Electrophysiology Effects of Cestrum nocturum n-Butyl Alcohol Extract on Isolated Guinea Pig Atria

摘要(Abstract):

[Objective]To study the electrophysiology effects of Cestrum nocturumn n-Butyl Alcohol Extract (CNAE) on isolated guinea pig atria.[Methods] The conventional experimental methods for electrophysiology of isolated guinea pig left and right atria were adopted,such as CNAE on contractility,spontaneous beat of right atrium,function refractory,post-rest potentiation and positive staircase phenomenon of left atrium were observed in isolated guinea pig.[Result] CNAE (0.1,0.1 × 3,1,1 × 3,10.0,10 × 3,100 mg/mL) prominently decreased spontaneous beat of right atrium.As for the contractibility of right atrium,it had a effect of biphasic manner,CNAE at a lower concentration (0.1,0.1 × 3,1 mg/mL) enhanced the contraction of left atria in a concentration-dependent manner,but at a higher concentration (1 × 3, 10.0,10 × 3,100 mg/mL) it exerted an inhibitory effect.And CNAE possessed the effect of depressing the excitability via increasing the threshold of excitability.And CNAE at a higher concentration (50.0,100.0 mg/mL) could shorten the function refractory period and restrain the positive staircase phenomenon of isolated guinea pig left atrium,but it had no effect on the post-rest potentiation phenomenon of myocardial contractility in left atrium.[Conclusion]The pharmacological effect of CNAE on isolated guinea pig atria may be related to the mechanism by restraining the voltage-dependent calcium channel (VDC) and natrium channel,or promoting the Potassium channel.
