  • Effects of Cadmium Stress on the Physiological and Ecological Characteristics of Cyperus malaccensis L.

    [Objective] The aim was to reveal the destructive effect of cadmium pollution on salt marsh plant from the perspective of physiological ecology,and to investigate the injured process and resistance mechanism of plant,in order to provide scientific basis for the ecotoxicology research on plants with heavy metal pollution.[Method] Taking Cyperus malaccensis L.as the test material,pot experiment was carried out to study the physiological response and resistance mechanism under cadmium stress.Plant height,diameter of aboveground stem,diameter of underground stem,fresh and dry weights of aboveground and underground parts were taken as the basic physiological indices.Leaf area,contents of chlorophyll a and b,soluble sugar and soluble protein,activities of CAT and POD were taken as biochemical indices.[Result] Low concentration of cadmium could promote the growth of Cyperus malaccensis L.,while high concentration of cadmium(≥5.0 mg/L) played an inhibitory role.Content of chlorophyll decreased with the increase of cadmium concentration,and cadmium stress had greater inhibited effect on chlorophyll a than chlorophyll b.The activity of root system and soluble protein content were improved under low cadmium concentration,and were inhibited under high cadmium concentration.Content of soluble sugar was on rise when cadmium concentration was smaller than 10.0 mg/L.The activity of CAT increased firstly and then decreased with the growth of cadmium concentration;while the activity of POD kept rising,which showed that when cadmium concentration was greater than 5.0 mg/L,the growth of Cyperus malaccensis L.was inhibited.[Conclusion] Cyperus malaccensis L.had realatively strong resistance.

  • Investigation on Caprifoliaceace Medicinal Plants in Taibai Mountain of Qinling Area

    Based on field collection,system investigation and literature reviewing,Caprifoliaceae medicinal plants in Taibai Mountain of Qinling Area were investigated and their medicinal efficacy was summarized.There were 5 genera and 21 species of Caprifoliaceae medicinal plants in Taibai Mountain,and the flowers,fruits or roots could be used as medicine and had high efficacy.Based on these,some suggestions for the utilization and protection of Caprifoliaceae medicinal plants in Taibai Mountain were put forward,according to the characteristics of these plants.

  • Distribution and Infection Situations of Mycorrhizal Fungi in 13 Traditional Chinese Medicines in Chongqing,China

    [Objective] The aim was to investigate the distribution and infection situations of VA mycorrhizal fungi in 13 traditional Chinese medicines produed in Chongqing City,China.[Method] Rhizosphere soils from 13 types of traditional Chinese medicinal materials were collected;and VA mycorrhizal fungi were isolated and identified.The infection situation was investigated as well.[Result] All the 13 types of traditional Chinese medicinal materials had spores of VA mycorrhizal fungi in the rhizosphere soil.7 types of VA mycorrhizal fungi were distributed in the rhizosphere soils of Scrophularia ningpoensis Hemsl,which were the most abundant.A.coreanu had the highest spore density of VA mycorrhizal fungi(118.5 per gram);while A.dahurica f.var.formosana showed the minimum spore density,which was 11.3 per gram.All the 13 traditional Chinese medicinal materials could be infected by VA mycorhiza;and the infection rate of Polygonatum odoratum(Mill.) Druce was the highest(82.29%).[Conclusion] There were abundant mycorrhizal fungi resources in 13 traditional Chinese medicinal materials produced in Chongqing;and the infection situation among different plant individuals differed a lot.

  • Effect of High Temperature Stress on Active Oxygen Metabolism and Activities of Protective Enzymes of Coptis chinensis Franch.Seedling

    [Objective] To study the effect of high temperature stress on active oxygen metabolism and activities of protective enzymes of Coptis cheinensis Franch.seedling.[Method] The correlative physiological and biochemical indices were determined when Coptis chinensis Franch.was under the high temperature(35±2) ℃.[Result] When suffering high temperature stress,Coptis chinensis Franch.was significant higher in MDA in comparison with CK(P<0.05).And the scavenging rate of superoxide anion decreased,which was highly significant lower than that in CK(P<0.01).In addition,the generating rate of hydroxyl radical was significant higher than that in CK(P<0.05),but it did not vary as a rule,the same as the activity of POD.Activity of SOD in Coptis cheinensis Franch.of different growth periods decreased with the increase of stress duration.[Conclusion] Coptis cheinensis Franch.had the ability to regulate the POD and SOD activities to reduce the injury degree produced by the high temperature stress,which,however,was reduced with the increase of high temperature stress duration.

