control group by randomized control method. Group A adopted D. officinale Kimura et Migo and metformin hydrochloride; group B adopted D. officinale Kimura et Migo; the control group adopted metformin hydrochloride. The fasting plasma glucose( FPG) of 3 groups was measured before the treatment and 2 weeks,4 weeks,6 weeks,8 weeks,10 weeks,and 12 weeks after the treatment,Hb A1 c was measured and diabetes symptom grade score was counted before the treatment and 12 weeks after treatment,observation indicators and clinical effect of 3 groups of patients were compared. [Results] The fasting plasma glucose level in group A was lower than that in other 2 groups,the difference was statistically significant( P < 0. 01),and there was no statistical significance in the difference between group B and the control group( P >0. 05); Hb A1 c of group A for 12 weeks after treatment was lower than that of other 2 groups,the difference was statistically significant( P < 0. 01),and there was no statistical significance in the difference between group B and the control group( P > 0. 05); the difference in diabetes symptom grade score of group A was greater than that in other 2 groups,the difference was statistically significant( P < 0. 01),and there was no statistical significance in the difference between group B and the control group( P > 0. 05). [Conclusions]D. officinale Kimura et Migo from Longhushan can effectively reduce the fasting plasma glucose level and Hb A1 c of type 2 diabetic patients and effectively improve the symptoms of patients; clinical application of D. officinale Kimura et Migo has equivalent effect with metformin hydrochloride,and combination of D. officinale Kimura et Migo with metformin hydrochloride has better effect than single use.

  • Effects of Soy Isoflavones on the Sex Hormone Level and Pathological Changes in the Perimenopausal Syndrome Animal Model

    [Objectives] To observe the effect of soy isoflavones on the perimenopausal syndrome model of female rats,and to find its mechanism,in order to provide experimental evidence for the clinical treatment of menopausal syndrome. [Methods]50 female SD rats were randomly divided into five groups,with 10 rats in each group,respectively,for the normal control group,sham operation group,model group,diethylstilbestrol group,soy isoflavone treated group,except the normal control group and sham operation group,the other groups were ovariectomized rats,the formation model of perimenopausal rats treated with normal saline,diethylstilbestrol,soy isoflavone by gavage,normal control group and sham operation group were given the same dose of normal saline and soy isoflavone by gavage. After 6 weeks of continuous treatment,abdominal aorta was taken blood,the serum was isolated,and radioimmunoassay method to measure E_2( Eestradiol),P( Progesterone),T( Testosterone). And removal of the pituitary gland,uterine and adrenal prepared pathological section,experimental effect was observed by light microscopy,using immunohistochemical method to observe the brain pituitary FSH( Follicle-Stimulating Hormone) and LH( Luteinizing Hormone) levels. [Results] Radiation immune analysis results showed that soy isoflavone could significantly improve serum E_2 level in ovariectomized rats,compared with the model group,the statistical analysis showed significant difference( P < 0. 05),but the serum P and T level had little effect. The pathological results showed that( i) uterus: the uterine wall and endometrial thickness of soy isoflavone group was between that of model group and sham operation group,the surface epithelium was columnar,glands increased;( ii) pituitary gland: soy isoflavone ovariectomized group cells were between model group and normal group;( iii) adrenal gland: soy isoflavone group adrenal tissue did not change significantly. Immunohistochemical results showed that soy isoflavone group pituitary tissues of FSH and LH cells compared with the model group decreased significantly. [Conclusions] The soy isoflavones could make the uterus wall and endometria of perimenopausal syndrome animal model thicken,improve the serum level of E_2 in ovariectomized rats,and decrease the level of FSH and LH,but had little effect on the adrenal tissues,level of T and P.

  • Preparation of Acute Myocardial Infarction Model for SD Rats and Ventricular Remodeling

    [Objectives] To prepare an acute myocardial infarction model for SD rats and conduct ventricular remodeling. [Methods] 45 male Sprague-Dawley( SD) rats were randomly selected in 60 male rats as the model group and the rest rats were sham operation group. For the model group,the left anterior descending coronary artery was ligated,while the sham operation group was not ligated. 4 weeks after the operation,hemodynamics was used to measure the cardiac function of both groups. HE staining and Masson staining were used to observe the changes in myocardial histopathology and myocardial fibrosis of both groups. [Results] Compared with the sham operation group,the model group rats showed significant decrease in the left ventricular systolic pressure( LVSP) and left ventricular pressure maximum rising and dropping rate( ± d_p/d_(tmax)) and significant increase in the left ventricular end-diastolic pressure( LVEDP). HE staining showed that a large number of myocardial cell necrosis occured in the model group,while the morphology of myocardial cells of rats in the sham operation group was normal. Masson staining showed that myocardial collagen fibers in the model group were significantly increased compared with those in the sham operation group. Collagen fibers were scattered in the sham operation group. [Conclusions] It can be concluded that cardiac function of rats changed and ventricular remodeling occurred in 4 weeks after myocardial infarction.