  • Quality Assessment of RADIX ANGELICAE SINENSIS Introduced to Dabieshan Mountains

    [Objective] To estimate the quality of RADIX ANGELICAE SINENSIS planted in two areas of Dabieshan Mountains.[Method] Contents of alcohol extracts,polysaccharide and ferulic acid in RADIX ANGELICAE SINENSIS planted in two areas of Dabieshan Mountains were determined,and were compared with those in genuine producing area(Minxian County,Ganshu Province)and with the standard of Chinese Pharmacopoeia(2005 edition).[Result] Genuine RADIX ANGELICAE SINENSIS,Dabieshan RADIX ANGELICAE SINENSIS Ⅰ and Dabieshan RADIX ANGELICAE SINENSIS Ⅱ were 50.800%,49.300% and 46.500% in alcohol extracts,25.500%,24.700% and 20.300% in polysaccharide and 0.055%,0.053% and 0.050% in ferulic acid,respectively.Content of alcohol extracts,polysaccharide and ferulic acid in introducing areas were lower than those in genuine area,but reached the quality standard ruled in Chinese Pharmacopoeia(2005 version).[Conclusion] RADIX ANGELICAE SINENSIS introduced to Dabieshan Mountains meets the quality standard ruled in Chinese Pharmacopoeia(2005 edition).

  • Factor Analysis on Trace Elements in 14 Kinds of Plants from Valley Area of Qinghai Lake

    [Objective] To analyze trace elements in 14 kinds of plants from valley area of Qinghai Lake.[Method] The characteristic of trace elements in plants from valley area of Qinghai Lake was analyzed by using factor analysis method.[Result] The contribution of matrix variance was more than 90% from the first 3 principal factors.Contents of Fe,Mn,Co and Ni were significantly related to the first principal factor,while Se content was significantly related to the second principal factor.Contents of trace elements were the highest in Astragalus polycladus.[Conclusion] This study could reveal the characteristics of trace elements in plants from valley area of Qinghai Lake,and provide important basis for studying the relationship between trace elements content and efficacy of traditional Chinese medicine.

  • Determination of the Content of Total Flavonoids in Crateva unilocularis in Different Production Areas of China

    [Objective] The aim of this research was to determine the content of total flavonoids in Crateva unilocularis from different production areas,which provided technical support for the selection and extension of C.unilocularis.[Method] Using rutin as the control substance,contents of total flavonoids in Crateva unilocularis from different production areas including Xishuang Banna,Pu'er,Shiping and Jianshui were determined at 510 nm by UV spectrophotometry with with sodium nitrite-aluminum nitrate-sodium hydroxide as the chromogenic reagent.[Result] Contents of total flavonoids in Crateva unilocularis from Xishuang Banna,Pu'er,Shiping and Jianshui were 1.89,2.22,2.02 and 1.49 mg/g,respectively.Among them,content of total flavonoids in Crateva unilocularis was the highest in Pu'er and lowest in Jianshui.[Conclusion] The method was accurate,reliable,repeatable and could be used to determine the content of total flavonoids in Crateva unilocularis.

  • Determination of Trace Elements in Eucommia ulmoides Oliv.by Combining Microwave Digestion with FAAS

    [Objective] To determine the contents of 4 trace elements in Eucommia ulmoides Oliv..[Method] 4 trace elements including Zn,Cu,Ca and Mg in leaves and barks of Eucommia ulmoides Oliv.were determined by combining microwave-assisted sample digestion with flame atomic absorption spectrometry.[Results] The recovery rates of 4 trace elements ranged from 97% to 100.9% with all RSD value of less than or equal to 3.5%.There were significant differences in contents of Zn,Cu,Ca and Mg and antihypertensive effects between barks and leaves of Eucommia ulmoides Oliv..[Conclusion] The method was accurate,fast and convenient.

  • Determination of Organochlorine Pesticides Residues in RADIX RHIZOMA GLYCYRRHIZAE by GC

    [Objective] The aim was to establish a new method to determine the organochlorine pesticides residues in RADIX RHIZOMA GLYCYRRHIZAE.[Method] Ultrasonic extraction of the tested samples was carried out by petroleum ether.After the sulfonation of concentrated sulfuric acid,the sample was determined by the GC chromatography with capillary column.[Result] the lowest detection limit of organochlorine pesticides by GC was 0.000 125-0.000 689 mg/kg,with the RSD being 0.78%-4.98% and recovery being 86.15%-102.12%.There were only γ-BHC in the samples and the quality was 0.001 mg/kg.[Conclusion] Compared with the traditional method,GC was more sensitive,accurate and favorable for the detection of organochlorine pesticides residues.

  • Determination of Contents of Liquiritin in RADIX RHIZOMA GLYCYRRHIZAE from Different Producing Areas by HPLC

    [Objective] To establish a method for determining contents of liquiritin in RADIX RHIZOMA GLYCYRRHIZAE from different producing areas by HPLC,so as to set up a sensitive and specific method for controlling its quality.[Method] The HPLC chromatographic conditions were as follows:column of Kromasil C18(4.6 mm×25 cm,5 μm),mobile phase of acetonitrile-0.5% acetic acid(V/V,1:4);flow rate of 1.0 ml/min,column temperature of 25 ℃;detection wavelength of 270 nm;external standard method was adopted in quantitative determination.[Result] The liquiritin presented a good linear relation with peak areas in a range of 0.1 to 1.2 μg(r=0.999 9),whose average recovery and RSD were 97.86% and 2.04%(n=6),respectively.[Conclusion] The method established in this study was simple,convenient and rapid with good repeatability which can be used in quantitative analysis of RADIX RHIZOMA GLYCYRRHIZAE.

  • Effects of HERBA CISTANCHES on the Ability of Resistance Exercise-induced Fatigue in Rats and Free Radical in Brain Tissue

    [Objective] To discuss the effects of HERBA CISTANCHES on the ability of resistance exercise-induced fatigue of rats and on the free radical in the brain tissue,so as to provide theoretical basis for the application of its resistance exercise-induced fatigue.[Method]Raised for four days,the rats were randomly subdivided into four groups(n=15) like static gavaged water group(C group),static gavaged HERBA CISTANCHES group(M group),movement gavaged water group(T group) and movement HERBA CISTANCHES group(TM group).Twenty-eight days later,the body weight of rats,swimming time when they were exhausted and the content of MDA,SOD,GSH-Px in rats' brain mitochondria in each group were determined.[Result] The rats in M group and C group had no significant differences in body weight,while those in T group were less than those in C group(P<0.01).The body weight of rats in TM group were less than those in M group(P<0.05).Rats in TM group was longer in exhaustive swimming time than T group(P<0.01).Rats in TM group was lower in MDA content(P<0.01) and was higher in SOD and GSH-Px activity compared with those in T group(P<0.01).[Conclusion] HERBA CISTANCHES is provided with effect of lessening the lipid peroxidation in brain tissue of rats and oxidative damage of mitochondrial by exercise-induced fatigue.

  • Study on Effect of Etnylicin on Keeping Seedling of Tussilago farfarae L.

    [Objective] To study keeping seedling effect of etnylicin under different concentration on Tussilago farfarae L..[Method] The local Tussilago farfarae L.stems were treated with low concentration(3 000 times),middle concentration(2 000 times) and high concentration(1 000 times) ethylicin,individually,and the ecdemic Tussilago farfarae was treated with high concentration ethylicin.Effects of different treating methods like root pouring and foliar spraying were comparatively studied.[Result] Before sowing,Tussilago farfarae L.treated with low concentration ethylicin was better in keeping seedling and ecdemic Tussilago farfarae L.showed higher seedling rate.At the early period of disease,Tussilago farfarae L.treated with middle concentration ethylicin using both root pouring and foliar spraying grew better.[Conclusion] Ethylicin showed keeping seedling effect on Tussilago farfarae L.,and foliar spraying would be better.

  • Determination of the Content of Total Lignans in Kadsura coccinea(Lem.) A.C.Smith.

    [Objective] The aim was to establish a method for determining the total lignans content in Kadsura coccinea(Lem.) A.C.Smith.[Method] With schisantherin A as the control,the ultraviolet spectrophotometry was adopted to measure the total lignans content in Kadsura coccinea(Lem.) A.C.Smith..[Result] Schisantherin A showed good linear relationship(r=0.999 1) within the range of 0.008 16-0.040 8 mg/ml.And the average recovery rate was 100.37%,and RSD value was 0.98%.Kadsura coccinea(Lem.) A.C.Smith.collected in Fangcheng of Guangxi had the highest content of total lignans.[Conclusion] Ultraviolet spectrophotometry was simple,stable and reliable,which was suitable for the determination of lignans content in Kadsura coccinea(Lem.) A.C.Smith.

  • Simultaneous Determination of Three Phenolic Compounds in the Fruit of Toona sinensis(A.Juss.) Roem.by RP-HPLC

    [Objective] To develop a high performance liquid chromatography(HPLC) method to simultaneously determine contents of three phenolic compounds including gallic acid,methyl gallate and 1,2,3,4,6-penta-O-galloyl-β-D-glucopyranose in the fruits of Toona sinensis(A.Juss.) Roem..[Method] The chromatographic separation was performed on a Hypersil C18 column(150 mm×6.0 mm,5 μm) with mobile phase of methanol and 0.1% formic acid in gradient elution whose flow rate was 1.0 ml/min.The temperature of sample manager was set at 30 ℃,and samples were detected at 280 nm and quantified by external standard method.[Result] The linear ranges of gallic acid,methyl gallate and 1,2,3,4,6-penta-O-galloyl-β-D-glucopyranose were respectively 0.048-0.48 μg,0.018-0.18 μg and 0.022-0.22 μg with correlation coefficient of more than or equal to 0.999 8.The average recoveries of 3 phenolic compounds were respectively 101.6%,99.5% and 99.1% with RSD of less than or equal to 2.9%.[Conclusion] The developed HPLC method was simple,sensitive and accurate with good separation,which was available for determining contents of phenolic compounds in the fruits of Toona sinensis(A.Juss.) Roem..

  • Hyperglycemic Effect of Black Buckwheat Powder on Alloxan-induced Diabetic Mice

    [Objective] To study the hypoglycemic effect of black buckwheat dry powder.[Method] The hypoglycemic effects were investigated in alloxan-induced diabetic mice after oral administration of black buckwheat powder with different dosage(0.50 and 0.25 mg/kg),different granularity(sieved from 120 and 200 meshes) and different drying ways(freeze-drying and drying) for 14 days.The blood glucose(GLU) levels of mice were determined.[Result] The high dosage,sieved from 200 screen and freeze-drying of black buckwheat powder lowered diabetic mice's glucose level obviously.[Conclusion] Black buckwheat powder possessed hypoglycemic activities to alloxan-induced diabetic mice.

  • Optimization of Extraction Process of Flavonoid Glycosides from HERBA LOBELIAE CHINENSIS

    [Objective] To study the optimal extraction technical parameters of flavonoids glycosides from HERBA LOBELIAE CHINENSIS by ultrasonic-assisted extraction.[Method] With ethanol as the extracting solvent,the orthogonal design was employed to test the effects of four factors like ethanol concentration,extraction time,extraction temperature and solid-liquid ratio.[Result] The optimum conditions for extracting flavonoids glycosides from HERBA LOBELIAE CHINENSIS were as follows:ethanol concentration of 50%,extraction time of 35 min,extraction temperature of 50 ℃ and solid-liquid ratio of 1:50.The content of falconoid glycosides could reach 4.209 2 mg/g under the optimal conditions.[Conclusion] High in extraction rate and good in stability,the method favored the determination of flavonoids glycosides from HERBA LOBELIAE CHINENSIS.

  • Preliminary Research on Extraction Process and Stability of Chlorophyll from Leaves of Camellia chrysantha(Hu) Tuyama.

    [Objective] To study the extraction process of chlorophyll from leaves of Camellia chrysantha(Hu) Tuyama.and effects of temperature,pH value and illumination time on the stability of chlorophyll.[Method] Effects of solvent type,extraction temperature,extraction time and solid-liquid ratio on the extraction of chlorophyll from Camellia chrysantha(Hu) Tuyama.leaves were investigated by orthogonal test design and single factor experiments.The stability of chlorophyll was investigated from 3 aspects including temperature,pH value and illumination time.[Result] The optimum extraction conditions of chlorophyll from Camellia chrysantha(Hu) Tuyama.leaves were as follows:extracting solvent of 95% ethanol and acetone in 1:1 volume ratio,extraction temperature of 50 ℃,solid-liquid ratio of 1:50 and extraction time of 3.0 h.The order of factors affecting the extraction of chlorophyll was extraction temperature>extraction time>solid-liquid ratio.Ultrasonic-assisted extraction had more advantage.Chlorophyll was unstable in condition of high temperature,strong acid,strong alkali and light.[Conclusion]The method used in this paper was simple,accurate and efficient that can be used for the extraction of chlorophyll from Camellia chrysantha(Hu) Tuyama.leaves.

  • Optimization of the Enzyme Matching Ratio by the Mixture Design in the Extraction of Lonicera edulis Pigment

    [Objective] The research aimed to find the optimum proportions of pectase,cellulase and papain in the extraction of Lonicera edulis pigment.[Method] The influences of different proportions of three enzymes on the absorbance value of Lonicera edulis pigment extraction solution were studied by the mixture design.The regression model between the enzyme matching ratio and the absorbance value was established,and the interaction effects of three enzymes were studied.Based on the model optimization,the optimum ratio was obtained.[Result] The optimum matching ratios of three enzymes were the pectase 46.6%,cellulase enzyme 17.6% and papain 36.0%.[Conclusion] The research provided the theory basis for the comprehensive utilization of Lonicera edulis resource.

  • Optimization of the Extraction Process of Geniposide from FRUCTUS GARDENIAE by Orthogonal Design

    [Objective] To optimize the extraction process of geniposide from FRUCTUS GARDENIAE.[Method] Taking the extraction rate of geniposide as index,orthogonal test designed with 3 investigation objects including solid-liquid ratio,extraction temperature and extraction time was performed to optimize the extraction process of geniposide from FRUCTUS GARDENIAE on the basis of single factor test.[Results] The optimal extraction conditions of geniposide were as follows:60% alcohol as extracting solvent,solid-liquid ratio of 1:4,extraction temperature of 75 ℃,extraction time of 3 h.Under the above conditions,the extraction rate of geniposide reached as high as 12.59%.[Conclusion] The efficiency and stability of geniposide could be improved by suitable extraction conditions.

  • Study on the Extraction Technology of Puerarin from RADIX PUERARIAE by Ultrasonic and Microwave Assisted Extraction

    [Objective] To explore the extraction technology of puerarin from RADIX PUERARIAE by ultrasonic and microwave assisted extraction(UMAE),so as to provide new strategies for its industrialize production.[Method] Taking extraction rate of puerarin as index,effects of microwave power,extraction time,ethanol concentration,solid-liquid ratio and soak time on the extraction of puerarin were investigated by singer-factor experiments and L9(33) orthogonal test.[Result] The optimum extraction technology of puerarin from RADIX PUERARIAE by UMAE were:microwave power of 250 W,extraction time of 25 s,ethanol concentration of 50%,solid-liquid ratio of 1:20 and soak time of 30 min.Under above conditions,the extraction rate of puerarin was up to 0.637 6%.[Conclusion] The extraction technology of puerarin from RADIX PUERARIAE by UMAE took advantages of high efficiency,saving time,saving solvent,saving energy,less pollution,etc.

  • Optimization of Extraction Process of Flavonoids from HERBA PHYLLANTHI URINARIAE by Response Surface Methodology

    [Objective] To optimize the extraction process of flavonoids from HERBA PHYLLANTHI URINARIAE by response surface methodology.[Method] Using extraction temperature,extraction time and solid-liquid ratio as independent variables and the yield of flavonoids as response value,response surface methodology with three-factors and three-levels was carried out for optimizing the extraction process of flavonoids from HERBA PHYLLANTHI URINARIAE on the basis of Box-Benhnken central composite design principles.[Result] The optimum extraction conditions of flavonoids were as follows:solid-liquid ratio of 1:18,extraction temperature of 66 ℃ and extraction time of 140 min.Under this condition,the yield of flavonoids was up to 2.43%.[Conclusion] This study could provide a theoretic basis and reference datum for extensive industrialization of HERBA PHYLLANTHI URINARIAE.

  • Study on the Extraction Technology of Flavonoids from Aloe arborescens Mill.by Microwave Method

    [Objective] To study extraction technology of flavonoids from Aloe arborescens Mill.by microwave method.[Method] Taking extraction rate of flavonoids as index,singer-factor experiments and orthogonal test were applied to optimize the extraction technology of flavonoids from Aloe arborescens Mill.by microwave method.[Results] The optimum extraction technology of flavonoids was as follows:solid-liquid ratio of 1:50,ethanol concentration of 80%,microwave power of 560 W and microwave treatment time of 40 s.Under this condition,the extraction rate of flavonoids was 5.604%.[Conclusion] Compared with ethanol extraction method,microwave method was characteristic of shorter extraction time,less ethanol volume and higher extraction rate,which was an ideal method to extract flavonoids from Aloe arborescens Mill..

  • Optimization of Extraction Process of Polysaccharides from RHIZOMA AREACTYLODIS LANCEAE by Response Surface Methodology

    [Objective] To optimize the extraction process of polysaccharides from RHIZOMA AREACTYLODIS LANCEAE by response surface methodology.[Method] Using extraction temperature,solid-liquid ratio and extraction time as independent variables and extraction rate of polysaccharides as response value,response surface methodology with three-factors and three-levels was carried out for optimizing the extraction process of polysaccharides from RHIZOMA AREACTYLODIS LANCEAE on the basis of central composite design principles.[Result] The optimum extraction conditions of polysaccharides were as follows:extraction temperature of 87 ℃,solid-liquid ratio of 1:22 and extraction time of 3.1 h.Under this condition,the extraction rate of polysaccharides was 2.12%.[Conclusion] The study could provide a certain theoretical direction for extracting polysaccharides from RHIZOMA AREACTYLODIS LANCEAE on a large scale.
